The presence of infective larvae (L 3 ) of gastrointestinal nematode (GIN) parasites in pastures directly contributes to the constant recurrence of infections in ruminant herds. This study aimed to evaluate the nematophagous fungus Duddingtonia flagrans (AC001) (proteolytic crude extract and/or conidia) in the in vitro control of GIN L 3 in coprocultures. To produce the proteolytic crude extract, a suspension (10 7 conidia/mL) of D. flagrans was inoculated into a liquid medium. After 6 days, the medium was filtered, centrifuged, and its proteolytic activity was measured. For the experimental assay, fecal samples were collected directly from the rectal ampulla of naturally infected sheep, and egg counts per gram of feces (EPG) were performed. Coprocultures were prepared using 10 g of fecal material with the groups defined as follows: control group G1 (1.0 mL of denatured proteolytic crude extract); treated group G2 (1.0 mL of active proteolytic crude extract); treated group G3 (1.0 mL of active proteolytic crude extract + 1.0 mL of AC001 conidia). The coprocultures were maintained at room temperature (25ºC), for 7 days, and then the L 3 larvae were recovered. The results demonstrated that AC001 successfully produced protease (56.34 U/mL). The treatments with active proteolytic crude extract (G2) and active proteolytic crude extract + AC001 conidia (G3) were significantly different (p < 0.01) from the control group with denatured proteolytic crude extract (G1). AC001 and its proteolytic crude extract acted concomitantly on helminths directly in the fecal environment, suggesting potential future applications in the field., (© 2024. Nematophagous, or helminthophagous fungi of the genera Duddingtonia , Arthrobotrys , Monacrosporium , Pochonia , Paecilomyces , and Mucor , have been used over the years in in vitro and in vivo experiments to control helminth parasites that are potentially zoonotic. These fungi have shown efficacy against the following helminth genera: Ancylostoma , Toxocara , Enterobius , Strongyloides , Angiostrongylus , Taenia , Fasciola , and Schistosoma . The results obtained from these experiments, together with studies on soil contamination, suggest the viability of their use as a sustainable and effective strategy to reduce environmental contamination by these zoonotic parasites. Therefore, the aim of this review was to address the role of helminthophagous fungi in the biological control of potentially zoonotic helminths. To this end, we describe (1) a brief history of helminthophagous fungi; (2) a discussion of some potentially zoonotic intestinal parasites; (3) the importance of helminthophagous fungi in the control of nematodes, cestodes, and trematodes; and (4) the potential of helminthophagous fungi as a practical and sustainable strategy.

We evaluated the efficacy of Bioverm®, a commercial product containing Duddingtonia flagrans, on the control of buffalo (Bubalus bubalis) gastrointestinal nematodes. We randomly divided 12 buffaloes into two groups of six animals. In the treated group, each animal received a Bioverm®`s single dose of 1g (10 5 chlamydospores of D. flagrans) to 10 kg of live weight; in the control group, each animal received 1g of corn bran for each 10 kg of live weight as a placebo. Fecal samples were individually collected from 12, 24, 36, 48, 60 and 72 h after treatments. To examine 1) viability of chlamydospores passed through the gastrointestinal tract, 2 g of faeces and 1000 infective larvae (L3) were added to Petri dishes with 2% water-agar, and 2) to examine larval predation by D. flagrans during fecal cultures, 2000 L3 were added. In the Petri dishes, were observed significant reductions (p < 0.01) in the treated group after 48 (56.7%) and 60 h (91.5%). In the fecal cultures, significant reductions (p < 0.01) occurred in the treated group from 36 h (75%), with larval reduction up to 72 h. High larval predation rate occurred 60 h after Bioverm® administration. This study aimed to evaluate the concomitant use of the nematophagous fungus Duddingtonia flagrans (AC001) and its protease-rich crude extract for the in vitro control of Panagrellus sp., Haemonchus spp., and Trichostrongylus spp. The nematicidal tests were carried out on larvae of the free-living nematode Panagrellus sp. and infective larvae of the gastrointestinal parasitic nematodes of domestic ruminants (Haemonchus spp. and Trichostrongylus spp). Five experimental groups were set: (1) one control group (G1) and (4) four treated groups -G2 - active crude extract; G3 - denatured crude extract; G4 - fungus, and G5 - fungus + active extract. Plates were incubated at 28 ºC for 24 h followed by the recovery of the larvae using the Baermann technique. The results showed a lower recovery of Panagrellus sp. larvae in the experimental groups compared to the control group, as follows: 52 % (G2), 16 % (G3), 46 % (G4), and 77 % (G5). An even greater reduction (77 ± 5 %) occurred in the group (G5). In addition, the authors observed lower averages of L 3 of Haemonchus spp. and Trichostrongylus spp. in the experimental groups compared to the control group, as follows: 59 % (G2), 0 % (G3), 86 % (G4), and 76 % (G5). In turn, there was a difference (p < 0.01) between (G5) and (G2). The control of gastrointestinal parasites in animals has become more challenging every year due to parasite resistance to conventional chemical control, which has been observed worldwide. Ovicidal or opportunistic fungi do not form traps to capture larvae. Their mechanism of action is based on a mechanical/enzymatic process, which enables the penetration of their hyphae into helminth eggs, with subsequent internal colonization of these. The biological control with the Pochonia chlamydosporia fungus has been very promising in the treatment of environments and prevention. When used in intermediate hosts of Schistosoma mansoni, the fungus promoted a high percentage decrease in the population density of aquatic snails. Secondary metabolites were also found in P. chlamydosporia. Many of these compounds can be used by the chemical industry in the direction of a commercial product. This review aims to provide a description of P. chlamydosporia and its possible use as a biological parasitic controller. The ovicidal fungus P. chlamydosporia is effective in the control of parasites and goes far beyond the control of verminosis, intermediate hosts, and coccidia. It can also be used not only as biological controllers in natura but also as their metabolites and molecules can have chemical action against these organisms. Toxoplasmosis is mainly transmitted to human beings through the consumption of contaminated food, and several outbreaks caused by infected meat consumption have been reported in Brazil. We performed a systematic literature review on the prevalence and risk factors for toxoplasmosis in slaughtered animals and performed a meta-analysis of its prevalence for different species and regions. Furthermore, we also discussed the infectivity of seropositive animals, risk factors, and preventive strategies. In the meta-analysis, the overall prevalence estimates for poultry, ostrich, goats, swine, equids, sheep, and bovines were 42.4, 40.4, 23.0, 19.9, 19.1, 17.3, and 16.2%, respectively. Regarding the Brazilian regions, the highest prevalence values were detected for bovines and equids in the South (32.3 and 34.4%, respectively) and swine, goats, sheep, and poultry in the Northeast (29.3, 23.0, 22.9, and 69.8%%, respectively). High proportions of Toxoplasma gondii viability in bioassay conducted on seropositive animals were seen for sheep (34/40 = 85%) and swine (12/15 = 80%). Toxoplasma gondii infections are widespread on Brazilian farms, and the summarized data allow the establishment of high-priority areas and/or species for the adoption of preventive strategies to control this parasite at different levels of the food chain.

The aim was to detect correlations of IgG antibodies against N. caninum in serum and colostrum samples from ewes, through the IFAT, and to evaluate the presence of this immunoglobulin in the serum of newborn lambs after colostrum ingestion. Blood samples from 162 ewes that did not show any disease in the general physical examination and from their newborn lambs, not more than five days postpartum, along with 162 colostrum samples and 182 blood samples from the neonates, were analyzed. In total, 27.8% (45/162) of the mothers were positive for anti- N. caninum IgG, among which antibodies were detected in the colostrum in 46.7% (21/45). All the ewes with positive colostrum had reactive offspring. The kappa agreement for the correlation between the serological tests on the ewes and the colostrum results was 0.558. This correlation increased as the antibody titers of the mothers increased, and reached 1.000 from the titer of 1:400 from the mothers. Comparison of the antibody detection results between the offspring's serum and colostrum showed a kappa agreement of 1.000. In conclusion, there was a good agreement regarding the detection of anti- N. caninum IgG between the colostrum samples and the lambs' serum; the use of colostrum forms a noninvasive alternative for diagnosing N. caninum in sheep herds.

Cattle parasitic otitis caused by the nematode Rhabditis spp. is a serious health problem in Brazil, a situation which is confounded by lack of effective control measures. In vitro studies associating biological and chemical control as an alternative method showed promising results. The objective was to evaluate the combined use of Duddingtonia flagrans (AC001), 10% dimethylsulfoxide (DMSO), and 1.9% ivermectin for the in vivo control of Rhabditis spp., in naturally infected Gyr cattle. For this purpose, 48 animals, whose infection in both ears was diagnosed, were randomly assigned to 6 groups: group 1 (ivermectin 1.9%); group 2 (10% DMSO); group 3 (AC001); group 4 (ivermectin 1.9% + 10% DMSO w/v); group 5 (1.9% ivermectin + AC001 w/v); group 6 (10% DMSO + AC001 v/v). The treatments were performed in a single dose, in the right ears, with the left ears remaining untreated, as a control group. There was a significant reduction (p < 0.01) in the number of nematodes in the treated groups in relation to the control, with the following best efficacies: groups 1 and 2, 47% and 52.9%, respectively, 7 days after treatment; groups 3, 4, and 5, 47.8%, 48.6% and 36.7%, respectively, 14 days post-treatment; group 6, 38.4%, 21 days post-treatment. We aimed to evaluate a Brazilian commercial formulation of the fungus Duddingtonia flagrans (Bioverm®) for controlling gastrointestinal nematodes in sheep kept on native pasture in the Caatinga biome, in the semiarid region of Brazil. Twenty ewes, aged between 12 and 18 months, were divided into two groups. In the treated group, each animal received 1 g of the Bioverm® product for each 10 kg of live weight, daily, together with commercial feed, for 6 months. In the control group, the animals received feed without Bioverm®. Each group remained throughout the experiment in a 1.2-ha paddock. Monthly counts of eggs per gram (EPG) of feces, fecal cultures, packed cell volume (PCV), weight measurements, and collection of leaf mass from the pasture were performed. There was greater reduction in EPG, greater weight gain, and less infestation by infective larvae in the paddock of the Bioverm® group, compared with the control group (p < 0.05). There was no significant difference (p ≥ 0.05) in the mean PCV percentage between the Bioverm® and control groups. In coprocultures, Haemonchus sp. was the most prevalent helminth.
