Summary: Diabetic kidney disease (DKD), the most common cause of kidney failure, is a frequent complication of diabetes and obesity, and yet to date, treatments to halt its progression are lacking. We analyze kidney single-cell transcriptomic profiles from DKD patients and two DKD mouse models at multiple time points along disease progression—high-fat diet (HFD)-fed mice aged to 90–100 weeks and BTBR ob/ob mice (a genetic model)—and report an expanding population of macrophages with high expression of triggering receptor expressed on myeloid cells 2 (TREM2) in HFD-fed mice. TREM2high macrophages are enriched in obese and diabetic patients, in contrast to hypertensive patients or healthy controls in an independent validation cohort. Trem2 knockout mice on an HFD have worsening kidney filter damage and increased tubular epithelial cell injury, all signs of worsening DKD. Together, our studies suggest that strategies to enhance kidney TREM2high macrophages may provide therapeutic benefits for DKD.

The renal actions of parathyroid hormone (PTH) promote 1,25-vitamin D generation; however, the signaling mechanisms that control PTH-dependent vitamin D activation remain unknown. Here, we demonstrated that salt-inducible kinases (SIKs) orchestrated renal 1,25-vitamin D production downstream of PTH signaling. PTH inhibited SIK cellular activity by cAMP-dependent PKA phosphorylation. Whole-tissue and single-cell transcriptomics demonstrated that both PTH and pharmacologic SIK inhibitors regulated a vitamin D gene module in the proximal tubule. SIK inhibitors increased 1,25-vitamin D production and renal Cyp27b1 mRNA expression in mice and in human embryonic stem cell–derived kidney organoids. Global- and kidney-specific Sik2/Sik3 mutant mice showed Cyp27b1 upregulation, elevated serum 1,25-vitamin D, and PTH-independent hypercalcemia. The SIK substrate CRTC2 showed PTH and SIK inhibitor–inducible binding to key Cyp27b1 regulatory enhancers in the kidney, which were also required for SIK inhibitors to increase Cyp27b1 in vivo. Finally, in a podocyte injury model of chronic kidney disease–mineral bone disorder (CKD-MBD), SIK inhibitor treatment stimulated renal Cyp27b1 expression and 1,25-vitamin D production. Together, these results demonstrated a PTH/SIK/CRTC signaling axis in the kidney that controls Cyp27b1 expression and 1,25-vitamin D synthesis. These findings indicate that SIK inhibitors might be helpful for stimulation of 1,25-vitamin D production in CKD-MBD.

TiO2 thin film coatings significantly improve catalyst separation in photocatalytic processes. They can be applied in heterogeneous photocatalysis under sunlight by mixing TiO2 with other oxides, such as CeO2, for the removal of pollutants in water. Here, TiO2-CeO2 thin films deposited on borosilicate slides were analyzed and applied in solar heterogeneous photocatalysis for the oxidation of pesticides. The films were synthesized by the sol-gel method with spin coating. The waste solutions from the synthesis were used to prepare TiO2 and TiO2-CeO2 powders. These were analyzed by XRD and XPS to explain the behavior of the films. The thin films were characterized by UV-Vis spectroscopy with transmittance, UV-Vis spectroscopy with RDS, profilometry, AFM and SEM. The addition of CeO2 to TiO2 caused a decrease in the average crystal size and an increase in the strain index. The addition of a second layer made the TiO2-CeO2 thin films thinner. The CeO2 created surface and electronic defects in the titania films, which enhanced their photocatalytic properties under sunlight in the mineralization of diuron and methyl parathion. The TiO2-CeO2-5.0% single-layer thin film samples were the most active in this study and will undoubtedly be applied in larger-scale reaction systems. [ABSTRACT FROM AUTHOR]

Saudi Arabia is forecast to develop a significant number of infrastructure projects, as recorded in its strategy Saudi Vision 2030. To implement this strategy, the country intends to leverage public–private partnerships. This research has various goals. Firstly, it intends to identify the determinant features (DFs) that may facilitate the population of private–public partnership (PPP) infrastructure projects in the Kingdom of Saudi Arabia (KSA) through the opinions of PPP experts. Second, it aims to analyse these opinions through a statistical approach. The research used the following methodologies: the development of a survey amongst sectorial experts, with a total of 45 valid responses; a statistical characterisation of the responses; and an exploratory factor analysis to determine the minimum number of factors explaining the proposed objectives. The findings of the research revealed that the five most relevant factors were the availability and effectiveness of a proper regulatory and legal framework; adequate risk allocation and sharing among project stakeholders; a straightforward project brief and client outcomes; the comprehensive and business viability of project feasibility studies; and finally, the assurance that proper project value management systems are in place during different project stages. As its principal contribution, this study finds, based on the experts’ opinions, that KSA should consider the mentioned determinant factors to ensure that PPP plays the expected role in the provision of infrastructure.

Management systems that are recognized as key tools to improve business management and the results associated with it have spread at the business level during the last 50 years. Regarding Safety Management Systems (SMSs), despite having specific international standards, there are no complete studies that analyze the degree of effectiveness of SMS, and even less in construction, a sector that concentrates a large part of registered labor accidents worldwide. The present investigation is an analysis of SMS effectiveness from an empirical study carried out over 48 months in five countries with a total of more than 23 million work hours between 2009 and 2012. Additionally, it is implied that the impact of SMS implementation in a certain organization must be complemented by a statistical qualitative analysis of its effect on the distribution of accidents. Both analyses are developed in the present study, thus contributing relevant implications when assessing both quantitatively and qualitatively the effects of developing and implementing a health and safety management system in this sector. The findings from this research can contribute to understanding how SMS implementation can help reduce accidents in this industry as well as to enhance SMS implementation in a high-risk sector.

A conceptual design of coffee stem biomass gasification was developed using the Aspen Plus process simulator, which was analyzed based on energy criteria such as the lower heating value of the produced synthesis gas and cold gas efficiency. The results yielded a lower heating value of 5.07 kJ/MJ, with a cold gas efficiency of 77.45% and a yield of 2.75 kg gas/kg biomass, all operating under optimal conditions with studied parameters including an air-to-biomass ratio between 1.8 and 2.5, gasifying agent temperature of 260 °C, and biomass moisture content below 10%. A sensitivity analysis was conducted on the formation of the most important compounds for synthesis gas quality (H2, CO, CO2, H2O, CH4). This involved understanding the reactions occurring throughout the gasification reactor and evaluating process parameters such as feed moisture content, gasification section temperature, and the ratio between feed and gasifying agent flows to optimize the process with the aim of improving product quality and reducing residue formation. The simulation scheme was validated against experimental data, yielding results consistent with reality. This contributed valuable information towards process optimization for projects targeting the Colombian coffee sector, paving the way for future gasifier designs tailored to the quantity of raw material to be processed.
