Zhenlong Xingnao Capsule (ZXC) is a Tibetan medicine used to treat ischemic stroke. In this study, we determined the in vitro and in vivo effects of ZXC on reactive oxygen species (ROS) in a mouse BV-2 microglial cell hypoxia-reoxygenation and rat middle cerebral artery occlusion infarction models. We aimed to clarify the role of ZXC in cerebral ischemia protection; reveal amino acid neurotransmitter changes in the frontal cortex after drug intervention; determine mRNA and protein expression changes in Bcl-2, Bax, caspase-3, P38, and nuclear factor (NF)-кB in the frontal cortex and changes in antioxidant indices in the brain; and elucidate the mechanisms underlying ZXC action. After hypoxia-reoxygenation, ROS levels were significantly increased in BV-2 cells, and their levels decreased after treatment with ZXC. ZXC had protective effects on ischemic/anoxic injury in vitro and in vivo by downregulating the expressions of caspase-3 and NF-кB mRNA during ischemia and reperfusion and that of p38 and caspase-3 during acute ischemia and reperfusion as well as the steady-state levels of excitatory amino acids/inhibitory amino acids and by improving the total antioxidant capacity and total superoxide dismutase activities during ischemia. These findings provide new molecular evidence for the mechanisms underlying ZXC action.

Abstract Background Inflammatory bowel disease (IBD) is a chronic disease in which macrophages play an important role in its pathogenesis. Platelet-derived growth factor-BB (PDGF-BB) secreted by macrophages is involved in the repair of vascular endothelial injury during inflammatory reactions. Methods The expression levels of M1 macrophages and PDGF-BB in serum and colonic mucosa of 30 patients with Crohn’s disease (CD) and 30 patients with ulcerative colitis (UC) were measured using enzyme-linked immunosorbent assays and immunohistochemistry. Logistic regression was used for univariate and multivariate analyses, and receiver operating characteristic curves were used to evaluate diagnostic value. Associations were evaluated using Spearman correlation analysis. Results The expression of serum PDGF-BB and M1 macrophages with positive CXCL9 expression in patients with active-stage IBD [206.55(160.41,262.90)and 337.30(217.73,472.28) pg/ml] was higher than that in patients with remission stage [153.42(107.02,219.68)and 218.37(144.49,347.33)pg/ml] and controls [156.19(91.16,216.08)and 191.20(121.42,311.76)pg/ml](P

Vertically aligned gold nanorod arrays have attracted much attention for their fascinating optical properties. Different from longitudinal surface plasmon wavelength (LSPW) and edge-to-edge spacing of gold nanorods, the role of gold nanorod diameter in plasmonic enhancement ability of vertical gold nanorod arrays has rarely been explored. In this work, we selected gold nanorods with similar LSPW but two different diameters (22 and 41 nm), the optical properties of which are dominated by absorption and scattering cross sections, respectively. The vertically aligned arrays of these gold nanorods formed by evaporation self-assembly are coupled with nonlinear ZnO nanocrystal films spin-coated on their surfaces. It was found that the gold nanorod array with a larger diameter can enhance the second harmonic generation (SHG) of ZnO nanofilm by a factor of 27.0, while it is about 7.3 for the smaller gold nanorod array. Theoretical simulations indicate that such stronger enhancement of the larger vertical gold nanorod array compared with the smaller one is due to its stronger scattering ability and greater extent of near-field enhancement at SHG fundamental wavelength. Our work shows that the diameter of gold nanorods is also an important factor to be considered in realizing strong plasmon enhancement of vertically aligned gold nanorod arrays.
