Abstract

Neuroscience research has expanded dramatically over the past 30 years by advancing standardization and tool development to support rigor and transparency. Consequently, the complexity of the data pipeline has also increased, hindering access to FAIR (Findable, Accessible, Interoperabile, and Reusable) data analysis to portions of the worldwide research community. brainlife.io was developed to reduce these burdens and democratize modern neuroscience research across institutions and career levels. Using community software and hardware infrastructure, the platform provides open-source data standardization, management, visualization, and processing and simplifies the data pipeline. brainlife.io automatically tracks the provenance history of thousands of data objects, supporting simplicity, efficiency, and transparency in neuroscience research. Here brainlife.io's technology and data services are described and evaluated for validity, reliability, reproducibility, replicability, and scientific utility. Using data from 4 modalities and 3,200 participants, we demonstrate that brainlife.io's services produce outputs that adhere to best practices in modern neuroscience research.

16. The Memory Binding Test Detects Early Subtle Episodic Memory Decline in Preclinical Alzheimer's Disease: A Longitudinal Study.

Abstract

Background: The asymptomatic at-risk phase might be the optimal time-window to establish clinically meaningful endpoints in Alzheimer's disease (AD). Objective: We investigated whether, compared with the Free and Cued Selective Reminding Test (FCSRT), the Memory Binding Test (MBT) can anticipate the diagnosis of emergent subtle episodic memory (EM) deficits to an at-risk phase. Methods: Five-year longitudinal FCSRT and MBT scores from 45 individuals matched for age, education, and gender, were divided into 3 groups of 15 subjects: Aβ-/controls, Aβ+/stable, and Aβ+/progressors (preclinical-AD). The MBT adds an associative memory component (binding), particularly sensitive to subtle EM decline. Results: In the MBT, EM decline started in the Aβ+/progressors (preclinical-AD) up to 4 years prior to diagnosis in delayed free recall (FR), followed by decline in binding-associated scores 1 year later. Conversely, in the FCSRT, EM-decline began later, up to 3 years prior to diagnosis, in the same group on both immediate and delayed versions of FR, while on total recall (TR) and intrusions decline started only 1 year prior to diagnosis. Conclusions: The MBT seems more sensitive than the FCSRT for early EM-decline detection, regarding the year of diagnosis and the number of scores showing AD-linked EM deficits (associated with the AD-characteristic amnesic hippocampal syndrome). Considering the MBT as a detection tool of early subtle EM-decline in an asymptomatic at-risk phase, and the FCSRT as a classification tool of stages of EM-decline from a preclinical phase, these tests ought to potentially become complementary diagnostic tools that can foster therapies to delay cognitive decline. Abstract

Problem‐solving often requires creativity and is critical in everyday life. However, the neurocognitive mechanisms underlying creative problem‐solving remain poorly understood. Two mechanisms have been highlighted: the formation of new connections among problem elements and insight solving, characterized by sudden realization of a solution. In this study, we investigated EEG activity during a modified version of the remote associates test, a classical insight problem task that requires finding a word connecting three unrelated words. This allowed us to explore the brain correlates associated with the semantic remoteness of connections (by varying the remoteness of the solution word across trials) and with insight solving (identified as a Eurêka moment reported by the participants). Semantic remoteness was associated with power increase in the alpha band (8–12 Hz) in a left parieto‐temporal cluster, the beta band (13–30 Hz) in a right fronto‐temporal cluster in the early phase of the task, and the theta band (3–7 Hz) in a bilateral frontal cluster just prior to participants' responses. Insight solving was associated with power increase preceding participants' responses in the alpha and gamma (31–60 Hz) bands in a left temporal cluster and the theta band in a frontal cluster. Source reconstructions revealed the brain regions associated with these clusters. Overall, our findings shed new light on some of the mechanisms involved in creative problem‐solving. Abstract

International audience; Despite being a promising tool for establishing direct communication and control from the brain over external effectors, non-invasive Brain-computer interface (BCI) systems yield suboptimal performance in ~30% of users. Measuring the dynamical features that are relevant to the execution of a task to improve BCI performance remains an open challenge. Functional imaging is dominated by aperiodic and scale-free perturbations called “Neuronal Avalanches”, spreading across the whole brain. The sequence of regions recruited by avalanches could convey the processes underpinning BCI performance. To test our hypothesis, we used source-reconstructed magnetoencephalography signals in a BCI framework, where 20 healthy subjects were compared in resting-state and while performing a motor imagery (MI) task, in order to track the dynamical features related to motor imagery. Each signal was z-scored over time. For each condition, we estimated the avalanche transition matrix (ATM), containing the probability that regions j would be active at time t+1, given region i was active at time t. We computed the difference between the ATMs obtained from the two conditions in each subject, and validated them via permutation analysis. Then, we correlated the difference of the probabilities to BCI performance. All the significantly different edges cluster upon the premotor areas, involved during the MI task, and the cunei, involved during visual processing. The differences in the probabilities associated with edges incident upon areas such as the right paracentral lobule and the caudal middle frontal bilaterally directly correlate with the BCI scores. Our results suggest that avalanches capture functionally-relevant processes which are of interest for alternative BCI designing. Future work will consist of investigating the use of avalanche transition matrices as potential alternative features for classification in BCI.

Abstract

International audience; Brain-Computer Interfaces (BCIs) constitute a promising tool for communication and control. However, mastering non-invasive BCI systems remains a learned skill difficult to develop for a non-negligible proportion of users. Even though similarities have been shown between MI-based BCI learning and motor sequence learning our understanding of the dynamical processes, and their reflection on brain signals during BCI performance is still incomplete. In particular, whole-brain functional imaging is dominated by a 'bursty' dynamics, "neuronal avalanches", with fast, fat-tailed distributed, aperiodic perturbations spreading across the whole brain. Neuronal avalanches evolve over a manifold during resting-state, generating a rich functional connectivity dynamics. In this work, we evaluated to which extent neuronal avalanches can be used as a tool to differentiate mental states in the context of BCI experiments.

Abstract

International audience; The reconfiguration of large-scale interactions among multiple brain regions is characterized by aperiodic perturbations, called “Neuronal Avalanches”, which can be tracked non-invasively as they expand across the brain. As such, learning a new task might affect the path of propagation of neuronal avalanches. Brain-Computer Interfaces (BCIs) constitute a promising tool for establishing direct communication and control from the brain over external effectors but mastering them remains a poorly-understood learned skill. Therefore, neuronal avalanche measures may constitute natural candidates to inform the underlying brain processes and their reflection on brain signals. To test this hypothesis, we used source-reconstructed magneto/electroencephalography, comparing resting-state to motor imagery conditions during a BCI protocol. For each experimental condition, we computed an individual avalanche transition matrix, to track the probability that an avalanche would spread across any two regions. We found a robust topography of the edges that were affected by the execution of the task, which mainly hinge upon the premotor regions. Finally, we related the individual differences to the task performance, showing that significant correlations are predominantly positive and involve edges connecting pre/motor regions to parietal ones. Our findings suggest that avalanches capture functionally-relevant processes crucial for alternative BCI designing.

Abstract

International audience; Non-invasive Brain-Computer Interfaces (BCIs) can exploit the ability of subjects to voluntary modulate their brain activity through mental imagery. Despite its clinical applications, controlling a BCI appears to be a learned skill that requires several weeks to reach relatively high-performance in control, without being sufficient for 15 to 30 % of the users. This gap has motivated a deeper understanding of mechanisms associated with motor imagery (MI) tasks. Here, we investigated dynamical changes in terms of cortical activations and network recruitment. We hypothesized that the associated characteristics would be able to predict the success of learning.

Abstract

International audience; Brain-Computer Interfaces (BCIs) constitute a promising tool for communication and control. However, mastering non-invasive BCI systems remains a learned skill difficult to develop for a non-negligible proportion of users. Even though similarities have been shown between MI-based BCI learning and motor sequence learning our understanding of the dynamical processes, and their reflection on brain signals during BCI performance is still incomplete. In particular, whole-brain functional imaging is dominated by a ‘bursty’ dynamics, “neuronal avalanches”, with fast, fat-tailed distributed, aperiodic perturbations spreading across the whole brain. Neuronal avalanches evolve over a manifold during resting-state, generating a rich functionalconnectivity dynamics. In this work, we evaluated to which extent neuronal avalanches can be used as a tool to differentiate mental states in the context of BCI experiments.

International audience; A progressive desynchronization of beta-band power (β-desync; 13-30 Hz) has been consistently observed before movement initiation. Although mounting evidence demonstrate a significant relationship between this β-desync and subsequent motor vigor, a neurophysiological foundation to explain this finding is still lacking. In the present study, we proposed that this association might come from changes in motor motivation and perceived effort. This hypothesis will be tested by recording the signal from electroencephalography and using it as neurofeedback to decrease or increase β-desync in two distinct sessions, in an intra-subject and double-blinded design. First, participants will be screened based on their ability to up- and down-regulate their β-desync. The selected participants will then perform the experimental test, in which each trial comprised a neurofeedback phase directly followed by an effort phase. The effort will consist in squeezing a dynamometer with the right hand during an allocated time of 10 seconds. Participants will be asked to apply enough force to cross a defined threshold, and that the more time the exerted force would exceed the threshold, the more money they would earn. Subjective perception of effort will be evaluated afterwards using an analog scale. Because effort exertion is associated with increased pupil dilation, which is itself modulated by noradrenergic activity, we will further investigate the influence of noradrenergic activity on changes in β-desync due to neurofeedback and effort exertion. Besides the contribution to unraveling the neurophysiological and functional bases of beta activity, the present work is of special interest to the study of Parkinson’s disease by helping to better understand the association between beta activity and motor symptoms.

Abstract

International audience; Motor Imagery-based BCI (MI-BCI) performances remain highly variable, with up to 30% of users unable to achieve control. Operating a BCI system requires users to learn to modulate their neural activity. This is a complex task, thus the training procedure is crucial. Our study draws on task-feedback transparency to improve the feedback given during training. MI-BCI protocols often feature abstract feedback (e.g., visual gauge) unrelated to the imagined movement. Transparent feedback, directly related to the task, may reinforce agency-the ability to self-attribute actions, such as feedback movements-and embodiment, facilitating kinesthetic MI. This could lessen complexity in BCI control and ultimately improve BCI performance.We tested this hypothesis in a beta-based MI neurofeedback task with electroencephalography (EEG). Twenty-three subjects participated in a single MIneurofeedback session that consisted in imagining clenching their right hand rhythmically. Feedback was based on online laplacian-filtered 8-30Hz power on C3 electrode computed with OpenViBE. There were 3 feedback conditions of expected increasing transparency: pendulum; virtual hand; virtual hand with additional motor illusion induced by vibrations to the participant's hand. The amplitude of the pendulum and virtual hand movements was proportional to beta desynchronisation. Each participant performed two runs of five 24s-trials per condition. Agency, embodiment, and user experience questionnaires were included. Repeated-measures ANOVA showed better BCI performance when using the virtual hand than the pendulum. However, participants had lower performance in the visuotactile condition compared to the virtual hand-only feedback. Agency showed a similar pattern.Feeling of success in the task did not vary with feedback conditions, but the virtual hand feedback felt easier to use. BCI performance correlated positively with agency for all conditions and with embodiment in conditions with a virtual hand.Thus, visual transparency—but not tactile feedback addition—increased BCI performance and this was associated with better agency and embodiment.

Abstract

International audience; The adoption of a technology can be defined as the decision made by a person or a society to accept and use it. It is based on two phases during which the willingness to use that technology is assessed. The first phase corresponds to the evaluation of the acceptability of the technology and takes place before the use while the second phase, the technology acceptance, comes afterwards. Assessing the acceptability of BCIs/NF is of utmost importance as it could provide i) insights on the propensity of people to use these technologies and ii) leads to facilitate their adoption.We reviewed the literature dealing with the acceptability of BCIs/NF and using validated questionnaires and models. Through a reproducible search method, we retrieved a list of 55 main references of acceptability questionnaires and models. Then, we searched Scopus, PubMed, and Web of Science to identify the articles in the fields of BCIs/NF that cited these references. We included in our review all the articles reporting experimental results on the acceptability of BCIs/NF that were based on one or several validated acceptability questionnaire(s).Four articles were included in our review. Two of them assessed the acceptability while the others assessed the acceptance of the BCIs. While there are too few articles included to conclude on the acceptability of those technologies, the results are encouraging. BCIs were perceived as useful at 78% by motor impaired individuals who used BCIs for pain management and at 84% by neurotypical individuals who used BCIs to browse an instruction manual. The perceived ease of use varied from 30% to 80%, which could be related to differences in the protocols. Among other factors, the presence of social support appeared to influence the acceptability.

Abstract

Background: Alzheimer's disease (AD) includes progressive symptoms spread along a continuum of preclinical and clinical stages. Although numerous studies uncovered the neuro-cognitive changes of AD, very little is known on the natural history of brain lesions and modifications of brain networks in elderly cognitively-healthy memory complainers at risk of AD for carrying pathophysiological biomarkers (amyloidopathy and tauopathy).Objective: We analyzed resting-state electroencephalography (EEG) of 318 cognitively-healthy subjective memory complainers from the INSIGHT-preAD cohort at the time of their first visit (M0) and two-years later (M24).Methods: Using 18F-florbetapir PET-scanner, subjects were stratified between amyloid negative (A-; n = 230) and positive (A+; n = 88) groups. Differences between A+ and A- were estimated at source-level in each band-power of the EEG spectrum.Results: At M0, we found an increase of theta power in the mid-frontal cortex in A+ compared to A-. No significant association was found between mid-frontal theta and the individuals' cognitive performance. At M24, theta power increased in A+ relative to A- individuals in the posterior cingulate cortex and the pre-cuneus. Alpha band revealed a peculiar decremental trend in posterior brain regions in the A+ relative to the A- group only at M24. Theta power increase over the mid-frontal and mid-posterior cortices suggests an hypoactivation of the default-mode network in the A+ individuals and a non-linear longitudinal progression at M24.Conclusion: We provide the first source-level longitudinal evidence on the impact of brain amyloidosis on the EEG dynamics of a large-scale, monocentric cohort of elderly individuals at-risk for AD. Categorization abilities are typically measured by the Wechsler Adult Intelligence Scale (WAIS) similarity subtest, which consists of naming the shared category of two items (e.g., 'How are beer and coffee alike'). Previous studies show that categorization, as measured by similarity tasks, requires executive control functions. However, other theories and studies indicate that semantic memory is organized into taxonomic and thematic categories that can be activated implicitly in semantic priming tasks. To explore whether categories can be primed during a similarity task, we developed a double semantic priming paradigm. We measured the priming effect of two primes on a target word that was taxonomically or thematically related to both primes (double priming) or only one of them (single priming). Our results show a larger and additive priming effect in the double priming condition compared to the single priming condition, as measured by both response times and, more consistently, event-related potentials. Our results support the view that taxonomic and thematic categorization can occur during a double priming task and contribute to improving our knowledge on the organization of semantic memory into categories. These findings show how abstract categories can be activated, which likely shapes the way we think and interact with our environment. Abstract

Response inhibition is a crucial component of executive control. Although mainly studied in upper limb tasks, it is fully implicated in gait initiation. Here, we assessed the influence of proactive and reactive inhibitory control during gait initiation in healthy adult participants. For this purpose, we measured kinematics and electroencephalography (EEG) activity (event‐related potential [ERP] and time‐frequency data) during a modified Go/NoGo gait initiation task in 23 healthy adults. The task comprised Go‐certain, Go‐uncertain, and NoGo conditions. Each trial included preparatory and imperative stimuli. Our results showed that go‐uncertainty resulted in delayed reaction time, without any difference for the other parameters of gait initiation. Proactive inhibition, that is, Go uncertain versus Go certain conditions, influenced EEG activity as soon as the preparatory stimulus. Moreover, both proactive and reactive inhibition influenced the amplitude of the ERPs (central P1, occipito‐parietal N1, and N2/P3) and theta and alpha/low beta band activities in response to the imperative—Go‐uncertain versus Go‐certain and NoGo versus Go‐uncertain—stimuli. These findings demonstrate that the uncertainty context; induced proactive inhibition, as reflected in delayed gait initiation. Proactive and reactive inhibition elicited extended and overlapping modulations of ERP and time‐frequency activities. Abstract

Introduction: Anticipating the diagnosis of Alzheimer's disease (AD) at an early asymptomatic at-risk stage, where therapeutics can more effectively delay conscious cognitive decline, is currently among the biggest challenges in the field. Herein, we aimed to compare the capacity of the Memory Binding Test (MBT) with the official diagnostic tool, the Free and Cued Selective Reminding Test (FCSRT), to anticipate AD diagnosis at an early preclinical stage based on the associative memory component of MBT (binding), suggested as more sensitive to the emergence of subtle episodic memory (EM) deficits (AD hallmark)., Methods: We assessed the tests performance longitudinally (over 5 years) in 263 cognitively-normal elderly individuals at risk of AD (>6 months of subjective memory complaints) using linear mixed-effect models controlled for age, sex, and education. We stratified participants in 2 models: amyloid-β (Aβ)/neurodegeneration (N) model, assessing Aβ burden and neurodegeneration effect [3 groups: controls (Aβ-/N-); stable/N- (Aβ+); stable/N+ (Aβ+)]; and the stable/progressors model, assessing progression to prodromal-AD effect [2 groups: stable (Aβ+); progressors (Aβ+)], based on 15 subjects who progressed to AD during follow-up (excluded once diagnosed)., Results: Aβ burden was associated with significantly less MBT-intrusions, while Aβ burden and neurodegeneration together, with the most. Progression status had a strong negative effect on both tests performance. 46. Measuring neuronal avalanches to inform brain-computer interfaces.

Abstract

Large-scale interactions among multiple brain regions manifest as bursts of activations called neuronal avalanches, which reconfigure according to the task at hand and, hence, might constitute natural candidates to design brain-computer interfaces (BCIs). To test this hypothesis, we used source-reconstructed magneto/electroencephalography during resting state and a motor imagery task performed within a BCI protocol. To track the probability that an avalanche would spread across any two regions, we built an avalanche transition matrix (ATM) and demonstrated that the edges whose transition probabilities significantly differed between conditions hinged selectively on premotor regions in all subjects. Furthermore, we showed that the topology of the ATMs allows task-decoding above the current gold standard. Neuroscience research has expanded dramatically over the past 30 years by advancing standardization and tool development to support rigor and transparency. Consequently, the complexity of the data pipeline has also increased, hindering access to FAIR data analysis to portions of the worldwide research community. brainlife.io was developed to reduce these burdens and democratize modern neuroscience research across institutions and career levels. Using community software and hardware infrastructure, the platform provides open-source data standardization, management, visualization, and processing and simplifies the data pipeline. brainlife.io automatically tracks the provenance history of thousands of data objects, supporting simplicity, efficiency, and transparency in neuroscience research. Here brainlife.io's technology and data services are described and evaluated for validity, reliability, reproducibility, replicability, and scientific utility. Abstract

Epilepsy presurgical investigation may include focal intracortical single-pulse electrical stimulations with depth electrodes, which induce cortico-cortical evoked potentials at distant sites because of white matter connectivity. Cortico-cortical evoked potentials provide a unique window on functional brain networks because they contain sufficient information to infer dynamical properties of large-scale brain connectivity, such as preferred directionality and propagation latencies.  Here, we developed a biologically informed modelling approach to estimate the neural physiological parameters of brain functional networks from the cortico-cortical evoked potentials recorded in a large multicentric database. Specifically, we considered each cortico-cortical evoked potential as the output of a transient stimulus entering the stimulated region, which directly propagated to the recording region. Both regions were modelled as coupled neural mass models, the parameters of which were estimated from the first cortico-cortical evoked potential component, occurring before 80 ms, using dynamic causal modelling and Bayesian model inversion. This methodology was applied to the data of 780 patients with epilepsy from the F-TRACT database, providing a total of 34 354 bipolar stimulations and 774 445 cortico-cortical evoked potentials. The cortical mapping of the local excitatory and inhibitory synaptic time constants and of the axonal conduction delays between cortical regions was obtained at the population level using anatomy-based averaging procedures, based on the Lausanne2008 and the HCP-MMP1 parcellation schemes, containing 130 and 360 parcels, respectively. To rule out brain maturation effects, a separate analysis was performed for older (>15 years) and younger patients (<15 years). In the group of older subjects, we found that the cortico-cortical axonal conduction delays between parcels were globally short (median = 10.2 ms) and only 16% were larger than 20 ms. This was associated to a median velocity of 3.9 m/s. Although a general lengthening of these delays with the distance between the stimulating and recording contacts was observed across the cortex, some regions were less affected by this rule, such as the insula for which almost all efferent and afferent connections were faster than 10 ms. Synaptic time constants were found to be shorter in the sensorimotor, medial occipital and latero-temporal regions, than in other cortical areas. Finally, we found that axonal conduction delays were significantly larger in the group of subjects younger than 15 years, which corroborates that brain maturation increases the speed of brain dynamics. To our knowledge, this study is the first to provide a local estimation of axonal conduction delays and synaptic time constants across the whole human cortex in vivo, based on intracerebral electrophysiological recordings., (© The Author(s) (2021). Abstract

Categorization - whether of objects, ideas, or events - is a cognitive process that is essential for human thinking, reasoning, and making sense of everyday experiences. Categorization abilities are typically measured by the Wechsler Adult Intelligence Scale (WAIS) similarity subtest, which consists of naming the shared category of two items (e.g., 'How are beer and coffee alike'). Previous studies show that categorization, as measured by similarity tasks, requires executive control functions. However, other theories and studies indicate that semantic memory is organized into taxonomic and thematic categories that can be activated implicitly in semantic priming tasks. To explore whether categories can be primed during a similarity task, we developed a double semantic priming paradigm. We measured the priming effect of two primes on a target word that was taxonomically or thematically related to both primes (double priming) or only one of them (single priming). Our results show a larger and additive priming effect in the double priming condition compared to the single priming condition, as measured by both response times and, more consistently, event-related potentials. Our results support the view that taxonomic and thematic categorization can occur during a double priming task and contribute to improving our knowledge on the organization of semantic memory into categories. These findings show how abstract categories can be activated, which likely shapes the way we think and interact with our environment. Our study also provides a new cognitive tool that could be useful to understand the categorization difficulties of neurological patients.
