Assessing the limitations of microbial metabolic resources is crucial for understanding plantation soil quality and enhancing fertility management. However, the variation of microbial resource limitations at the aggregate level in response to changes in stands remains unclear. This research explores carbon (C), nitrogen (N), and phosphorus (P) limitations affecting microbial metabolism in bulk soils and aggregates in two mixed and one pure Chinese fir stands in subtropical China, analyzing resource limitations concerning soil carbon, nutrients, and microbial indicators. The results revealed that microbes in all aggregates of the pure stands and in the micro aggregates (<0.25 mm) of the three stands were relatively limited by C and P. In contrast, microbial metabolism was more N-limited in macroaggregates (>2 mm) and small aggregates (2–0.25 mm) in the mixed stands. Additionally, in the mixed stands the proportion of soil macroaggregates increased, and that of micro aggregates decreased, resulting in a shift from C and P limitation to N limitation for bulk soil microbial metabolism. Redundancy analysis identified soil aggregate organic carbon and nutrient content as the main factors affecting microbial resource limitation, rather than their stoichiometric ratios. Pathway analysis further confirmed that soil nutrients and their stoichiometric ratios indirectly influenced soil microbe resource limitation by regulating microbial biomass, microbial respiration, and extracellular enzyme activities. Thus, the impact of mixed plantations on soil nutrients and microbial activity at the aggregate level may be crucial for maintaining land fertility and achieving sustainability. Procambarus clarkii is the most widely distributed freshwater shrimp in China, with important economic value and great potential for development. The forkheadboxL2 (Foxl2) gene has been found to be involved in the reproductive development of many crustaceans. To understand the role of the Foxl2 gene in the gonad development of P. clarkii, we designed CDS-specific primers for the P. clarkii Foxl2 (PcFoxl2) gene and cloned its CDS sequence using RT-PCR. The nucleotide and protein sequence information was then analyzed through bioinformatics analysis. The expression and subcellular localization of PcFoxl2 in various tissues were detected using qRT-PCR and in situ hybridization. The effects of PcFoxl2 knockdown on gonad development were investigated using RNA interference. The results showed that the CDS length of the PcFoxl2 gene was 1614 bp and encoded 537 amino acids. Protein sequence comparison and phylogenetic analysis showed that PcFoxl2 was the closest relative to Crayfish. qRT-PCR analysis indicated that the expression level of PcFoxl2 in the testis was significantly higher (>40 fold) than that in the ovary (p < 0.01). The in situ hybridization results showed that PcFoxl2 was expressed in both the cytoplasm and the nucleus of egg cells, and that the expression was strongest in egg cells at the early stage of yolk synthesis, while weak in the secondary oocytes. The positive signal was strongest in the spermatocyte nucleolus, while only a trace signal was observed in the cytoplasm. After interfering with the PcFoxl2 gene using dsRNA, the expression of PcFoxl2 in the RNA interference group was significantly lower than that in the control group, and this interference effect lasted for one week. Moreover, the gonad index of the experimental group was significantly lower than that of the control group (p < 0.05) after 10 days of P. clarkii cultivation following PcFoxl2 knockdown. The expression levels of the nanos and S3a genes, which are related to gonad development, decreased significantly after PcFoxl2 gene interference. The results suggest that the Foxl2 gene is involved in the growth and development of gonads, particularly in the development of testis, and is related to the early development of oocytes. Low-rank coal accounts for over half of China's proven coal reserves. The possibility of coal and gas outbursts in low-rank coal is higher, especially in the m·agmatic thermal evolution area. The complexity of coal's molecular structure is one of the reasons for problems during the process of mining operations. Different analysis techniques, including XRD, FTIR and Raman spectroscopy, were used to obtain the molecular characteristics of magmatic thermal evolution coal samples and normal coal samples, so that a comparative study could be carried out to investigate the influence of the magmatic thermal evolution effect on the molecular structure of low-rank coal. The ranges of the aromatic interlayer spacing (d002), average stacking heights (Lc) and stacking layer number (Nave) of the thermally evolved coal samples are 3.41–3.51 Å, 22.76–27.02 Å, and 6.68–7.70, respectively. The ranges of the full width at half maximum ratio (FD1/FG) and the peak integral intensity ratio (ID1/IG) are 2.16–2.19 and 1.55–1.84, respectively. Compared with the normal coal samples, those affected by magmatic thermal evolution have smaller d002, ID1/IG, and FD1/FG values, but larger Lc values. The results indicate that the thermally evolved coal samples have more ordered structures and more developed microcrystalline structure sizes than normal coal samples. [ABSTRACT FROM AUTHOR]

Simple Summary: Sika deer is a rare livestock resource in China, and the main purpose of breeding is to obtain antlers that can be used as medicinal herbs. As an important limiting amino acid for ruminants, the dietary level of methionine greatly affects the utilization efficiency of protein in feed. In this study, we investigated the effects of different dietary methionine levels on antler-bearing sika deer by supplementing methionine. The results of the study showed that methionine supplementation improved the quality of Sika deer antlers and increased the efficiency of the animals utilizing acid detergent fiber and neutral detergent fiber in the feed. In addition, methionine supplementation altered the composition of free amino acids in the rumen fluid and the composition of fecal bacteria in sika deer. Amino acid balance is central to improving the efficiency of feed protein utilization and for reducing environmental pollution caused by intensive farming. In previous studies, supplementation with limiting amino acids has been shown to be an effective means of improving animal nutrient utilization and performance. In this experiment, the effects of methionine on the apparent digestibility of nutrients, antler nutrient composition, rumen fluid amino acid composition, fecal volatile fatty acids and intestinal bacteria in antler-growing sika deer were investigated by randomly adding different levels of methionine to the diets of three groups of four deer at 0 g/day (CON), 4 g/day (LMet) and 6 g/day (HMet). Methionine supplementation significantly increased the apparent digestibility of organic matter, neutral detergent fiber (NDF) and acid detergent fiber (ADF) in the LMet group (p < 0.05). The crude protein and collagen protein of antlers were significantly higher in the LMet and HMet groups compared to the CON group and also significantly higher in the HMet group compared to the LMet group, while the calcium content of antlers was significantly lower in the HMet group (p < 0.05). Ruminal fluid free amino acid composition was altered in the three groups of sika deer, with significant changes in aspartic acid, citrulline, valine, cysteine, methionine, histidine and proline. At the phylum level, Firmicutes and Bacteroidetes were highest in the rectal microflora. Unidentified bacterial abundance was significantly decreased in the HMet group compared to the CON group. Based on the results of principal coordinate analysis (PCoA) and Adonis analysis, there was a significant difference in the composition of the intestinal flora between the CON and HMet groups (p < 0.05). At the genus level, compared with the CON group, the abundance of Rikenellaceae_RC9_gut_group and Lachnospiraceae_UCG-010 in the LMet group increased significantly (p < 0.05), the abundance of dgA-11_gut_group in the HMet group decreased significantly (p < 0.05) and the abundance of Lachnospiraceae_UCG-010, Saccharofermentans and Lachnospiraceae_NK3A20_group increased significantly. Taken together, the results showed that methionine supplementation was beneficial in increasing the feed utilization efficiency and improving antler quality in sika deer, while affecting the composition of fecal bacteria. Simple Summary: Enrofloxacin (EF) is an important antibiotic in global aquaculture. Due to the abuse and the emergence of drug resistance, the excess use of EF has occurred in recent years to result in residue exceeding in the edible tissues of aquatic animals. Withdrawal time (WT) is a red line to ensure food safety for humans. However, there is no related report of WT for EF in yellow catfish (Pelteobagrus fulvidraco). In addition to yellow catfish being sensitive to temperature, it can alter the disposition and elimination of EF and its main metabolite in fish bodies, ciprofloxacin (CF), thereby changing the WT in tissues. Therefore, we will conduct a WT estimation of EF in yellow catfish at different temperatures to establish related regulations to help residue surveillance in fish products. The objective of the present study was to investigate the residue depletion of EF and CF in yellow catfish to estimate its WTs in plasma and tissues after multiple oral doses for 3 days at 20 mg/kg at 15, 20, and 25 °C. Samples were collected at pre-designed time points after oral doses. A validated method was performed to quantify EF and CF in plasma and tissues by high-performance liquid chromatography. Statistical differences were conducted using one-way ANOVA analysis. According to the maximum residue limit of China and Europe considering 95% percentile with 95% confidence, the WTs were estimated to be 44, 72, 66, 99, and 95 days at 15 °C; 32, 66, 65, 86, and 73 days at 20 °C; and 32, 61, 64, 55, and 59 days at 25 °C in the plasma, muscle and skin, gill, liver, and kidney, respectively. We found that increased temperature shortened the WTs in plasma and tissues. This study provides a comprehensive assessment of the potential ecological and health risks in the area of the antimony mine spill in Longnan, Northwest China, and identifies the sources of potentially toxic elements (PTEs) in the soil as a result of the spill. The geo-accumulation index and enrichment factor show that the study area is highly contaminated with arsenic (As), mercury (Hg) and antimony (Sb). The ecological risk index in the tailings spill area ranged from 320.43 to 5820.46 (mean: 1489.82), indicating a very-high potential ecological risk, with mean values of 104.86, 1118.87 and 248.84 for As, Hg and Sb, respectively. The multivariate statistical analysis suggested that Sb and Hg come from tailings leakage, while copper (Cu), nickel (Ni) and zinc (Zn) may be imported from natural sources, and As and lead (Pb) originate from agricultural activities. In addition As and Sb pose a high health risk. With the exception of the non-carcinogenic risk in adults, all other risks are significantly exceeded in other populations, with children being the highest-risk group. Ma bamboo (Dendrocalamus latiflorus Munro) is a major bamboo species cultivated in southern China with high economic, ecological, and social value. However, highly dense forests and reasonable structures in bamboo forests have unclear and adverse effects on light transmittance and forest productivity that are not adequately understood. Here, we investigated varied light-intensity treatments during different phases of shoot emergence and development on Ma bamboo shoots. The amount of total chlorophyll, carotenoids, gas exchange indicators, and biosynthetic products were also compared to explore the response mechanism of shoot germination on downstream biochemical pathways. We found that compared to the L0 treatment (full sunlight), the number of germinated bamboo shoots under the L1 treatment (40% light) increased significantly by 44.07% and 101.32% in the shooting initial-phase and metaphase, respectively (p < 0.05). Additionally, the net photosynthetic rate (Pn) during the shooting initial-phase and metaphase was the highest in the L1 treatment, while the L4 (10% light) and L0 treatments inhibited chlorophyll synthesis. Further, the accumulation of leaf carbon (C) and nitrogen (N) was higher in the L1 treatment than in other treatments. Ma bamboo showed rich carbohydrate contents under L0 and L1 treatments in the shooting initial-phase and metaphase. Principal component analysis (PCA) also revealed that the L1 treatment positively correlated with bamboo shoot germination and biochemical activity during the shooting periods. Ultimately, our data suggest that the L1 treatment is the most optimal for promoting bamboo shoot germination, providing a scientific basis for cultivating shoot-used bamboo forests in southern China. Food authenticity has become increasingly important as a result of food adulteration. To identify the authenticity of sweet potato starch noodles, the ladder-shape melting temperature isothermal amplification (LMTIA) method of determining cassava (Manihot esculenta Crantz) DNA in sweet potato starch noodles was used. A set of primers targeted at the internal transcription spacer (ITS) of cassava was designed, genomic DNA was extracted, the LMTIA reaction temperature was optimized, and the specificity of the primer was verified with the genomic DNAs of cassava, sweet potato (Ipomoea batatas L.), Solanum tuberosum L., Zea mays L., Vigna radiate L., Triticum aestivum L., and Glycine max (L.) Merr. The sensitivity with the serially diluted genomic DNA of cassava and the suitability for the DNA extracted from sweet potato starch adulterated with cassava starch were tested. The LMTIA assay for identifying the cassava component in sweet potato starch noodles was established. At the optimal temperature of 52 °C, the primers could specifically distinguish a 0.01% (w/w) cassava component added to sweet potato starch. Additionally, the LMTIA method was applied to the cassava DNA detection of 31 sweet potato starch noodle samples purchased from retail markets in China. Of these, 14 samples were positive. The LMTIA assay could be a reliable method for the rapid detection of cassava components in sweet potato starch noodles, to protect the rights of consumers and to regulate the sale market order of starch noodles. Little is known on how karst plants adapt to highly heterogeneous habitats via adjusting leaf anatomical structures. Phyllostachys glauca McClure is a dominant species that grow across different microhabitats in the limestone mountains of Jiangxi Province, China. We investigated the leaf anatomical structures, plant biomass, soil water content, soil total nitrogen (TN), and soil total phosphorus (TP) from three habitats characterized by different rock exposure, including high rock exposure (HRE), medium rock exposure (MRE) and low rock exposure (LRE), and aimed to discern the relationships between the leaf anatomical plasticity and edaphic factors. The leaves of P. glauca in different habitats showed significant anatomical plasticity in two aspects. First, the leaves adjusted cuticle thickness, papillae length, bulliform cell size and mesophyll thickness to lower water loss and then adapt to the water-deficient habitats (HRE). Second, the leaves enlarged vessels and vascular bundles (first-order and second-order parallel veins) to improve water and nutrient transportation and then enhance plant growth in nitrogen-rich habitats (HRE). Soil water and soil nutrients purely explained the total variation of leaf anatomical traits by 21.7% and 15.7%, respectively, and had a shared proportion of 15.8%. Our results indicated that the leaf anatomical variations in different habitats were associated with both soil water and soil nutrients. Moreover, we found that leaf anatomical structures were more affected by TN than TP. The present study advanced the current understanding of the strategies employed by karst plants to cope with highly heterogeneous habitats via leaf anatomical plasticity. [ABSTRACT FROM AUTHOR]

The H9N2 subtype avian influenza viruses (AIVs) have been circulating in China for more than 20 years, attracting more and more attention due to the potential threat of them. At present, vaccination is a common prevention and control strategy in poultry farms, but as virus antigenicity evolves, the immune protection efficiency of vaccines has constantly been challenged. In this study, we downloaded the hemagglutinin (HA) protein sequences of the H9N2 subtype AIVs from 1994 to 2019 in China—with a total of 5138 sequences. The above sequences were analyzed in terms of time and space, and it was found that h9.4.2.5 was the most popular in various regions of China. Furthermore, the prevalence of H9N2 subtype AIVs in China around 2006 was different. The domestic epidemic branch was relatively diversified from 1994 to 2006. After 2006, the epidemic branch each year was h9.4.2.5. We compared the sequences around 2006 as a whole and screened out 15 different amino acid positions. Based on the HA protein of A/chicken/Guangxi/55/2005 (GX55), the abovementioned amino acid mutations were completed. According to the 12-plasmid reverse genetic system, the rescue of the mutant virus was completed using A/PuertoRico/8/1934 (H1N1) (PR8) as the backbone. The cross hemagglutination inhibition test showed that these mutant sites could transform the parental strain from the old to the new antigenic region. Animal experiments indicated that the mutant virus provided significant protection against the virus from the new antigenic region. This study revealed the antigenic evolution of H9N2 subtype AIVs in China. At the same time, it provided an experimental basis for the development of new vaccines. [ABSTRACT FROM AUTHOR]
