Chitin (Ct) is a crucial biopolymer present in fungi, algae, arthropods, and is usually obtained from crustacean shells. Chitosan (Cs) is a derivative from Ct deacetylation, and possesses numerous uses in various agro-industrial fields. Research on fungal-derived Ct and Cs is mostly focused on pharmaceutical uses, however their uses for plant disease control remain less explored. The main objective of the current study is to evaluate the possibility of using chitosan obtained from mushroom Pleurotus eryngii (Cs-Pe) for controlling some phytopathogens compared to commercial chitosan (C.Cs). This study is focused on the following key areas: (i) extracting Ct from P. eryngii mycelium and converting it to Cs through deacetylation, using both bleaching and non-bleaching methods; (ii) conducting a physico-chemical characterization and in vitro evaluation of the antimicrobial activity of the obtained Cs; (iii) performing an in vivo assessment of the phytotoxic and cytotoxic effects of Cs; and (iv) investigating in vivo the impact of the studied chitosan on fruit quality and its biocontrol efficacy against Monilinia laxa infections in plum fruits. Results showed that Cs-Pe, especially the unbleached one, displayed promising in vitro antimicrobial activity against the majority of tested pathogens. Regarding the cytotoxicity, the highest significant increase in cell abnormality percentage was observed in the case of C.Cs compared to Cs-Pe. In the in vivo study, Cs-Pe acted as a protective barrier, lowering and/or preventing moisture loss and firmness of treated plums. The studied Cs-Pe demonstrated notable efficacy against M. laxa which decreased the fruits' percentage decline. These results strongly suggest that Cs derived from P. eryngii is a potential candidate for increasing plums' shelf-life. Introduction: Many lactic acid bacteria (LAB) strains are currently gaining attention in the food industry and various biological applications because of their harmless and functional properties. Given the growing consumer demand for safe food, further research into potential probiotic bacteria is beneficial. Therefore, we aimed to characterize Pediococcus pentosaceus DSPZPP1, a LAB strain isolated from traditional fermented sausages from the Basilicata region of Southern Italy. Methods: In this study, we analyzed the whole genome of the P. pentosaceus DSPZPP1 strain and performed in silico characterization to evaluate its applicability for probiotics and use in the food industry. Results and Discussion: The whole-genome assembly and functional annotations revealed many interesting characteristics of the DSPZPP1 strain. Sequencing raw reads were assembled into a draft genome of size 1,891,398 bp, with a G + C content of 37.3%. Functional annotation identified 1930 protein-encoding genes and 58 RNAs including tRNA, tmRNA, and 16S, 23S, and 5S rRNAs. The analysis shows the presence of genes that encode water-soluble B-group vitamins such as biotin, folate, coenzyme A, and riboflavin. Furthermore, the analysis revealed that the DSPZPP1 strain can synthesize class II bacteriocin, penocin A, adding importance to the food industry for bio-enriched food. The DSPZPP1 genome does not show the presence of plasmids, and no genes associated with antimicrobial resistance and virulence were found. In addition, two intact bacteriophages were identified. Importantly, the lowest probability value in pathogenicity analysis indicates that this strain is non-pathogenic to humans. 16 s rRNA-based phylogenetic analysis and comparative analysis based on ANI and Tetra reveal that the DSPZPP1 strain shares the closest evolutionary relationship with P. pentosaceus DSM 20336 and other Pediococcus strains. Analysis of carbohydrate active enzymes (CAZymes) identified glycosyl transferases (GT) as a main class of enzymes followed by glycoside hydrolases (GH). Our study shows several interesting characteristics of the isolated DSPZPP1 strain from fermented Italian sausages, suggesting its potential use as a promising probiotic candidate and making it more appropriate for selection as a future additive in biopreservation. Kiwifruit vine decline syndrome (KVDS) is an emerging challenge related to various factors, including water stagnation, soil compaction, root hypoxia, imbalanced soil redox potential, soil nutrient imbalance and fungal pathogens, yet its definitive causes remain unconfirmed. This study compared soils and roots of healthy plants (CTRL group) and plants affected by KVDS (KVDS group) at a kiwifruit orchard (Actinidia chinensis cv. 'Zesy002') in Sermoneta (Lazio region), Italy. Our findings indicate that soil redox potential, a parameter related to soil oxidation, and soil macroporosity were significantly lower in the KVDS soils. The water table was also found to be higher in the KVDS block of the orchard, demonstrating a higher soil water content. This suggests less aeration and more water content in KVDS‐affected soils compared to the control. Root analyses showed that KVDS led to browning and decomposition of rhizodermis, decay of the central stele, detachment of the cortex, reduced cell size and the presence of starch granules in the parenchyma, with significant variations in root morphometric parameters. To mitigate KVDS, sustainable agronomic practices, including precision irrigation, green manure, compost addition and water drainage root pruning, were carried out on some KVDS‐symptomatic vines (SUST group). Molecular analyses revealed the presence of 10 fungal species from treated root plants, with Paraphaeosphaeria michotii, Fusarium oxysporum and Ilyonectria vredenhoekensis being frequently isolated from symptomatic KVDS‐affected plants but less from SUST plants. Also, Beauveria bassiana and Bacillus amyloliquefaciens, showed an effect against I. vredenhoekensis and P. michotii, and may be suitable candidates as biocontrol agents. The adoption of sustainable agronomic practices resulted in significantly more healthy plants (46% increase) by the end of the trial, demonstrating that a sustainable soil use and water management at kiwifruit orchards can mitigate KVDS through restoration of natural growth conditions. In recent years, the Asian chestnut gall wasp (ACGW) Dryocosmus kuriphilus has been reported to have a high incidence in Italy and other Mediterranean basin countries. In 2021-2022, a study was undertaken in the Basilicata Region (Southern Italy) to investigate the relationship between the galls produced by ACGW on sweet chestnut (Castanea sativa Mill.) and fungal pathogens. In particular, the fungal diversity from green and necrotic galls collected from two important sweet chestnut sites (Melfi and Rionero in Vulture) was investigated. Nineteen fungal taxa were identified based on their morphological and molecular traits. In both localities, the most frequent species isolated from green and necrotic galls were Gnomoniopsis castaneae, Colletotrichum acutatum, and Pestalotiopsis sp. It is essential to understand the role played by the galls as an inoculum source for sweet chestnut fungal pathogens, particularly for G. castaneae, an emerging pathogen of which biology is still poorly understood. Findings from the present study stressed that the complex relationship between host-insectmicrobial community needs to be elucidated to be able to control the pathogenic fungi and consequently maintain sweet chestnut trees' health as they play a key role in the local agriculture (horticulture, forestry) and subsidiary economy. Genetic diversity in crop plants is the conditio sine qua non for sustainable agriculture and long-term food security. Our research carried out the morphological, agronomic, and physico-chemical characterization and resistance to late blight of 35 tomato landraces from seven countries. These landraces have been approved and appear in the Official Catalog of Varieties. The International Plant Genetic Resources Institute (IPGRI) descriptors have been used to describe the tomato's morphological and agronomic characteristics. For the physico-chemical characteristics, the dry matter, the pH, and the carotenoid content (lycopene, lutein, and β-carotene)) were analyzed. Carotenoids were monitored by high-performance liquid chromatography (HPLC). The results showed that the morphological diversity of landraces was very high. Three landraces of remarkable commercial value have shown increased resistance to late blight caused by Phytophthora infestans, one of the most damaging diseases of tomato. Also, six landraces had a lycopene content exceeding 100 µg/g sample. The carotenoid content ranged between 0.769 (Marmande-FR 166) and 140.328 mg kg−1 FW (Răscruci). The landrace with the highest β carotene content was PT 308 with 65.499 mg kg−1 FW, while the lowest values were registered for Marmande-FR 166 with 0.105 mg kg−1 FW. The present study provides essential information on the morphological and agronomic qualities of these tomato landraces and their lycopene and other carotenoid content. The results are discussed in light of the importance of tomato landraces in meeting the preferences of different producers and consumers, the choice of the most suitable landraces for specific pedoclimatic conditions, and the supply of carotenoid pigment sources for the pharmaceutical industry. Our research responds to humanity's great global challenges: preserving agricultural biodiversity, protecting the environment by identifying pest-resistant varieties, and also protecting consumer health by finding important sources of antioxidants. The investigation focused on the deterioration of the walls in the hypogeum of "San Pietro Barisano" rupestrian church, located in the Matera-Sassi (Southern Italy), one of the UNESCO World Heritage sites. The study evaluated the biocide activity of a mixture of natural glycoalkaloids (GAs) extracted from the unripe fruit of Solanum nigrum and applied to clean a hypogeum wall surface in the church affected by bio-patinas. The analyzed bio-patina, collected before treatment and, at pre-established times, after treatment, showed changes in chemical composition detected by XPS, accompanied by visible discoloration and biological activity variation. The biocidal action of the glycoalkaloids mixture, directly employed on the wall surface, was effective after about four weeks for most bio-patina colonizers but not for the fungal species that can migrate and survive in the porosities of the calcarenite. Consequently, the cleaning procedure requires the integration of fungicidal actions, combined with the consolidation of the surfaces, to obtain complete bioremediation and avoid subsequent biological recolonization. SEM images and associated microanalysis of pretreated bio-patina have revealed the biocalcogenity of some autochthonous microorganisms, thus preluding to their eventual isolation and reintroduction on the wall surface to act as consolidants once the bio-cleaning phase has been completed.
