To explore perceptions of community dwelling caregivers of preschool-aged children regarding the influence of social determinants of health (SDOH) on children's school readiness. Parents' perspectives regarding solutions to enhance school readiness in preschool-aged children are also explored. This study employed a qualitative, descriptive design and a community based participatory research (CBPR) approach. An academic institution collaborated with parents, teachers, and administrators at a community-based preschool learning center. Ten young-adult to middle-aged mothers and caregivers attended two separate focus groups and completed open-ended questionnaires. Inductive and deductive thematic analysis of text were employed. Three themes emerged 1) Families described the vast lack of appropriate community resources and inability to access those resources that are available to prepare their children for school 2). Family members need help processing information about social resources 3) Community, individual and systemic level solutions to enhance school readiness. Academic-Community partnerships provide an opportunity to (1) identify solutions to remove systemic barriers that impede children's readiness for school, and (2) design interventions to support families through that process. Interventions to enhance school readiness should be family-focused and can be informed by understanding the influence of SDOH during the planning stages. SDOH create barriers and prevent parents from prioritizing their children's school, health-care, and developmental needs. Pollution from pesticides is an increasing concern for human health and biodiversity conservation. However, there is lack of knowledge about some emerging molecules such as SDHI fungicides (succinate dehydrogenase inhibitors) that are widely used but potentially highly toxic for vertebrates. Boscalid, fluopyram, and bixafen are 3 frequent SDHI molecules commonly detected in surface waters, which may pose risks to aquatic species. This study aimed to (1) test the in vitro effects of SDHI on mitochondrial activities (inhibition of succinate dehydrogenase SDH, also named respiratory chain complex II) and (2) assess the in vivo effects of sublethal SDHI concentrations on fish physiology and behaviour over 96 hours of exposure, using Carassius auratus fish as a model species. Results show that bixafen and boscalid inhibited complex II activities in vitro as expected (bixafen > boscalid), while fluopyram had no in vitro effects. In contrast, in vivo studies showed that fluopyram strongly altered fish behaviour (enhanced activity, social and feeding behaviours), likely explained by reduced AChE enzymatic activity. In addition, fluopyram increased muscle lipid content, suggesting metabolic disruption. These findings raise serious concern about the toxic effects of SDHI pesticides, especially fluopyram, although its underpinning molecular mechanisms remain to be explored. We thus encourage further research on the long-term impacts of SDHI pesticides to improve existing regulation and prevent adverse effects on wildlife. Individuals in remission from depression (MDDR) tend to experience lingering cognitive and emotional processing alterations. However, little is known about the neural profiles underlying these features. Using simultaneous EEG+fMRI, we assessed neural profiles during the emotional word Stroop task (eStroop) in people with MDDR and healthy volunteers (HVs). Event-related potentials (ERPs) were extracted (N450, N2 & P3). Assessments of brain activation, as modulated by ERPs, were carried out, as were fMRI-informed ERP analyses. A trend for greater P3 amplitudes in MDDR versus HV groups existed. HV versus MDDR groups had greater brain activation to emotional versus neutral words in various regions, including the left amygdala, inferior frontal gyrus (IFG); this appeared to be driven by elevated activity to neutral words in the MDDR group (neutral > emotional). HVs showed greater activation (emotional > neutral) modulated by N450 amplitude in various regions, while MDDRs showed greater neutral > emotional activation modulated by N450 amplitude in the left IFG and left precuneus. Our EEG+fMRI findings indicate that people with MDDR appear to have blunted neural differentiation to emotional versus neutral stimuli or elevated neural responses to neutral information processing. This might represent altered neuronal processing (i.e., underlying attention and conflict processing) during a cognitive task with an emotional component in individuals remitted from depression, or elevated neural responses to ambiguous or neutral information. Succinate dehydrogenase inhibitors (SDHIs) are widely-used fungicides, to which humans are exposed and for which putative health risks are of concern. In order to identify human molecular targets for these agrochemicals, the interactions of 15 SDHIs with expression and activity of human cytochrome P-450 3A4 (CYP3A4), a major hepatic drug metabolizing enzyme, were investigated in vitro. 12/15 SDHIs, i.e. , bixafen, boscalid, fluopyram, flutolanil, fluxapyroxad, furametpyr, isofetamid, isopyrazam, penflufen, penthiopyrad, pydiflumetofen and sedaxane, were found to enhance CYP3A4 mRNA expression in human hepatic HepaRG cells and primary human hepatocytes exposed for 48 h to 10 µM SDHIs, whereas 3/15 SDHIs, i.e., benzovindiflupyr, carboxin and thifluzamide, were without effect. The inducing effects were concentrations-dependent for boscalid (EC 50 =22.5 µM), fluopyram (EC 50 =4.8 µM) and flutolanil (EC 50 =53.6 µM). They were fully prevented by SPA70, an antagonist of the Pregnane X Receptor, thus underlining the implication of this xenobiotic-sensing receptor. Increase in CYP3A4 mRNA in response to SDHIs paralleled enhanced CYP3A4 protein expression for most of SDHIs. With respect to CYP3A4 activity, it was directly inhibited by some SDHIs, including bixafen, fluopyram, fluxapyroxad, isofetamid, isopyrazam, penthiopyrad and sedaxane, which therefore appears as dual regulators of CYP3A4, being both inducer of its expression and inhibitor of its activity. The inducing effect nevertheless predominates for these SDHIs, except for isopyrazam and sedaxane, whereas boscalid and flutolanil were pure inducers of CYP3A4 expression and activity. Most of SDHIs appear therefore as in vitro inducers of CYP3A4 expression in cultured hepatic cells, when, however, used at concentrations rather higher than those expected in humans in response to environmental or dietary exposure to these agrochemicals. • SDHIs induce CYP3A4 mRNA and protein level in human hepatic HepaRG cells. • Inhibition of the Pregnane-X-receptor prevents SDHIs-mediated CYP3A4 induction. • Some SDHIs act as direct in vitro inhibitors of CYP3A4 activity. • CYP3A4 mRNA induction predominates over activity inhibition for various SDHIs. • SDHIs were however predicted to poorly induce in vivo CYP3A4 expression.
