Abstract The mixed-valence compound YbB12 displays paradoxical quantum oscillations in electrical resistivity and magnetic torque in a regime with a well-developed insulating charge gap and in the absence of an electronic Fermi surface. However, signatures of such unusual fermionic quasiparticles in other bulk thermodynamic observables have been missing. Here we report the observation of a series of sharp double-peak features in the specific heat as a function of the magnetic field. The measured Hall resistivity evolves smoothly across the field values at which the characteristic anomalies appear in the thermodynamic response and rules out the possibility of conventional electrons as their origin. Our observations of thermodynamic anomalies in a bulk three-dimensional electrical insulator provide the evidence for the presence of emergent dispersing fermionic excitations within the insulating bulk, which sets the stage for further investigation of electron fractionalization in other correlated mixed-valence compounds.

Abstract Due to the increasing power system capacity, the interruption of large fault currents has gradually become a major challenge in power systems. At the moment, such currents are interrupted mainly via circuit breakers based on the method of active current commutation. However, these types of circuit breakers tend to be costly and oversized. One possible alternative—a fuse‐based circuit breaker with current commutation process enhanced by an external transverse magnetic field (ETMF)—is proposed by the authors. Its main advantage lies in the fast current commutation achieved by an ETMF‐enhanced three‐stage increase of the vacuum arc voltage. The study of the current commutation process is mainly represented by the authors, and the influencing factors of the current commutation—contact opening speed and ETMF current—are discussed. At last, the proposed circuit breaker is verified by an experiment, whose results show that a short‐circuit current of 42 kA can be interrupted within just 2 ms.

Abstract Fuel consumption is one of the main concerns for heavy-duty trucks. Predictive cruise control (PCC) provides an intriguing opportunity to reduce fuel consumption by using the upcoming road information. In this study, a real-time implementable PCC, which simultaneously optimizes engine torque and gear shifting, is proposed for heavy-duty trucks. To minimize fuel consumption, the problem of the PCC is formulated as a nonlinear model predictive control (MPC), in which the upcoming road elevation information is used. Finding the solution of the nonlinear MPC is time consuming; thus, a real-time implementable solver is developed based on Pontryagin’s maximum principle and indirect shooting method. Dynamic programming (DP) algorithm, as a global optimization algorithm, is used as a performance benchmark for the proposed solver. Simulation, hardware-in-the-loop and real-truck experiments are conducted to verify the performance of the proposed controller. The results demonstrate that the MPC-based solution performs nearly as well as the DP-based solution, with less than 1% deviation for testing roads. Moreover, the proposed co-optimization controller is implementable in a real-truck, and the proposed MPC-based PCC algorithm achieves a fuel-saving rate of 7.9% without compromising the truck’s travel time.

Thalassemia is a hemoglobin disease characterized by reduced or complete absence of the production of the α/β globin gene. Currently, the detection of β-thalassemia carriers is based on differences in blood cell parameters. However, β-thalassemia carriers cannot be distinguished from α- and β-thalassemia co-inherited carriers based solely on hematological findings, and the differential diagnosis must rely on molecular diagnosis. We report a 32-year-old male from Yunnan Province, who had abnormal hemoglobin E without obvious anemia. A rare αCS (CD142, TAA→CAA) combined with a βE (CD26, GAG→AAG) double heterozygous mutation was identified in the proband by PCR-reverse dot blot (PCR-RDB) and DNA sequencing. Additionally, a family lineage analysis was performed. This study complements the spectrum of rare thalassemia gene variants and is critical for clinical genetic counseling.

Abstract Background Osteoarthritis (OA) is a prevalent global health concern associated with the loss of articular cartilage and subchondral bone. The lack of disease-modifying drugs for OA necessitates the exploration of novel therapeutic options. Our previous study has demonstrated that traditional Chinese medical herb Trachelospermum jasminoides (Lindl.) Lem. extract suppressed osteoclastogenesis and identified trachelogenin (TCG) as a representative compound. Here, we delved into TCG’s potential to alleviate OA. Methods We initially validated the in vivo efficacy of TCG in alleviating OA using a rat OA model. Subsequently, we isolated primary bone marrow-derived macrophages in vitro to investigate TCG's impact on osteoclastogenesis. We further employed a small molecule pull-down assay to verify TCG's binding target within osteoclasts. Finally, we isolated primary mouse chondrocytes in vitro to study TCG's regulatory effects and mechanisms on chondrocyte survival. Results TCG preserved subchondral bone integrity and protected articular cartilage in a rat OA model. Subsequently, in vitro experiments unveiled TCG's capability to inhibit osteoclastogenesis and function through binding to Ras association proximate 1 (Rap1) and inhibiting its activation. Further study demonstrated that TCG inhibited Rap1/integrin αvβ3/c-Src/Pyk2 signaling cascade, and consequently led to failed F-actin ring formation. Besides, TCG promoted the proliferation of mouse primary chondrocytes while suppressing apoptosis in vitro. This is attributed to TCG's ability to upregulate HIF1α, thereby promoting glycolysis. Conclusion TCG exerted inhibitory effects on osteoclastogenesis through binding to Rap1 and inhibiting Rap1 activation, consequently preventing subchondral bone loss. Moreover, TCG enhanced chondrocyte survival by upregulating HIF1α and promoting glycolysis. These dual mechanisms collectively provide a novel approach to prevented against cartilage degradation.

Abstract Background Cayratia albifolia C.L.Li (CAC), commonly known as “Jiao-Mei-Gu” in China, has been extensively utilized by the Dong minority for several millennia to effectively alleviate symptoms associated with autoimmune diseases. CAC extract is believed to possess significant anti-inflammatory properties within the context of Dong medicine. However, an in-depth understanding of the specific pharmaceutical effects and underlying mechanisms through which CAC extract acts against rheumatoid arthritis (RA) has yet to be established. Methods Twenty-four Sprague–Dawley rats were divided into four groups, with six rats in each group. To induce the collagen-induced arthritis (CIA) model, the rats underwent a process of double immunization with collagen and adjuvant. CAC extract (100 mg/kg) was orally administered to rats. The anti-RA effects were evaluated in CIA rats by arthritis score, hind paw volume and histopathology analysis. Pull-down assay was conducted to identify the potential targets of CAC extract from RAW264.7 macrophage lysates. Moreover, mechanism studies of CAC extract were performed by immunofluorescence assays, real-time PCR and Western blot. Results CAC extract was found to obviously down-regulate hind paw volume of CIA rats, with diminished inflammation response and damage. 177 targets were identified from CAC extract by MS-based pull-down assay. Bioinformatics analysis found that these targets were mainly enriched in macrophage activation and neutrophils extracellular traps (NETs). Additionally, we reported that CAC extract owned significant anti-inflammatory activity by regulating PI3K-Akt-mTOR signal pathway, and inhibited NETosis in response to PMA. Conclusions We clarified that CAC extract significantly attenuated RA by inactivating macrophage and reducing NETosis via a multi-targets regulation. Graphical Abstract

Abstract Objective Acute pancreatitis (AP) stands as a frequent cause for clinical emergency hospital admissions. The X‐box binding protein 1 (XBP1) was found to be implicated in pancreatic acinar cell apoptosis. The objective is to unveil the potential mechanisms governed by XBP1 and SIRT6 in the context of AP. Methods Caerulein‐treated human pancreatic duct epithelial (HPDE) cells to establish an in vitro research model. The levels and regulatory role of SIRT6 in the treated cells were evaluated, including its effects on inflammatory responses, oxidative stress, apoptosis, and endoplasmic reticulum stress. The relationship between XBP1 and SIRT6 was explored by luciferase and ChIP experiments. Furthermore, the effect of XBP1 overexpression on the regulatory function of SIRT6 on cells was evaluated. Results Caerulein promoted the decrease of SIRT6 and the increase of XBP1 in HPDE cells. Overexpression of SIRT6 slowed down the secretion of inflammatory factors, oxidative stress, apoptosis level, and endoplasmic reticulum stress in HPDE cells. However, XBP1 negatively regulated SIRT6, and XBP1 overexpression partially reversed the regulation of SIRT6 on the above aspects. Conclusion Our study illuminates the role of XBP1 in downregulating SIRT6 in HPDE cells, thereby promoting cellular injury. Inhibiting XBP1 or augmenting SIRT6 levels holds promise in preserving cell function and represents a potential therapeutic avenue in the management of AP.

There is an evident requirement for a rapid, efficient, and simple method to screen the authenticity of milk products in the market. Fourier transform infrared (FTIR) spectroscopy stands out as a promising solution. This work employed FTIR spectroscopy and modern statistical machine learning algorithms for the identification and quantification of pasteurized milk adulteration. Comparative results demonstrate modern statistical machine learning algorithms will improve the ability of FTIR spectroscopy to predict milk adulteration compared to partial least square (PLS). To discern the types of substances utilized in milk adulteration, a top-performing multiclassification model was established using multi-layer perceptron (MLP) algorithm, delivering an impressive prediction accuracy of 97.4 %. For quantification purposes, bayesian regularized neural networks (BRNN) provided the best results for the determination of both melamine, urea and milk powder adulteration, while extreme gradient boosting (XGB) and projection pursuit regression (PPR) gave better results in predicting sucrose and water adulteration levels, respectively. The regression models provided suitable predictive accuracy with the ratio of performance to deviation (RPD) values higher than 3. The proposed methodology proved to be a cost-effective and fast tool for screening the authenticity of pasteurized milk in the market.

With the rise of social networks, various types of information have emerged in the vision field in a complex manner, making it crucial to analyze the propagation patters of online public opinion to effectively guide information dissemination. To elucidate the dynamics of information dissemination, this paper proposes a directed network information based on self-organized network information dissemination scenario. This model takes into account the influence of networks formed by users spontaneously and distinguishes the dissemination population based on the in- and out-degree of user nodes in the network. To assess the model’s performance, it is evaluated using real retweets from Chinese Sina Weibo, considering the impact of user interactions on information dissemination. Comparing real data with model-fitted data, the proposed model-based evaluation and numerical analysis demonstrate that the forwarding and transfer probabilities align with actual information dissemination. Furthermore, the evaluation sensitivity analyses describe the key factors influencing information dissemination, aiding decision-making in formulating strategies to guide public opinion. To quantify the importance of these factors, assessment metrics are introduced, such as the propagation regeneration number.

Abstract Transcription-replication conflicts (TRCs), especially Head-On TRCs (HO-TRCs) can introduce R-loops and DNA damage, however, the underlying mechanisms are still largely unclear. We previously identified a chloroplast-localized RNase H1 protein AtRNH1C that can remove R-loops and relax HO-TRCs for genome integrity. Through the mutagenesis screen, we identify a mutation in chloroplast-localized primase ATH that weakens the binding affinity of DNA template and reduces the activities of RNA primer synthesis and delivery. This slows down DNA replication, and reduces competition of transcription-replication, thus rescuing the developmental defects of atrnh1c. Strand-specific DNA damage sequencing reveals that HO-TRCs cause DNA damage at the end of the transcription unit in the lagging strand and overexpression of ATH can boost HO-TRCs and exacerbates DNA damage. Furthermore, mutation of plastid DNA polymerase Pol1A can similarly rescue the defects in atrnh1c mutants. Taken together these results illustrate a potentially conserved mechanism among organisms, of which the primase activity can promote the occurrence of transcription-replication conflicts leading to HO-TRCs and genome instability.

Abstract Quantum oscillation phenomenon is an essential tool to understand the electronic structure of quantum matter. Here we report a systematic study of quantum oscillations in the electronic specific heat C el in natural graphite. We show that the crossing of a single spin Landau level and the Fermi energy give rise to a double-peak structure, in striking contrast to the single peak expected from Lifshitz-Kosevich theory. Intriguingly, the double-peak structure is predicted by the kernel term for C el/T in the free electron theory. The C el/T represents a spectroscopic tuning fork of width 4.8k B T which can be tuned at will to resonance. Using a coincidence method, the double-peak structure can be used to accurately determine the Landé g-factors of quantum materials. More generally, the tuning fork can be used to reveal any peak in fermionic density of states tuned by magnetic field, such as Lifshitz transition in heavy-fermion compounds.

Crimped wool in Tan sheep gradually transitions to straight wool after 35 days (the er-mao stage), which reduces its commercial value. To investigate the changes in wool proteins during this stage, we performed comparative proteomic analysis of the straight and crimped wool using tandem mass tag (TMT)-based quantification. The mean fur curvature (MFC) of crimped wool was significantly greater than that of straight wool (p < 0.001). We identified 1218 proteins between the two types of wool, including 50 keratins (Ks) and 10 keratin-associated proteins (KAPs). There were 213 differentially expressed proteins, including 13 Ks and 4 KAPs. Crimped wool showed relatively high abundances of KAP24-1, K84, K32, K82, and intermediate filament rod domain-containing protein (IRDC), whereas straight wool had relatively high abundances of K6A, K27, K80, KAP16-1, KAP27-1, and trichohyalin (TCHH). The expression levels of KAP16-1, KAP24-1, and KAP27-1 were related to the ratio of paracortex, which may be associated with wool crimp formation. Additionally, high expressions of TCHH, K27, and K6A in the inner root sheath (IRS) were linked to fiber fineness in straight wool. These findings provide insight into the overall expression and distribution patterns of Ks and KAPs, offering opportunities to improve wool quality and enhance its economic potential in the textile industry.

Mastitis (MAS), endometritis (MET), and ketosis (KET) are prevalent diseases in dairy cows that result in substantial economic losses for the dairy farming industry. This study gathered 26,014 records of the health and sickness of dairy cows and 99,102 data of reproduction from 13 Holstein dairy farms in Central China; the milk protein and milk fat content from 56,640 milk samples, as well as the pedigree data of 37,836 dairy cows were obtained. The logistic regression method was used to analyze the variations in the prevalence rates of MAS, MET, and KET among various parities; the mixed linear model was used to examine the effects of the three diseases on milk production, milk quality, and reproductive traits. DMU software (version 5.2) utilized the DMUAI module in conjunction with the single-trait and two-trait animal model, as well as best linear unbiased prediction (BLUP), to estimate the genetic parameters for the three diseases, milk production, milk quality, and reproductive traits in dairy cows. The primary findings of the investigation comprised the following: (1) The prevalence rates of MAS, MET, and KET in dairy farms were 20.04%, 10.68%, and 7.33%, respectively. (2) MAS and MET had a substantial impact (p < 0.01) on milk production, resulting in significant decreases of 112 kg and 372 kg in 305-d Milk Yield (305-d MY), 4 kg and 12 kg in 305-d Protein Yield (305-d PY), and 6 kg and 16 kg in 305-d Fat Yield (305-d FY). As a result of their excessive 305-d MY, some cows were diagnosed with KET due to glucose metabolism disorder. The 305-d MY of cows with KET was significantly higher than that of healthy cows (205 kg, p < 0.01). (3) All three diseases resulted in an increase in the Interval from Calving to First Service (CTFS, 0.60–1.50 d), Interval from First Service to Conception (FSTC, 0.20–16.20 d), Calving Interval (CI, 4.00–7.00 d), and Number of Services (NUMS, 0.07–0.35). (4) The heritabilities of cows with MAS, MET, and KET were found to be low, with values of 0.09, 0.01, and 0.02, respectively. The genetic correlation between these traits ranged from 0.14 to 0.44. This study offers valuable insights on the prevention and control of the three diseases, as well as feeding management and genetic breeding.

Probiotic biofilms have been beneficial in the fight against infections, restoring the equilibrium of the host’s gut microbiota, and enhancing host health. They are considered a novel strategy for probiotic gut colonization. In this case, we evaluated the effects of various active substances from traditional Chinese medicine on Escherichia coli Nissle 1917 (EcN) to determine if they promote biofilm formation. It was shown that 8–64 μg/mL of oleanolic acid increased the development of EcN biofilm. Additionally, we observed that oleanolic acid can effectively suppress biofilm formation in pathogenic bacteria such as Salmonella and Staphylococcus aureus. Next, we assessed the amount of EcN extracellular polysaccharides, the number of live bacteria, their metabolic activity, the hydrophobicity of their surface, and the shape of their biofilms using laser confocal microscopy. Through transcriptome analysis, a total of 349 differentially expressed genes were identified, comprising 134 upregulated and 215 downregulated genes. GO functional enrichment analysis and KEGG pathway enrichment analysis revealed that oleanolic acid functions are through the regulation of bacterial motility, the iron absorption system, the two-component system, and adhesion pathways. These findings suggest that the main effects of oleanolic acid are to prevent bacterial motility, increase initial adhesion, and encourage the development of EcN biofilms. In addition, oleanolic acid interacts with iron absorption to cooperatively control the production of EcN biofilms within an optimal concentration range. Taking these results together, this study suggests that oleanolic acid may enhance probiotic biofilm formation in the intestines, presenting new avenues for probiotic product development.

Abstract Background Meiotic recombination is initiated by Spo11-dependent programmed DNA double-strand breaks (DSBs) that are preferentially concentrated within genomic regions called hotspots; however, the factor(s) that specify the positions of meiotic DSB hotspots remain unclear. Results Here, we examined the frequency and distribution of R-loops, a type of functional chromatin structure comprising single-stranded DNA and a DNA:RNA hybrid, during budding yeast meiosis and found that the R-loops were changed dramatically throughout meiosis. We detected the formation of multiple de novo R-loops in the pachytene stage and found that these R-loops were associated with meiotic recombination during yeast meiosis. We show that transcription-replication head-on collisions could promote R-loop formation during meiotic DNA replication, and these R-loops are associated with Spo11. Furthermore, meiotic recombination hotspots can be eliminated by reversing the direction of transcription or replication, and reversing both of these directions can reconstitute the hotspots. Conclusions Our study reveals that R-loops may play dual roles in meiotic recombination. In addition to participation in meiotic DSB processing, some meiotic DSB hotspots may be originated from the transcription-replication head-on collisions during meiotic DNA replication.

Prostate cancer is one of the most prevalent malignant tumors in men worldwide, and early diagnosis is essential to improve patient survival. This review provides a comprehensive discussion of recent advances in prostate cancer biomarkers, including molecular, cellular, and exosomal biomarkers. The potential of various biomarkers such as gene fusions (TMPRSS2-ERG), noncoding RNAs (SNHG12), proteins (PSA, PSMA, AR), and circulating tumor cells (CTCs) in the diagnosis, prognosis, and targeted therapies of prostate cancer is emphasized. In addition, this review systematically explores how multi-omics data and artificial intelligence technologies can be used for biomarker discovery and personalized medicine applications. In addition, this review provides insights into the development of specific probes, including fluorescent, electrochemical, and radionuclide probes, for sensitive and accurate detection of prostate cancer biomarkers. In conclusion, this review provides a comprehensive overview of the status and future directions of prostate cancer biomarker research, emphasizing the potential for precision diagnosis and targeted therapy.

Abstracts Affected by the burden reduction policy, out-of-school hours care places have become a hot issue of social concern. Taking Xi’an out-of-school hours care places as a research case, this paper discusses its location choice, spatial relationships and influencing factors using methods such as text analysis, spatial analysis, and mathematical statistics. The results show that: (1) the distribution of out-of-school hours care places in Xi’an is closely related to the community and schools. The names mostly use words such as “sunshine,” “teacher,” and “love,” which are mainly distributed on the lower floors (one to three floors), of which the first floor accounts for the largest proportion. (2) The high-value areas of out-of-school hours care places are mainly concentrated in the Lianhu District, Yanta District, Xincheng District, and the north Chang’an District. Their distribution has obvious directionality, showing a “northeast-southwest” trend, and the global spatial autocorrelation is positively correlated. (3) The spatial pattern of out-of-school hours care places is basically consistent with that of primary and secondary schools, and most of them are located within 1000 m of it. (4) The influencing factors mainly include the distribution of primary and secondary schools, residential areas, population density, house rent, and policies.

A new UHPC pre-stress transfer device is proposed for pre-stressed anti-floating anchor rods. To investigate the axial compression performance of the device during pre-stressing, physical experiments and a finite element verification were conducted on four different types of full-scale device specimens. The changes in the axial displacements and ultimate bearing capacities under axial pressure for the different types of devices were analyzed. The results indicated that bending failures occurred in the upper plate portions of all the types of specimens, with transverse cracks appearing near the upper plate portions of the short columns. The axial compression ultimate bearing capacity of each specimen exceeded the design value. Among them, the axial compression ultimate bearing capacity of the HM18 device increased the most relative to its design value by 87%, while axial compression ultimate bearing capacity of the HM45 device increased the least relative to its design value by only 2%. The new UHPC transfer device exhibits good applicability in pre-stressed anti-floating anchor rods.

Manganese toxicity is a major obstacle to agriculture in acid soils. Dark pericarp disease (DPD) is a newly spread physiological disorder induced by excess Mn in litchi, leading to undesirable fruit appearance and substantial economic loss. In this work, broadcast of alkaline soil conditioner in winter, followed by foliar sprays of ascorbic acid and sulfur solution at fruit development, was adopted to examine the effect of these combinations on DPD alleviation in a litchi orchard, with DPD morbidities of 70~85% in recent ten years. The combination of soil conditioner broadcast and foliar water spray was used as the control. At harvest, DPD incidence was significantly decreased by sulfur spray (3.3 ± 1.0%) and slightly reduced by ascorbic acid spray (10.7 ± 8.0%) compared to the control (12.9 ± 7.6%). Soil pH and available Mn were significantly increased and reduced by the soil conditioner broadcast. Sulfur spray significantly inhibited Mn uptake but enhanced the accumulation of Mg, Ca, sugars and cyanidin-3-rutinoside in the pericarp, leading to improved fruit pigmentation. Antioxidase activities were regulated to resist Mn stress by sulfur spray. The spray of ascorbic acid could not mitigate DPD as expected, probably due to the dose used. Conclusively, this study provides a practicable approach to mitigate Mn phytoavailability in acid soils.

We introduce a novel method that combines satellite data, advanced clustering techniques, machine learning feature extraction, and statistical models to enhance fishery forecasting accuracy. Focusing on jumbo flying squid in the southeast Pacific Ocean near Peru, we utilize MODIS-Aqua and MODIS-Terra satellite data on sea surface temperature (SST) to construct a deep convolutional embedded clustering (DCEC) model and extract the monthly SST features (FM) based on an optimized number of clusters determined by the Davies–Bouldi index (DBI). We use the extracted FM to construct a series of Generalized Additive Models (GAM) to forecast the catch per unit effort (CPUE) of jumbo flying squid within a spatial resolution of 0.5° × 0.5°. Our results demonstrate the following findings: (1) The SST feature clusters obtained through the DCEC model could capture the SST monthly variations; (2) The GAM models with FM outperform the models with the traditional monthly average SST in terms of predictive accuracy; (3) Using both FM and average SST together can further improve model performance. This study demonstrates the effectiveness of the DCEC combined with DBI in extracting marine environmental features and highlights the ocean environment feature extraction method to enhance the precision and reliability of fishery forecasting models.

The evaluation of coronary morphology provides important guidance for the treatment of coronary heart disease (CHD). A chaotic Gaussian mutation antlion optimizer algorithm (CGALO) is proposed in the paper, and it is combined with SVM to construct a classification prediction model for Fractional flow reserve (FFR). To overcome the limitations of the original antlion optimizer (ALO) algorithm, the chaotic Gaussian mutation strategy is introduced, which leads to an improvement in its convergence speed and accuracy. To evaluate the proposed algorithm's performance, comparative experiments were conducted on 23 benchmark functions alongside 12 other cutting-edge optimization algorithms. The experimental outcomes demonstrate that the proposed algorithm achieves superior convergence accuracy and speed compared to the alternative comparison algorithms. Additionally, it is combined with SVM and FS to construct a hierarchical FFR classification model, which is utilized to make effective predictions for 84 patients at the affiliated hospital of medical school, Ningbo university. The experimental results demonstrate that the proposed model achieves an average accuracy of 92%. Moreover, it concludes that smoking history, number of lesion vessels, lesion location, diffuse lesions and ST segment changes, and other factors are the most critical indicators for FFR. Therefore, the model that has been established is a new FFR intelligent classification prediction technology that can effectively assist doctors in making corresponding decisions and evaluation plans.

Depression has several negative effects on emotion as well as learning and memory abilities. Previous studies showed that depression could exacerbate inflammation, which in turn further aggravated depression. Deferoxamine (DFO) is a chelating agent binding iron and aluminium, and is clinically applied to treat acute ion poisoning and hemochromatosis. Researches showed that it could reduce inflammation via increasing the expression of hypoxia-inducible factor-1alpha (HIF-1α). Here, we established a chronic unpredictable mild stress (CUMS) model to investigate whether DFO exerted a neuroprotective function in depression. The results demonstrated that CUMS (4 weeks) effectively induced depression-like behaviors in mice based on sucrose preference test (SPT), forced swim test (FST), tail suspension test (TST), open field test (OFT), and elevated plus-maze test (EPT). It also brought cognitive deficits based on Morris water maze (MWM) test and the impairment of synaptic plasticity based on in vivo electrophysiological recordings. Additionally, CUMS exposure significantly decreased the expression of hippocampal synapse related proteins and the spine density of neurons in the DG region, accompanied by increasing the expression of hippocampal inflammatory cytokines, and promoted the activation of microglia in the hippocampus. The expression of HIF-1α was down-regulated as expected. However, DFO distinctly reversed the CUMS-induced impairments. The mechanism is associated with the DFO inhibition of inflammation by upregulating HIF-1 expression, thereby alleviating a series of pathology changes. Together, these findings suggest that DFO likely plays a protective role in cognitive impairments and synaptic plasticity deficits resulting from depression.

Abstract Osteoblasts play an important role in the regulation of bone homeostasis throughout life. Thus, the damage of osteoblasts can lead to serious skeletal diseases, highlighting the urgent need for novel pharmacological targets. This study introduces chemical genetics strategy by using small molecule forskolin (FSK) as a probe to explore the druggable targets for osteoporosis. Here, this work reveals that transglutaminase 2 (TGM2) served as a major cellular target of FSK to obviously induce osteoblast differentiation. Then, this work identifies a previously undisclosed allosteric site in the catalytic core of TGM2. In particular, FSK formed multiple hydrogen bonds in a saddle‐like domain to induce an “open” conformation of the β‐sandwich domain in TGM2, thereby promoting the substrate protein crosslinks by incorporating polyamine. Furthermore, this work finds that TGM2 interacted with several mitochondrial homeostasis‐associated proteins to improve mitochondrial dynamics and ATP production for osteoblast differentiation. Finally, this work observes that FSK effectively ameliorated osteoporosis in the ovariectomy mice model. Taken together, these findings show a previously undescribed pharmacological allosteric site on TGM2 for osteoporosis treatment, and also provide an available chemical tool for interrogating TGM2 biology and developing bone anabolic agent.

Albinism is characterized by a variable degree of hypopigmentation affecting the skin and the hair, and causing ophthalmologic abnormalities. Its oculocutaneous, ocular and syndromic forms follow an autosomal or X-linked recessive mode of inheritance, and 22 disease-causing genes are implicated in their development. Our aim was to clarify the genetic background of a Hungarian albinism cohort. Using a 22-gene albinism panel, the genetic background of 11 of the 17 Hungarian patients was elucidated. In patients with unidentified genetic backgrounds (n = 6), whole exome sequencing was performed. Our investigations revealed a novel, previously unreported rare variant (N687S) of the two-pore channel two gene (TPCN2). The N687S variant of the encoded TPC2 protein is carried by a 15-year-old Hungarian male albinism patient and his clinically unaffected mother. Our segregational analysis and in vitro functional experiments suggest that the detected novel rare TPCN2 variant alone is not a disease-causing variant in albinism. Deep genetic analyses of the family revealed that the patient also carries a phenotype-modifying R305W variant of the OCA2 protein, and he is the only family member harboring this genotype. Our results raise the possibility that this digenic combination might contribute to the observed differences between the patient and the mother, and found the genetic background of the disease in his case.

Cucurbitacins, oxygenated tetracyclic triterpenoids that are found mainly in the Cucurbitaceae family, play essential roles as defensive compounds, serving as allomones against herbivores and pathogens and as signals for insect–parasite recognition. These compounds also exhibit various pharmacological effects. The biosynthesis of cucurbitacins is largely regulated by the bitter (Bi) gene, encoding an oxidosqualene cyclase, which catalyzes the conversion of 2,3-oxidosqualene into cucurbitadienol, a common precursor for cucurbitacin synthesis. Previous studies focused on uncovering the Bi gene clusters in Cucurbitaceae, but their presence in other cucurbitacin-producing plants remained unexplored. Here, the evolutionary history of Bi genes and their clusters were investigated in twenty-one plant genomes spanning three families based on chemotaxonomy. Nineteen Bi genes were identified in fourteen Cucurbitaceae, four Begoniaceae, and one Aquilaria species. Phylogenetic analysis suggested that the genome of Aquilaria sinensis contained the earliest Bi gene clusters in this dataset. Moreover, the genomic analysis revealed a conserved microsynteny of pivotal genes for cucurbitacin biosynthesis in Cucurbitaceae, while interspersed Bi gene clusters were observed in Begoniaceae, indicating rearrangements during plant Bi gene cluster formation. The bitter gene in A. sinensis was found to promote cucurbitadienol biosynthesis in the leaves of Nicotiana benthamiana. This comprehensive exploration of plant Bi genes and their clusters provides valuable insights into the genetic and evolutionary underpinnings of cucurbitacin biosynthesis. These findings offer prospects for a deeper understanding of cucurbitacin production and potential genetic resources for their enhancement in various plants.

NAC is a class of plant-specific transcription factors that are widely involved in the growth, development and (a)biotic stress response of plants. However, their molecular evolution has not been extensively studied in Malvales, especially in Aquilaria sinensis, a commercial and horticultural crop that produces an aromatic resin named agarwood. In this study, 1502 members of the NAC gene family were identified from the genomes of nine species from Malvales and three model plants. The macroevolutionary analysis revealed that whole genome duplication (WGD) and dispersed duplication (DSD) have shaped the current architectural structure of NAC gene families in Malvales plants. Then, 111 NAC genes were systemically characterized in A. sinensis. The phylogenetic analysis suggests that NAC genes in A. sinensis can be classified into 16 known clusters and four new subfamilies, with each subfamily presenting similar gene structures and conserved motifs. RNA-seq analysis showed that AsNACs presents a broad transcriptional response to the agarwood inducer. The expression patterns of 15 AsNACs in A. sinensis after injury treatment indicated that AsNAC019 and AsNAC098 were positively correlated with the expression patterns of four polyketide synthase (PKS) genes. Additionally, AsNAC019 and AsNAC098 were also found to bind with the AsPKS07 promoter and activate its transcription. This comprehensive analysis provides valuable insights into the molecular evolution of the NAC gene family in Malvales plants and highlights the potential mechanisms of AsNACs for regulating secondary metabolite biosynthesis in A. sinensis, especially for the biosynthesis of 2-(2-phenyl) chromones in agarwood.

Abstract Developing low‐cost and high‐voltage manganese (Mn)‐based Na superionic conductor (NASICON) cathode materials have attracted extensive interest. The low capacity and cycling instability of Na4MnAl(PO4)3 (NMAP), however, limits its performance in sodium‐ion batteries (SIBs). Herein, a binary Na4Mn0.5Fe0.5Al(PO4)3 (MNFAP) is fabricated to ease the structural instability and, in turn, deliver an improved reversible capacity of 102 mAh g−1 at 0.1 C and a high energy density of 287.7 Wh kg−1. The synergistic interaction of Fe and Mn in Na4Mn0.5Fe0.5Al(PO4)3/C composite leads to a one‐phase solid‐solution reaction mechanism with high structural reversibility. Theoretical calculations have also been performed to explain the upshifted voltage platform of both Fe2+/Fe3+ and Mn3+/Mn4+ redox potentials. The rational design of NASICON‐type cathodes by regulating their composition with dual metal ions provides new perspectives for developing high‐performance SIBs.

While most broadly neutralizing antibodies (bnAb) against Influenza virus target conserved conformational epitopes of the glycoprotein hemagglutinin (HA), Sun et al. characterize a lineage of bnAbs that neutralize group 1 and 2 strains. Structural characterization shows that antibody 28-12 binds a continuous epitope within H3 (group 2) but requires a conformational epitope for H1 (group 1) binding. Comparison of germline-reverted Ab and intermediate mutants provides evidence for an evolutionary adaptation from group 2 to group 1 strain.

The thermal performance of a novel exterior coating material for commonly used grain and food-grain oil structures was investigated. Grain structures included a concrete squat silo and a concrete warehouse while the edible oil structure was a concrete sided tank. The exterior coating provided excellent moisture runoff and solar reflectance properties and is best described as a superamphiphobic self-cleaning passive subambient daytime radiative cooling (SSC-PSDRC) coating. The coating exhibited a remarkable subambient daytime cooling effect in various structures in different climatic regions. Compared with the roof surface temperatures of a cool white-coated concrete grain silo and a gray carbon iron-based edible oil storage tank, those of the PSDRC coated top surfaces could be reduced by 37 °C and 33 °C, respectively. The roof surface temperature of a warehouse painted with a cool-white coating—with a solar reflectance of 0.9 and an emissivity of 0.85—and that of a warehouse with the roof installed with aluminised polymer waterproof membranes were 19 °C and 18 °C higher than that of the PSDRC warehouse, respectively. Consequently, the interior temperature of the wheat pile in the PSDRC grain silo was 10 °C lower than that in the control squat silo. With the inner loop flow temperature control system operating, the interior air temperatures of the PSDRC west-facing separate space were 6 °C and 3 °C higher than those of the cool-white coated and control west-facing separate spaces, respectively. Even after the application of PSDRC coating for only a few days, the interior air temperature of the PSDRC oil storage tank was reduced by 38 °C, and the interior temperature of the oil storage tank was reduced by 4 °C. Furthermore, in practical applications, the coating showed impressive superamphiphobic self-cleaning capabilities and super aging resistance. The wide applications of the coating would have far-reaching, global implications for maintaining grain and edible oil products, particularly in the sub-tropical climates.

Background and aimsCompared with self-prepared LRD, a prepackaged low-residue diet (LRD) can improve patient compliance, but whether it can further improve the quality of bowel preparation is uncertain. The study aimed to compare the application of the prepackaged formula LRD with self-prepared LRD in bowel preparation for colonoscopy.MethodsA multicenter randomized controlled trial was conducted in 15 centers. The eligible subjects were randomly assigned to one of two groups: the formula LRD group and the self-prepared LRD group. On the day before the colonoscopy, subjects in the self-prepared LRD group were instructed to consume a restricted LRD prepared by themselves, while subjects in the formula LRD group were given six bags of prepackaged formula LRD and instructed to consume them according to their individual need. The primary outcome was an adequate bowel preparation rate. Secondary outcomes mainly included Boston Bowel Preparation Scale (BBPS) scores, dietary restriction compliance rate, tolerance, satisfaction, adenoma detection rate (ADR), and adverse reactions. The trial was registered at ClinicalTrials.gov under the identifier NCT03943758.ResultsA total of 550 subjects were recruited. Compared with the self-prepared LRD group, the formula LRD group showed a higher adequate bowel preparation rate (94.5 vs. 80.4%; P < 0.01), BBPS scores (7.87 ± 1.13 vs. 6.75 ± 1.47; P < 0.01), dietary compliance rate (92.4 vs. 78.9%; P < 0.01), tolerance (P < 0.01 in degree of hunger, intensity of physical strength, and negative influence on daily activities), satisfaction (8.56 ± 1.61 vs. 7.20 ± 2.02; P < 0.01), and ADR (25.6 vs. 16.0%; P < 0.01). There was no significant difference in adverse reactions.ConclusionCompared with self-prepared LRD, the formula LRD showed similar safety and higher bowel preparation quality, compliance, and tolerance in bowel preparation. More formula LRDs could be designed according to different dietary habits and ethnic populations, and further researches are warranted to confirm their effect.Clinical trial registrationhttps://register.clinicaltrials.gov, identifier: NCT03943758.

Adulteration of higher priced milks with cheaper ones to obtain extra profit can adversely affect consumer health and the market. In this study, pure buffalo milk (BM), goat milk (GM), camel milk (CM), and their mixtures with 5–50% (vol/vol) cow milk or water were used. Mid-infrared spectroscopy (MIRS) combined with modern statistical machine learning was used for the discrimination and quantification of cow milk or water adulteration in BM, GM, and CM. Compared to partial least squares (PLS), modern statistical machine learning—especially support vector machines (SVM), projection pursuit regression (PPR), and Bayesian regularized neural networks (BRNN)—exhibited superior performance for the detection of adulteration. The best prediction models for the different predictive traits are as follows: The binary classification models developed by SVM resulted in differentiation of CM-cow milk, and GM/CM-water mixtures. PLS resulted in differentiation of BM/GM-cow milk and BM-water mixtures. All of the above models have 100% classification accuracy. SVM was used to develop multi-classification models for identifying the high and low proportions of cow milk in BM, GM, and CM, as well as the high and low proportions of water adulteration in BM and GM, with correct classification rates of 94%, 100%, 100%, 99%, and 100%, respectively. In addition, a PLS-based model was developed for identifying the high and low proportions of water adulteration in CM, with correct classification rates of 100%. A regression model for quantifying cow milk in BM was developed using PCA + BRNN, with RMSEV = 5.42%, and RV2 = 0.88. A regression model for quantifying water adulteration in BM was developed using PCA + PPR, with RMSEV = 1.70%, and RV2 = 0.99. Modern statistical machine learning improved the accuracy of MIRS in predicting BM, GM, and CM adulteration more effectively than PLS.

The injection of large volumes of wastewater has induced earthquakes from June 2010 to July 2011 along the Guy–Greenbrier fault in Arkansas. We measure time-dependent changes in seismic velocities underneath three temporary seismic stations, using the daily autocorrelations of ambient seismic noise. We do not find any effect of the pumping rate or volume injected in the changes in velocity. However, the induced earthquakes’ ground motions are associated with an extreme reduction in seismic velocities. The magnitude of the changes surpasses ten times the values typically found in other earthquake settings. Our full-waveform investigation suggests that the changes mainly occurred around the top of the Ozark aquifer. We interpret these extreme changes as a result of dynamic shaking in the elevated pore pressure system of the Ozark aquifer.

Knowledge innovation is a key factor in industrial development and regional economic growth. Understanding regional knowledge innovation and its dynamic changes is one of the fundamental tasks of regional policy-makers and business decision-makers. Although many existing studies have been conducted to support in understanding knowledge innovation patterns, data-driven and intuitive visual analysis of georeferenced knowledge innovation has not been sufficiently studied. In this work, we analysed knowledge innovation by visually exploring big georeferenced scholarly data. More specifically, we first applied network analysis and statistical methods to derive key measures (e.g., the number of publications and academic collaborations) of knowledge innovation with multiple spatial scales. We then designed geovisualizations to explicitly represent the multiscale spatiotemporal patterns and relations. We integrated the analytical methods and geovisualizations into an interactive tool to facilitate stakeholders’ visual learning and analysis of knowledge innovation with a spatial focus. Our work shows that geovisualizations have great potential in supporting complex geoinformation communication in knowledge innovation.

Aim: This study evaluates the cost–effectiveness of imipenem/cilastatin/relebactam (IMI/REL) for treating hospital-acquired bacterial pneumonia and ventilator-associated bacterial pneumonia (HABP/VABP) in an ‘early adjustment prescribing scenario’. Methods: An economic model was constructed to compare two strategies: continuation of empiric piperacillin/tazobactam (PIP/TAZ) versus early adjustment to IMI/REL. A decision tree was used to depict the hospitalization period, and a Markov model used to capture longterm outcomes. Results: IMI/REL generated more quality-adjusted life years than PIP/TAZ, at an increased cost per patient. The incremental cost–effectiveness ratio of $17,529 per QALY is below the typical US willingness-to-pay threshold. Conclusion: IMI/RELmay represent a cost-effective treatment for payers and a valuable option for clinicians, when considered alongside patient risk factors, local epidemiology, and susceptibility data.

Ethyl methanesulfonate (EMS)-induced mutagenesis is a powerful tool to generate genetic resource for identifying untapped genes and characterizing the function of genes to understand the molecular basis of important agronomic traits. This review focuses on application of contemporary EMS mutagenesis in the field of plant development and abiotic stress tolerance research, with particular focuses on reviewing the mutation types, mutagenesis site, mutagen concentration, mutagenesis duration, the identification and characterization of mutations responsible for altered stress tolerance responses. The application of EMS mutation breeding combined with genetic engineering in the future plant breeding and fundamental research was also discussed. The collective information in this review will provide good insight on how EMS mutagenesis is efficiently applied to improve abiotic stress tolerance of crops with the utilization of Next-generation sequencing (NGS) for mutation identification.

BackgroundThe progression process of lung cancer can be accelerated by M2 macrophages. However, genes that affect M2 macrophage polarization remain unidentified.MethodsThe Cancer Genome Atlas, Gene Expression Omnibus, and Arrayexpress databases were used to obtain open-access data. The analysis of public data was mostly performed with R studio. The RNA levels of specific genes were detected using quantitative real-time PCR. The proliferation ability of the cells was assessed by CCK8, colony formation, and EdU assays.ResultsBased on the multiple datasets, we noticed a poor prognosis in patients with high M2 macrophage infiltration. There were 114 genes differentially expressed between high and low M2 macrophages infiltrated samples, regarded as M2 macrophage-related genes. Subsequently, a prognosis prediction signature consisting of ABHD5, HS3ST2, TM6SF1, CAPZA2, LEPROT, HNMT, and MRO was identified and presented a satisfactory performance. The pathway enrichment results revealed a positive correlation between riskscore and enrichment scores for most immunotherapy-related positive terms. Also, there might be an increase in genomic instability among patients at high risk. Interestingly, low risk patients are most likely to benefit from PD-1 therapy, while high risk patients may benefit from CTLA-4 therapy. Meanwhile, the estimated IC50 of seven drugs differs significantly between two risk groups, including Cisplatin, Docetaxel, Doxorubicin, Gefitinib, Paclitaxel, Sunitinib and Vinorelbine. Moreover, further experiments indicated that HNMT was overexpressed and can enhance the proliferation ability in lung cancer cells.ConclusionsIn summary, our study identified the molecules significantly affecting M2 macrophage infiltration and identified a prognosis signature that robustly indicated patients prognosis. Moreover, we validated the cancer-promoting effect of HNMT using in vitro experiments.

Summary: Background: The E2F family of transcription factors play a crucial role in the development of various cancers. However, E2F members lack targetable binding pockets and are typically considered “undruggable”. Unlike canonical small-molecule therapeutics, molecular glues mediate new E3 ligase–protein interactions to induce selective proteasomal degradation, which represents an attractive option to overcome these limitations. Methods: Human proteome microarray was utilized to identify a natural product-derived molecular glue for targeting E2F2 degradation. Co-IP analysis with stable isotope labeling of amino acids in cell culture (SILAC)-based quantitative proteomics was carried out to further explore the E3 ligase for E2F2 degradation. Findings: In this study, we identified a molecular glue bufalin, which significantly promoted E2F2 degradation. Unexpectedly, E2F2 underwent ubiquitination and proteasomal degradation via a previously undisclosed atypical E3 ligase, zinc finger protein 91 (ZFP91). In particular, we observed that bufalin markedly promoted E2F2-ZFP91 complex formation, thereby leading to E2F2 polyubiquitination via K48-linked ubiquitin chains for degradation. E2F2 degradation subsequently caused transcriptional suppression of multiple oncogenes including c-Myc, CCNE1, CCNE2, MCM5 and CDK1, and inhibited hepatocellular carcinoma growth in vitro and in vivo. Interpretation: Collectively, our findings open up a new direction for transcription factors degradation by targeting atypical E3 ligase ZFP91. Meanwhile, the chemical knockdown strategy with molecular glue may promote innovative transcription factor degrader development in cancer therapy. Abstract Background The receptor-binding domain (RBD) variants of SARS-CoV-2 could impair antibody-mediated neutralization of the virus by host immunity; thus, prospective surveillance of antibody escape mutants and understanding the evolution of RBD are urgently needed. Methods Using the single B cell cloning technology, we isolated and characterized 93 RBD-specific antibodies from the memory B cells of four COVID-19 convalescent individuals in the early stage of the pandemic. Then, global RBD alanine scanning with a panel of 19 selected neutralizing antibodies (NAbs), including several broadly reactive NAbs, was performed. Furthermore, we assessed the impact of single natural mutation or co-mutations of concern at key positions of RBD on the neutralization escape and ACE2 binding function by recombinant proteins and pseudoviruses. Results Thirty-three amino acid positions within four independent antigenic sites (1 to 4) of RBD were identified as valuable indicators of antigenic changes in the RBD. The comprehensive escape mutation map not only confirms the widely circulating strains carrying important immune escape RBD mutations such as K417N, E484K, and L452R, but also facilitates the discovery of new immune escape-enabling mutations such as F486L, N450K, F490S, and R346S. Of note, these escape mutations could not affect the ACE2 binding affinity of RBD, among which L452R even enhanced binding. Furthermore, we showed that RBD co-mutations K417N, E484K, and N501Y present in B.1.351 appear more resistant to NAbs and human convalescent plasma from the early stage of the pandemic, possibly due to an additive effect. Conversely, double mutations E484Q and L452R present in B.1.617.1 variant show partial antibody evasion with no evidence for an additive effect. Conclusions Our study provides a global view of the determinants for neutralizing antibody recognition, antigenic conservation, and RBD conformation. The in-depth escape maps may have value for prospective surveillance of SARS-CoV-2 immune escape variants. Special attention should be paid to the accumulation of co-mutations at distinct major antigenic sites. Finally, the new broadly reactive NAbs described here represent new potential opportunities for the prevention and treatment of COVID-19.

Abstract Topological Dirac semimetals have emerged as a platform to engineer Berry curvature with time-reversal symmetry breaking, which allows to access diverse quantum states in a single material system. It is of interest to realize such diversity in Dirac semimetals that provides insight on correlation between Berry curvature and quantum transport phenomena. Here, we report the transition between anomalous Hall and chiral fermion states in three-dimensional topological Dirac semimetal KZnBi, which is demonstrated by tuning the direction and flux of Berry curvature. Angle-dependent magneto-transport measurements show that both anomalous Hall resistance and positive magnetoresistance are maximized at 0° between net Berry curvature and rotational axis. We find that the unexpected crossover of anomalous Hall resistance and negative magnetoresistance suddenly occurs when the angle reaches to ~70°, indicating that Berry curvature strongly correlates with quantum transports of Dirac and chiral fermions. It would be interesting to tune Berry curvature within other quantum phases such as topological superconductivity.

Placenta accreta spectrum (PAS) is a high-risk obstetrical complication associated with significant morbidity and mortality. Here the authors discover a uniquely high prevalence of circulating trophoblasts clusters in PAS and explore their diagnostic potential to augment current diagnostic modalities for the early detection of PAS.

MXene-based composites have been widely used in electric energy storage device. As a member of MXene, niobium carbide (Nb2C) is a good electrode candidate for energy storage because of its high specific surface area and electronic conductivity. However, a pure Nb2C MXene electrode exhibits limited supercapacitive performance due to its easy stacking. Herein, sodium anthraquinone-2-sulfonate (AQS) with high redox reactivity was employed as a tailor to enhance the accessibility of ions and electrolyte and enhance the capacitance performance of Nb2C MXene. The resulting Nb2C–AQS composite had three-dimensional porous layered structures. The supercapacitors (SCs) based on the Nb2C–AQS composite exhibited a considerably higher electrochemical capacitance (36.3 mF cm−2) than the pure Nb2C electrode (16.8 mF cm−2) at a scan rate of 20 mV s−1. The SCs also exhibited excellent flexibility as deduced from the almost unchanged capacitance values after being subjected to bending. A capacitance retention of 99.5% after 600 cycles was observed for the resulting SCs, indicating their good cycling stability. This work proposes a surface modification method for Nb2C MXene and facilitates the development of high-performance SCs.

Abstract Previous simulation studies of dust particles movement behavior in open-pit coal mines only aimed at a single operation link, and the macro simulation is lacking. This study seeks to explore microscopic migration and macroscopic diffusion of dust particles in the mining area by numerical simulation method. The movement characteristics revealed, and the distribution law of the dust particles in a stable state with the corresponding migration paths in the mining area were obtained respectively. The results showed that the increase amplitude of dust particles diffusion velocity was inversely proportional to particle size, which was vital for dust pollution in the mine. And the larger the particle size in the mining pit was, the lower the escape rate of the dust particles was. Dust particles distributed over a wide range that were not limited by space height, and the distribution characteristics of dust particles at different heights were basically the same. Meanwhile, it is found that the dust distribution in the two places is relatively concentrated due to the circulation phenomenon of the mining pit and the west dump. And wind action would accelerate the moving dust particles to reach a stable distribution state.

Abstract Rechargeable lithium/sodium–sulfur batteries working at room temperature (RT‐Li/S, RT‐Na/S) appear to be a promising energy storage system in terms of high theoretical energy density, low cost, and abundant resources in nature. They are, thus, considered as highly attractive candidates for future application in energy storage devices. Nevertheless, the solubility of sulfur species, sluggish kinetics of lithium/sodium sulfide compounds, and high reactivity of metallic anodes render these cells unstable. As a consequence, metal–sulfur batteries present low reversible capacity and quick capacity loss, which hinder their practical application. Investigations to address these issues regarding S cathodes are critical to the increase of their performance and our fundamental understanding of RT‐Li/S and RT‐Na/S battery systems. Metal–sulfur interactions, recently, have attracted considerable attention, and there have been new insights on pathways to high‐performance RT‐Li/Na sulfur batteries, due to the following factors: (1) deliberate construction of metal–sulfur interactions can enable a leap in capacity; (2) metal–sulfur interactions can confine S species, as well as sodium sulfide compounds, to stop shuttle effects; (3) traces of metal species can help to encapsulate a high loading mass of sulfur with high‐cost efficiency; and (4) metal components make electrodes more conductive. In this review, we highlight the latest progress in sulfide immobilization via constructing metal bonding between various metals and S cathodes. Also, we summarize the storage mechanisms of Li/Na as well as the metal–sulfur interaction mechanisms. Furthermore, the current challenges and future remedies in terms of intact confinement and optimization of the electrochemical performance of RT‐Li/Na sulfur systems are discussed in this review.

Metabolic reprogramming is a hallmark of glioma, and sterol O-acyltransferase 1 (SOAT1) is an essential target for metabolic therapy. However, the prognostic value of SOAT1 and its association with immune infiltration has not been fully elucidated. Using RNA-seq and clinical data of glioma patients from The Cancer Genome Atlas (TCGA), SOAT1 was found to be correlated with poor prognosis in glioma and the advanced malignancy of clinicopathological characteristics. Next, the correlation between SOAT1 expression and tumor-infiltrating immune cells was performed using the single-sample GSEA algorithm, gene expression profiling interactive analysis (GEPIA), and tumor immune estimation resource version 2 (TIMER2.0); it was found that SOAT1 expression was positively correlated with multiple tumor-infiltrating immune cells. To further verify these results, immunofluorescence was conducted on paraffin-embedded glioma specimens, and a positive trend of the correlation between SOAT1 expression and Treg infiltration was observed in this cohort. Finally, differentially expressed gene analysis, and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were performed to explore the biological processes and signaling pathways that SOAT1 may be involved in during glioma pathogenesis. A protein-protein interaction network was established, and co-expression analysis was conducted to investigate the regulatory mechanism of SOAT1 in glioma. To the best of our knowledge, this is the first comprehensive study reporting that SOAT1 may serve as a novel prognostic biomarker associated with immune infiltrates, providing a novel perspective for glioma metabolic therapy.
