Abstract The sustainable development of Chinese agriculture needs to coordinate the quality and quantity of agricultural products. To verify whether government regulation can promote agricultural development by controlling pesticides, the indices of government regulation, quantity and quality of agricultural products, and pesticide use per area of cropland were calculated, and a comprehensive evaluation index and the coupling degree and coordination degree of agricultural products were analyzed by coupling function. The results showed that the coupling and coordination degree of the quality and quantity of agricultural products and the comprehensive index are improving. The regulation of pesticide use per area of cropland is a key mechanism in promoting the coordinated development of agricultural product safety. The regulation trend of agricultural development in China is to reduce pesticide usage appropriately and expand the production-possibility frontier of agricultural products. Graphical abstract

Abstract Vacuum ultraviolet (VUV) light sensing shows great potential applications in the space science, materials, biophysics, and plasma physics. In this work, an all-optical detection method is proposed for VUV sensing by constructing an optical fiber-end Fabry-Pérot interferometer based on a single aluminum nitride (AlN) microwire. Compared with the traditional electrical devices, this all-optical detection method overcomes the difficulties like the fast response and electromagnetic interference immunity in detecting VUV bands at the present stage, and improves the response speed. The proposed device shows the excellent performance of VUV detection, with the static sensitivity of 1.03 nm/(W·cm−2), response rise time of down to 10 µs, and decay time of 0.64 ms. Beneficial from the excellent radiation resistance of AlN microwires and UV resistance of silica fibers, the proposed device is expected to have the good stability and potential applications in the fields of the solar physics and space exploration.

Summary: Objectives: The exostosins (EXT), which are responsible for heparan sulfate backbone synthesis and play a vital role in tissue homeostasis, have been reported to be correlated with prognosis of various cancers. However, the expression, prognostic value, and immune infiltration of EXT1 and EXT2 in head and neck squamous cell carcinoma (HNSC) remain uncertain. Methods: GEPIA, UALCAN, and Xiantao bioinformatics tools were used to explore the EXT1 and EXT2 expression level in HNSC. GEPIA and Sangerbox were utilised to obtain the prognostic value of EXT1 and EXT2 in HNSC. Genetic alterations, immune cell infiltration, and single-cell analysis were conducted in cBioPortal, TIMER, and TISCH2. In addition, the expressions of EXT1 and EXT2 were validated by real-time polymerase chain reaction (PCR) in HNSC samples. Results: EXT1 and EXT2 were highly expressed in HNSC, especially in malignant cells. Only EXT2 was significantly negatively correlated to the prognosis of patients with HNSC. EXT1 and EXT2 were found to be associated with focal adhesin and cell adhesin molecule binding. EXT1 expression levels were considerably connected with CD8+ T cell infiltrating levels, whilst EXT2 expression levels were considerably negatively connected with infiltrating levels of CD4+ T cells, macrophages, neutrophils, and dendritic cells in HNSC. The gene mutation rates of EXT1 and EXT2 in HNSC were 7% and 2.8%, respectively. Moreover, EXT2 was validated to be highly expressed in HNSC samples by real-time PCR. Conclusion: EXT2 was highly expressed and presented negative correlation with the prognosis and immune infiltration of HNSC, which might be a potential biomarker for HNSC.

Abstract A new type of human immunoglobulin G (IgG) sensors based on the surface plasmon resonance (SPR) in the low refractive index (RI) plastic optical fiber (POF) and an antibody immobilization method is presented. A 50-nm-thick gold film was formed on the polished D-shaped fiber surface by magnetron sputtering. The RI response of the POF sensor is 30 049.61 nm/RIU, which is 26.5 times higher than that of single mode fiber (SMF) SPR sensors. The proposed SPR biosensor can be developed by simple and rapid modification of the gold film with 11-mercapto undecanoic acid (MUA). Upon immobilization of the goat anti-human IgG antibody, the resonance wavelength shifts by 11.2 nm. The sensor can be used to specifically detect and quantify the human IgG at concentrations down to 245.4 ng/mL with the sensitivity of 1.327 7 nm per µg/mL, which offers an enhancement of 12.5-fold compared to that of the conventional SMF based SPR sensors. The proposed device may find the potential applications in the case of use at the point of care.

Abstract Stripe rust fungus Puccinia striiformis f. sp. tritici (Pst) is a destructive pathogen of wheat worldwide. Pst has a macrocyclic-heteroecious lifecycle, in which one-celled urediniospores are dikaryotic, each nucleus containing one haploid genome. We successfully generated the first fully haplotype-resolved and nearly gap-free chromosome-scale genome assembly of Pst by combining PacBio HiFi sequencing and trio-binning strategy. The genome size of the two haploid assemblies was 75.59 Mb and 75.91 Mb with contig N50 of 4.17 Mb and 4.60 Mb, and both had 18 pseudochromosomes. The high consensus quality values of 55.57 and 59.02 for both haplotypes confirmed the correctness of the assembly. Of the total 18 chromosomes, 15 and 16 were gapless while there were only five and two gaps for the remaining chromosomes of the two haplotypes, respectively. In total, 15,046 and 15,050 protein-coding genes were predicted for the two haplotypes, and the complete BUSCO scores achieved 97.7% and 97.9%, respectively. The genome will lay the foundation for further research on genetic variations and the evolution of rust fungi.

Pasteurella multocida toxin (PMT) is one of the most important virulence factors of Pasteurella multocida type D. Pasteurella multocida infection has caused enormous economic losses in the pig farming industry. Although it is well known that this bacterial infection causes progressive atrophic rhinitis, its effects on other organ tissues in pigs are unclear. In this study, PMT was expressed and purified, and the cytotoxic effects of PMT on four types of swine cells, LLC-PK1, PAM, IPEC, and ST, were investigated. LLC-PK1 exhibited the highest sensitivity to the cytotoxic effects of PMT. Our studies revealed that a PMT concentration of 0.1 μg/kg can lead to weight loss, whereas a PMT concentration of 0.5 μg/kg can lead to death in mice. PMT causes damage to the intestines, kidneys, lungs, livers, and spleens of mice. Furthermore, PMT caused acute death in pigs at treatment concentrations greater than 5 μg/kg; at PMT concentration of 2.5 μg/kg, weight loss occurred until death. PMT mainly caused damage to the hearts, lungs, livers, spleens and kidneys of pigs. The organ coefficient showed that damage to the heart and kidneys was the most severe and caused the renal pelvis and renal pyramid to dissolve and become cavitated. Pathology revealed hemorrhage in the lungs, liver, and spleen, and the kidneys were swollen and vacuolated, which was consistent with the damaged target organs in the mice. In conclusion, these findings demonstrate that PMT is extremely toxic in vitro and in vivo, causing damage to various organs of the body, especially the kidneys and lungs. This study provides a theoretical basis for the in-depth exploration of the cytotoxic effects of PMT on target organs.

Abstract Benefiting from the great advances of the femtosecond laser two-photon polymerization (TPP) technology, customized microcantilever probes can be accurately 3-dimensional (3D) manufactured at the nanoscale size and thus have exhibited considerable potentials in the fields of microforce, micro-vibration, and microforce sensors. In this work, a controllable microstructured cantilever probe on an optical fiber tip for microforce detection is demonstrated both theoretically and experimentally. The static performances of the probe are firstly investigated based on the finite element method (FEM), which provides the basis for the structural design. The proposed cantilever probe is then 3D printed by means of the TPP technology. The experimental results show that the elastic constant k of the proposed cantilever probe can be actively tuned from 2.46 N/m to 62.35 N/m. The force sensitivity is 2.5 nm/µN, the Q-factor is 368.93, and the detection limit is 57.43 nN. Moreover, the mechanical properties of the cantilever probe can be flexibly adjusted by the geometric configuration of the cantilever. Thus, it has an enormous potential for matching the mechanical properties of biological samples in the direct contact mode.

α-RuCl3, a narrow-band Mott insulator with a large work function, offers intriguing potential as a quantum material or as a charge acceptor for electrical contacts in van der Waals devices. In this work, we perform a systematic study of the optical reflection contrast of α-RuCl3 nanoflakes on oxidized silicon wafers and estimate the accuracy of this imaging technique to assess the crystal thickness. Via spectroscopic micro-ellipsometry measurements, we characterize the wavelength-dependent complex refractive index of α-RuCl3 nanoflakes of varying thickness in the visible and near-infrared. Building on these results, we simulate the optical contrast of α-RuCl3 nanoflakes with thicknesses below 100 nm on SiO2/Si substrates under different illumination conditions. We compare the simulated optical contrast with experimental values extracted from optical microscopy images and obtain good agreement. Finally, we show that optical contrast imaging allows us to retrieve the thickness of the RuCl3 nanoflakes exfoliated on an oxidized silicon substrate with a mean deviation of −0.2 nm for thicknesses below 100 nm with a standard deviation of only 1 nm. Our results demonstrate that optical contrast can be used as a non-invasive, fast, and reliable technique to estimate the α-RuCl3 thickness.

A 33-year-old patient presented with a chief complaint of patent ductus arteriosus (PDA) persisting for over 30 years. Physical examination revealed bilateral facial angiofibromas, multiple nail fibromas, intraoral fibromas, and a ’shagreen patch’ on the left lumbar region. Genetic testing was performed using a peripheral venous blood sample, which confirmed the diagnosis of Tuberous Sclerosis Type 2 (TSC2). Subsequently, the patient underwent cardiac color Doppler ultrasound and chest computed tomography angiography, which confirmed the presence of PDA. Tuberous sclerosis complex (TSC) is associated with cardiovascular diseases. The initial clinical manifestation of TSC is usually cardiac rhabdomyoma in children, and it is rarely reported in adults with PDA. In this case, the patient was diagnosed with PDA when he was young, and the genetic test showed heterozygous variation of TSC2 gene. The purpose of this article is to explore the correlation between TSC and PDA at the gene level through literature review.

The gene encoding 9-cis-epoxycarotenoid dioxygenase 3 (NCED3) functions in abscisic acid (ABA) biosynthesis, plant growth and development, and tolerance to adverse temperatures, drought and saline conditions. In this study, three rice lines were used to explore the function of OsNCED3, these included an OsNCED3-overexpressing line (OsNCED3-OE), a knockdown line (osnced3-RNAi) and wild-type rice (WT). These rice lines were infested with the brown plant hopper (BPH; Nilaparvata lugens) and examined for physiological and biochemical changes, hormone content, and defense gene expression. The results showed that OsNCED3 activated rice defense mechanisms, which led to an increased defense enzyme activity of superoxide dismutase, peroxidase, and polyphenol oxidase. The overexpression of OsNCED3 decreased the number of planthoppers and reduced oviposition and BPH hatching rates. Furthermore, the overexpression of OsNCED3 increased the concentrations of jasmonic acid, jasmonyl-isoleucine and ABA relative to WT rice and the osnced3-RNAi line. These results indicate that OsNCED3 improved the stress tolerance in rice and support a role for both jasmonates and ABA as defense compounds in the rice-BPH interaction.

Textbooks have a crucial role in shaping students' knowledge, behaviors, and attitudes in different school subjects. This study compares the structure and content of science textbooks of grade nine in Egypt and China to reveal the common and different features in the textbook design. It opts for a horizontal analysis of four science textbooks in the associated countries. The results revealed that the distribution of science subjects has partial similarities to some extent among the preparatory stage between the Chinese and Egyptian science textbooks besides the overlapping in the associated topics, presenting Biology as a common subject of interest. Moreover, the number of activities distributed within units and subjects have the highest shares in the Chinese textbooks, and most of the activities in the Egyptian textbooks focused on Chemistry and Physics subjects. In addition to the structure analysis, this study also explored the textbooks content in both countries, covering three dimensions: (1) cognitive expectations, (2) learning goals, and (3) efficiency of illustration. The results provide valuable insights for textbook designers and curriculum developers to enhance the quality of science curricula and textbooks. Therefore, the study recommends considering instructional design and lesson plans when distributing the learning activities and developing international standards for designing school science textbooks.

BackgroundThe interaction between pyroptosis—a form of programmed cell death—and tumor immunity represents a burgeoning field of interest. Pyroptosis exhibits a dual role in cancer: it can both promote tumor development and counteract it by activating immune responses that inhibit tumor evasion and encourage cell death. Current tumor immunotherapy strategies, notably CAR-T cell therapy and immune checkpoint inhibitors (ICIs), alongside the potential of certain traditional Chinese medicinal compounds, highlight the intricate relationship between pyroptosis and cancer immunity. As research delves deeper into pyroptosis mechanisms within tumor therapy, its application in enhancing tumor immune responses emerges as a novel research avenue.PurposeThis review aims to elucidate the mechanisms underlying pyroptosis, its impact on tumor biology, and the advancements in tumor immunotherapy research.MethodsA comprehensive literature review was conducted across PubMed, Embase, CNKI, and Wanfang Database from the inception of the study until August 22, 2023. The search employed keywords such as “pyroptosis”, “cancer”, “tumor”, “mechanism”, “immunity”, “gasdermin”, “ICB”, “CAR-T”, “PD-1”, “PD-L1”, “herbal medicine”, “botanical medicine”, “Chinese medicine”, “traditional Chinese medicine”, “immunotherapy”, linked by AND/OR, to capture the latest findings in pyroptosis and tumor immunotherapy.ResultsPyroptosis is governed by a complex mechanism, with the Gasdermin family playing a pivotal role. While promising for tumor immunotherapy application, research into pyroptosis’s effect on tumor immunity is still evolving. Notably, certain traditional Chinese medicine ingredients have been identified as potential pyroptosis inducers, meriting further exploration.ConclusionThis review consolidates current knowledge on pyroptosis’s role in tumor immunotherapy. It reveals pyroptosis as a beneficial factor in the immunotherapeutic landscape, suggesting that leveraging pyroptosis for developing novel cancer treatment strategies, including those involving traditional Chinese medicine, represents a forward-looking approach in oncology.

Sirtuins are pivotal in orchestrating numerous cellular pathways, critically influencing cell metabolism, DNA repair, aging processes, and oxidative stress. In recent years, the involvement of sirtuins in tumor biology has garnered substantial attention, with a growing body of evidence underscoring their regulatory roles in various aberrant cellular processes within tumor environments. This article delves into the sirtuin family and its biological functions, shedding light on their dual roles—either as promoters or inhibitors—in various cancers including oral, breast, hepatocellular, lung, and gastric cancers. It further explores potential anti-tumor agents targeting sirtuins, unraveling the complex interplay between sirtuins, miRNAs, and chemotherapeutic drugs. The dual roles of sirtuins in cancer biology reflect the complexity of targeting these enzymes but also highlight the immense therapeutic potential. These advancements hold significant promise for enhancing clinical outcomes, marking a pivotal step forward in the ongoing battle against cancer.

Given the increasing demands for quality assurance in the food industry, a significant challenge emerges in the form of expensive integration of food sensors into packaging. This integration is crucial for strengthening food safety measures and ensuring the impeccable quality of food products. Official laboratory food safety testing heavily relies on expensive and bulky equipment. This article presents a new chemical sensing platform and a comparative study of in-house built novel designs for a robust multimode chemical sensor head probed by highly sensitive light-sheet skew rays for addressing cost and footprint issues. The sensing mechanism is the interaction between evanescent field mediated by refined skew rays propagating through a structured coreless multimode fiber and external chemicals, resulting in probe light absorption. The sensitivity is enhanced by the controlled excitation of skew rays using a light sheet and four specially engineered coreless multimode fiber structure, including uniform, tapered, microstub and microbubble designs. The sensitivity was demonstrated to be as high as 0.046 (dB/cm) / dB(1 ng/ml) and the limit of detection as low as 1.028 ng/ml for the microbubble structure. The results of our research pave the groundwork for a new range of chemical sensors suitable for food safety monitoring.

Emerging research into metabolic dysfunction-associated steatotic liver disease (MASLD) up until January 2024 has highlighted the critical role of cuproptosis, a unique cell death mechanism triggered by copper overload, in the disease's development. This connection offers new insights into MASLD's complex pathogenesis, pointing to copper accumulation as a key factor that disrupts lipid metabolism and insulin sensitivity. The identification of cuproptosis as a significant contributor to MASLD underscores the potential for targeting copper-mediated pathways for novel therapeutic approaches. This promising avenue suggests that managing copper levels could mitigate MASLD progression, offering a fresh perspective on treatment strategies. Further investigations into how cuproptosis influences MASLD are essential for unraveling the detailed mechanisms at play and for identifying effective interventions. The focus on copper's role in liver health opens up the possibility of developing targeted therapies that address the underlying causes of MASLD, moving beyond symptomatic treatment to tackle the root of the problem. The exploration of cuproptosis in the context of MASLD exemplifies the importance of understanding metal homeostasis in metabolic diseases and represents a significant step forward in the quest for more effective treatments. This research direction lights path for innovative MASLD management and reversal.

The most important optical component in an optical fiber endoscope is its objective lens. To achieve a high imaging performance level, the development of an ultra-compact objective lens is thus the key to an ultra-thin optical fiber endoscope. In this work, we use femtosecond laser 3D printing to develop a series of micro objective lenses with different optical designs. The imaging resolution and field-of-view performances of these printed micro objective lenses are investigated via both simulations and experiments. For the first time, multiple micro objective lenses with different fields of view are printed on the end face of a single imaging optical fiber, thus realizing the perfect integration of an optical fiber and objective lenses. This work demonstrates the considerable potential of femtosecond laser 3D printing in the fabrication of micro-optical systems and provides a reliable solution for the development of an ultrathin fiber endoscope.

Abstract Spatial omics technologies can help identify spatially organized biological processes, but existing computational approaches often overlook structural dependencies in the data. Here, we introduce Smoother, a unified framework that integrates positional information into non-spatial models via modular priors and losses. In simulated and real datasets, Smoother enables accurate data imputation, cell-type deconvolution, and dimensionality reduction with remarkable efficiency. In colorectal cancer, Smoother-guided deconvolution reveals plasma cell and fibroblast subtype localizations linked to tumor microenvironment restructuring. Additionally, joint modeling of spatial and single-cell human prostate data with Smoother allows for spatial mapping of reference populations with significantly reduced ambiguity.

A novel surface plasmon resonance (SPR) refractive index (RI) sensor based on the D-type dual-mode photonic crystal fiber (PCF) is proposed. The sensor employs a side-polished few-mode PCF that facilitates the transmission of the fundamental and second-order modes, with an integrated microfluidic channel positioned directly above the fiber core. This design minimizes the distance to the analyte and maximizes the interaction between the optical field and the analyte, thereby enhancing the SPR effect and resonance loss for improved sensing performance. Au-TiO2 dual-layer material was coated on the surface of a microfluidic channel to enhance the penetration depth of the core evanescent field and tune the resonance wavelength to the near-infrared band, meeting the special needs of chemical and biomedical detection fields. The finite element method was utilized to systematically investigate the coupling characteristics between various modes and surface plasmon polariton (SPP) modes, as well as the impact of structural parameters on the sensor performance. The results indicate that the LP11b_y mode exhibits greater wavelength sensitivity than the HE11_y mode, with a maximum sensitivity of 33,000 nm/RIU and an average sensitivity of 8272.7 nm/RIU in the RI sensing range of 1.25–1.36, which is higher than the maximum sensitivity of 16,000 nm/RIU and average sensitivity of 5666.7 nm/RIU for the HE11b_y mode. It is believed that the proposed PCF-SPR sensor features both high sensitivity and high resolution, which will become a critical device for wide RI detection in mid-infrared fields.

Glaesserella parasuis cytolethal distending toxin (GpCDT) can induce cell cycle arrest and apoptosis. Our laboratory’s previous work demonstrated that GTPase 4b (Rab4b) is a key host protein implicated in GpCDT-induced cytotoxicity. This study investigated the probable involvement of Rab4b in the process. Our study used CRISPR/Cas9 technology to create a Rab4b-knockout cell line. The results showed greater resistance to GpCDT-induced cell cytotoxicity. In contrast, forced Rab4b overexpression increased GpCDT-induced cytotoxicity. Further immunoprecipitation study reveals that GpCDT may bind with Rab4b. In PK-15 cells, GpCDT is transported to the early endosomes and late endosomes, while after knocking out Rab4b, GpCDT cannot be transported to the early endosome via vesicles. Rab4b appears essential for GpCDT-induced cytotoxicity in PK-15 cells.

Tumor cells invade normal surrounding tissues through continuous division. In this study, we hypothesized that cell cycle regulation changes the immune efficacy of ovarian cancer. To investigate this hypothesis, a Förster resonance energy transfer (FRET) sensor was constructed to characterize the cell activity in real time. Cell shrinkage caused by apoptosis induces the aggregation of proteins on the cell membrane, leading to variations in the fluorescence lifetime of FRET sensors. Moreover, we tracked cell activity across various cycles following co-culture with an immune checkpoint inhibitor. Consequently, we assessed how cell cycle regulation influences immunotherapy in a tumor mouse model. This approach, which involves inhibiting typical cell cycle processes, markedly enhances the effectiveness of immunotherapy. Our findings suggest that modulating the cycle progression of cancer cells may represent a promising approach to enhance the immune response of ovarian cancer cells and the efficacy of immunotherapy based on immune checkpoint inhibitors.

The deep integration of communication and sensing technology in fiber-optic systems has been highly sought after in recent years, with the aim of rapid and cost-effective large-scale upgrading of existing communication cables in order to monitor ocean activities. As a proof-of-concept demonstration, a high-degree of compatibility was shown between forward-transmission distributed fiber-optic vibration sensing and an on–off keying (OOK)-based communication system. This type of deep integration allows distributed sensing to utilize the optical fiber communication cable, wavelength channel, optical signal and demodulation receiver. The addition of distributed sensing functionality does not have an impact on the communication performance, as sensing involves no hardware changes and does not occupy any bandwidth; instead, it non-intrusively analyzes inherent vibration-induced noise in the data transmitted. Likewise, the transmission of communication data does not affect the sensing performance. For data transmission, 150 Mb/s was demonstrated with a BER of 2.8 × 10−7 and a QdB of 14.1. For vibration sensing, the forward-transmission method offers distance, time, frequency, intensity and phase-resolved monitoring. The limit of detection (LoD) is 8.3 pε/Hz1/2 at 1 kHz. The single-span sensing distance is 101.3 km (no optical amplification), with a spatial resolution of 0.08 m, and positioning accuracy can be as low as 10.1 m. No data averaging was performed during signal processing. The vibration frequency range tested is 10–1000 Hz.

Although the Japanese encephalitis virus (JEV) infects various cell types, its receptor molecules are still not clearly understood. In our laboratory’s prior research, Rab4b was identified as a potential host factor that facilitates JEV infection in PK15 cells, utilizing a genome-wide CRISPR/Cas9 knockout library (PK-15-GeCKO). To further explore the effect of Rab4b on JEV replication, we used the Rab4b knockout PK15 cell line using the CRISPR/Cas9 technology and overexpressing the Rab4b PK15 cell line, with IFA, RT–qPCR, and Western blot to study the effect of Rab4b on viral replication in the whole life cycle of the JEV. The results show that the knockout of Rab4b inhibited the replication of the JEV in PK15 cells, and the overexpression of Rab4b promoted the replication of the JEV in PK15 cell lines. Furthermore, we demonstrated for the first time that host factor Rab4b facilitates the adsorption, internalization, assembly, and release of the JEV, thereby promoting JEV replication. This study enriches the regulatory network between the JEV and host factors and lays the experimental foundation for further understanding of the function of the Rab4b protein.

For distributed fiber-optic sensors, slowly varying vibration signals down to 5 mHz are difficult to measure due to low signal-to-noise ratios. We propose and demonstrate a forward transmission-based distributed sensing system, combined with a polarization-generated carrier for detection bandwidth reduction, and cross-correlation for vibration positioning. By applying a higher-frequency carrier signal using a fast polarization controller, the initial phase of the known carrier frequency is monitored and analyzed to demodulate the vibration signal. Only the polarization carrier needs to be analyzed, not the arbitrary-frequency signal, which can lead to hardware issues (reduced detection bandwidth and less noise). The difference in arrival time between the two detection ends obtained through cross-correlation can determine the vibration position. Our experimental results demonstrate a sensitivity of 0.63 mrad/με and a limit of detection (LoD) of 355.6 pε/Hz1/2 at 60 Hz. A lock-in amplifier can be used on the fixed carrier to achieve a minimal LoD. The sensing distance can reach 131.5 km and the positioning accuracy is 725 m (root-mean-square error) while the spatial resolution is 105 m. The tested vibration frequency range is between 0.005 Hz and 160 Hz. A low frequency of 5 mHz for forward transmission-based distributed sensing is highly attractive for seismic monitoring applications.

In the quest to address the critical shortage of donor organs for transplantation, xenotransplantation stands out as a promising solution, offering a more abundant supply of donor organs. Yet, its widespread clinical adoption remains hindered by significant challenges, chief among them being immunological rejection. Central to this issue is the role of the complement system, an essential component of innate immunity that frequently triggers acute and chronic rejection through hyperacute immune responses. Such responses can rapidly lead to transplant embolism, compromising the function of the transplanted organ and ultimately causing graft failure. This review delves into three key areas of xenotransplantation research. It begins by examining the mechanisms through which xenotransplantation activates both the classical and alternative complement pathways. It then assesses the current landscape of xenotransplantation from donor pigs, with a particular emphasis on the innovative strides made in genetically engineering pigs to evade complement system activation. These modifications are critical in mitigating the discordance between pig endogenous retroviruses and human immune molecules. Additionally, the review discusses pharmacological interventions designed to support transplantation. By exploring the intricate relationship between the complement system and xenotransplantation, this retrospective analysis not only underscores the scientific and clinical importance of this field but also sheds light on the potential pathways to overcoming one of the major barriers to the success of xenografts. As such, the insights offered here hold significant promise for advancing xenotransplantation from a research concept to a viable clinical reality.

Early-stage infective endocarditis (IE) can lead to severe complications, including infarctions and metastatic infections caused by inflammatory embolus shedding. Common embolism sites include the brain, spleen, kidneys, lungs, and intestines. Additionally, acute heart failure (AHF) can occur in up to 40% of cases, and its presence can impact the clinical outcomes of patients with IE. Cardiogenic shock (CGS) is often more likely to occur after AHF has taken place. If bacteria invade the blood, infectious shock can occur. Patients with IE can experience simple CGS, septic shock, or a combination of the two. Extracorporeal membrane oxygenation (ECMO) typically serves as a Bridge for Heart failure and Cardiogenic shock. Previous research indicates that there are limited reports of ECMO support for patients with IE after CGS has occurred. Because CGS may occur at any time during IE treatment, it is important to understand the timing of ECMO auxiliary support and how to carry out comprehensive treatment after support. Timely treatment can help to reduce or avoid the occurrence of serious complications and improve the prognosis of patients with IE. Our work combines a case study to review the ECMO support of IE patients after CGS through a literature review. Overall, we suggest that when patients with IE have large bacterial thrombosis and a greater risk of shedding, it is recommended to carefully evaluate the indications and contraindications for ECMO after discussion by a multidisciplinary team (MDT). Still, active surgical treatment at an early stage is recommended.

A highly sensitivity balloon-like fiber interferometer based on ethanol coating is presented in this paper. The Mach–Zehnder interferometer is formed by bending a single-mode fiber to a balloon-like structure and nested in the Teflon tube. Then, an ethanol solution was filled into the tube of the balloon-like fiber interferometer by the capillary effect. Due to the high sensitivity of the refractive index (RI) of ethanol solutions to temperature, when the external temperature varies, the optical path difference changes. The change in temperature can be detected by the shift in the interference spectrum. Limited by the size of the balloon-like structure, three kinds of these structures with different sensitive lengths were prepared to select the best parameters. The sensitive lengths were 10, 15 and 20 mm, respectively, and the RI detection performance of each structure in 10~26% NaCl solutions was investigated experimentally. The results show that when the sensitive length is 20 mm, the RI sensitivity of the sensor is the highest, which is 212.88 nm/RIU. Ultimately, the sensitive length filled with ethanol is 20 mm. The experimental results show that the temperature sensitivity of the structure is 1.145 nm/°C in the range of 28.1 °C~35 °C, which is 10.3 times higher than that of an unfilled balloon-like structure (0.111 nm/°C). The system has the advantages of low cost and easy fabrication, which can potentially be used in high-precision temperature monitoring processes.

Afforestation plays a critical role in ecosystem restoration and carbon sequestration. However, there continues to be insufficient knowledge about the long-term effects of different tree species on the forest soil in central subtropical China. In this study, five indigenous afforestation tree species commonly used in the region, including Bretschneidera sinensis, Liriodendron chinense, Schima superba, Phoebe bournei, and Cunninghamia lanceolata, were selected to explore their long-term effects on the forest soil. The soil’s physicochemical properties, organic carbon content, enzyme activity, and respiration were investigated. Our results revealed significant differences in the soil physicochemical properties, enzyme activity, organic carbon content, and soil respiration among the different tree species even with the same tree species types. Broad-leaved species, particularly L. chinense and P. bournei, exhibited superior soil physicochemical properties, higher amounts of organic carbon contents, enzyme activity, and soil respiration compared to coniferous species C. lanceolata. Notably, for the two studied evergreen tree species, P. bournei seemed to improve the forest soil quality more than S. superba. Hence, increasing the proportion of broad-leaved tree species may have a beneficial effect on the soil’s physicochemical properties and microecology. Furthermore, considering tree species’ compositions in afforestation will help to optimize soil quality and ecosystem health.

Tsoongiodendron odorum Chun is a large evergreen tree in the Magnoliaceae family and an ancient relict species represented by small wild populations. It has excellent material quality, high ornamental value, and scientific significance. However, due to the complicated natural reproduction and notable habitat destruction, its wild populations must be urgently conserved. We used genotyping-by-sequencing to examine 17 natural populations of T. odorum in China, the species’ primary habitat, to better understand the genetic diversity of this species and use its germplasm resources. T. odorum had a very low level of genetic diversity; its mean values for Ho, He, Pi, and PIC were 0.175, 0.123, 0.160, and 0.053, respectively. With an average within-population Fst of 0.023 and an inter-population gene flow Nm of 10.918, population genetic variation was primarily found within populations, demonstrating minute genetic divergence between populations. The 17 natural populations of T. odorum were divided into two major categories: the Fujian populations in eastern China and the Jiangxi, Guangdong, Hunan, and Guangxi populations in central and western China. Our research contributes to the understanding of T. odorum’s genetic diversity and organization and offers a theoretical framework for the species’ conservation, breeding, and selection.

Totally video-guided thorascopic cardiac surgery (TVTCS) represents one of the most minimally invasive access routes to the heart. Its feasibility and safety can be guaranteed by an experienced surgeon with skilled operative techniques under the guidance of a video signal via thoracoscopy and the imaging from transesophageal echocardiography. At present, this surgical approach has been applied for atrioventricular valve disease, atrial septum defects plus and partial anomalous pulmonary venous drainage, cardiac tumors, hypertrophic obstructive cardiomyopathy, aortic valve disease, and atrial fibrillation. Multimodality cardiovascular imaging, including echocardiography, X-ray, computed tomography (CT), magnetic resonance imaging (MRI) and cardiac catheterization, provides morphologic characteristics and function status of the cardiovascular system and a comprehensive view of the target anatomy. In this review, the benefits of multimodality cardiovascular imaging are summarized for the clinical practice of TVTCS, including the preoperative preparation, intraoperative guidance and postoperative supervision. The disease categories are also individually reviewed on the basis of multimodality cardiovascular imaging, to ensure the feasibility and safety for TVTCS. Cardiovascular imaging technologies not only confirm who is a candidate for this surgical technique, but also provide technical support during the procedure and for postop follow to assess the clinical outcomes. Multimodality cardiovascular imaging is instrumental to provide the requirements to solve the problems for conduction of TVTCS; and to provide individualized protocols with high-resolution and real-time dynamic imaging fusion.

Abstract Background Vesicle trafficking is a highly important process in numerous human diseases, especially in the central nervous system dysfunctions. However, as a key component of vesicle trafficking‐related genes (VRGs), Cornichon family AMPA receptor auxiliary protein 4 (CNIH4) has not been systematically elucidated in glioma so far. Methods Differentially expressed VRGs were selected using molecular signatures database (MSigDB), The Cancer Genome Atlas (TCGA), and Genotype‐Tissue Expression (GTEx) mRNA expression profiles. Further exploration of CNIH4 was determined using LASSO‐Cox regression algorithms. Then Kaplan–Meier (K‐M) plotter, receiver operating characteristic (ROC) curves, and multivariate Cox regression analyses were utilized to assess the independent significance of CNIH4 in the CGGA validation cohort. Functional exploration was performed with Gene Set Enrichment Analysis (GSEA) and then verified using a series of functional experiments in glioma cells. Finally, the consensus clustering algorithm was applied to identify clusters in glioma samples. After that, differences in prognosis, the tumor immune microenvironment (TIME), and therapy response were evaluated between clusters. Results CNIH4 was shown to be overexpressed in malignant glioma variants and was frequently observed in GCSs and TMZ‐resistant cell lines. Higher CNIH4 levels were significantly related to poor outcomes and positively correlated with adverse clinicopathological characteristics. Survival analyses revealed CNIH4 as an independent risk factor that outperformed traditional measures. Enrichment analysis indicated that overactive CNIH4 significantly gathered in stem cell processes. Furthermore, functional assays of silencing CNIH4 expression suppressed stem cell‐like properties in vitro and inhibited tumorigenicity in vivo. Finally, the CNIH4‐enriched subgroup negatively modulated immunotherapeutic response and reflected lower chemotherapy sensitivity for glioma patients. Conclusion Our study identified CNIH4 as a potential VRG that regulates tumor stemness, microenvironment immunity, and chemotherapy sensitivity. It may serve as a novel prognostic factor and a promising target against glioma therapy.

Solar energy may now be captured in two ways: heat extraction in solar collectors and electricity generation in photovoltaic panels. It may effectively reduce the degradation of PV cell efficiency caused by overheating of the battery by collecting the heat from the PV/T system. The heat dissipation cooling fin is directly mounted to the rear of the solar panel in this study and coated with an insulating layer to create an air-cooling channel. The heat dissipation fin transfers heat from the back of the photovoltaic panel to a certain mass flow of air in the flow channel, resulting in combined heat and power. The electrical and thermal capabilities of three distinct fin numbers were studied by monitoring electrical power and temperature data. A fixed mass flow rate (0.011 kg/s) for air, three different numbers of fins (0-5-10) and solar irradiance (200 W/m2–600 W/m2) are the experimental conditions. After the system was run stably for 10 min, the average temperature of the back of the PV panel, the device’s inlet and outlet temperatures, and the PV panel’s electric power were recorded. These figures are used to calculate the PV/T system’s electrical and thermal efficiency. When the median electrical efficiency of the three types of fins is compared with varying solar irradiation, it is discovered that the electric efficiency value of the three types of fins is approximately 16%, with a difference of less than 0.6%. The comprehensive efficiency of five groups of fins and ten groups of fins was 61.08% and 69.85%, respectively, based on the median comparison of photoelectric photothermal comprehensive efficiency. The overall efficiency of the zero groups of fins was 26.27%, which was only 14.59% greater than the control plate, and there was a considerable difference between the five and ten groups of fins. This suggests that selecting the right number of fins has a substantial impact on improving PV/T thermal performance. This research will assist in further optimizing the design of PV/T systems for heat collection and will provide valuable information for PV/T system application in building integration.

ABSTRACTCoxsackievirus A16 (CA16), a main causative agent of hand, foot, and mouth disease (HFMD), has become a serious public health concern in the Asia-Pacific region. Here, we generated an anti-CA16 monoclonal antibody, DMA2017, derived from an epidemic strain CA16. Surprisingly, although DMA2017 could not neutralize the original and circulating CA16 strains in vitro, the passive transfer of DMA2017 (10 μg/g) could protect suckling mice from a lethal challenge with CA16 in vivo. Then, we confirmed the protective effect of DMA2017 relies on the Fc-dependent effector functions, such as antibody-dependent cellular cytotoxicity (ADCC). The linear epitope of DMA2017 was mapped by phage display technique to a conserved patch spanning residues 143–148 (NSHPPY) of the VP2 EF-loop of CA16. DMA2017 could inhibit the binding of the antibodies present in the sera of naturally infected children to CA16, indicating that the epitope of DMA2017 is immunodominant for CA16. Our results confirm, for the first time, that a potential preventive and therapeutic effect could be mediated by a non-neutralizing antibody elicited against CA16. These findings bring a hitherto understudied protective role of non-neutralizing antibodies during viral infections into the spotlight and provide a new perspective on the design and evaluation of CA16 vaccines.

Fractional‐order vector vortex beams (VVBs) have recently been demonstrated as new carriers of fractional‐strength optical vortices. However, the inherent mechanism of the new vortex beams, formed by the combination of both unstable states, to propagate stably in free space requires clarification. In this study, this scientific problem is solved by revealing the physical essence of propagable fractional‐strength optical vortices in free space. Therefore, three new viewpoints regarding these unique vortex beams are proposed: Abbe's diffraction limit, the phase evolution of the vortex beam, and the binary time vector property of phase. First, due to Abbe's diffraction limit, the inherent polarization modes are intertwined, thereby maintaining the entire unique vortex beam in free space. In the second case, the phase evolution of the vortex beam is the physical cause for the polarization rotation of fractional‐order VVBs. Third, the phase is not merely a scalar attribute of the light beam and demonstrates a binary time vector property. This study provides entirely different physical viewpoints on the phase of the vortex beam and Abbe's diffraction limit, which may deepen the knowledge on the behavior of light beams in classical optics.

It is a big challenge to in situ monitor the health status of high-temperature magnetic equipment such as electric motors and generators since it is difficult to achieve a high-temperature magnetoelectric sensor. Here, the Pb(Zr0.52Ti0.48)O3 film with a ferroelectric Curie temperature of 400 °C and the Metglas alloy slice with a magnetic Curie temperature of 430 °C were combined by using a high-temperature inorganic glue to achieve a high-temperature magnetoelectric sensor. The magnetoelectric coefficient αE of the flexible sensor is as high as 104 V/(cm Oe) at 25 °C, 63.6 V/(cm Oe) at 200 °C, and 39.7 V/(cm Oe) at 330 °C. Besides, the magnetic sensor has a detection accuracy of ∼0.3 nT at 25–330 °C. Most importantly, the high-temperature sensor is flexible, high precision, low cost, light weight, and low power consumption simultaneously.

Purpose – The electrical properties of piezoelectric vibrators have a crucial influence on the operating state of ultrasonic motors. In order to solve the problem that the current piezoelectric vibrator generates a large amount of heat during vibration to degrade its performance, which in turn affects the normal operation of ultrasonic motors, this paper prepares a novel piezoelectric vibrator and tests its maximum vibration velocity under the working condition, which is more than twice as much as that of the current commercial PZT-8. Design/methodology/approach – The crystal structures of the samples were analyzed by using an X-ray diffractometer. For microstructure observation, samples were observed by scanning electron microscope (SEM). The quasi-static piezoelectric coefficient meter (ZJ-3AN) was used for piezoelectric measurement. Dielectric properties were measured by utilizing an impedance analyzer (Agilent 4294A) with a laboratory heating unit. Ferroelectric hysteresis loops were obtained using a ferroelectric analyzer (Radiant, Multiferroic 100). A Doppler laser vibrometer (Polytec PSV-300F, Germany) and a power amplifier were used for piezoelectric vibration measurements, during which the temperature rise was determined by an infrared radiation thermometer (Victor 303, China). Findings – The ceramics exhibit enhanced piezoelectric performance at 0.1–0.4 mol% of Yb doping contents. The ceramic of 0.4 mol% Yb reaches the maximal internal bias field and presents a larger mechanical quality factor of 1,692 compared with that of 0.2 mol% Yb-doped ceramic, in spite of a slightly decreased dielectric constant of 439 pC/N, the unit of the piezoelectric constant, which is the ratio of the local charge (pC) to the frontal force (N) and electromechanical coupling coefficient of 0.63. The vibrator with this large mechanical quality factor ceramic displays a vibration velocity of up to 0.81 m/s under the constraint of 20 °C temperature rising, which is much higher than commercial high-power piezoelectric ceramics PZT-8. Originality/value – The enhanced high-power properties of the piezoelectric vibrator by Yb doping may provide a potential application for the high-performance USM and offer the possibility of long-term stable operation under high power for special equipment like USM. In the subsequent phase of research, the novel PZT-based high-power piezoelectric vibrator can be utilized in the USM, and the motor's performance will be evaluated under aerospace conditions to objectively assess the reliability of the piezoelectric vibrator.

The combustion behavior of various propellant samples, including double-base propellants, pressed nitramine powders, and modified double-base propellants containing nitramine, was examined using OH-PLIF technology. The combustion process took place within a combustion chamber, and images capturing the flame at the moment of stable combustion were selected for further analysis. The distribution and production rate of OH radicals in both the double-base propellant and the nitramine-modified double-base propellant were simulated using Chemkin-17.0 software. The outcomes from both the experimental and simulation studies revealed that the concentration of OH radicals increased with a higher content of NG in the double-base propellant. In the modified double-base propellant containing RDX, the OH radical concentration decreased as the RDX content increased, with these tendencies of change aligning closely with the simulation results.

Despite various treatment methods, the remission rate of membranous nephropathy remains limited. Refractory membranous nephropathy especially lacks effective treatment plans. Telitacicept achieves comprehensive inhibition of CD20-positive B cells, plasma cells, and T cells, thereby bringing new hope to the treatment of membranous nephropathy and refractory membranous nephropathy. Here, we report a case of a 46-year-old man with membranous nephropathy. Although the combined treatment with glucocorticoid, tacrolimus, mycophenolate mofetil, cyclophosphamide, and rituximab was not successful, the patient achieved complete remission of urinary protein after glucocorticoid combined with telitacicept. This is the first report on the application of telitacicept in the treatment of membranous nephropathy, especially refractory membranous nephropathy. The application of telitacicept in the treatment of membranous nephropathy deserves further attention.

Background and Aims: Although type 2 innate lymphoid cells (ILC2s) were originally found to be liver-resident lymphocytes, the role and importance of ILC2 in liver injury remains poorly understood. In the current study, we sought to determine whether ILC2 is an important regulator of hepatic ischaemia/reperfusion injury (IRI). Methods: ILC2-deficient mice (ICOS-T or NSG) and genetically modified ILC2s were used to investigate the role of ILC2s in murine hepatic IRI. Interactions between ILC2s and eosinophils or macrophages were studied in coculture. The role of human ILC2s was assessed in an immunocompromised mouse model of hepatic IRI. Results: Administration of IL-33 prevented hepatic IRI in association with reduction of neutrophil infiltration and inflammatory mediators in the liver. IL-33-treated mice had elevated numbers of ILC2s, eosinophils, and regulatory T cells. Eosinophils, but not regulatory T cells, were required for IL-33-mediated hepatoprotection in IRI mice. Depletion of ILC2s substantially abolished the protective effect of IL-33 in hepatic IRI, indicating that ILC2s play critical roles in IL-33-mediated liver protection. Adoptive transfer of ex vivo-expanded ILC2s improved liver function and attenuated histologic damage in mice subjected to IRI. Mechanistic studies combining genetic and adoptive transfer approaches identified a protective role of ILC2s through promoting IL-13-dependent induction of anti-inflammatory macrophages and IL-5-dependent elevation of eosinophils in IRI. Furthermore, in vivo expansion of human ILC2s by IL-33 or transfer of ex vivo-expanded human ILC2s ameliorated hepatic IRI in an immunocompromised mouse model of hepatic IRI. Conclusions: This study provides insight into the mechanisms of ILC2-mediated liver protection that could serve as therapeutic targets to treat acute liver injury. Impact and Implications: We report that type 2 innate lymphoid cells (ILC2s) are important regulators in a mouse model of liver ischaemia/reperfusion injury (IRI). Through manipulation of macrophage and eosinophil phenotypes, ILC2s mitigate liver inflammation and injury during liver IRI. We propose that ILC2s have the potential to serve as a therapeutic tool for protecting against acute liver injury and lay the foundation for translation of ILC2 therapy to human liver disease.

Mechanical metamaterials can adjust mechanical properties of structures flexibly through a mechanical structural design based on the premise that the materials remain unchanged. Here, a cantilever probe microstructure is designed using mechanical metamaterials for an optical fiber microforce sensor tip that can be prepared by femtosecond laser-induced two-photon polymerization. The elastic constant k of the fabricated fiber-optic microforce sensor has been adjusted by two orders of magnitude from 0.165 to 46 N/m, and the geometric configuration of the cantilever beam can be tailored to match the mechanical properties of biological specimens. This fiber microforce sensor shows an ultra-high force sensitivity of 154 nm/µN and a force resolution of up to 130 pN. The optical fiber microforce sensor that shows the lowest force resolution in a direct-contact mode has high potential for biosensing applications, and the results reveal a potential design strategy for special scanning tunneling microscope probes with unique physical properties.

BackgroundPost-operative etiological studies are critical for infection prevention in lung transplant recipients within the first year. In this study, mNGS combined with microbial culture was applied to reveal the etiological characteristics within one week (ultra-early) and one month (early) in lung transplant recipients, and the epidemiology of infection occurred within one month.MethodsIn 38 lung transplant recipients, deep airway secretions were collected through bronchofiberscope within two hours after the operation and were subjected to microbial identification by mNGS and microbial culture. The etiologic characteristics of lung transplant recipients were explored. Within one month, the infection status of recipients was monitored. AimTo construct and validate a risk prediction model for the development of peritoneal dialysis-associated peritonitis (PDAP) in patients undergoing peritoneal dialysis (PD).MethodsThis retrospective analysis included patients undergoing PD at the Department of Nephrology, the First Affiliated Hospital of Anhui University of Chinese Medicine, between January 2016 and January 2021. Baseline data were collected. The primary study endpoint was PDAP occurrence. Patients were divided into a training cohort (n = 264) and a validation cohort (n = 112) for model building and validation. Least Absolute Shrinkage and Selection Operator (LASSO) regression was applied to optimize the screening variables. Predictive models were developed using multifactorial logistic regression analysis with column line plots. Receiver operating characteristic (ROC) curves, calibration curves, and Hosmer-Lemeshow goodness-of-fit tests were used to verify and evaluate the discrimination and calibration of the prediction models. Decision curve analysis (DCA) was used to assess the clinical validity of the prediction models.ResultsFive potential predictors of PDAP after PD catheterization were screened using LASSO regression analysis, including neutrophil-to-lymphocyte ratio (NLR), serum ALBumin (ALB), uric acid (UA), high sensitivity C-reactive protein (hsCRP), and diabetes mellitus (DM). Predictive models were developed by multi-factor logistic regression analysis and plotted in columns. The area under the ROC curve (AUC) values were 0.891 (95% confidence interval [CI]: 0.829–0.844) and 0.882 (95% CI: 0.722–0.957) for the training and validation cohorts, respectively. The Hosmer-Lemeshow test showed a good fit (p = 0.829 for the training cohort; p = 0.602 for the validation cohort). The DCA curves indicated that the threshold probabilities for the training and validation cohorts were 4–64% and 3–90%, respectively, predicting a good net gain for the clinical model.ConclusionNLR, ALB, UA, hsCRP, and DM are independent predictors of PDAP after PD catheterization. The column line graph model constructed based on the abovementioned factors has good discriminatory and calibrating ability and helps to predict the risk of PDAP after PD catheterization.

Abstract Background The major histocompatibility complex (MHC) and the killer cell immunoglobulin-like receptors (KIR) are key regulators of immune responses. The cynomolgus macaque, an Old World monkey species, can be applied as an important preclinical model for studying human diseases, including coronavirus disease 2019 (COVID-19). Several MHC-KIR combinations have been associated with either a poor or good prognosis. Therefore, macaques with a well-characterized immunogenetic profile may improve drug evaluation and speed up vaccine development. At present, a complete overview of the MHC and KIR haplotype organizations in cynomolgus macaques is lacking, and characterization by conventional techniques is hampered by the extensive expansion of the macaque MHC-B region that complicates the discrimination between genes and alleles. Methods We assembled complete MHC and KIR genomic regions of cynomolgus macaque using third-generation long-read sequencing approach. We identified functional Mafa-B loci at the transcriptome level using locus-specific amplification in a cohort of 33 Vietnamese cynomolgus macaques. Results This is the first physical mapping of complete MHC and KIR gene regions in a Vietnamese cynomolgus macaque. Furthermore, we identified four functional Mafa-B loci (B2, B3, B5, and B6) and showed that alleles of the Mafa-I*01, -B*056, -B*034, and -B*001 functional lineages, respectively, are highly frequent in the Vietnamese cynomolgus macaque population. Conclusion The insights into the MHC and KIR haplotype organizations and the level of diversity may refine the selection of animals with specific genetic markers for future medical research.

Abstract Molecular combing technology (MCT) is an effective means for stretching DNA molecules and making them thus accessible for in situ studies. MCT uses the force exerted in the process of liquid flow via surface tension to stretch DNA molecules and spread them on solid surfaces, i.e. glass cover slips. Many DNA molecules can be stretched at the same time in parallel and neatly arranged side-by-side, making the approach convenient for statistical analysis. Accordingly, DNA replication and transcription can be studied at the single molecule level. In this paper, the principle, experimental methods, important applications, advantages and shortcuts of MCT in medical field are presented and discussed.

Abstract We propose a new Cu–Al dual-ion battery that aqueous solution composed of LiCl, CuCl and AlCl3 (LiCuAl) is used as the electrolyte, CuS is used as the cathode of aqueous aluminum ion battery for the first time and copper foil is used as the anode. The assembled Cu–Al dual-ion battery yields a reversible capacity of 538 mA h/g at 200 mA/g, and exhibits longterm cycling stability of over 200 cycles with 88.6% capacity retention at 1000 mA/g. Above excellent performance is inseparable from the three components of LiCuAl electrolyte and electrode materials. The Al-storage mechanism of CuS is proposed that the S–S bond in CuS lattice interacts with aluminum ions during the aluminum storage process. In addition, the charging and discharging process does not cause irreversible damage to the S–S bond, thus Cu–Al dual-ion battery with CuS as cathode shows great cycle stability.

PF-543 is a sphingosine kinase 1（SPHK1）inhibitor developed by Pfizer and is currently considered the most potent selective SPHK1 inhibitor. SPHK1 catalyses the production of sphingosine 1-phosphate (S1P) from sphingosine. It is the rate-limiting enzyme of S1P production, and there is substantial evidence to support a very important role for sphingosine kinase in health and disease. This review is the first to summarize the role and mechanisms of PF-543 as an SPHK1 inhibitor in anticancer, antifibrotic, and anti-inflammatory processes, providing new therapeutic leads and ideas for future research and clinical trials.

Anthocyanins have important physiological functions and are beneficial to the improvement of fruit quality in strawberry. Light is important for anthocyanin biosynthesis, and specific light quality was identified to promote anthocyanin accumulation in many fruits. However, research on the molecular mechanisms of anthocyanin accumulation regulated by light quality in strawberry remains limited. Here we described the effects of red- and blue-light irradiation on anthocyanin accumulation in strawberry. The results showed that blue light, rather than red light, could lead to the rapid accumulation of anthocyanins after exposure to light for 48 hours. The transcriptional levels of anthocyanin structural and regulatory genes displayed similar trend to the anthocyanin content. To investigate the mechanism of blue light-induced anthocyanin accumulation, the homologs of Arabidopsis blue light signal transduction components, including the blue light photoreceptor FaCRY1, an E3 ubiquitin ligase FaCOP1 and light-responsive factor FaHY5, were cloned from the strawberry cultivar ‘Benihoppe’. The protein-protein interaction of FaCRY1-FaCOP1-FaHY5 was revealed by yeast two-hybrid and fluorescence signal assays. Functional complementation analysis showed that overexpression of either FaCOP1 or FaHY5 restored the anthocyanin content and hypocotyl length in corresponding Arabidopsis mutants under blue light. Moreover, dual-luciferase assays showed that FaHY5 could increase the activity of FaRAP (anthocyanin transport gene) promoter and that this function relied on other, likely B-box protein FaBBX22, factors. The overexpression of FaHY5-VP16 (chimeric activator form of FaHY5) and FaBBX22 promoted the accumulation of anthocyanins in transgenic strawberry plants. Further, transcriptomic profiling indicated that the genes involved in the phenylpropanoid biosynthesis pathway were enriched in both FaHY5-VP16-OX and FaBBX22-OX strawberry plants. In summary, our findings provide insights into a mechanism involving the regulation of blue light-induced anthocyanin accumulation via a FaCRY1-FaCOP1-FaHY5 signal transduction module in strawberry.

Klotho (KL) is a renal protein with aging-suppression properties that mediates its regulatory effect during cardiac fibroblast aging. However, to determine whether KL can protect aged myocardial cells by attenuating ferroptosis, this study aimed to investigate the protective effect of KL on aged cells and to explore its potential mechanism. Cell injury of H9C2 cells was induced with D-galactose (D-gal) and treated with KL in vitro. This study demonstrated that D-gal induces aging in H9C2 cells. D-gal treatment increased β-GAL(β-galactosidase) activity, decreased cell viability, enhanced oxidative stress, reduced mitochondrial cristae, and decreased the expression of solute carrier family 7 member 11 (SLC7A11), glutathione peroxidase-4 (GPx4), and P53, which are primary regulators of ferroptosis. The results showed that KL can eliminate D-gal-induced aging in H9C2 cells, likely due to its ability to increase the expression of the ferroptosis-associated proteins SLC7A11 and GPx4. Moreover, pifithrin-α, a P53-specific inhibitor, enhanced the expression of SLC7A11 and GPx4. These results suggest that KL may be involved in D-gal-induced H9C2 cellular aging during ferroptosis, mainly through the P53/SLC7A11/GPx4 signaling pathway.

We propose a humidity sensor using an excessively tilted fiber grating (Ex-TFG) coated with agarose fabricated using femtosecond laser processing. The processed grating showcases remarkable differentiation between TE and TM modes, achieving an exceptionally narrow bandwidth of approximately 1.5 nm and an impressive modulation depth of up to 15 dB for both modes. We exposed the agarose-coated TFG sensor to various relative humidity levels and monitored the resonance wavelength to test its humidity sensing capability. Our findings demonstrated that the sensor exhibited a rapid response time (2–4 s) and showed a high response sensitivity (18.5 pm/%RH) between the humidity changes and the resonant wavelength shifts. The high sensitivity, linearity, repeatability, low hysteresis, and excellent long-term stability of the TFG humidity sensor, as demonstrated in our experimental results, make it an attractive option for environmental monitoring or biomedical diagnosis.

Fiber-optic biosensors have garnered significant attention and witnessed rapid development in recent years owing to their remarkable attributes such as high sensitivity, immunity to electromagnetic interference, and real-time monitoring. They have emerged as a potential tool in the realm of biomarker detection for low-concentration and small molecules. In this paper, a portable and cost-effective optical fiber biosensor based on surface plasmon resonance for the early detection of breast cancer is demonstrated. By utilizing the aptamer human epidermal growth factor receptor 2 (HER2) as a specific biomarker for breast cancer, the presence of the HER2 protein can be detected through an antigen-antibody binding technique. The detection method was accomplished by modifying a layer of HER2 aptamer on the flat surface of a gold-coated D-shaped polymer optical fiber (core/cladding diameter 120/490 μm), of which the residual thickness after side-polishing was about 245 μm, the thickness of the coated gold layer was 50 nm, and the initial wavelength in pure water was around 1200 nm. For low-concentration detection of the HER2 protein, the device exhibited a wavelength shift of ~1.37 nm with a concentration of 1 μg/mL (e.g., 5.5 nM), which corresponded to a limit of detection of ~5.28 nM. Notably, the response time of the biosensor was measured to be as fast as 5 s. The proposed biosensor exhibits the potential for early detection of HER2 protein in initial cancer serum and offers a pathway to early prevention of breast cancer.

The subtropical region of China possesses abundant broad-leaf tree species resources; however, the anatomical properties and microstructure of the wood are still unclear, which restricts the processing and utilization of wood. In this study, 14 broad-leaf trees and four coniferous trees were selected. Wood anatomical indices and wood microanatomy were used to evaluate the wood properties using a comprehensive index method. The results have shown that Dalbergia assamica exhibited the highest wood basic density among the 14 broad-leaved tree species, accompanied by a significant fiber proportion and vessel lumen diameter but a small vessel proportion and a high number of wood rays. Conversely, Parakmeria lotungensis and Michelia chapensis had relatively low wood basic densities, rendering them less suitable as valuable broad-leaved wood sources. Altingia chinensis, Castanopsis kawakamii, and the remaining 11 tree species exhibited medium-level wood basic densities. The 14 broad-leaved tree species had medium-length fibers. Phoebe bournei, Dalbergia assamica, and Castanopsis kawakamii demonstrated relatively high fiber proportion. Altingia chinensis, Dalbergia assamica, and Castanopsis kawakamii exhibited a large number of wood rays, making their wood more susceptible to cracking, whereas other broad-leaved tree species possessed fewer wood rays. The findings have provided a scientific basis for the exploration of precious broad-leaved tree resources and wood use.
