This study aimed to provide scientific data on the anti-Alzheimer’s disease (AD) effects of phenolic compounds from Drynariae Rhizoma (DR) extract using a multi-component approach. Screening of DR extracts, fractions, and the ten phenolic compounds isolated from DR against the key AD-related enzymes acetylcholinesterase (AChE), butyrylcholinesterase (BChE), β-site amyloid precursor protein cleaving enzyme 1 (BACE1), and monoamine oxidase-B (MAO-B) confirmed their significant inhibitory activities. The DR extract was confirmed to have BACE1-inhibitory activity, and the ethyl acetate and butanol fractions were found to inhibit all AD-related enzymes, including BACE1, AChE, BChE, and MAO-B. Among the isolated phenolic compounds, compounds (2) caffeic acid 4-O-β-D-glucopyranoside, (6) kaempferol 3-O-rhamnoside 7-O-glucoside, (7) kaempferol 3-o-b-d-glucopyranoside-7-o-a-L-arabinofuranoside, (8) neoeriocitrin, (9) naringin, and (10) hesperidin significantly suppressed AD-related enzymes. Notably, compounds 2 and 8 reduced soluble Amyloid Precursor Protein β (sAPPβ) and β-secretase expression by over 45% at a concentration of 1.0 μM. In the thioflavin T assay, compounds 6 and 7 decreased Aβ aggregation by approximately 40% and 80%, respectively, and degraded preformed Aβ aggregates. This study provides robust evidence regarding the potential of DR as a natural therapeutic agent for AD, highlighting specific compounds that may contribute to its efficacy.

Alzheimer’s disease (AD) is a progressive neurodegenerative condition characterized by memory and cognitive decline in older individuals. Beta-amyloid (Aβ), a significant component of senile plaques, is recognized as a primary contributor to AD pathology. Hence, substances that can inhibit Aβ production and/or accumulation are crucial for AD prevention and treatment. Agrimonia pilosa LEDEB. (A. pilosa) (Rosaceae), specifically its aerial parts, was identified in our previous screening study as a promising candidate with inhibitory effects on Aβ production. Therefore, in this study, A. pilosa extract was investigated for its anti-amyloidogenic effects, and its bioactive principles were isolated and identified. The ethanol extract of A. pilosa reduced the levels of sAPPβ and β-secretase by approximately 3% and 40%, respectively, compared to the DMSO-treated control group in APP-CHO cells (a cell line expressing amyloid precursor protein), which were similar to those in the positive control group. In addition, the ethanol extract of A. pilosa also hindered Aβ’s aggregation into fibrils and facilitated the disaggregation of Aβ aggregates, as confirmed by a Thioflavin T (Th T) assay. Subsequently, the active constituents were isolated using a bioassay-guided isolation method involving diverse column chromatography. Eleven compounds were identified—epi-catechin (1), catechin (2), (2S, 3S)-dihydrokaempferol 3-O-β-D-glucopyranoside (3), (-)-epiafzelechin 5-O-β-D-glucopyranoside (4), kaempferol 3-O-β-D-glucopyranoside (5), apigenin 7-O-β-D-glucopyranoside (6), dihydrokaempferol 7-O-β-D-glucopyranoside (7), quercetin 3-O-β-D-glucopyranoside (8), (2S, 3S)-taxifolin 3-O-β-D-glucopyranoside (9), luteolin 7-O-β-D-glucopyranoside (10), and apigenin 7-O-β-D-methylglucuronate (11)—identified through 1D and 2D NMR analysis and comparison with data from the literature. These compounds significantly decreased Aβ production by reducing β- and γ-secretase levels. Moreover, none of the compounds affected the expression levels of sAPPα or α-secretase. Further, compounds 1, 2, 4, 8, and 10 demonstrated a dose-dependent reduction in Aβ aggregation and promoted the disaggregation of pre-formed Aβ aggregates. Notably, compound 8 inhibited the aggregation of Aβ into fibrils by about 43% and facilitated the disassembly of Aβ aggregates by 41% compared to the control group containing only Aβ. These findings underscore the potential of A. pilosa extract and its constituents to mitigate a crucial pathological aspect of AD. Therefore, A. pilosa extract and its active constituents hold promise for development as therapeutics and preventatives of AD.

The seasonal dynamics of phytoplankton communities in Korean coastal waters (KCWs) are influenced by complex interactions between ocean currents and nearshore human activities. Despite these influences, the understanding of seasonal phytoplankton changes and their environmental relationships in KCWs remains limited. We investigate the influence of the distinct characteristics of the three seas surrounding the KCWs (the Yellow Sea, the South Sea, and the East Sea) on seasonal phytoplankton communities based on field surveys conducted at 23 stations between 2020 and 2021. The East Sea exhibited higher winter temperatures due to the Jeju and Tsushima warm currents, while summer temperatures were lower compared to the other regions, highlighting the role of currents and deeper oceanic waters. The Yellow Sea showed significant freshwater influence with low salinity levels from major rivers, contrasting with the higher salinity in the East Sea. These differences led to a disparity in the productivity of the two regions: the highest value of Chl. a was observed to be 6.05 µg L−1 in the Yellow Sea in summer. Diatoms dominated in nutrient-rich conditions, particularly in the Yellow Sea, where they comprised up to 80–100% of the phytoplankton community in summer, winter, and spring. PCA analysis revealed positive correlations between diatoms and Chl. a, while cryptophytes, which thrive in the absence of diatom proliferation, showed no such correlation, indicating their opportunistic growth in nutrient-limited conditions. This study highlights the significant impact of region-specific hydrographic factors on phytoplankton communities in KCWs, with diatoms dominating in summer and cryptophytes and dinoflagellates showing seasonal and regional variations. Understanding these dynamics is crucial for predicting phytoplankton bloom dynamics and their ecological implications in coastal ecosystems.

Dokdo, a volcanic island located in offshore waters, is significantly influenced by various currents and the island effect resulting from upwelling events. Despite these factors, there is a limited understanding of the seasonal changes in phytoplankton populations and their relationship with the environmental factors in the waters around Dokdo, even during dramatic shifts in phytoplankton dynamics. We focused on seasonal oceanographic features over three years (2018, 2019, and 2020) to understand the phytoplankton community structure and seasonal species succession. Winter, characterized by thorough mixing, results in high nutrient levels, leading to increased phytoplankton biomass. The dominance of the large-sized diatom Chaetoceros spp. contributes to relatively low diversity (H’: 1.14 ± 0.31). In contrast to the typical coastal waters, spring exhibits dominance by the small nano-flagellates and Cryptomonas spp. associated with a lack of surface nutrients due to increased water temperature. Summer, characterized by strong stratification, shows low phytoplankton biomass but high Chl. a concentrations, possibly influenced by picoplankton and the emergence of dinoflagellates, such as Gyrodinium sp. and Katodinium sp., which increases diversity (H’: 2.18 ± 0.28). In autumn, there is typically a phytoplankton bloom, but in 2019, an unusually low biomass occurred. This was likely due to the intrusion of deep, cold water from the bottom and low-salinity Changjiang diluted water (CDW) from the surface, increasing the water’s stability. This, in turn, led to nutrient depletion, contributing to a rise in diversity (H’: 1.14 ± 0.31). These environmentally complex waters around Dokdo result in a distinct pattern of biodiversity indices, with the highest in summer and the lowest in winter, differing from typical temperate waters. In conclusion, this research highlights the substantial influence of distinctive oceanographic features and nutrient dynamics on the phytoplankton biomass and biodiversity in the Ulleung Basin and Dokdo region. Understanding these patterns is vital for the effective management of marine ecosystems and fisheries resources, emphasizing the necessity for continued long-term monitoring in the vicinity of the Dokdo area.

Beta-amyloid (Aβ) proteins, major contributors to Alzheimer’s disease (AD), are overproduced and accumulate as oligomers and fibrils. These protein accumulations lead to significant changes in neuronal structure and function, ultimately resulting in the neuronal cell death observed in AD. Consequently, substances that can inhibit Aβ production and/or accumulation are of great interest for AD prevention and treatment. In the course of an ongoing search for natural products, the roots of Davallia mariesii T. Moore ex Baker were selected as a promising candidate with anti-amyloidogenic effects. The ethanol extract of D. mariesii roots, along with its active constituents, not only markedly reduced Aβ production by decreasing β-secretase expression in APP–CHO cells (Chinese hamster ovary cells which stably express amyloid precursor proteins), but also exhibited the ability to diminish Aβ aggregation while enhancing the disaggregation of Aβ aggregates, as determined through the Thioflavin T (Th T) assay. Furthermore, in an in vivo study, the extract of D. mariesii roots showed potential (a tendency) for mitigating scopolamine-induced memory impairment, as evidenced by results from the Morris water maze test and the passive avoidance test, which correlated with reduced Aβ deposition. Additionally, the levels of acetylcholine were significantly elevated, and acetylcholinesterase levels significantly decreased in the brains of mice (whole brains). The treatment with the extract of D. mariesii roots also led to upregulated brain-derived neurotrophic factor (BDNF) and phospho-cAMP response element-binding protein (p-CREB) in the hippocampal region. These findings suggest that the extract of D. mariesii roots, along with its active constituents, may offer neuroprotective effects against AD. Consequently, there is potential for the development of the extract of D. mariesii roots and its active constituents as effective therapeutic or preventative agents for AD.

Osteoporosis, one of the most common bone diseases, results from an imbalance between bone formation and resorption. Osteoblasts are primarily involved in bone formation, whereas osteoclasts are involved in bone resorption. Therefore, any substances that can increase osteoblastic differentiation would be beneficial for the prevention or treatment of osteoporosis. In this study, peanut sprouts, a germinated product of peanuts, significantly enhanced osteoblastic differentiation of pre-osteoblastic MC3T3-E1 cells, as determined by the alkaline phosphatase (ALP) assay. The ethyl acetate fraction of peanut sprout extract was subjected to diverse column chromatographies using silica gel, ODS, and Sephadex LH-20 as stationary phases, and three nucleobases, namely, adenine, uracil, and thymine, and three phenolic acids, including caffeic acid, coumaric acid, and ferulic acid, were isolated as active constituents. In particular, adenine at 4 μg/mL and caffeic acid at 1 μg/mL increased ALP activity by 40 and 30%, respectively, compared with the osteoblastic differentiation medium-treated group, and these two compounds were set as marker compounds. Furthermore, extraction of peanut sprouts grown for 14–17 days with 60% ethanol was the best condition to obtain a high quantity of extract for peanut sprouts by analyzing the contents of marker compounds through HPLC. Together, these results suggest that peanut sprouts and their isolated compounds significantly enhance osteoblastic differentiation. Therefore, peanut sprouts have the potential to be developed as functional foods against osteoporosis.

Ship biofouling is recognized as a significant pathway for the introduction and spread of invasive organisms. The in-water cleaning of ship hulls generates wastewater that includes antifouling paint residues and biofouling organisms, which inevitably leak into the marine environments, resulting in substantial adverse effects on marine ecosystems. To assess the impact of hull cleaning wastewater (HCW) on microalgae, we conducted microcosm experiments using HCW including attached microalgae. The experiments consisted of a total of 12 combined trials, including the following groups: ambient seawater as a control, the 5% HCW group (HCW), and the 5% HCW + nutrient addition group (HCW+N), conducted at temperatures of 15 and 20 °C, respectively. The Chl. a concentrations in the water column in the control group exhibited maximum values on day 1 (5.24 μg L−1 at 15 °C and 12.37 μg L−1 at 20 °C), but those of the treatments were at low levels, below 2 μg L−1 at both temperatures. On the other hand, the Chl. a concentrations on plastic plates were higher in the treatments than in the control group. Specifically, the Fv/Fm ratio in the water column, which indicates photosynthetic activity, was significantly higher in the control group compared to both the HCW and HCW+N groups at 15 and 20 °C (p < 0.05). This suggests that the growth of water column phytoplankton was inhibited following HCW inoculation. However, there were no significant differences in the Fv/Fm on plastic plates between the control and HCW treatment groups, implying that the periphyton maintained a high photosynthetic capacity even in the presence of HCW treatments. The elution of particulate copper in HCW was observed, which was considered as the main reason for the growth of phytoplankton. Our study results suggest that the runoff of HCW in the marine environment has a greater negative effect on phytoplankton than on periphyton, which can lead to changes in microalgae community composition and a decrease in productivity in the marine environment. Therefore, it is crucial to manage HCW runoff based on scientific assessments to minimize the ecological risks associated with the removal of biofilm or slime from ship biofouling during in-water hull cleaning.

Graesiella emersonii is a commercially exploitable source of bioactive compounds and biofuels with potential applications in microalgae-based industries. Despite this, little taxonomical information is available. Therefore, proper identification and characterization are needed for the sustainable utilization of isolated microalgae. In this study, an axenically isolated unicellular green alga from the Geumgang Estuary, Korea was investigated for its morphological, molecular, and biochemical characteristics. The morphological characteristics were typical of G. emersonii. Molecular phylogenetic analysis of the 18S rDNA sequence verified that the isolate belonged to G. emersonii and was subsequently named G. emersonii GEGS21. It was isolated from brackish water, and its optimal growth temperature, salinity, and light intensity were at 28–32 °C, 0 M NaCl, and 130–160 µmol m−2 s−1, respectively. The strain thrived over a range of temperatures (5–40 °C) and withstood up to 0.5 M NaCl. The isolate was rich in omega-6 linoleic acid (C18:2 n-6, 26.3%) and palmitic acid (C16:0, 27.5%). The fuel quality properties were determined, and biodiesel from GEGS21 could be used as a biodiesel blend. Value-added carotenoids lutein (1.5 mg g−1 dry cell weight, DCW) and neoxanthin (1.2 mg g−1 DCW) were biosynthesized as accessory pigments by this microalga. The biomass of this microalga may serve as feedstock for biodiesel production as well as producing valuable ω-6 and carotenoids.

The amount of CO2 absorbed by the oceans continues to rise, resulting in further acidification, altering some functional traits of phytoplankton. To understand the effect of elevated partial pressures of CO2 (pCO2) on functional traits of dinoflagellates Alexandrium affine and A. pacificum, the cardinal temperatures and chain formation extent were examined under two pCO2 (400 and 1,000 µatm) over the range of temperature expected to be associated with growth. The growth rate and chain formation extent of A. affine increased with higher pCO2, showing significant changes in cardinal temperatures and a substantial increase in middle chain-length (4--8 cells) fractionation under elevated pCO2 condition. By contrast, there were no significant differences in specific growth rate and any chain-length fractionation of A. pacificum between ambient and elevated pCO2 conditions. The observed interspecies variation in the functional traits may reflect differences in ability of species to respond to environmental change with plasticity. Moreover, it allows us to understand the shifting biogeography of marine phytoplankton and predict their phenology in the Korea Strait. [ABSTRACT FROM AUTHOR]
