Contains fulltext : 291657.pdf (Publisher’s version ) (Open Access) BACKGROUND: Anti-TNF drugs, such as infliximab, are associated with attenuated antibody responses after SARS-CoV-2 vaccination. We aimed to determine how the anti-TNF drug infliximab and the anti-integrin drug vedolizumab affect vaccine-induced neutralising antibodies against highly transmissible omicron (B.1.1.529) BA.1, and BA.4 and BA.5 (hereafter BA.4/5) SARS-CoV-2 variants, which possess the ability to evade host immunity and, together with emerging sublineages, are now the dominating variants causing current waves of infection. METHODS: CLARITY IBD is a prospective, multicentre, observational cohort study investigating the effect of infliximab and vedolizumab on SARS-CoV-2 infection and vaccination in patients with inflammatory bowel disease (IBD). Patients aged 5 years and older with a diagnosis of IBD and being treated with infliximab or vedolizumab for 6 weeks or longer were recruited from infusion units at 92 hospitals in the UK. In this analysis, we included participants who had received uninterrupted biological therapy since recruitment and without a previous SARS-CoV-2 infection. The primary outcome was neutralising antibody responses against SARS-CoV-2 wild-type and omicron subvariants BA.1 and BA.4/5 after three doses of SARS-CoV-2 vaccine. We constructed Cox proportional hazards models to investigate the risk of breakthrough infection in relation to neutralising antibody titres. The study is registered with the ISRCTN registry, ISRCTN45176516, and is closed to accrual. FINDINGS: Between Sept 22 and Dec 23, 2020, 7224 patients with IBD were recruited to the CLARITY IBD study, of whom 1288 had no previous SARS-CoV-2 infection after three doses of SARS-CoV-2 vaccine and were established on either infliximab (n=871) or vedolizumab (n=417) and included in this study (median age was 46·1 years [IQR 33·6-58·2], 610 [47·4%] were female, 671 [52·1%] were male, 1209 [93·9%] were White, and 46 [3·6%] were Asian). Abstract

When the eleven- and twelve-year-olds on the Cannon Street YMCA All-Star team registered for a baseball tournament in Charleston, South Carolina, in June 1955, it put the team and the forces of integration on a collision course with segregation, bigotry, and the southern way of life. White teams refused to take the field with the Cannon Street All-Stars, the first Black Little League team in South Carolina. The Cannon Street team won the tournament by forfeit and advanced to the state tournament. When all the white teams withdrew in protest, the Cannon Street team won the state tournament. If the team had won the regional tournament in Rome, Georgia, it would have advanced to the Little League World Series. But Little League officials ruled the team ineligible to play in the tournament because it had advanced by winning on forfeit and not on the field, denying the boys their dream of playing in the Little League World Series. Little League Baseball invited the Cannon Street All-Stars to be the organization's guests at the World Series, where they heard spectators yell, “Let them play! Let them play!” when the ballplayers were introduced. This became a national story for a few weeks but then faded and disappeared as Americans read of other civil rights stories, including the torture and murder of fourteen-year-old Emmett Till.Stolen Dreams is the story of the Cannon Street YMCA All-Stars and of the early civil rights movement. It's also the story of centuries of bigotry in Charleston, South Carolina—where millions of enslaved people were brought to this country and where the Civil War began, where segregation remained for a century after the war ended and anyone who challenged it did so at their own risk.

Abstract

The site-specific administration of neuroactive substances or pharmacological agents is a routine practice in neurological studies. To facilitate chronic treatments intercranial cannulae are often implanted in the skull. These surgical procedures are widely described and well-documented for rodents, as the most common animal model, and even refined over the years. However, precise descriptions of proper procedure for third ventricle cannulation in young broiler chicks is not readily available. This absence is surprising, as chicken models are often used in the investigation of physiological control of the metabolism and ingestion. Furthermore, as a commercial animal, there is also a particular interest in elucidating the central regulation of feed intake and metabolism to optimize feeding and living conditions for broilers. The brain nuclei that serve as the regulatory centers of feeding and metabolism are the arcuate nucleus, the ventromedial hypothalamic nucleus, and the lateral hypothalamus, which are all located near the third ventricle. Here, we provide a full protocol for intracerebroventricular (i.c.v.) cannulation of 7-day old broiler chicks, as well as a review of some of the advances made in rodent i.c.v. cannulations and whether these advances are applicable to cannulations in chickens. Using the surgical procedure described here, we were able to achieve a success rate of proper implantation of 88%, with a mortality rate of less than 1% (n = 224). • Detailed procedure for i.c.v. canula implantation in the third ventricle of 7d-old broiler chicks. • Cement cap with anchoring screws is necessary in broilers, glue does not provide sufficient stability. • Carboxylate cement and self-adhesive resin cement were tested as an alternative for methyl methacrylate cement.
