Plants adapt to balance growth–defense tradeoffs in response to both biotic and abiotic stresses. Green leaf volatiles (GLVs) are released after biotic and abiotic stresses and function as damage-associated signals in plants. Although, GLVs enter plants primarily through stomata, the role of stomatal regulation on the kinetics of GLV uptake remains largely unknown. Here, we illustrate the effect of stomatal closure on the timing and magnitude of GLV uptake. We closed stomata by either exposing maize (Zea mays) plants to darkness or applying abscisic acid, a phytohormone that closes the stomata in light. Then, we exposed maize seedlings to (Z)-3-hexen-1-ol and compared its dynamic uptake under different stomatal conditions. Additionally, we used (E)-3-hexen-1-ol, an isomer of (Z)-3-hexen-1-ol not made by maize, to exclude the role of internal GLVs in our assays. We demonstrate that closed stomata effectively prevent GLV entry into exposed plants, even at high concentrations. Furthermore, our findings indicate that reduced GLV uptake impairs GLV-driven induction of biosynthesis of sesquiterpenes, a group of GLV-inducible secondary metabolites, with or without herbivory. These results elucidate how stomata regulate the perception of GLV signals, thereby dramatically changing the plant responses to herbivory, particularly under water stress or dark conditions. Lepidopterans commonly feed on plant material, being the most significant insect herbivores in nature. Despite plant resistance to herbivory, such as producing toxic secondary metabolites, herbivores have developed mechanisms encoded in their genomes to tolerate or detoxify plant defensive compounds. Recent studies also highlight the role of gut microbiota in mediating detoxification in herbivores; however, convincing evidence supporting the significant contribution of gut symbionts is rare in Lepidoptera. Here, we show that the growth of various lepidopteran species was inhibited by a mulberry-derived secondary metabolite, 1-deoxynojirimycin (DNJ); as expected, the specialist silkworm Bombyx mori grew well, but interestingly, gut microbiota of early-instar silkworms was affected by the DNJ level, and several bacterial species responded positively to enriched DNJ. Among these, a bacterial strain isolated from the silkworm gut (Pseudomonas fulva ZJU1) can degrade and utilize DNJ as the sole energy source, and after inoculation into nonspecialists (e.g., beet armyworm Spodoptera exigua), P. fulva ZJU1 increased host resistance to DNJ and significantly promoted growth. We used genomic and transcriptomic analyses to identify genes potentially involved in DNJ degradation, and CRISPR-Cas9- mediated mutagenesis verified the function of ilvB, a key binding protein, in metabolizing DNJ. Furthermore, the ilvB deletion mutant, exhibiting normal bacterial growth, could no longer enhance nonspecialist performance, supporting a role in DNJ degradation in vivo. Therefore, our study demonstrated causality between the gut microbiome and detoxification of plant chemical defense in Lepidoptera, facilitating a mechanistic understanding of host-microbe relationships across this complex, abundant insect group. Plants perceive environmental stresses as whole organisms via distant signals conveying danger messages through their vasculature. In parallel to vascular transport, airborne plant volatile compounds, including green leaf volatiles (GLVs), can bypass the lack of vascular connection. However, some small volatile compounds move through the vasculature; such vascular transport is little known about GLVs. Here we illustrate GLV alcohols as solutes move within xylem vessels in Zea mays. We describe GLV alcohols, including Z‐3‐hexen‐ol and its isomer E‐3‐hexen‐ol, which is not synthesized in maize, moving through the transpiration stream via xylem vessels. Since transpiration is mediated by the stomatal aperture, closing stomata by two independent methods diminishes the transport of GLV alcohol and its isomer. In addition, the lower transport of GLV alcohols impairs their function in inducing terpenoid biosynthesis, suggesting that xylem transport of GLV alcohols plays a significant role in their systemic function. Our study suggests that GLV alcohols, in addition to airborne signals, are transported through xylem vessels. Our findings can be critical in future studies about the perception and function of these compounds in plants. Water is essential to plant growth and drives plant evolution and interactions with other organisms such as herbivores. However, water availability fluctuates, and these fluctuations are intensified by climate change. How plant water availability influences plant–herbivore interactions in the future is an important question in basic and applied ecology. Here we summarize and synthesize the recent discoveries on the impact of water availability on plant antiherbivore defense ecology and the underlying physiological processes. Water deficit tends to enhance plant resistance and escape traits (i.e. early phenology) against herbivory but negatively affects other defense strategies, including indirect defense and tolerance. However, exceptions are sometimes observed in specific plant–herbivore species pairs. We discuss the effect of water availability on species interactions associated with plants and herbivores from individual to community levels and how these interactions drive plant evolution. Although water stress and many other abiotic stresses are predicted to increase in intensity and frequency due to climate change, we identify a significant lack of study on the interactive impact of additional abiotic stressors on water–plant–herbivore interactions. Abiotic stressors, such as salt stress, can reduce crop productivity, and when combined with biotic pressures, such as insect herbivory, can exacerbate yield losses. However, salinity-induced changes to plant quality and defenses can in turn affect insect herbivores feeding on plants. This study investigates how salinity stress in tomato plants (Solanum Lycopersicum cv. Better Boy) impacts the behavior and performance of a devastating insect pest, the tomato fruitworm caterpillar (Helicoverpa zea). Through choice assays and performance experiments, we demonstrate that salt-stressed tomato plants are poor hosts for H. zea, negatively affecting caterpillar feeding preferences and growth rates. While changes in plant nutritional quality were observed, the primary factor influencing insect performance appears to be direct ionic toxicity, which significantly impairs multiple life history parameters of H. zea including survival, pupation, adult emergence, and fecundity. Plant defense responses show complex interactions between salt stress and herbivory, with two proteinase inhibitor genes - PIN2 and AspPI, showing a higher induced response to insect herbivory under salt conditions. However, plant defenses do not seem to be the main driver of reduced caterpillar performance on salt-treated plants. Furthermore, we report reduced oviposition by H. zea moths on salt-treated plants, which was correlated with altered volatile emissions. Our findings reveal that H. zea exhibits optimal host selection behaviours for both larval feeding and adult oviposition decisions, which likely contribute to its success as an agricultural pest. This research provides insights into the complex interactions between abiotic stress, plant physiology, and insect behaviour, with potential implications for pest management strategies in saline agricultural environments., (© 2025 The Author(s).
