This study aimed to investigate the effect of an essential oil/fumaric combination, mannan-oligosaccharide, galactooligosaccharide, and a mannan-oligosaccharide/galactooligosaccharide combination on the dry matter disappearance (DMD), gas production, greenhouse gasses, volatile fatty acid, and microbial community of a total mixed ration using a 24 h in vitro batch culture technique. The study design was a completely randomized design with four treatments as follows: a control treatment without any additives, the control treatment supplemented with galactooligosaccharide at 3% (Gos treatment), a galactooligosaccharide and mannan-oligosaccharide mixture at 1:1 at 3% (Gosmos treatment), or an essential oil blend (200 μL/g feed) and fumaric acid at 3% combination (Eofumaric treatment). The Gosmos treatment had the highest (p < 0.05) DMD (63.8%) and the numerical lowest acetate–propionate ratio (p = 0.207), which was 36.9% higher compared to the control. The lowest Shannon index, Simpson's index, and all the diversity indices were recorded for the Eofumaric treatment, while the other treatments had similar Shannon index, Simpson's index, and diversity index. The Z-score differential abundance between the Eofumaric and the control indicated that the inclusion of the Eofumaric treatment differentially increased the abundance of Patescibacteria, Synergistota, Chloroflexi, Actinobacteriota, Firmicutes, and Euryarchaeota while Verrucomicrobiota, WPS-2, Fibrobacterota, and Spirochaetota were decreased. The Random Forest Classification showed that the lower relative abundance of Fibrobacterota, Spirochaetota, and Elusimicrobiota and the higher relative abundance of Firmicutes and Chloroflexi were most impactful in explaining the microbial community data. Overall, the essential oil blend showed great potential as a methane gas mitigation strategy by modifying rumen fermentation through changes in the microbial community dynamics. Simple Summary: Essential oil blends show promise as natural ruminant feed additives, potentially modulating rumen fermentation and mitigating greenhouse gas emissions. The present study investigated the metabolite profiles, methane production, and fermentation dynamics of five novel essential oil blends in Black Angus beef cattle. The blends, composed of four distinct essential oils in varying proportions, were directly infused into the rumen to simulate practical application. Advanced metabolomics techniques were employed to analyze and elucidate the EOB-induced changes in the rumen fluid. Results demonstrated that the infusion of the blends modified the rumen metabolism, fermentation patterns, and metabolite production. The study provides insights into how the essential oil blends influenced the specific metabolic processes and fermentation characteristics in the rumen. This research contributes to the development of bio-based strategies for optimizing rumen function, offering valuable information for animal nutritionists and producers aiming to improve feed efficiency and minimize the environmental footprint of ruminant livestock production. GHG emissions were not significantly affected, but a numerical decrease was observed in the animals infused with the EOB5 treatment. Ammonia nitrogen concentrations remained within the suitable range for rumen microbes' growth, indicating a normal internal environment for microbial crude protein synthesis. In conclusion, the study has demonstrated that the direct infusion of EOBs significantly improved the generation of VFAs and impacted the energy production, protein synthesis, and microbial activity of the animals. In this study, the chemical compositions of rosemary (Rosmarinus officinalis Spenn.) and sweet marjoram (Origanum majorana L.) essential oils, both individually and in various blend ratios, were investigated by using Gas Chromatography-Mass Spectrometry. Their antifungal activities were also determined against the potato rubbery rot disease agent, Geotrichum candidum, under in vitro and in vivo conditions. Camphor (20.69%) and terpinen-4-ol (35.13%) were determined as primary constituents in rosemary and sweet marjoram essential oils, respectively. Sweet marjoram and rosemary essential oils completely inhibited mycelial growth of G. candidum at 55.0 and 65.0 µL/Petri concentrations, respectively. Essential oils at these concentrations were found as fungicidal. Notably, a synergistic fungicidal antifungal effects were also observed in essential oil blends. Blend of sweet marjoram and rosemary essential oil in a 3:1 ratio (O75R25) significantly enhanced antifungal activity at the relatively lower concentration (45.0 µL/Petri). Microscopic observations revealed structural deformations in exposed hyphae, including cytoplasmic coagulation and vacuolization. Essential oil blend (O75R25), sweet marjoram and rosemary essential oils completely inhibited the disease lesion caused by G. candidum on potato tubers at concentrations of 100.0, 125.0, and 150.0 µL/L air, respectively. These findings emphasized that essential oil blends of rosemary and sweet marjoram have synergistic antifungal potential and can be used as effective biofungicides against plant fungal diseases. The present study evaluated the inclusion of fumaric acid and essential oil blends (EOBs) containing anise, cedarwood, clove, cumin, eucalyptus, garlic, ginger, lavender, lemongrass, nutmeg, oregano, and peppermint at different proportions on in vitro dry matter (DM) disappearance (DMD), fiber fraction disappearance, the efficiency of microbial production, and the total volatile fatty acids (VFAs). Ten treatments without (control treatment) or with different EOB/fumaric combinations were used in the study with eight replicates. The EOB inclusion level was 200 μL/g of feed (total mixed ration, (TMR)) while fumaric acid was administered at 3% of the TMR (DM basis). The highest DMD, in vitro true degradable DM, partitioning factor (PF24), and in vitro apparent degradable DM were recorded for the fumaric only treatment and the control. Neutral detergent fiber disappearance was reduced with the inclusion of EOB/fumaric combinations. The production of microbial mass and undegraded DM were higher (p < 0.001) for all EOBs and EOB and fumaric treatments. The inclusion of EOB and fumaric combinations reduced (p < 0.001) the total gas production, methane, and ammonia, with a higher PF24 value noted for EOB3 treatment. The inclusion of individual EOB1 containing garlic, lemongrass, cumin, lavender, and nutmeg in a ratio of 4:2:2:1:1 or combined with fumaric acid yielded the highest propionate concentration across all treatments. We concluded that EOBs decreased methane production and nutrient degradability with better results with the individual EOB1 or EOB1/fumaric combination, which showed a potential enhancement in energy production. A simple green method for the quantification of sweet orange and lavender essential oils (EOs) content in blends was developed using synchronous fluorescence of the first derivative spectra and sample preparation by surfactant-free microemulsions (SFMEs). Excitation and emission wavelengths pairs (λex/λem) were determined for both EOs (at 336/436 nm for sweet orange and at 330/388 nm for lavender). Optimization was conducted to establish the condition to prepare the SFMEs systems: 50 μL of oily phase containing EO diluted in octan-1-ol (1:4, v/v), 2.0 mL of water and propan-1-ol up to 5.0 mL final volume. Different proportions of EO in the oily phase were evaluated for analytical curves construction. Synchronous fluorescence measurements were performed and the acquired data were corrected by inner filter effect. Considering the first derivative of the synchronous scanning, data were extracted at 316.8 nm (Δλ = 100 nm) and 352.0 nm (Δλ = 58 nm) for sweet orange and lavender EOs, respectively, with correlation coefficients above 0.99. Limits of quantification of 7.97 μg mL−1 and 65.80 μg mL−1 were obtained indicating that 0.46% of sweet orange and 3.69% of lavender EOs can be quantified in mixtures. Simulated blends (25:75%, 50:50% and 75:25%, volume proportions) were also evaluated with recoveries of 71.5 – 128.9% and coefficient of variation between 3.1% – 11.6%. Additionally, the method's greenness was evaluated through three green metrics (Analytical Eco-Scale, GAPI, and AGREE) with consistent results.

Palm leaf manuscripts (PLMs) are traditional carriers that contain a wealth of knowledge from different disciplines written by ancient scholars. However, PLMs are susceptible to deterioration due to moisture, light, fungi and bacteria. An existing method of applying essential oils (EOs) to preserve PLMs does not currently appear successful, hence the study presented here which aimed to maximise the preservation characteristics of six different EO blends, prepared and applied to palm leaves, which were then tested for stiffness, brightness and weight gain. The interpretation of the efficiency of EO blends in the preservation of PLMs is attributed to the compatibility between the oils due to the various constituents present in the individual EOs. FTIR spectroscopy was used to understand the compatibility of the oils in a blend and any EO blend rendering a higher stiffness and brightness was identified as having a high preservation performance. Based on the investigation, good compatibility is observed between the FTIR spectrum of Blend-E with its contributing EOs of citronella, neem and mustard. On the other hand, Blend-B exhibits poor preservation characteristics and incompatibility. The study concludes with the recommendation that Blend-E is suitable for the treatment and regular maintenance of PLMs as it can help reduce physical and chemical damage. [ABSTRACT FROM AUTHOR]
