Tomato is a short shelf-life climacteric fruit. The alcobaça (alc) mutation delays fruit ripening but has negative pleiotropic effects in fruit colour (red colour) and firmness, while also provoking the loss of tomato-like climacteric behaviour. Previous studies of this mutant described the effect of alc on fruit colour using qualitative descriptors (yellow, orange, and pale red), but a quantitative approach to understand the impact of alc on colour is still lacking. The main objective of this thesis was studying the relation of shelf-life and fruit colour intensity in alc mutant homozygous and heterozygous genotypes, using a quantitative approach. For this purpose, an F2 segregating population for the alc mutation was developed from an alc mutant parent (LC215, Penjar tomato landrace) and a non-mutant parent (LC854, processing tomato). Yield-related traits, colour, and shelf life (measured up to 5 months postharvest) were studied in a per plant basis in an experimental field developed in Agropolis-UPC (Viladecans, Spain). LC854 showed a red colour and short shelf-life, while LC215 showed a pale red colour and a long shelf-life. The hybrid showed a lower red colour intensity, but increased shelf-life compared with LC854; however, F1 shelf-life was much lower than the alc mutant parent, signalling the recessive behaviour of the alc allele. In the F2 population, a broad diversity was found for all the traits, including shelf life and fruit colour intensity; some individuals showed fruits with a long shelf-life, similar than LC215, while other individuals showed a red colouration similar than LC854. However, long shelf-life F2-individuals had also a low red colour intensity, and high yellow colour intensity, signalling the pleiotropic negative effect of the alc allele on the fruit colour. Furthermore, a positive relation between shelf-life and yellow colour intensity was found, as well as negative correlation between red colour intensity and shelf-life. La mutació alcobaça (alc) retarda la maduració del fruit amb efectes pleiotròpics en la intensitat de color vermell, la fermesa i la pèrdua del comportament climatèric del fruit. L'efecte de la mutació alc sobre la coloració del fruit ha estat descrita a la bibliografia mitjançant descriptors qualitatius, mancant informació quantitativa. L'objectiu principal d'aquest treball va ser estudiar la co-segregació de la postcollita i la coloració en una població segregant F2. Per fer-ho, es va desenvolupar una població segregant F2 utilitzant un parental mutant alc (LC215, varietat tradicional de tomàquet de Penjar) i un no mutant (LC854, tomàquet d'indústria). Les característiques de rendiment, qualitat i conservació postcollita (avaluada fins als 5 mesos) van ser estudiades per planta en un assaig realitzat a Agropolis-UPC (Viladecans, Espanya). El parental LC854 va mostrar un color vermell i una postcollita curta, mentre que LC215 va mostrar un color vermell pàl·lid i una postcollita llarga. L'híbrid va mostrar una coloració vermella menys intensa i una postcollita més llarga que el parental LC854, però molt inferior que el parental mutant alc, suggerint un efecte parcialment recessiu de l'al·lel alc sobre la conservació postcollita. A la població F2, tots els caràcters inclosos postcollita i intensitat de color, van mostrar una àmplia diversitat; alguns individus van presentar fruits amb una postcollita llarga, similar a la de LC215, mentre que altres individus van mostrar una coloració vermella similar a LC854. Els individus F2 que van mostrar una postcollita llarga van tenir també una baixa intensitat de color vermell i una alta intensitat de color groc, assenyalant un efecte pleiotròpic negatiu de l'al·lel alc en la coloració del fruit. A més, es va trobar una relació positiva entre postcollita i intensitat de coloració groga, així com una correlació negativa entre la intensitat de coloració vermell i la conse, El tomate es un fruto climatérico de postcosecha corta. Las características de rendimiento, calidad y postcosecha (evaluada hasta los 5 meses) fueron estudiados por planta en un ensayo de campo desarrollado en Agropolis-UPC (Viladecans, España). El parental LC854 mostró un color rojo y una postcosecha corta, mientras que LC215 mostró un color rojo pálido y una postcosecha larga. El híbrido mostró un color rojo menos intenso y una postcosecha más larga que el parental LC854, pero muy inferior que el parental mutante alc, sugiriendo un efecto parcialmente recesivo del alelo alc sobre la conservación postcosecha. En la población F2, todos los caracteres incluidos postcosecha e intensidad de color, mostraron una amplia diversidad; algunos individuos presentaron frutos con una postcosecha larga, similar a la de LC215, mientras que otros individuos mostraron una coloración roja similar a LC854. Los individuos de F2 con postcosecha larga, tuvieron además una baja intensidad de color rojo y una alta intensidad de color amarillo, señalando un efecto pleiotrópico negativo del alelo alc en la coloración del fruto. Además, se encontró una relación positiva entre postcosecha e intensidad de coloración amarilla, así como una correlación negativa entre la intensidad de coloración roja y conservación

Non-trellised culture of tomato is gaining interest among farmers cultivating long shelf-life (LSL) landraces because of the reduction in production costs. This cropping system can benefit from the selection of determinate growth genotypes, mirroring the advancements achieved in processing tomatoes. With this aim, and profiting from the natural variation found within the Penjar landrace, we selected traditional determinate genotypes and breed novel lines carrying both the self-pruning (sp) and the compound inflorescence (s) mutations. Traditional genotypes and breeding lines were compared to commercial controls in a three multi-locality trial and under two irrigation regimes (normal watered, 100% ETc; and water deficit, 50% ETc). Water use efficiency, harvest index, yield and fruit quality traits were studied at harvest. During postharvest, the effects of genotype and management practices on shelf life were assessed under different storage conditions (controlled conditions/cold storage/farmer facilities). All genotypes used water more efficiently under water deficit irrigation than under normal irrigation. Harvest index was improved in the determinate genotypes and was neither affected by locality nor by irrigation factors. Breeding lines showed yields similar to the controls and the traditional ones; however, they displayed a plant architecture that facilitated their management. They also presented higher postharvest shelf life than controls and traditional lines. Shelf life was significantly affected by genotype (G), locality (L, at early stages), and irrigation regime (I, at later stages), with the contribution of the genotype to the phenotypic variance increasing along the postharvest. Low watering increased shelf life in some genotypes while cold storage and high humidity conditions impeded the long shelf-life trait.
