Most of advanced hypopharyngeal squamous cell carcinoma (HSCC) are resistant to chemotherapy, and there is still lack of effective treatment for HSCC now. The present study aimed to investigate whether downregulation of RNA-binding motif protein 17 (RBM17) could enhance cisplatin sensitivity and inhibit cell invasion in HSCC and the underlying mechanism. We observed that RBM17 was upregulated in tumor tissues and associated with poor progression. Treatment of FaDu cells with cisplatin increased RBM17 expression in mRNA levels. Downregulation of RBM17 enhanced cisplatin-mediated inhibition of FaDu cells. In addition, downregulation of RBM17 effectively suppressed tumor cell migration and invasion through the reversion of epithelial–mesenchymal transition. Moreover, downregulation of RBM17 could significantly slow tumor growth in FaDu xenograft tumor model. Liquid chromatography–mass spectrometry/mass spectrometry detection and independent PRM analysis showed that 21 differentially expressed proteins were associated with the downregulation of RBM17. Taken together, our study implied that downregulation of RBM17 could serve as a novel approach to enhance cisplatin sensitivity in HSCC.

Objective: The objective of this retrospective cohort study is to investigate the impact of monitoring serum estradiol (E2) levels before progesterone administration within hormone replacement therapy (HRT) on pregnancy outcomes in women undergoing frozen-thawed embryo transfer (FET). Methods: Analyzed HRT-FET cycles conducted at a reproductive center from 2017 to 2022. Serum E2 levels were measured prior to progesterone administration. Multivariate stratified and logistic regression analyses were performed on 26,194 patients grouped according to terciles of serum E2 levels before progesterone administration. Results: The clinical pregnancy rate (CPR) and live birth rate (LBR) exhibited a gradual decline with increasing serum E2 levels across the three E2 groups. Even after controlling for potential confounders, including female age, body mass index, infertility diagnosis, cycle category, number of embryos transferred, fertilization method, indication for infertility, and endometrial thickness, both CPR and LBR persistently showed a gradual decrease as serum E2 levels increased within the three E2 groups. The same results were obtained by multivariate logistic regression analysis. Conclusions: This large retrospective study indicates that elevated serum E2 levels before progesterone administration during HRT-FET cycles are associated with reduced CPR and LBR post-embryo transfer. Natural killer (NK) cell-based therapies have made great progress in treating both hematological and solid tumors. Their unique mechanism of action does not rely on antigen presentation to recognize and eliminate tumor cells, making them a promising approach for cancer immunotherapy. Objectives: Ants are ecologically dominant insects in most terrestrial ecosystems, with more than 14,000 extant species in about 340 genera recorded to date. However, genomic resources are still scarce for most species, especially for species endemic in East or Southeast Asia, limiting the study of phylogeny, speciation and adaptation of this evolutionarily successful animal lineage. Here, we assemble and annotate the genomes of Odontoponera transversa and Camponotus friedae, two ant species with a natural distribution in China, to facilitate future study of ant evolution. Data description: We obtained a total of 16 Gb and 51 Gb PacBio HiFi data for O. transversa and C. friedae, respectively, which were assembled into the draft genomes of 339 Mb for O. transversa and 233 Mb for C. friedae. Genome assessments by multiple metrics showed good completeness and high accuracy of the two assemblies. Gene annotations assisted by RNA-seq data yielded a comparable number of protein-coding genes in the two genomes (10,892 for O. transversa and 11,296 for C. friedae), while repeat annotations revealed a remarkable difference of repeat content between these two ant species (149.4 Mb for O. transversa versus 49.7 Mb for C. friedae). Simple Summary: Recently, the escalating prices of feedstuff resources have posed significant challenges to animal husbandry, necessitating the creation of innovative approaches to mitigate its effects. GAA (also known as N-amidinoglycine) serves as the sole precursor for creatine synthesis. GAA could enhance growth performance and regulate fat deposition to improve meat quality. Current studies on GAA mainly center on pigs and chickens, with limited reports on its effects on sheep. In our study, we used weaned Hu sheep as subjects. We aimed to explore the impact of dietary GAA supplementation on production performance, serum biochemistry, meat quality and rumen fermentation. This study provides a theoretical foundation for utilizing GAA in sheep meat production. Our findings indicate that GAA enhances growth performance, rumen fermentation and fat accumulation. In addition, GAA might enhance the quality of sheep meat, which presents a promising prospect for meat sheep breeding. Guanidinoacetic acid (GAA) can effectively improve the metabolism of energy and proteins by stimulating creatine biosynthesis. We present a study exploring the impact of GAA on production performance, serum biochemistry, meat quality and rumen fermentation in Hu sheep. A total of 144 weaned male Hu sheep (body weight 16.91 ± 3.1 kg) were randomly assigned to four groups with three replicates of twelve sheep in each group. The diets were supplemented with 0 (CON), 500 (GAA−1), 750 (GAA−2) and 1000 mg/kg (GAA−3) of GAA (weight of feed), respectively. After a comprehensive 90-day experimental period, we discovered that the supplementation of GAA had a remarkable impact on various muscle parameters. Specifically, it significantly enhanced the average daily growth (ADG) of the animals and improved the shear force and fiber diameter of the muscle, while also reducing the drip loss and muscle fiber density. Furthermore, the addition of GAA to the feed notably elevated the serum concentrations of high-density lipoprotein cholesterol (HDL−C), total protein (TP) and globulin (GLB), as well as the enzyme activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH−Px). Concurrently, there was a decrease in the levels of triglycerides (TG) and malondialdehyde (MDA) in the serum. In addition, GAA decreased the pH and the acetate-to-propionate ratio and increased the total volatile fatty acids (TVFA) and ammoniacal nitrogen (NH3−N) levels of rumen fluid. Additionally, GAA upregulated acetyl-CoA carboxylase (ACC) gene expression in the Hu sheep's muscles. In conclusion, our findings suggest that GAA supplementation not only enhances muscle quality but also positively affects serum biochemistry and ruminal metabolism, making it a potential candidate for improving the overall health and performance of Hu sheep. Background: The aberrant expression of phosphofructokinase-platelet (PFKP) plays a crucial role in the development of various human cancers by modifying diverse biological functions. However, the precise molecular mechanisms underlying the role of PFKP in head and neck squamous cell carcinoma (HNSCC) are not fully elucidated. Methods: We assessed the expression levels of PFKP and c-Myc in tumor and adjacent normal tissues from 120 HNSCC patients. A series of in vitro and in vivo experiments were performed to explore the impact of the feedback loop between PFKP and c-Myc on HNSCC progression. Additionally, we explored the therapeutic effects of targeting PFKP and c-Myc in HNSCC using Patient-Derived Organoids (PDO), Cell Line-Derived Xenografts, and Patients-Derived Xenografts. Results: Our findings indicated that PFKP is frequently upregulated in HNSCC tissues and cell lines, correlating with poor prognosis. Our in vitro and in vivo experiments demonstrate that elevated PFKP facilitates cell proliferation, angiogenesis, and metastasis in HNSCC. Mechanistically, PFKP increases the ERK-mediated stability of c-Myc, thereby driving progression of HNSCC. Moreover, c-Myc stimulates PFKP expression at the transcriptional level, thus forming a positive feedback loop between PFKP and c-Myc. Additionally, our multiple models demonstrate that co-targeting PFKP and c-Myc triggers synergistic anti-tumor effects in HNSCC. Conclusion: Our study demonstrates the critical role of the PFKP/c-Myc positive feedback loop in driving HNSCC progression and suggests that simultaneously targeting PFKP and c-Myc may be a novel and effective therapeutic strategy for HNSCC. An energy-saving control strategy for wheel loaders is proposed in this paper to address the issue of high energy consumption during their operation. The strategy is based on the intelligent identification of working stages, allowing for staged power matching and resulting in reduced energy consumption. Each work stage of the loader is identified by matching it to the main pump pressure waveform and actuator pilot pressure waveform. Using a sliding time window method, pressure waveforms from each working stage are subjected to feature extraction. A bidirectional long short-term memory neural network (BILSTM) algorithm is then used to establish an intelligent recognition model. Based on work stage identification, an energy-saving control strategy based on power matching is proposed for the shoveling stage of the loader, and the Grey Wolf optimization (GWO)-PID algorithm is utilized for control parameter tuning. Finally, the effectiveness of the energy-saving control strategy based on work stage identification is verified through experiments. The research results indicate that the BILSTM recognition model outperforms other models with a recognition accuracy of 96.1%. The optimal time window width is 0.6 s, and the proposed energy-saving control strategy achieves a fuel-saving rate of 6.81%. Purpose: The study aims to analyze how the characteristics of intellectual capital (IC) facilitate green entrepreneurship development in the context of ecology, environment and sustainability. Specifically, the evolution of IC and green entrepreneurship was explored through a systematic review, including the relationships and interactions between human, structural and relational capital and green entrepreneurship. Design/methodology/approach: Meticulously combing the Web of Science Core Collection, the researcher conducted a bibliometric analysis of 800 English-language articles from 2002 to 2023. Employing co-word analysis and visualization, the literature on IC and green entrepreneurship was synthesized and systematized, exploring core topics, knowledge architectures and their evolutionary trajectories. Findings: The IC elements such as human, structural and relational capital interact with green entrepreneurship; IC enhances the innovation and competitiveness of green entrepreneurship, while green entrepreneurship orientation influences the accumulation and reshaping of IC. The flow of IC impacts the establishment of green start-ups and the emergence of green industries, promoting sustainable growth. Originality/value: The dynamic interplay between IC and green entrepreneurship is marked by intricate relationships and diverse attributes. Currently, no comprehensive theoretical model has been established to address the complexities intrinsic to this study. The evidence suggests that the green entrepreneurial orientation influences corporate initiatives to bolster human and structural capital, with structural capital serving as both a constraint and catalyst for human capital. The paper presents an embryonic framework of IC for green entrepreneurship, highlighting its critical role in the aggregation and reconfiguration of IC or venture creation and industry evolution. Modulation of light underpins a central part of modern optoelectronics. Conventional optical modulators based on refractive‐index and absorption variation in the presence of an electric field serve as the workhorse for diverse photonic technologies. However, these approaches based on electro‐refraction or electro‐absorption effect impose limitations on frequency converting and signal amplification. Lanthanide‐activated phosphors offer a promising platform for nonlinear frequency conversion with an abundant spectrum. Here, we propose a novel approach to achieve frequency conversion and digital modulation of light signal by coupling lanthanide luminescence with an electrically responsive ferroelectric host. The technological benefits of such paradigm‐shifting solution are highlighted by demonstrating a quasi‐continuous and enhancement of the lanthanide luminescence. The ability to locally manipulate light emission can convert digital information signals into visible waveforms, and visualize electrical logic and arithmetic operations. The proof‐of‐concept device exhibits perspectives for developing light‐compatible logic functions. Photothermal therapy (PTT) is a promising cancer treatment method due to its ability to induce tumor-specific T cell responses and enhance therapeutic outcomes. However, incomplete PTT can leave residual tumors that often lead to new metastases and decreased patient survival in clinical scenarios. This is primarily due to the release of ATP, a damage-associated molecular pattern that quickly transforms into the immunosuppressive metabolite adenosine by CD39, prevalent in the tumor microenvironment, thus promoting tumor immune evasion. This study presents a photothermal nanomedicine fabricated by electrostatic adsorption among the Fe-doped polydiaminopyridine (Fe-PDAP), indocyanine green (ICG), and CD39 inhibitor sodium polyoxotungstate (POM-1). The constructed Fe-PDAP@ICG@POM-1 (FIP) can induce tumor PTT and immunogenic cell death when exposed to a near-infrared laser. Significantly, it can inhibit the ATP-adenosine pathway by dual-directional immunometabolic regulation, resulting in increased ATP levels and decreased adenosine synthesis, which ultimately reverses the immunosuppressive microenvironment and increases the susceptibility of immune checkpoint blockade (aPD-1) therapy. With the aid of aPD-1, the dual-directional immunometabolic regulation strategy mediated by FIP can effectively suppress/eradicate primary and distant tumors and evoke long-term solid immunological memory. Li‐O2 batteries, known for their impressive theoretical specific energy, face significant practical challenges, including slow kinetics, voltage hysteresis, and reduced cycle life. In this study, a novel approach is introduced utilizing a bimetallic metal‐organic framework known as Fe‐UiO‐66 as a multifunctional photocatalyst to enhance the oxygen‐related reactions in photo‐assisted Li‐O2 batteries. In contrast to UiO‐66, Fe‐UiO‐66 exhibits an extended response to visible light, as confirmed through UV–vis and photoluminescence spectra. Electron paramagnetic resonance analysis provides further validation of the pivotal role played by O2− radicals in the electrochemical processes. Additionally, computations are employed to elucidate the distinct function of Fe centers and the underlying reaction mechanisms. Li‐O2 batteries with Fe‐UiO‐66 cathodes have a reduced charge/discharge voltage difference of 0.14 V under illumination and operate stably for 500 cycles. A formidable challenge in the multi-label text classification (MLTC) context is that the labels often exhibit a long-tailed distribution, which typically prevents deep MLTC models from obtaining satisfactory performance. To alleviate this problem, most existing solutions attempt to improve tail performance by means of sampling or introducing extra knowledge. Data-rich labels, though more trustworthy, have not received the attention they deserve. In this work, we propose a multiple-stage training framework to exploit both model- and feature-level knowledge from the head labels, to improve both the representation and generalization ability of MLTC models. Moreover, we theoretically prove the superiority of our framework design over other alternatives. Comprehensive experiments on widely used MLTC datasets clearly demonstrate that the proposed framework achieves highly superior results to state-of-the-art methods, highlighting the value of head labels in MLTC. The concept of sustainable development has gained significant traction in recent years, leading to a growing emphasis on green consumption. Previous studies have rarely emphasized the important role of sustainability awareness in influencing green consumption. This study explores the mechanism of the influence of sustainability awareness on green purchase intention, using the theory of planned behavior as a conceptual framework. The aim is to contribute to the promotion of environmental protection and green consumption. The research used an online five-point Likert scale questionnaire and collected data from university students in China. Useable data were collected from 419 respondents and were then validated and analyzed using structural equation modeling (SEM) and the PROCESS macro for SPSS 22.0. The findings suggest that attitudes toward green purchases, subjective norms, and perceived behavioral control have a positive effect on green purchase intention; sustainability awareness significantly and positively moderates the direct effect of attitudes toward green purchases on green purchase intention and the predictive effect of subjective norms on green purchase intention; and sustainability awareness significantly and negatively moderates the direct effect of perceived behavioral control on green purchase intention. In silicon photonics, BaTiO3 (BTO) emerges as a promising electro‐optic (EO) integrated material due to its excellent EO modulation capabilities. Nevertheless, the EO response of multi‐domain BTO thin films tends to be lower than that of bulk materials. BTO thin films are fabricated with strong EO responses utilizing the pulsed laser deposition technique. Considering the importance of the SrTiO3 (STO) buffer layer, a series of epitaxial BTO thin films are deposited on STO substrates with engineered domain alignment. Given the contribution of domain variants in BTO thin films to their EO response, specific electrode patterns are designed to enhance their performance. An effective Pockels coefficient of 286 pm V−1 is determined for the BTO thin film via the transmission geometry method, which surpasses recent reports. Through domain engineering in the BTO thin film, the remarkable enhancement in the EO response arising from in‐plane polarization (a‐axis) characteristics is confirmed. Simultaneously, utilizing the Second Harmonic Generation (SHG) optical probe, deep exploration into the domain dynamics within BTO thin films is conducted. These findings provide insights into the contribution of domain variants to the EO response and shed light on further developing silicon‐based heterogeneously integrated devices. Versatile all-in-one nanoplatforms, which combine a variety of therapeutic and diagnostic capabilities, possess interesting physiochemical and biological characteristics, making them highly attractive in cancer-related biomedical applications. However, up to now, the precise design of nanocomposites with multiple functions has remained an ongoing challenge. Herein, this study constructed a tumor microenvironment (TME)-responsive nanocomposite therapeutic platform with a core–shell structure using bismuth (Bi) as the core and iron-porphyrin-containing covalent–organic polymers (COPs) as the shell. Under the irradiation of an 808 nm laser, Bi efficiently inhibits tumor cell growth through photothermal therapy (PTT), and porphyrin-based COPs can produce reactive oxygen species (ROS) by photodynamic therapy (PDT) under the irradiation of a 660 nm laser combined with chemodynamic therapy (CDT). Meanwhile, Bi as a green metal with good biocompatibility also has the potential for use in X-ray computed tomography (CT) imaging and near-infrared (NIR) thermal imaging. Therefore, this nanocomposite integrated multifunctional treatment pathways (PTT, PDT, and CDT) and multimodel (CT and NIR) imaging to cope with the complexity and diversity of tumor cells, effectively improving therapeutic efficacy and becoming a potential candidate for tumor diagnosis and treatment. Skeletal muscle myogenesis hinges on gene regulation, meticulously orchestrated by molecular mechanisms. While the roles of transcription factors and non-coding RNAs in myogenesis are widely known, the contribution of RNA-binding proteins (RBPs) has remained unclear until now. Therefore, to investigate the functions of post-transcriptional regulators in myogenesis and uncover new functional RBPs regulating myogenesis, we employed CRISPR high-throughput RBP-KO (RBP-wide knockout) library screening. Through this approach, we successfully identified Eef1a1 as a novel regulatory factor in myogenesis. Using CRISPR knockout (CRISPRko) and CRISPR interference (CRISPRi) technologies, we successfully established cellular models for both CRISPRko and CRISPRi. Our findings demonstrated that Eef1a1 plays a crucial role in promoting proliferation in C2C12 myoblasts. Through siRNA inhibition and overexpression methods, we further elucidated the involvement of Eef1a1 in promoting proliferation and suppressing differentiation processes. RIP (RNA immunoprecipitation), miRNA pull-down, and Dual-luciferase reporter assays confirmed that miR-133a-3p targets Eef1a1. Co-transfection experiments indicated that miR-133a-3p can rescue the effect of Eef1a1 on C2C12 myoblasts. In summary, our study utilized CRISPR library high-throughput screening to unveil a novel RBP, Eef1a1, involved in regulating myogenesis. Eef1a1 promotes the proliferation of myoblasts while inhibiting the differentiation process. Additionally, it acts as an antagonist to miR-133a-3p, thus modulating the process of myogenesis. Objective: The objective of this scoping review was to investigate the effectiveness and limitations of risk prediction models for postpartum glucose intolerance in women with gestational diabetes mellitus (GDM). The aim was to provide valuable insights for healthcare professionals in the development of robust risk prediction models. Methods: A comprehensive literature search was conducted across multiple databases, including PubMed, EBSCO, Web of Science Core Collection, Ovid Full-Text Medical Journal Database, ProQuest, Elsevier ClinicalKey, China National Knowledge Infrastructure, China Biology Medicine, and WanFang Database, spanning from January 1990 to July 2023. To assess the quality of the included models, the Predictive Model Risk of Bias Assessment Tool (PROBAST) was employed. Results: Fourteen relevant studies were identified and included in the final review, all focusing on model development. The discrimination ability of the included models ranged from 0.725 to 0.940, indicating satisfactory prediction accuracy. However, a notable limitation was that nine of these models (64.3%) did not provide clear guidelines on the selection of potential predictors. Furthermore, only six models (42.86%) underwent internal validation, with none undergoing external validation. A high risk of bias was observed across the included models. Logistic regression, Cox regression, and machine learning were the primary methods employed in the construction of these models. Conclusion: The risk prediction models included in this review demonstrated favorable prediction accuracy. However, due to variations in construction methodologies, direct comparison of their performance is challenging. These models exhibited certain shortcomings, such as inadequate handling of missing data and a lack of internal and external validation, resulting in a high risk of bias. Therefore, it is recommended that these models be updated and externally validated. The development of prospective, multi-center studies is encouraged to construct predictive models with low risk of bias and high clinical applicability, ultimately guiding evidence-based clinical practice. STUDY QUESTION Could actin-related protein T1 (ACTRT1) deficiency be a potential pathogenic factor of human male infertility? SUMMARY ANSWER A 110-kb microdeletion of the X chromosome, only including the ACTRT1 gene, was identified as responsible for infertility in two Chinese males with sperm showing acrosomal ultrastructural defects and fertilization failure. WHAT IS KNOWN ALREADY The actin-related proteins (e.g. ACTRT1, ACTRT2, ACTL7A, and ACTL9) interact with each other to form a multimeric complex in the subacrosomal region of spermatids, which is crucial for the acrosome-nucleus junction. Actrt1 -knockout (KO) mice are severely subfertile owing to malformed sperm heads with detached acrosomes and partial fertilization failure. There are currently no reports on the association between ACTRT1 deletion and male infertility in humans. STUDY DESIGN, SIZE, DURATION We recruited a cohort of 120 infertile males with sperm head deformations at a large tertiary hospital from August 2019 to August 2023. Genomic DNA extracted from the affected individuals underwent whole exome sequencing (WES), and in silico analyses were performed to identify genetic variants. Morphological analysis, functional assays, and ART were performed in 2022 and 2023. PARTICIPANTS/MATERIALS, SETTING, METHODS The ACTRT1 deficiency was identified by WES and confirmed by whole genome sequencing, PCR, and quantitative PCR. Genomic DNA of all family members was collected to define the hereditary mode. Papanicolaou staining and electronic microscopy were performed to reveal sperm morphological changes. Western blotting and immunostaining were performed to explore the pathological mechanism of ACTRT1 deficiency. ICSI combined with artificial oocyte activation (AOA) was applied for one proband. MAIN RESULTS AND THE ROLE OF CHANCE We identified a whole-gene deletion variant of ACTRT1 in two infertile males, which was inherited from their mothers, respectively. The probands exhibited sperm head deformations owing to acrosomal detachment, which is consistent with our previous observations on Actrt1 -KO mice. Decreased expression and ectopic distribution of ACTL7A and phospholipase C zeta were observed in sperm samples from the probands. ICSI combined with AOA effectively solved the fertilization problem in Actrt1 -KO mice and in one of the two probands. LIMITATIONS, REASONS FOR CAUTION Additional cases are needed to further confirm the genetic contribution of ACTRT1 variants to male infertility. WIDER IMPLICATIONS OF THE FINDINGS Our results reveal a gene–disease relation between the ACTRT1 deletion described here and human male infertility owing to acrosomal detachment and fertilization failure. This report also describes a good reproductive outcome of ART with ICSI-AOA for a proband.
