International audience; Infant formula (IF) is a complex matrix requiring numerous ingredients and processing steps. The objective was to understand how the quality of protein ingredients impacts IF structure and, in turn, their kinetics of digestion. Four powdered IFs (A/B/C/D), based on commercial whey protein (WP) ingredients, with different protein denaturation levels and composition (A/B/C), and on caseins with different supramolecular organisations (C/D), were produced at a semi-industrial level after homogenization and spray-drying. Once reconstituted in water (13 %, wt/wt), the IF microstructure was analysed with asymmetrical flow field-flow fractionation coupled with multi-angle light scattering and differential refractometer, transmission electron microscopy and electrophoresis. The rehydrated IFs were subjected to simulated infant in vitro dynamic digestion (DIDGI®). Digesta were regularly sampled to follow structural changes (confocal microscopy, laser-light scattering) and proteolysis (OPA, SDS-PAGE, LC-MS/MS, cation-exchange chromatography). Before digestion, different microstructures were observed among IFs. IF-A, characterized by more denatured WPs, presented star-shaped mixed aggregates, with protein aggregates bounded to casein micelles, themselves adsorbed at the fat droplet interface. Non-micellar caseins, brought by non-micellar casein powder (IF-D) underwent rearrangement and aggregation at the interface of flocculated fat droplets, leading to a largely different microstructure of IF emulsion, with large aggregates of lipids and proteins. During digestion, IF-A more digested (degree of proteolysis + 16 %) at 180 min of intestinal phase than IF-C/D. The modification of the supramolecular organisation of caseins implied different kinetics of peptide release derived from caseins during the gastric phase (more abundant at G80 for IF-D). Bioactive peptide release kinetics were also different during digestion with IF-C presenting a maximal abundance for a large proportion of them. Overall, the present study highlights the importance of the structure and composition of the protein ingredients (WPs and caseins) selected for IF formulation on the final IF structure and, in turn, on proteolysis. Whether it has some physiological consequences remains to be investigated.

The objective was to understand how protein structure and quality within IFs impact plasma amino acid (AA) kinetics, gut physiology and microbiota.Four IFs containing whey proteins (WPs) with different origins and denaturation rates (IFs-A/-B/-C) or caseins with different organizations (IFs-C/-D) were produced as isonitrogenous despite slight differences of protein profile and AA concentrations. Both structure and in vitro digestion kinetics differed among IFs. Ten Yucatan mini-piglets (12- to 27-day-old) used as an infant model, received each IF during three days according to a Williams Latin square, followed by a two days wash-out period. Jugular plasma was regularly sampled from 10 min pre-prandial to 4 hours post-prandial on the third experimental day to measure free AA, urea, insulin and glucose concentrations. Data were analysed using a linear mixed model for repeated measures.IFs-A and -B, with higher WP denaturation rates, had significantly higher plasma total and essential AAs concentrations compared to IFs-C and -D, regardless of the time. Plasma threonine concentration was significantly different regardless of the time (A>B>C=D), probably resulting from glycomacropeptides presence in IFs-A and -B, known to cause increased threoninemia in infants. WP denaturation (IF-A) and casein supramolecular organization (IF-D) seemed to reduce AA catabolism. All these differences were present before meal ingestion indicating that a metabolic adjustment occurred during the dietary adaptation. Overall, this highlights the importance of the protein ingredient quality (WPs and caseins) within IFs on the plasma AA delivery, which can further impact protein metabolism, including the anabolic pathway of high importance at the infant stage. Whether it has consequences on the gut maturation and on the microbiota, an important actor within the microbiota-gut-brain axis is currently being investigated.

International audience; Exclusive breastfeeding in the first months of life has protective effects on infant health compared to formula-fed infants including a lower risk of gastrointestinal and respiratory infections and of metabolic and immune disorders. These observable differences in 'health effect' are likely due to differences in composition between breast milk and infant formula. In particular, breast milk contains a wide variety of bacterial species that are present at low dose. This microbial counterpart contributes to the development of the newborn's gut microbiota after digestion of milk matrix. It was also suggested to influence more largely intestinal homeostasis, acting on the gut immunity and intestinal barrier, and thus to contribute to breastmilk health promoting effects [1]. Several studies have investigated the impact of commensal bacteria on gut homeostasis. However, they generally do not include the different phases of digestion, like gastric (G) or intestinal (I) phases, which could have an impact on the physiological state and properties of the bacteria. The aim of this study was to evaluate the impact of newborn digestion on the physiology of breastmilk bacteria and on their mmunomodulatory potential. For this study, six strains representative of the prevalent bacteria in breast milk were selected. The strains were digested in a static in vitro model of digestion, at full-term infant stage, in a milk matrix. Following the G and I phases, bacterial cultivability was measured and the immunomodulation properties were determined through the quantification of IL-10 and TNF-α released by macrophages (THP-1 line: human monocytic cell line differentiated into macrophages). The impact of the G and I digestion phases on both viability and immunomodulation properties was strain-dependent, pointing out the interest to consider these steps for the determination of ingested bacteria properties.This study is part of the PROLIFIC project and was financially supported by the Régions Bretagne and Pays de Loire and the Milkvalley-BBA consortium.

International audience; Introduction et but de l’étudeLes préparations pour nourrissons (PPN), seuls substituts adéquats au lait maternel (LM), sont des matrices complexes qui nécessitent de nombreux ingrédients et étapes de transformation. L'objectif de ces essais était de comprendre comment la structure et la composition protéiques des préparations pour nourrissons modulent leur cinétique de digestion et d’absorption en utilisant des modèles de digestion in vitro et in vivo.Matériel et MéthodesQuatre PPNs contenant des protéines de lactosérum avec différents taux de dénaturation (A/B/C) ou des caséines de structures micellaires différentes (C/D) ont été fabriquées. Leur profil protéique variait légèrement mais leur composition nutritionnelle et leur taux d'azote aminé étaient similaires. Elles ont été digérées (1) à l'aide d'un modèle de digestion in vitro dynamique (DIDGI) pour suivre la structure des digestas (microscopie), la protéolyse globale (OPA) et la cinétique d'apparition des peptides (LC-MS/MS) au cours de la digestion et (2) par des mini-porcelets Yucatan (n=10, 12-27 jours de vie) afin de suivre la cinétique postprandiale (4h) d'apparition plasmatique des acides aminés (AA). Les données ont été analysées par un modèle linéaire mixte pour données répétées.Résultats et Analyses statistiquesAvant digestion, les structures lipoprotéiques étaient différentes entre les PPNs. La PPN A présentait des structures lipoprotéiques étoilées avec la présence d'agrégats de protéines du lactosérum à la surface des caséines pour un taux de dénaturation global des protéines du lactosérum de 44%. Les PPNs B et C ne présentaient pas de structures particulières et avaient des taux de dénaturation respectifs de 36 et 31%. La PPN D contenant des caséines non micellaires présentait de grandes structures avec de nombreuses structures caséiques à l'interface de gouttelettes lipidiques partiellement floculées. Son taux de dénaturation des PS était de 29%. La PPN A, caractérisée par des taux de protéines du lactosérum glyquées et dénaturées plus élevés, avait un degré de protéolyse significativement plus élevé à 180 min de phase gastrique que la PPN D et en phase intestinale que les PPNs C et D. Les peptides (en particulier ceux issus des caséines) apparaissaient plus tôt en phase gastrique pour la PPN D (à 80 mincontre 180 min pour les PPNs A et B). La PPN C présentait une abondance maximale pour près de la moitié des peptides bioactifs. Chez le porcelet, les PPNs A et B présentaient des concentrations plasmatiques en AA totaux et essentiels significativement plus élevées que les PPNs C et D. La dénaturation des protéines du lactosérum (PPN A) ou la modification structurelle des caséines (PPN D) semblerait induire une diminution du catabolisme oxydatif des acides aminés.ConclusionLes approches complémentaires in vitro et in vivo ont permis de mettre en évidence l'influence de la structure des PPNs sur la digestion des protéines et l'absorption des acides aminés. La structure micellaire de l'ingrédient caséique avait un impact majeur sur la structure protéique des PPNs, leur cinétique de protéolyse avec une libération plus précoce et plus abondante de peptides pour les structures caséiques modifiées ainsi que sur l'oxydation des acides aminés plasmatiques. La structure de la fraction protéique du lactosérum a induit des différences de cinétiques de protéolyse in vitro et in vivo, avec une augmentation de la protéolyse pour les protéines du lactosérum les plus dénaturées induisant des concentrations en acides aminés plasmatiques postprandiales plus importantes. Au vue de la littérature, la digestion de la PPN C (présentant une faible dénaturation,des caséines micellaires et induisant des cinétiques postprandiales plus faibles pour de nombreux acides aminés comparés aux PPNs A et B) se rapprocherait davantage de celle du lait maternel. L'impact physiologique de ces PPNs sera évalué au cours d'une prochaine étude sur le porcelet Yucatan

International audience; Introduction et but de l’étudeLa formulation des préparations pour nourrissons (PPN) est très réglementée et doit mimer au mieux l’aminogramme du lait humain (LH), premier élément essentiel de la qualité nutritionnelle des protéines. Peu d’études in vivo se sont intéressées à la digestibilité des acides aminés (AA) du LH et des PPNs, permettant d’estimer leur biodisponibilité, autre indicateur important de la qualité nutritionnelle des protéines. L’objectif de cette étude était de mesurer la digestibilité iléale vraie (DIV) des protéines du LH et d’une PPN, afin de calculer leur DIAAS (Digestible Indispensable Amino Acid Score ; FAO, 2013). Un modèle mini-porcelet a été utilisé comme modèle du nourrisson.Matériel et MéthodesDes porcelets âgés de 19 jours ont reçu pendant 6 jours du LH (n=7) ou une PPN (n=9). Un troisième groupe de porcelets a reçu un régime sans protéines (n=6), permettant d’évaluer les pertes en protéines endogènes. Les trois régimes contenaient du Cobalt-EDTA comme marqueur de transit. Le dernier jour, les repas ont été distribués toutes les heures pendant six heures avant sacrifice des animaux et récupération des contenus iléaux pour mesurer la digestibilité vraie de l’azote total et des AA. Le DIAAS a été calculé à partir des digestibilités et des teneurs en AA des aliments au regard du profil de référence, correspondant au profil en AA du LH (FAO, 2007). Des hydrolyses de durées multiples (0 à 120 h) des échantillons ont été réalisées afin d’évaluer les pertes en AA au cours de cette étape et corriger les valeurs classiquement obtenues à 24 h d’hydrolyse acide ou 16 h d’hydrolyse basique pour le tryptophane. Une ANOVA multifactorielle a permis l’analyse et la comparaison des données.Résultats et Analyses statistiquesLes hydrolyses multiples ont permis de corriger les teneurs en AA des aliments et des digestas, en particulier pour la tyrosine et la valine (+10% de la valeur mesurée à 24 h d’hydrolyse). Les profils en AA du LH étaient en accord avec la méta-analyse de Zhang et al. (2003). La DIV de l’azote aminé n’était pas significativement différente (LM : 96,7 ± 1,0 % et PPN : 98,0 ± 0,7 for IF, p > 0,05) tandis que la DIV de l’azote total était significativement plus faible pour le LH (91,3 ± 1,2 %) que pour la PPN (98,0 ± 0,9 %). Cette différence s’expliquait par la plus forte proportion d’azote non protéique dans le LH (LH : 31,0 ± 0,7 % et PPN :4,4 ± 0,2 % de l’azote total), partiellement non digestible et non-absorbable. Quelques AA présentaient des différences de digestibilité. Ainsi, la lysine était significativement plus digestible dans le LH que dans la PPN (98,4 ± 0,7 % vs. 96,0 ± 0,5 %), probablement lié à l’absence de glycation des protéines (réaction de Maillard) dans le LH. A l’inverse, les DIVs de la phénylalanine, la thréonine, la valine, l’alanine, la proline et la sérine, étaient significativement légèrement plus faibles dans le LH que dans la PPN (-2,6 à -7,3 points). La DIV moyenne dutryptophane était proche entre le LH et la PPN (96,8 ± 0,5 %, p = 0,079). Le DIAAS était plus élevé pour le LH que pour la PPN (+10%) et était, pour les deux aliments, liés à une teneur en AA aromatiques (phénylalanine + tyrosine) plus basse que celle du profil de référence.ConclusionCette étude a démontré que le LH, comparé à la PPN, avait une DIV plus faible uniquement pour l’azote total et non pas pour l’azote aminé. Ceci suggère qu’une fraction d’azote non protéique du LH est sous forme non digestible et non absorbable, tel que l’urée, et est donc transférée vers le colon. Cette fraction azotée, bien que peu considérée dans la formulation des PPNs, peut cependant avoir un rôle sur la composition du microbiote. Par ailleurs, la digestibilité plus élevée de la lysine dans le LH que la PPN, est certainement dû à l’absence de réaction de Maillard dans le LH contrairement aux PPNs, qui reçoivent de nombreux traitements thermiques

Le réseau GALACTINNOV vient structurer une collaboration fructueuse entre un ensemble de dix unités de recherche sous tutelle des établissements français partenaires, et de quatre universités québécoises, regroupées au sein du regroupement « Op+lait ».; International audience; Les préparations pour nourrissons (PPNs) sont des matrices complexes, issues de nombreuses étapes de transformation, qui peuvent potentiellement altérer la structure et la composition des protéines qu’elles contiennent. L’objectif était de comprendre comment la structure et la composition des ingrédients protéiques laitiers impactaient la structure des PPNs et leurs cinétiques de digestion. Quatre PPNs ont été formulées à partir d’ingrédients commerciaux dont la structure protéique variait selon leur origine et leurs procédés d’obtention, avec des protéines du lactosérum (PS) de degrés de dénaturation et glycation différents (PPNs-A/B/C) et des caséines ayant une organisation supramoléculaire différente (PPNs-C/D). La structure des PPNs a été étudiée à différentes échelles avant et pendant la digestion ainsi que leur protéolyse en conditions in vitro dynamiques.Les structures lipoprotéiques des PPNs avant digestion étaient différentes, avec la PPN-A qui présentait des structures lipoprotéiques étoilées et la PPN-D de larges structures composées de caséines agrégées et de matière grasse floculée. A l’issye de chacune des phases de digestion (gastrique et intestinale), le degré d’hydrolyse de la PPN-A était plus élevé que celui des PPNs-C/D. La cinétique de relargage des peptides issus de caséines était également différente, en particulier pour la PPN-D, dont l’organisation supramoléculaires des caséines étaient modifiée avant digestion, par comparaison aux autres PPNs.Ces résultats soulignent que la qualité initiale des ingrédients protéiques laitiers, dépendante de leur origine et de leur transformation, influe sur la structure finale des PPNs et sur leur protéolyse. Des études in vivo complémentaires ont été réalisées et permettront d’évaluer l’impact physiologique de ces différences.

Early nutrition plays a dominant role in infant development and health. It is now understood that the infant diet impacts the gut microbiota and its relationship with gut function and brain development. However, its impact on the microbiota-gut-brain axis has not been studied in an integrative way. The objective here was to evaluate the effects of human milk (HM) or cow’s milk based infant formula (IF) on the relationships between gut microbiota and the collective host intestinal-brain axis. Eighteen 10-day-old Yucatan mini-piglets were fed with HM or IF. Intestinal and fecal microbiota composition, intestinal phenotypic parameters, and the expression of genes involved in several gut and brain functions were determined. Unidimensional analyses were performed, followed by multifactorial analyses to evaluate the relationships among all the variables across the microbiota-gut-brain axis. Compared to IF, HM decreased the α-diversity of colonic and fecal microbiota and modified their composition. Piglets fed HM had a significantly higher ileal and colonic paracellular permeability assessed by ex vivo analysis, a lower expression of genes encoding tight junction proteins, and a higher expression of genes encoding pro-inflammatory and anti-inflammatory immune activity. In addition, the expression of genes involved in endocrine function, tryptophan metabolism and nutrient transport was modified mostly in the colon. These diet-induced intestinal modifications were associated with changes in the brain tissue expression of genes encoding the blood-brain barrier, endocrine function and short chain fatty acid receptors, mostly in hypothalamic and striatal areas. The integrative approach underlined specific groups of bacteria (Veillonellaceae, Enterobacteriaceae, Lachnospiraceae, Rikenellaceae, and Prevotellaceae) associated with changes in the gut-brain axis. There is a clear influence of the infant diet, even over a short dietary intervention period, on establishment of the microbiota-gut-brain axis.

L’objectif était de comprendre comment la structure des ingrédients protéiques impactait la structure des PPNs et leur cinétique de digestion. Quatre PPNs, formulées à partir de protéines sériques (PS) présentant des degrés de dénaturation et de glycation différents (A/B/C) et de caséines avec une organisation supramoléculaire différente (C/D), ont été digérées à l’aide d’un modèle in vitro dynamique (DIDGI®) au stade infantile. Les digestas ont été échantillonnés à intervalle régulier afin de suivre les modifications structurales, la protéolyse (OPA, SDS-PAGE, LC-MS/MS) et la lipolyse (GC-MS). Les données ont été analysées par ANOVA pour mesures répétées. Avant digestion, les structures lipoprotéiques des PPNs étaient différentes. La PPN-A, caractérisée par des taux élevés de glycation et de dénaturation de ses PS, tendait à être davantage digérée à 180min de la phase intestinale que les PPNs-C/D (augmentation du degré de protéolyse et lipolyse : +16% et + 5%). Les peptides apparaissaient plus rapidement en phase gastrique pour la PPN-D (dès 80min vs. 180min pour les PPN-A/B), suggérant que les grosses structures lipoprotéiques initialement présentes dans la matrice étaient moins denses et donc plus accessibles par la pepsine. Ces résultats soulignent l’importance de la structure des ingrédients protéiques sur la structure des PPNs et leur digestion. Une étude in vivo complémentaire est en cours.

International audience; Human milk (HM) and Infant formula (IF) are assumed to present different digestion kinetics, while rarely directly compared. The study aimed to address this question using a dynamic digestion model (DIDGI®) at the infant stage.Fresh HM (pool from 50 mothers) and standard IF with similar total nitrogen content were digested in triplicate. Digesta were sampled regularly in gastric and intestinal phase from 0 to 180 min to evaluate structural changes (confocal microscopy and laser light scattering), proteolysis (SDS-PAGE and densitometry, OPA), and nitrogen digestibility (µ-Kjeldahl). Differences between groups were assessed using a two-way repeated measures ANOVA.The microstructure of the digesta differed between HM and IF along the gastric digestion. Proteolysis of the common HM and IF proteins, caseins and α-lactalbumin, was significantly slower for HM than for IF. The intestinal degree of proteolysis was lower for HM than IF, at least during the first two hours. Total N digestibility was lower with HM, such as observed in vivo. Peptidomic and lipolysis data will complete the dataset.Despite nutritional similarity, this study highlights the influence of the matrix on the digestion kinetics and gives some further understanding to the global value of digestibility, such as determined in vivo.

Many new-borns are fed with substitutes of breast milk. These products undergo several heat treatments, necessary to insure their microbiologic safety but probably modifying their organo-leptic properties. High temperature can damage milk proteins and form Maillard reaction prod-ucts with lactose. Microfiltration – to replace the heat treatments for a milk debacterization – combined with a soft spray-drying is currently an alternative little exploited to produce infant formulas (IFs) although microbiologically safe. The aim of this study was to determine the im-pact of such processing route on the organoleptic properties of the IFs. Three experimental IFs were manufactured at a semi-industrial scale, from fresh bovine skimmed milk: one with no heat treatments (T-), one with one moderate heat treatment (T+) and another one with successive and higher heat treatments (T+++) such as in industries. Sensory evaluations were performed by a panel of 50 untrained adults to characterise overall sensory differences (triangular tests) and the organoleptic properties (check-all-that-apply (CATA) de-scription) of the 3 experimental IFs, in comparison with 2 commercial ones (Blédilait, Guigoz). Volatile organic compounds (VOCs) contained in T- and T+++ were extracted with the solvent assisted flavour evaporation technique. The water phase was extracted with dichloromethane and concentrated in Kuderna-Danish apparatus. Samples were analysed by gas chromatography – electronic ionisation – mass spectrometry (GC-EI-MS), equipped with a polar capillary DB-WAX column to perform VOCs identification and relative quantification. Significant sensory differences were found in the triangular tests, between the experi-mental/commercial IFs, T-/T+++ and the both commercial brands. The CATA method supplied a global description of the 5 IFs, with 20 descriptors ticked by the panellists – related to colour, texture, taste, odour. A principal component analysis (PCA) was performed on quotations fre-quencies for each descriptor: there was a clear difference between off-flavour and pleasant de-scriptors. Another PCA shows an opposition between Guigoz – close to off-flavour descriptors – and the 4 other IFs – near to pleasant ones (“sweet”, “dairy”, “fat”, “butter”). These 4 IFs could be discriminated: T+++ was close to the same descriptors as Blédilait, as T- was with T+. Around 40 VOCs belonging to various chemical classes were identified. They are similar to T- and T+++, but in different proportions. These VOCs have olfactory connotations bound to a “fat in-mouth” sensation, “grass” and “fish” odours. A simple and saturated aldehydes group (pentanal, hexanal) comes probably from lipid oxidation, with a formation of hydroperoxides altering food flavour. Decomposition of these peroxides drives to a complex mixture of second-ary products (aldehydes, ketones…). VOCs quantities found in the IFs are in the range of the commercial ones. The IFs produced by different processing routes were different from a sensory point of view, with the less treated IF characterised by pleasant descriptors. The VOCs characterisation con-firms some flavour differences between the both experimental IFs and reveals presence of alde-hydes coming from the lipid peroxidation, which is commonplace for such IFs. Overall, these experimental IFs with a minimal processing route fit well withn IFs market, from a sensory point of view.

International audience; Introduction et but de l’étude: Les nombreux Introduction et but de l’étude: Les nombreux traitements thermiques appliqués aux préparations pour nourrissons (PPN) sont susceptibles de dénaturer et agréger les protéines, modifiant potentiellement leur digestibilité et la biodisponibilité des acides aminés. Récemment, un procédé technologique innovant, sans traitement thermique, basé sur la microfiltration membranaire a permis la production de PPN native, i.e. avec un très faible taux de protéines dénaturées (6%, Yu et al. 2021 Journal of Dairy Science). L’objectif de cette étude est de comparer la digestibilité des protéines et des acides aminés d’une même PPN, traitée thermiquement de manière classique (T+++ ; 58% de protéines dénaturées) ou native (T-) chez le raton. Matériel et méthodes: Des ratons Wistar mâles âgés de 21 jours en début d’expérimentation ont été nourris durant 14 jours avec soit un régime de croissance témoin (n = 8 ; 20% P/E, 54%G/E, 26%L/E), soit un régime contenant 40% (m.s.) de PPN T- (n = 30) ou T+++ (n = 30 ; 20% P/E, 54%G/E, 26%L/E). Les ratons témoins ont été euthanasiés 6h après l’ingestion d’un repas calibré protéiprive afin d’évaluer les pertes digestives d’azote et d’acides aminés endogènes. Les ratons T- et T+++ ont été euthanasiés 0, 1, 2, 3 ou 6h après l’ingestion d’un repas calibré contenant leur régime respectif (n = 6/temps). Les contenus digestifs et le plasma ont été collectés. Résultats et Analyse statistique: Aucune différence de poids et de prise alimentaire n’a été notée entre les groupes. La cinétique de vidange gastrique des ratons nourris avec le régime T- était comparable à celle des ratons nourris avec le régime T+++. La digestibilité orocécale vraie de l’azote tendait à être légèrement supérieure pour T- (96,6 ± 3,1%) par rapport à T+++ (91,9 ± 5,4%, P = 0,09). La digestibilité orocécale vraie des acides aminés variait de 90,0 ± 5,7% (serine) à 98,6 ± 1,7% (arginine) pour T- et de 85,5 ± 7,8% (glycine) à 95,9 ± 4,5% (histidine) pour T+++. Aucune différence de digestibilité moyenne des acides aminés n’a été mise en évidence entre T- (95,0 ± 3,5%) et T+++ (91,6 ± 5,1%). Les cinétiques postprandiales plasmatiques d’acides aminés étaient significativement différentes entre les 2 régimes (P = 0,0002), avec une augmentation plus importante et plus rapide des concentrations d’acides aminés totaux pour les rats T- par rapport aux rats T+++. Conclusion: Les PPN native (T-) et traitée thermiquement (T+++) permettent de supporter de manière comparable la croissance des ratons. Les 2 PPN présentent une bonne digestibilité (> 90%) mais la dénaturation/agrégation des protéines induites par les traitements thermiques (PPN T+++) semble diminuer leur digestibilité et limiter l’augmentation postprandiale des acides aminés plasmatiques. Nos résultats suggèrent ainsi que la production de PPN native, avec un minimum de procédés de transformation, permettrait d’optimiser la qualité nutritionnelle des protéines.

Un impact physiologique différent entre lait humain (LH) et préparations pour nourrisson (PPN) est démontré sans que les mécanismes d’actions, en particulier sur le rôle de l’axe microbiote-intestin-cerveau, ne soient réellement connus. L’objectif de cette étude a été de préciser son rôle chez le mini-porcelet Yucatan utilisé comme modèle du nourrisson. Matériel et méthodes : Deux groupes de 9 porcelets âgés de 19 jours ont été nourris avec un pool de LH (provenant de 50 mères) ou une PPN pendant 6 jours puis ont été sacrifiés. Le microbiote (séquençage ARNr 16S) et la digestibilité iléale standardisée (DIS) du tryptophane (Trp) ont été analysés. La perméabilité iléale et colique a été mesurée en chambre d’Ussing. L’expression de gènes impliqués dans les fonctions barrière et immunitaire, le métabolisme, ou codant les récepteurs de nutriments, de neurotransmetteurs et d’hormones a été analysée par PCR (SmartChip) dans le cerveau (69 gènes) et l’intestin (106 gènes). Résultats et analyse statistique : Une baisse de l’α-diversité associée à une plus faible abondance des phyla Bacteroidetes, Fusobacteria et Deferribacteres ont été observées dans le colon des porcelets LH. Le nombre de gènes différemment exprimés entre LH et PPN était variable selon les tissus : 35% dans le colon, 10% dans l’iléon, 16% dans hypothalamus, 14% dans le cortex préfrontal, 10% dans l’hippocampe et seulement 3% dans le striatum. Dans les colons du groupe LH, les gènes impliqués dans la réponse immunitaire pro- et anti-inflammatoire (BAFF, IL10, IL10Rα, TNFαR1 et TLR2) étaient significativement plus exprimés (+28% à +70%, P0,05). Dans le cerveau, les gènes impliqués dans les fonctions barrière hémato-encéphalique (Cdh2 et Marveld2) et endocrine (NPY et GLP1r) et de la neuro-/synaptogenèse (BDNF, SHH, CYPFIP2) étaient moins exprimés chez les porcelets LH (-52% à -211%, P

Fortification of human milk (HM) is often necessary to meet the nutritional requirements of preterm infants. This study sought to establish whether HM supplemented with an experimental donkey milk-derived fortifier (DMF) or a commercial bovine milk-derived fortifier (BMF) affected digestion, using an in vitro dynamic system at the preterm stage. Particle size in gastric phase was higher in DMF than in BMF, due to protein aggregates surrounding lipid globules. Before digestion, BMF, with its extensively hydrolysed proteins, had a higher degree of proteolysis (30%) than DMF (11%), which contained intact proteins. After digestion, this difference was reduced concomitantly to a similar net degree of proteolysis (33%). DMF, with a higher proportion of ω3, resulted in a lower ω6/ω3 free PUFA ratio than BMF throughout digestion, although the final degree of lipolysis was similar (54%). In summary, DMF could represent a better source of proteins and lipids for the preterm infant.

International audience; The impact of protein structure in model infant milk formulas (IMFs) on digestion was evaluated using an in vitro dynamic digestion model. IMFs differed by their whey protein profile and the heating conditions. Digesta microstructure was followed by laser light scattering and confocal laser scanning microscopy. Proteolysis kinetics were monitored by SDS-PAGE and OPA assay and peptide release kinetics by tandem mass spectrometry. The gastric digesta microstructure depended on the protein structure within IMFs, with the highest protein particle size in IMF containing native casein micelles. Among whey proteins, denatured/aggregated lactoferrin was the most sensitive to pepsin hydrolysis. The peptidomic profile during digestion varied among IMFs. More numerous and resistant intestinal bioactive peptides were observed for the unheated IMF having a whey protein profile closer to that of human milk. Further physiological studies are required to investigate the benefits of adapting the IMF whey protein profile on infant health.

International audience; High Temperature-Short Time (HTST) pasteurization was proposed as an alternative to Holder pasteurization (HOP) to increase the retention of specific human milk (HM) bioactive proteins. The present study explored whether HTST and HOP differently affect peptide release during simulated preterm infant gastrointestinal digestion. Raw (RHM), HOP-and HTST-pasteurized HM were digested using an in vitro dynamic system, and the identified peptides were analyzed by mass spectrometry and multivariate statistics. Before digestion, 158 peptides were identified in either RHM, HTST-or HOP-HM, mostly (84.4%) originating from β-casein (CASB). During gastric digestion, HOP-HM presented a greater number and more abundant specific CASB peptides. A delayed release of peptides was observed in RHM during the intestinal phase, with respect to both pasteurized HM. Although limited to gastric digestion, the HM peptidomic profile differed according to the pasteurization type, and the pattern of the HTST peptides showed a greater similarity with RHM.
