This review paper examines acid and alkaline pretreatments on perennial grasses for second-generation (2G) bioethanol production, a relatively unexplored area in this field. It compares the efficiency of these pretreatments in producing fermentable sugar and bioethanol yield. This study finds that alkaline pretreatment is more effective than acidic pretreatment in removing lignin and increasing sugar yield, leading to higher ethanol yields. However, it is costlier and requires longer reaction times than acidic pretreatment, while acidic pretreatment often leads to the formation of inhibitory compounds at higher temperatures, which is undesirable. The economic and environmental impacts of lignocellulosic biomass (LCB) are also assessed. It is revealed that LCB has a lower carbon but higher water footprint and significant costs due to pretreatment compared to first-generation biofuels. This review further explores artificial intelligence (AI) and advanced technologies in optimizing bioethanol production and identified the gap in literature regarding their application to pretreatment of perennial grasses. This review concludes that although perennial grasses hold promise for 2G bioethanol, the high costs and environmental challenges associated with LCB necessitate further research. Excessive use of fossil fuels has accelerated climate change and global warming necessitates the need for renewable energy sources that have a lower environmental impact. In the recent decade, lignocellulosic biomass has become a prominent alternative to renewable energy resources for the production of bioenergy. The pretreatment procedure is considered a pivotal step for transforming biomass into value-added products such as sugars, biofuels, etc. Therefore, the present work aims to study the effect of different pretreatment approaches on rice husk with acids (H2SO4 and HCl), alkalis (NaOH and KOH), and organic solvents (ethanol and methanol) utilizing different concentrations like (2, 4 and 6% in case of acids), (2,4 and 6% for alkalis) and (50% and 70% for organic solvents) with different residence time (1, 3, 6, and 24 h). The most effective results obtained from the aforementioned steps were further adopted for enzymatic hydrolysis. Further, the changes in structural properties of biomass were assessed in relation to the pretreatment process employing scanning electron microscopy (SEM), X-ray diffraction (XRD), and Fourier Transform Infrared (FTIR) analyses. This paper also highlights the techno-economic analysis of alkali pretreatment. Additionally, the operational targets for the process were identified by using a modeling software-SuperPro Designer. Results obtained from the study showed a maximum yield of reducing sugar i.e., 1.906 ± 0.2 mg/ml (4% NaOH with 6 h of incubation). The present paper discusses response surface methodology (RSM) as an efficient tactic for predictive model and optimization of the whole experimental methods of reducing sugar and energy. In this work, the application of RSM presented for optimizing reducing sugar and energy as compared with production between chemical and biological pretreatments. All experiments applied statistical designs in order to develop a statistic multivariate analysis model that provides to consider the effect of different parameters on a process and describe the optimum values of these variables to optimize the response. Dred sunflower stalks were pretreated by sodium hydroxide (NaOH) and Trichoderma reesei as a function of two variables: concentration of NaOH (%) and T. reesei (%) and time for pretreatment (Day) to receive reducing sugar and energy. The chemical pretreatment model was characterized by 13 runs, varying the variables at two factors, NaOH (1, 1.5, 2%) and Day (1, 2, 3). The biological pretreatment model was characterized by 13 runs, varying the variables at two factors, T. reesei (1, 1.5, 2%) and Day (1, 2, 3), by central composite design experimental design. In the chemical pretreatment, experiments performed at 2% (w/v) of NaOH for 3 days were used. The chemical pretreatment model at 2% NaOH for a 3-day release reduced sugar by 5.812 g/L and energy by 92.992 kJ/L; on the other hand, biological pretreatment model at 2% T. reesei for a 3-day release reduced sugar by 3.891 g/L and energy by 62.256 kJ/L, reducing sugar starter for fermentation by 49.0670 ± 6.4660 g/L and fermentation efficiency by 71.60% at 48 h fermented time. The objective of this study was to develop an efficient method for enhancing the production of bioethanol in a single‐reactor system (SRS) by implementing acid–base pretreatment and enzymatic hydrolysis techniques, thereby eliminating differentiation and removal processes. The aim was to establish a process for bioethanol synthesis using hydrolyzed palm oil mill effluent (POME) and a locally sourced cellulase enzyme. The pretreatment and enzymatic hydrolysis methods were successfully optimized, resulting in a maximum yield of reducing sugars of 26.6 g/L (53.14%). To achieve the highest bioethanol yield, fermentation was carried out using both the one‐factor‐at‐a‐time (OFAT) and face‐centered central composite design (FCCCD) approaches. The OFAT approach was employed to obtain the maximum bioethanol production, which yielded 6.75% v/v of bioethanol from the hydrolyzed POME using the same bioreactor. In the case of the FCCCD process, the optimal conditions led to a bioethanol production of 7.64% v/v during the fermentation stage. Kinetic analyses of the bioethanol produced revealed a specific growth rate (μ) of 0.198 h−1 and a specific product formation rate (qp) of 0.239 h−1 after 3 days of fermentation. These findings highlight a promising strategy for efficient management of POME through the production of biofuels, which could contribute to the economic growth of the country. The organic selenium (Se), particularly in the form of selenoamino acids, in non-edible sections or by-products of Se-enriched plants, has the potential to generate Maillard reaction products (MRPs) during thermal treatment or fermentation. To elucidate the recycling process of organic selenium in foods and improve the utilization rate of Se, the biotransformation of organic selenium was studied by the cultivation of edible radish sprouts with Se-MPRs. Maillard reactions were simulated using selenoamino acids (SeAAs; selenomethionine and methylselenocysteine) and reducing sugars (glucose and fructose) for preparing Se-MRPs. The structures of the possible dehydrated Se-MRPs were analyzed using a HPLC-ESI-MS/MS system based on their fragmentation patterns and Se isotopic characteristics. Se absorption by the radish sprouts cultivated using Se-MRPs was estimated by the corresponding Se in the SeAAs and the total Se contents. The capabilities of SeAA transformation and total Se assimilation by the sprouts were related to the substrate composition during the Se-Maillard reaction. A particular Se-MRP (selenomethionine + fructose) increased SeAAs transformation by 33.8% compared to selenomethionine. However, glucose and fructose seemed to inhibit the transformation of the Se-MRPs to SeAAs by 10.0 to 59.1% compared to purified Se-MRPs. Ultrasonication is a potential and highly efficient green technique to extract bioactive elements from plants. In this study, ultrasonication technology is used to extract reducing sugar and polyphenolic compounds from burdock root waste (Arctium lappa L.) using the response surface approach. For this, the ultrasonic extraction parameters, solid-to-water ratio (20–40 g/mL), sonication time (60–100 min), and sonication temperature (40–60 °C) were optimized through response surface methodology (RSM), where the reducing sugar and total phenolic content were selected as the responses. The results indicated that all the parameters considered had a significant impact (p<0.05) on reducing sugar (ranging from 4.38 to 6.12%) and total phenolic content (ranging between 4.94 and 6.98 mg GAE/g). The optimized condition was solid to liquid ratio of 38.52 g/mL, sonication time of 90.06 min, and sonication temperature of 59.86 °C, which predicted reducing sugar of 6.12% (6.48% after validation) and total phenolic content of 7.04 mg GAE/g dw (7.44 mg GAE/g dw after validation). Furthermore, in vitro biological activities of burdock root extracts exhibited excellency in retaining the reducing sugar (6.48–6.97%), total flavonoid content (40.66—42.36 mg RE/g), DPPH (33.70—43.45 μmol TE/g), and ABTS+ (28.10—29.83 μmol TE/g) radical scavenging activity, ferric-reducing antioxidant power (54.53—58.70 μmol TE/g), and α-glucosidase inhibitory activity (>20%) both in ultrasonic and conventionally extracted samples with ultrasonication showed the higher activity. The results suggest that ultrasonication is an effective green technology and that burdock root waste has significant antioxidant and antidiabetic potential and could be used as a promising source of functional components. The experiment was conducted during 2023 at Bioenergy Laboratory, Department of RBEE, College of Agricultural Engineering and Technology, CCSHAU, Hisar, Haryana, India. Compositional changes in sugarcane bagasse subjected to varying concentrations of sodium hydroxide (0.3% to 1.2%), revealing significant increases in glucan content (from 37.13% to 53.81%) alongside decreases in xylan, acidinsoluble lignin, acid-soluble lignin, ash, and other extractives. These changes were validated using microscopic technique SEM, confirming the efficacy of the pretreatment process. The utilization of a customized cellulase cocktail derived from Trichoderma harzianum and Trichoderma viride holds significant promise in enhancing the saccharification from alkali-pretreated sugarcane bagasse. This study investigates the synergistic effects of cellulase enzymes produced by these fungi on the hydrolysis of lignocellulosic biomass. The enzymatic hydrolysis process is optimized by varying enzyme dosages, reaction conditions, and incubation times to maximize the release of fermentable sugars. Results indicate a substantial improvement in saccharification efficiency with the customized cellulase cocktail, highlighting its potential for sustainable biofuel production. The pretreated sugarcane bagasse, when saccharified with Trichoderma harzianum and Trichoderma viride individually, released 254.43 mg g-1 and 325.53 mg g-1 of reducing sugars, respectively, after 40 h of incubation. In contrast, the combined enzymatic cocktail achieved a substantial increase in glucose yields (345.12 mg g-1) at 40 h, showcasing the synergistic effect of the combined enzymatic activity. The intention of this post-harvest analysis was to see how the interaction of wrapping materials and storage requirements influenced the shelf life and chemical fruit quality of the ‘Khirsapat’ mango. ‘Khirsapat’ mangoes were harvested when they were fully mature and wrapped in various materials, viz. Low-density polyethylene (LDPE) + No perforation, LDPE + 5 % perforation, LDPE + Blotting paper inside, LDPE + 5 % perforation+ Blotting paper inside, Corrugated Fiber Board (CFB) Box (5% ventilation), LDPE (5% perforation) + CFB Box (5% ventilation), Plastic crate & Control (without any wrapping). Biochemical parameters such as TSS, titratable acidity, reducing sugars, and total sugars were measured in the fruits. According to the data, the physicochemical characteristics of a ‘Khirsapat’ mango were significantly impacted by the wrapping materials and storage period. Wrapping of ‘Khirsapat’ mango in LDPE 5% perforation + CFB Box 5% ventilation at temperature 12±1°C has shown that, up to the 12th day the most successful treatment approach showed a more progressive increase in TSS and total sugar than other treatments. The cultivar's optimum shelf life and nutritional value retention was found by this study. However, in terms of sensory quality, the highest score for ‘Khirsapat’ mango was achieved in a CFB Box with 5% ventilation.

The destructive effects of burning conventional fossil fuels on the environment have created the realisation of the significance of using alternative fuels as a mitigating measure to reduce greenhouse gas emissions. One of the most promising and environmentally friendly substitutes for petroleumbased gasoline fuel is bioethanol. This study uses agricultural residue (bagasse), halophytes and non-edible biomass as feedstocks to produce bioethanol, through lignin content extraction from Panicum antidotale, Phragmites karka, Sugarcane bagasse, Jatropha curcas husk and Conocarpus erectus leaves and then cellulose reduced by acid pretreatment followed by hydrolysis. The cellulose content was found higher in Panicum antidotale (43.39 percent) and lowest in Jatropha curcas husk (35.10 percent), while moderate values of (42.81 percent) in Sugarcane bagasse, Phragmites karka (37.48 percent), and Conocarpus erectus (38.53 percent). At the beginning and end of the fermentation process, the total carbohydrate contents of the hydrolysed biomass were analysed, and their bioethanol yields were measured. Maximum reducing sugar was found 19 gm/L (1.186 lb/ft3 ) in Panicum antidotale followed by Phragmites karka, sugarcane bagasse, Jatropha curcas husk, and Conocarpus erectus leaves as 15.89 gm/L (0.991 lb/ft³ ) and 12.09 gm/L (0.754 lb/ft3 ), 10.52 gm/L (0.656 lb/ft3 ), 4.51 gm/L (0.281 lb/ft3 ), respectively. Bioethanol percentage yield by volume was found as 4.36 percent (Panicum antidotale), 3.93 percent (Phragmites karka), 2.87 percent (Jatropha curcas husk), 2.52 percent (sugarcane bagasse) and 1.64 percent (Conocarpus erectus leaves), respectively. The results showed the potential of producing eco-friendly bioethanol as biofuels from halophytes that will help ease the strain on food crops by providing inexpensive and abundant feedstock. The demand for better lignocellulosic solvents has prompted the search for new materials for polysaccharide processing. Ionic liquids have been studied as alternatives to evaluate solubilization mechanisms. In this study, 1-butyl-3-methylimidazolium chloride ([Bmim][Cl]) ionic liquid has been used for the dissolution of Typha latifolia (TL) biomass. The effects of temperature (X1, °C), contact time (X2, h), and solids loading (X3, %w/v) on the reducing sugar concentration (Y, mg/g) were determined by the response surface methodology (RSM). The pretreatment conditions were optimized using the Box-Behnken design, and it was found that the maximum production of reducing sugars was 214.14 mg of sugar/g of dry biomass, with a temperature of 120 °C, a time of 1 h, and a solids loading of 6%w/v. Additionally, analysis by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and thermal analysis (TG-DTG) suggested that pretreatment of biomass with IL could significantly alter the lignocellulosic structure associated with recalcitrance and, therefore, promote the dissolution of the cellulosic fraction. While the treatment of red grapes with pulsed electric fields (PEFs) has been widely investigated, white grapes have been little studied to date. This work, therefore, investigates the PEF treatment of white grapes (Thompson Seedless) by applying different field strengths (2 to 6 kV/cm), resulting in different specific energy inputs (2.5 to 22.5 kJ/kg). Numerous parameters of the whole grapes and the extracted juice were analyzed, including the total phenolic index (TPI), reducing sugars, juice yields, and changes in color and texture. Changes in the aroma profile were investigated using instrument-based and sensory analysis. The most striking effect seen in the PEF-treated samples was the visual change in the grape skin, specifically browning. Depending on the field strength, the treated grapes showed a 26% to 50% reduction in crunchiness. No significant changes were measured in the juice yields compared with the control, although at E = 4.5 kV/cm there was an increase in reducing sugars of up to 11% (220.97 g/L) and a 30% higher TPI (398.90 mg/L). Aroma differences were not detected between the control and the treated grapes, meaning that consumers should not expect negative sensory effects in the juice of treated white grapes. This study investigates the impact of various pre-drying treatments and drying methods on the physicochemical, phytochemical, and sensorial quality of dried bananas. To achieve this goal, four pretreatment solutions were used before drying to enhance the quality characteristics of banana slices: citric acid 1% and ascorbic acid 1%, sucrose 10%, sodium bisulphate 0.5%, and lemon juice 5% and acetic acid 0.5%. Two drying methods were applied, sun drying (SD) and tunnel drying (TD). The dried slices were evaluated for moisture, total acidity, water activity (aw), browning index, rehydration ratio (RR), reducing sugar, ascorbic acid, total phenol, and DPPH %, as well as sensory attributes (color, odor, taste, texture, and overall acceptability). The results showed that the retention of acid content, ascorbic acid, total phenolics, and DPPH in slice samples dried in TD was higher than that in SD. The browning index values in the TD samples were lower than those in the SD samples. Moreover, the TD samples showed a higher RR than the SD samples. It is noted that the pretreatment solutions used have an important role in improving the final quality characteristics of dried banana slices. The results of the sensory evaluation revealed a significant difference in the degrees of color and texture for the slices pretreated with sodium bisulphate. Meanwhile, the rates of taste and odor were significantly higher for the sucrose-pretreated slices. Preservation and shelf life extension of mature coconut water (MCW) is paramount importance for the production of flavored beverages. The present study aims to evaluates the effects of packaging materials (glass bottles, PET bottles and stand-up pouches) and storage temperatures such as atmospheric (28 ± 6 °C) and cold storage (5 ± 1 °C) temperatures on microbial growth and quality parameters (pH, Total Soluble Solids (TSS), total sugar, reducing sugar, and non-reducing sugar) of ultrasound treated MCW during storage. The results showed that pH, total soluble solids, total sugar, reducing sugar and non-reducing sugars decreased significantly (p < 0.05) during storage, irrespective of packaging materials. It was observed that the microbial load increased significantly (p < 0.05) during storage of ultrasonicated samples and control samples. The present study concluded that ultrasonication of MCW at 60% amplitude for 10 min treatment time increased their shelf life up to 12 days under cold storage condition (5 ± 2 °C) with a safe limit of microbial load and maximum retention of nutrients. Even though all the packaging materials were found to be statistically on par (p > 0.05) in maintaining quality parameters and microbial load, glass bottles were observed to be most effective packaging followed by PET bottles and stand-up pouches. • The effects of storage temperature and packaging materials on coconut water was investigated. • Maximum shelf life of 12 days was observed at cold storage. • Samples stored in glass bottles having better quality coconut water. Wheat straw, a lignocellulosic waste biomass, is an inexpensive source of fermentable sugars for bacterial conversion into value-added products. Thermotolerant lactic acid-producing Bacillus sonorenesis was isolated from red soil of brick kiln, which showed remarkable capability to ferment xylose (10 g L− 1) producing 9.24 g L− 1 lactic acid with an overall productivity of 0.128 g L− 1 h− 1 and yield of 0.92 g g− 1 at 50 ∘C. Separate hydrolysis of wheat straw was done by employing response surface methodology (RSM) to optimize pretreatment with sulphuric acid, alkali and hot water. FTIR spectrogram showed removal of lignin due to reduction in peak at 1640 cm− 1 after pretreatment. Scanning electron microscopy (SEM) revealed that the ordered and compact surface morphology of wheat straw was destroyed upon pretreatment. X-ray diffractogram (XRD) revealed 9.71% increase in crystallinity index (CrI) in pretreated biomass. 57.6 g L− 1 reducing sugar was obtained upon pretreatment of 100 g wheat straw biomass. Fermentation of wheat straw hydrolysate by inhibitor (furfural and 5-hydroxymethylfurfural) tolerant Bacillus sonorenesis DGS15 yielded 55.9 g L− 1 lactic acid with an overall productivity of 0.77 g L− 1 h− 1 and yield of 0.97 g g− 1. Bacillus sonorenesis strain DGS15 being thermotolerant as well as inhibitor tolerant appears to be a novel, attractive candidate for producing lactic acid from wheat straw hydrolysate, which can have wide range of commercial applications. Lytic polysaccharide monooxygenases (LPMOs) can oxidatively break the glycosidic bonds of crystalline cellulose, providing more actionable sites for cellulase to facilitate the conversion of cellulose to cello-oligosaccharides, cellobiose and glucose. In this work, a bioinformatics analysis of BaLPMO10 revealed that it is a hydrophobic, stable and secreted protein. By optimizing the fermentation conditions, the highest protein secretion level was found at a IPTG concentration of 0.5 mM and 20 h of fermentation at 37 °C, with a yield of 20 mg/L and purity > 95%. The effect of metal ions on the enzyme activity of BaLPMO10 was measured, and it was found that 10 mM Ca2+ and Na+ increased the enzyme activity by 47.8% and 98.0%, respectively. However, DTT, EDTA and five organic reagents inhibited the enzyme activity of BaLPMO10. Finally, BaLPMO10 was applied in biomass conversion. The degradation of corn stover pretreated with different steam explosions was performed. BaLPMO10 and cellulase had the best synergistic degradation effect on corn stover pretreated at 200 °C for 12 min, improving reducing sugars by 9.2% compared to cellulase alone. BaLPMO10 was found to be the most efficient for ethylenediamine-pretreated Caragana korshinskii by degrading three different biomasses, increasing the content of reducing sugars by 40.5% compared to cellulase alone following co-degradation with cellulase for 48 h. The results of scanning electron microscopy revealed that BaLPMO10 disrupted the structure of Caragana korshinskii, making its surface coarse and poriferous, which increased the accessibility of other enzymes and thus promoted the process of conversion. Molasses, a sugar industry waste product with low cost and high sugar content, can be used as a high-quality carbon source for microbial fermentation. In this study, molasses was pretreated by means of acid–base treatment combined with 0.6% mass fraction of activated carbon in order for its sugar concentration (78.42 g/L) and composition to meet the needs of Clostridium tyrobutyricum fermentation. C. tyrobutyricum was able to produce butyric acid from the sugars of treated molasses by microbial adaptive laboratory evolution. After 120 h of shake flask fermentation, high-performance liquid chromatography and gas chromatography showed that 46.39% of fructose and 32.56% of glucose in molasses after treatment were converted to 31.52 g/L butyric acid by C. tyrobutyricum. The results showed that the two-step process, combined with microbial adaptive laboratory evolution technology used to hydrolyze and reuse sugar, is an adapted method for the utilization of molasses, a by-product of the sugarcane industry. The objective of this work was to valorize a waste from cardinal vine shoot into a hydrolysate rich in reducing sugars. Plackett–Burman design was considered to identify the significant factors, while a Box Behnken design was considered to optimize the extraction in the following experimental conditions: 100 °C, 750 rpm, trifluoracetic acid (CF3O2H) concentration (TFA) in the range (1–10%), for 20 to 180 min and considering the following solid–liquid (S/V) ratios (1:1, 3:1, 5:1). The optimal result was 2.53% in sugars equivalent to a yield of 50.64% per gram of dry matter. Shoot vine waste was characterized by attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD), simultaneous thermal analysis (STA), and X-ray fluorescence (XRF). The chemical composition was 43.38% cellulose, 23.58% hemicellulose, 21.22% lignin, 2.53% ash, 5.82% crude protein, 11.7% moisture, and extractives (0.81% fat, 0.56% total sugars, 2.3% extractive (hexane-ethanol)). A This study aimed to employ three reducing sugar (fructose, galactose and glucose)-lysine mode reaction systems (FLMRS, GaLMRS and GLMRS) in examining the influences of reaction conditions, such as initial pH value, reaction temperature, and time on the Nε -(1-Carboxymethyl)-L-lysine (CML) formation. Results indicated that the initial pH value were positively correlated with the CML content in the system. However, the influence of reaction temperature and time on the CML content in the system displayed an initial increasing trend followed by a decline. A temperature of 100.58/125.24/123.24 ℃ and a heating time of 30.00/24.66/25.38 min maximized the formation of CML on FLMRS/GaLMRS/GLMRS and resulted in a CML production of 2.11/1.39/1.34 mmol/mol lysine of response surface methodology. Hydrothermal treatment (HT) is envisaged as a promising technology to treat the lignocellulosic biomass. HT temperature is an important parameter influencing the hydrolysate compositions such as organic compounds and potential inhibitors, and therefore affect the subsequential anaerobic digestion (AD) process. Herein, HT-AD was employed to treat the wheat straw-derived digestate. The HT temperature of 190 °C was proved to be the best performance with a higehst reducing sugar yield (45.05 mg g−1) in the hydrolysate and a highest methane yield (120.8 mL gTS−1) from the AD of the hydrolysate, which was 42.5% higher than the methane yield in the control without the hydrolysate addition (84.8 mL gTS−1). 3-Furaldehyde was the dominant organic in the hydrolysates. The HT temperature of 210 °C led to the presence of AD inhibitory moieties (e.g., phenols and furans) in the hydrolysate, resulting in a low methane yield. Although the treatments with the addition of 100% hydrolysate outperformed those of 50% hydrolysate in the methane yields in the late stage, the latter had higher methane yields in the first stage, suggesting that the additional ratios of hydrolysates should be carefully considered in AD, especially the detrimental effects of inhibitors and adaptability issues of AD consortia. The MiSeq sequencing showed that the hydrolysis/acidogenesis was dominant in the first stage, while methanogenesis became dominant in the late stage with the acetoclastic/hydrogenotrophic methanogens (Methanosarcina and Methanobacterium) enriched in the hydrolysate-feeding reactors. These findings demonstrated that a integration of HT-AD was a promising approach for the digestate valorization and to reduce the potential carbon emission from waste treatments. This study aims to obtain the results of the optimization of biological hydrolysis of oil palm empty fruit bunches (OPEFB) using Aspergillus niger (A. niger) InaCC F57. The optimized hydrolysis parameters include incubation temperature, substrate pH, incubation time, and additional parameters in the form of substrate concentration and the addition of nutrient salt solution (NSS) in alkaline pretreatment and non-pretreatment OPEFB. Pretreatment of OPEFB is alkaline generated from the delignification process with 10% NaOH at a temperature of 150°C and given a pressure of 5 kg/cm2 for 30 min in a chemical explosive reactor (CHIMEX), and then the NREL method is carried out where the OPEFB is chemically hydrolyzed using H2SO4 72% then followed by HPLC analysis and UV spectrophotometric analysis to determine the content of cellulose, hemicellulose, and lignin. These results indicate that the performance of A. niger InaCC F57 in the hydrolysis process is influenced by the pretreatment stage. Optimization of hydrolysis was carried out by inoculation of OPEFB samples using 10% A. niger isolate under sterile conditions. A. niger InaCC F57 isolate was obtained by regenerating cultures grown on sterile agar media at 28°C for 5 days and followed by inserting 10 mL of sterile distilled water into culture tubes and then scraping them off to separate them from the media. The optimum conditions for the enzymatic hydrolysis of OPEFB based on A. niger InaCC F5 were at a temperature of 40°C, pH 5, incubation time of 48 h (2 days), 10% substrate concentration, and the addition of nutrient salt solution (NSS). The alkaline pretreatment OPEFB hydrolysis yielded 1.40 mg/mL reducing sugar, 14.20% hydrolysis percent, and 0.56 U/mL cellulase activity, while non-pretreated OPEFB produced 1.90 mg/mL reducing sugar, 17.10% hydrolysis percent, and 0.76 U/mL cellulase activity. Based on the results obtained indicate the potential for biological hydrolysis between OPEFB and A. niger InaCC F57 as an important part of the OPEFB pretreatment in the future. The evolution of green technology for the simple and ecological formation of silver nanoparticles (AgNPs) inspired the present work for simple and efficient detection of reducing sugars (RS) in foods. The proposed method relies on gelatin as the capping and stabilizing agent and the analyte (RS) as the reducing agent. This work may attract significant attention, especially in the industry, for testing the sugar content using gelatin-capped silver nanoparticles as it not only detects the sugar in food, but also determines the content (%), which could be an alternative technique to the conventionally used DNS colorimetric method. For this purpose, a certain amount of maltose was mixed with a gelatin-silver nitrate. Different conditions that may affect the color changes at 434 nm owing to the in situ formed AgNPs, such as gelatin-silver nitrate ratio, PH, time, and temperature, have been investigated. The 1:3 mg/mg ratio of gelatin-silver nitrate dissolved in 10 mL distilled water was most effective in color formation. The development of AgNPs color increases within 8–10 min at PH 8.5 as the selected optimum value and at the optimum temperature of 90 °C for the evolution of the gelatin-silver reagent's redox reaction. The gelatin-silver reagent showed a fast response (less than 10 min) with a detection limit for maltose at 46.67 µM. In addition, the selectivity of maltose was checked in the presence of starch and after its hydrolysis with α-amylase. Compared with the conventionally used dinitrosalicylic acid (DNS) colorimetric method, the proposed method could be applied to commercial fresh apple juice, watermelon, and honey to prove its viability for detecting RS in fruits; the total reducing sugar content was 287, 165, and 751 mg/g, respectively. Cold-induced sweetening (CIS) is an unwanted physiological phenomenon in which reducing sugars (RS) get accumulated in potato (Solanum tuberosum) upon cold storage. High RS content makes potato commercially unsuitable for processing due to the unacceptable brown color in processed products like chips, fries, etc., and the production of a potential carcinogen, acrylamide. UDP-glucose pyrophosphorylase (UGPase) catalyzes the synthesis of UDP-glucose towards the synthesis of sucrose and is also involved in the regulation of CIS in potato. The objective of the present work was RNAi-mediated downregulation of the StUGPase expression level in potato for the development of CIS tolerant potato. Hairpin RNA (hpRNA) gene construct was developed by placing UGPase cDNA fragment in sense and antisense orientation intervened by GBSS intron. Internodal stem explants (cv. Kufri Chipsona-4) were transformed with hpRNA gene construct, and 22 transgenic lines were obtained by PCR screening of putative transformants. Four transgenic lines showed the highest level of RS content reduction following 30 days of cold storage, with reductions in sucrose and RS (glucose & fructose) levels of up to 46% and 57.5%, respectively. Cold stored transgenic potato of these four lines produced acceptable chip colour upon processing. The selected transgenic lines carried two to five copies of the transgene. Northern hybridization revealed an accumulation of siRNA with a concomitant decrease in the StUGPase transcript level in these selected transgenic lines. The present work demonstrates the efficacy of StUGPase silencing in controlling CIS in potato, and the strategy can be employed for the development of CIS tolerant potato varieties. The selection of suitable pretreatment strategy that facilitates saccharification is the most critical step in the valorisation of seaweed biomass into bioethanol. Considering the scarcity of data on the comparative efficacy of different pretreatments particularly in tropical seaweed species, 50 pretreatment strategies were analysed on three major species representing all the three broad seaweed categories, viz. Ulva fasciata (green algae) , Sargassum wightii (brown algae) , and Gracilaria corticata (red algae). An improved release of reducing sugars was observed in all the tested pretreatments, with different conversion ratios for different species in each pretreatment. Ultrasonication and magnetic stirring showed maximum yield among physical pretreatments. In thermal methods, microwave oven incubation resulted in maximum yield from both U. fasciata and S. wightii , while it was autoclaving for G. corticata. Sulphuric acid was the best chemical for U. fasciata and G. corticata, while it was orthophosphoric acid for S. wightii. Hydrochloric acid was the best chemical for physicothermochemical pretreatment in all species. Maximum conversion ratios of 2.58, 2.11, and 2.0 were recorded in U. fasciata , G. corticata, and S. wightii respectively. In short, the paper sketches the best pretreatment method for three major tropical seaweeds along with their proximate composition. The results can be applied for aiding the valorisation of biomass of these seaweeds through a cost-effective manner. The effects of hydrolysis methods (hydrothermal, acid, alkali, hydrothermal-enzyme, acid-enzyme, and alkali-enzyme) on hydrolysate characteristics and photo fermentative hydrogen production (PFHP) of corn straw (CS) and sorghum straw (SS) were investigated. The optimum production of reducing the sugar of straw in different solvent environments was studied by one-step hydrolysis and co-enzymatic hydrolysis pretreatment through a 3,5-dinitrosalicylic acid method. The hydrogen production process by photolytic fermentation of hydrolysates of Rhodobacter sphaeroides HY01 was further analyzed through a gas chromatograph, including the differences in accumulated PFHP yield, chemical oxygen consumption (COD), and volatile fatty acid (VFA) composition. The results showed that the highest reducing sugar yield was obtained by the acid method among one-step hydrolysis. In contrast, acid-enzyme hydrolysis can further increase the reducing sugar yield, which reached 0.42 g·g−1-straw of both straws. Both CS and SS had the highest hydrogen yield from acid-enzyme hydrolysate, 122.72 ± 3.34 mL·g−1-total solid of straw (TS) and 170.04 ± 4.12 mL·g−1-TS, respectively, compared with their acid hydrolysates with 40.46% and 10.53% higher hydrogen yields, respectively. The use of enzymatic hydrolysis showed a significantly higher hydrogen yield for CS compared to SS, indicating that acid hydrolysis was more suitable for SS and acid-enzyme hydrolysis was more suitable for CS. This study evaluated the effects of sodium alginate, pectin and chitosan addition (0.5–1.5%) on the physicochemical properties including pH, water activity, moisture content, color values, hardness, diameter, thickness, spread ratio, antioxidant activities and sensory scores of biscuits in air frying processing. In addition, the formation of acrylamide and hydroxymethylfurfural (HMF) were discussed. Physicochemical properties of biscuits including water content, water activity, hardness, appearance, shape, color, flavor, texture, overall acceptability, and DPPH radical scavenging activity of biscuits were not influenced significantly by the addition (0.5–1.0%) of three food hydrocolloids. The data showed that the biscuits with hydrocolloids addition had lower acrylamide contents than that of the control biscuit without hydrocolloids addition, and the reducing power of biscuits increased after adding the hydrocolloids. The highest mitigation of acrylamide formation was obtained by the chitosan addition formulation. The formation of acrylamide showed a negative correlation with the content of sodium alginate and chitosan addition, and they were effective ingredients in terms of mitigating the formation of acrylamide in biscuit formulation.
