Abstract

In the quest for early cancer diagnosis, early identification and treatment are paramount. Recently, ctDNA detection has emerged as a viable avenue for early screening of cancer. The examination of ctDNA in fluid biopsies has gained substantial attention in tumor diagnosis and therapy. Both the scientific community and industry are actively exploring this field. However, developing cost-effective, portable, and real-time ctDNA measurement methods using conventional gene detection equipment poses a significant challenge. This challenge has led to the exploration of alternative approaches. Electrochemical biosensors, distinguished by their heightened sensitivity, remarkable specificity, affordability, and excellent portability, have emerged as a promising avenue for ctDNA detection. This review is dedicated to the specific focus on ctDNA detection, highlighting recent advancements in this evolving detection technology. We aimed to reference previous studies related to ctDNA-targeted cancer detection using electrochemical biosensors to advocate the utilization of electrochemical biosensors in healthcare diagnostics. Further research is imperative for the effective integration of ctDNA analysis into point-of-care cancer testing. 21. Molecular characterization of sequence variants of Chrysanthemumchlorotic mottle viroidfrom Himachal Pradesh, India

Abstract

Chrysanthemumis an important cut flower crop but is known to be infected with viruses and viroids. Here, we have identified and characterized two sequence variants of Chrysanthemum chlorotic mottle viroid(CCHMVd) from chrysanthemumvarieties Yellow Star and Orange Daisy that showed stunting, mottling and chlorosis-like symptoms, respectively. The viroid sequences were sequenced which upon BLAST analysis showed 97–98% sequence similarity to other CCHMVd isolates reported from India and world. CCHMVd disease incidence was found to be 15% that was higher in comparison to 8–10% in South Indian states. In phylogenetic analysis, the Palampur isolate grouped with other Indian isolates while isolate YS showed different ancestral origin and grouped separately with other world isolates.

22. Molecular detection and characterization of Potato spindle tuber viroid(PSTVd) infecting Tomato (Solanum lycopersicumL.) in Karnataka State of India

Abstract

Potato Spindle Tuber Viroid (PSTVd) is a non-coding, infectious, small, circular RNA known to cause disease in agricultural and horticultural plants. In the present work, an investigation was conducted in the southern districts of Karnataka state to assess the possible pospiviroid infections on tomato plants that are considered natural hosts for viroids. A total of 83 tomato samples showing disease symptoms (virus or viroid-like) along with healthy ones were collected and subjected to viroid detection by conventional Reverse Transcriptase Polymer Chain Reaction (RT-PCR) using universal (Pospi1-RE/Pospi1-FW) and a specific set of primers (3H1/2H1). The study confirmed the presence of PSTVd in one of the samples of tomato collected from Banghatta village of Mandy District, with an expected amplicon of ~ 361 bp. The bioassay conducted on tomato plants (cv. Rutgers) proved the association of PSTVd, which was further confirmed by RT-PCR. The amplicons were cloned, sequenced, and the representative sequences were deposited in the NCBI GenBank. The sequence alignment and secondary structure analysis of the isolated viroid with other reference sequences revealed the variations in the pathogenicity, central conserved region, and Terminal right domains. The variations observed between the isolated PSTVd with that of other Indian isolates support that viroid may have been transmitted among the crop plants, possibly through seed or mechanical means.

Abstract

Apple stem grooving virus (ASGV) is a highly variable RNA virus and recombination is one of the mechanisms that lead to variations in the viral genome. Here, we report the biological and molecular properties of two apple isolates of ASGV (p12 and MK) isolated from two different locations in India. Complete genome of both isolates consisted of 6493 nucleotides excluding the poly (A) tail and contained two open reading frames (ORFs). The isolates shared 97% sequence identity with each other. Phylogenetic analyses revealed neighbouring association of these isolates, but their infection on known herbaceous host, Phaseolus vulgaris, showed different phenotypes. Recombination analyses suggest isolate MK and p12 as non‐recombinants, but recombination events increased with increasing host diversity. Two recombination hotspots were identified in the ORF1 region spanning nucleotides 4500–4800 and 6200–6500, which were designated as recombination hot spots I and II, respectively. The findings here also suggest that this virus is emerging with expanding host range breadth and diversified host adaptability driven by recombination. [ABSTRACT FROM AUTHOR]
