The human pathogen Bacillus cereus is responsible of many food poisoning due to the ingestion of contaminated vegetables or processed foods. For the survival and colonization, the bacteria form a biofilm on biotic and abiotic surfaces in which a specific subpopulation differentiates to produce a multifunctional extracellular matrix (ECM), mainly composed by exopolysaccharides, proteins and extracellular DNA. One of the most attractive components of the ECM are the amyloid proteins due to its high tendency to fibrillate and the multifunctional role in the bacterial physiology. In the B. cereus genome there is a specific genomic region that contains two orthologues genes to the tasA gene in Bacillus subtilis, initially described as essential in the assembly of amyloid fibers, named tasA and calY respectively. The deletion of these genes gives a different phenotype related with a deficient biofilm formation in both cases. In the tasA mutant, the biofilm finally detaches at 72 hours, and the calY mutant shows a thinner ring phenotype in comparison with the wild type strain. The gene expression dynamic during the biofilm development reveals that there are three different subpopulations: both genes are expressed, only calY or none of them. Furthermore, transcriptomic analysis by RNA-Seq show that the mutants have different changes in the gene regulation and the calY mutant is the most affected, with 227 genes deregulated that affect to several important pathways such as the synthesis of extracellular DNA, an important component of the ECM. The results mentioned above indicate that TasA and CalY might have complementary roles in the biopellicle development but also evidence that this physiological process is more complex in B. cereus in comparison with the model described in B. subtilis. Comunicación oral a congreso Las comunidades microbianas presentes en la rizosfera de árboles de aguacate sanos presentan una elevada presencia de bacterias del filo Proteobacteria. Destacan aquellas especies pertenecientes al complejo Pseudomonas fluorescens que presentan actividades relacionadas con la promoción del crecimiento de plantas (Plant Growth Promotion: PGP), así como con el control biológico de diferentes fitopatógenos como hongos, bacterias e insectos. Para estudiar el comportamiento y las interacciones que tienen lugar entre distintas cepas de P. chlororaphis aisladas de la rizosfera de aguacate, se ha construido una comunidad sintética de tres cepas y se han analizado sus fenotipos. Los genomas de estas cepas están secuenciados y se realizó un estudio comparativo de sus genomas. Por un lado, se han realizado estudios de compatibilidad y competitividad in vitro e in vivo entre las distintas cepas, y se determinó el fenotipo dominante en las diferentes combinaciones de cepas para la morfología de colonias y formación de biofilm. Por otro lado, se observó mediante microscopía confocal la formación de biofilm in vitro, así como la distribución de las cepas en solitario y combinadas sobre la raíz de aguacate. Por último, se ha estudiado, el control biológico y la estabilidad del consorcio en la rizosfera de aguacate, determinando su capacidad de colonización y persistencia. Las proteínas amiloides tienen la tendencia natural de formar fibras insolubles a partir de monómeros solubles, un proceso que implica la formación de agregados de distintos tamaños, y con distintas conformaciones. Una de características definitorias de las fibras amiloides es el enriquecimiento en láminas beta que confieren propiedades tintoriales, bioquímicas y biológicas. A pesar de preservar la denominación patológica, los amiloides median funciones que son esenciales para la homeostasis celular o la adaptación al entorno. En Bacillus subtilis, la proteína amiloide TasA desempeña varias de estas funciones: i) estructural, contribuyendo al ensamblaje de la matriz extracelular, ii) celular, preservando la fluidez y funcionalidad de la membrana, iii) señalización, manteniendo ciertas subpoblaciones celulares. Por lo tanto, resulta esencial conocer los residuos o dominios de la proteína que hacen posible a TasA transitar entre las distintas conformaciones, como paso previo a entender esta diversidad de funciones. En trabajos previos (1), estudiamos las propiedades estructurales de TasA en su forma fibrilar, y mediante la digestión con proteínasa K demostramos la existencia de una región resistente en la mitad amino terminal y que denominamos el núcleo amiloide. En este trabajo se ha estudiado esta región mediante el uso de diversas aproximaciones, genética, bioinformáticas, bioquímicas o biofísicas. Como resultado hemos detectado la existencia de residuos aminoácidos y regiones conservadas que parecen ser esenciales al menos en el proceso de amiloidogenesis, y por tanto en el papel estructural involucrado en la formación de biofilm. The extracellular matrix and sporulation are defensive mechanisms used by Bacillus cells when they interact with Pseudomonas strains expressing a type VI secretion system (T6SS). Here, we define Tse1 as the main toxin mobilized by the Pseudomonas T6SS that triggers sporulation in Bacillus. We characterize Tse1 as a peptidoglycan hydrolase and electron microscopy analysis and the use of diverse chemical probes, let us visualize malfunction of the Bacillus cell membrane. By performing RNA-seq and immunocytochemistry analyses, we also delineate the response of Bacillus cells to Tse1, which through the coordinated actions of the extracellular sigma factor σW and the cytoplasmic histidine kinases KinA and KinB, culminates in activation of the sporulation cascade. We propose that this cellular developmental response is conserved in Bacilli to defend against the toxicity of T6SS-mobilized Tse1 effector. Universidad de Málaga. Bacterial communities are continuously adapting and evolving for survival. They produce and secrete a broad range of molecules that kill, defend, or mediate communication between cells of different lineages, thus shaping the final structure of the microbial community. In this work, with the combination of -omics approaches, molecular biology and microscopic techniques, we expand our knowledge on the chemical interplay and specific mutations that modulate the transition from antagonism to co-existence between two plant-beneficial bacteria, Pseudomonas chlororaphis and Bacillus amyloliquefaciens. We demonstrate that bacillaene, a secondary metabolite with bacteriostatic activity produced by Bacillus, interacts with the protein elongation factor FusA of P. chlororaphis to arrest its growth and population advancement. Point mutations in this protein lead to tolerance to bacillaene and other inhibitors of protein translation. Additionally, we describe the key role of the glycerol kinase GlpK from B. amyloliquefaciens in its unspecific tolerance against P. chlororaphis. Mutations in GlpK provoked by a decrease of Bacillus cell membrane permeability among other pleiotropic responses. We conclude that nutrient specialization and mutations in basic biological functions are bacterial adaptive dynamics that lead to the coexistence of two primary competitive bacterial species rather than their mutual eradication. Comunicación a congreso en formato póster La cepa modelo Pseudomonas chlororaphis PCL1606 (PcPCL1606) es una bacteria asociada a la rizosfera de las plantas de aguacate. Esta rizobacteria muestra una marcada capacidad de antagonismo y control biológico frente a diferentes hongos fitopatógenos de suelo, incluido Rosellinia necatrix, agente causal de la podredumbre blanca radicular en cultivos de aguacate en el área Mediterránea. La actividad de control biológico viene mediada, principalmente, por la producción de la molécula antifúngica 2-hexil, 5-propil resorcinol (HPR). Además, HPR también interviene en procesos como la colonización de las raíces, la formación de biopelículas y las interacciones multitróficas en la rizosfera, lo que indica una actividad reguladora adicional. Datos previos obtenidos en un estudio transcriptómico para identificar los genes regulados por HPR, revelaron la inducción de un clúster de genes durante la interacción con la rizosfera de aguacate. Este clúster presenta homología con genes que codifican un posible pili tipo IV Flp/ Tad (por sus siglas en inglés Tight Adherence), descrito previamente por su implicación en la adhesión temprana. Para estudiar el papel de esta región génica en la biología de PcPCL1606, se construyó un mutante dirigido por deleción en los genes que codifican para la principal proteína estructural de este pili y se validó su fenotipo con respecto a la cepa silvestre. Los resultados obtenidos evidencian la importancia de este clúster de genes en la adhesión y la formación de la biopelícula, ambos procesos relacionados con el control biológico y la colonización de PcPCL1606 en las raíces de la planta de aguacate. Comunicación a congreso en formato póster Las biopelículas bacterianas están formadas por agregados celulares embebidos en una matriz extracelular de producción propia, formada principalmente por exopolisacáridos (EPS), proteínas y ADN extracelular. Existen datos recientes que sugieren que entre el 40-80% de las bacterias se encuentran formando biopelículas en la naturaleza, y que la formación de las biopelículas se ve afectada por factores ambientales. Pseudomonas syringae pv. syringae (Pss) es una bacteria fitopatógena causante de la necrosis apical del mango (NAM), enfermedad limitante de la producción de este cultivo en la región mediterránea. En Pss se ha descrito una conexión entre factores ambientales y la incidencia y severidad de la NAM, jugando los EPS celulosa y uno tipo-Psl, un papel relevante en la transición entre un estilo de vida epífito o patogénico. En este trabajo, se ha analizado el papel de algunos factores ambientales relevantes como son la luz y la temperatura en la formación de la biopelícula in vitro en una colección de cepas de Pss aisladas de mango. Los resultados obtenidos sugieren que la temperatura y particularmente la luz pueden influir en la formación de las biopelículas a través de la biosíntesis de los EPS, principalmente celulosa. Además, también se ha observado como niveles más bajos de producción de celulosa podrían estar asociados con una mayor virulencia. Estos resultados indican que los EPS, y principalmente celulosa juegan un papel clave en la ecología de Pss sobre la planta de mango a través de la formación de la biopelícula. Beneficial microbes are known to stimulate the germination of the seeds; however, the exact mechanisms mediating these interactions are only beginning. Bacillus subtilis is a commonly detected member of the plant holobiont and provides multifaceted traits to the plant health. In this work, we demonstrated that B. subtilis triggered genetic and physiological responses in seeds that resulted in changes in the metabolic and developmental status of adult plants. A multidisciplinary approach based on microscopy, transcriptomics and metabolomics demonstrated that the chemically diverse extracellular matrix of Bacillus structurally cooperate in bacterial colonization of the seed storage tissues. The amyloid protein TasA and fengycin, two components of the extracellular matrix, differentially stimulated levels of ROS inside seeds after imbibition and targeted the oil bodies of the seed endosperm, provoking specific changes in lipid metabolism or accumulation of glutathione-related molecules that resulted in two different plant growth programs: the development of seed radicles or major growth and immunization of adult plants. The need to implement sustainable agricultural models has led to the search for alternatives to deal with different plant diseases, with the use of biocontrol agents being one of the most versatile options. A clear example of these agents are the strains belonging to the Bacillus velezensis group, Grampositive bacteria capable of colonizing the different structures of plants and synthesizing a variety of compounds with various activities, ranging from growth promotion to antagonism against to different phytopathogens due to the synthesis of molecules such as lipopeptides (surfactins, fengycins and iturins) among others. Previous studies carried out in our research group have shown how the B. velezensis UMAF6639 strain has a great biocontrol capacity in plants belonging to the Cucurbitaceae family, derived mainly from the production of lipopeptides. The objective of this work focuses on the genetic improvement by random mutagenesis of said strain to obtain mutants with greater antimicrobial activity. Once the derivatives with the greatest antimicrobial activity have been identified, their characterization is being carried out to determine the genetic changes that justify the increase in their biocontrol capacity, as well as the changes in the production profile of secondary metabolites, implementing genomic and metabolomic tools to decipher the different mechanisms responsible for the antagonistic activity of these strains.
