Background: The grief that accompanies witnessing the death of a child puts health care professionals at risk of secondary trauma, burnout, and turnover when left unaddressed. Objective: Support staff well-being and promote resiliency. Methods: Descriptive implementation of a structured, peer-to-peer bereavement support program for intensive care unit (ICU) staff at a tertiary children's hospital. Results: Thirty-five virtual sessions were held over the period of one year.Through these sessions, participants shared perspectives and normalized reactions, and explored potential coping strategies. Post-session feedback surveys demonstrated the negative impact of a death on the personal or work life of ICU staff. Additionally, nearly all reported some level of burnout. Conclusions: The sessions were feasible and positively impacted staff coping and well-being. Barriers and facilitators to session attendance, as well as suggestions for improvement, were also explored. Implications for practice and future research are discussed. Alzheimer's disease (AD), an age-related progressive neurodegenerative disorder, exhibits reduced cognitive function with no cure to date. One of the reasons for AD is the accumulation of Amyloid-beta 42 (Aβ42) plaque(s) that trigger aberrant gene expression and signaling, which results in neuronal cell death by an unknown mechanism(s). Misexpression of human Aβ42 in the developing retina of Drosophila exhibits AD-like neuropathology. Small non-coding RNAs, microRNAs (miRNAs), post-transcriptionally regulate the expression of their target genes and thereby regulate different signaling pathways. In a forward genetic screen, we identified miR-277 (human ortholog is hsa-miR-3660) as a genetic modifier of Aβ42-mediated neurodegeneration. Loss-of-function of miR-277 enhances the Aβ42-mediated neurodegeneration. Whereas gain-of-function of miR-277 in the GMR > Aβ42 background downregulates cell death to maintain the number of neurons and thereby restores the retinal axonal targeting defects indicating the functional rescue. In addition, gain-of-function of miR-277 rescues the eclosion- and climbing assays defects observed in GMR > Aβ42 background. Thus, gain-of-function of miR-277 rescues both structurally as well as functionally the Aβ42-mediated neurodegeneration. Furthermore, we identified head involution defective (hid), an evolutionarily conserved proapoptotic gene, as one of the targets of miR-277 and validated these results using luciferase- and qPCR -assays. In the GMR > Aβ42 background, the gain-of-function of miR-277 results in the reduction of hid transcript levels to one-third of its levels as compared to GMR > Aβ42 background alone. Here, we provide a novel molecular mechanism where miR-277 targets and downregulates proapoptotic gene, hid, transcript levels to rescue Aβ42-mediated neurodegeneration by blocking cell death. These studies shed light on molecular mechanism(s) that mediate cell death response following Aβ42 accumulation seen in neurodegenerative disorders in humans and provide new therapeutic targets for neurodegeneration., (© 2024. The pathology of Alzheimer's disease involves a long preclinical period, where the characteristic clinical symptoms of the changes in the brain are undetectable. During the preclinical period, homeostatic mechanisms may help prevent widespread cell death. Evidence has pointed towards selective cell death of diseased neurons playing a potentially protective role. As the disease progresses, dysregulation of signaling pathways that govern cell death contributes to neurodegeneration. Aberrant activation of the c-Jun N-terminal kinase pathway has been established in human and animal models of Alzheimer's disease caused by amyloid-beta 42- or tau-mediated neurodegeneration. Clonal mosaic studies in Drosophila that examine amyloid-beta 42 in a subset of neurons suggest complex interplay between amyloid-beta 42-expressing and wild-type cells. This review examines the role of c-Jun N-terminal kinase signaling in the context of cell competition and short-range signaling interactions between amyloid-beta 42-expressing and wild-type neurons. Cell competition is a conserved phenomenon regulating tissue integrity by assessing the fitness of cells relative to their neighbors and eliminating suboptimal cells. Somatic clones of amyloid-beta 42 that juxtapose genetically distinct neuronal cell populations show promise for studying neurodegeneration. Generating genetic mosaics with labeled clones of amyloid-beta 42- or tau-expressing and wild-type neurons will allow us to understand how short-range signaling alterations trigger cell death in neurons and thereby contribute to the progression of Alzheimer's disease. Structure-activity relationship studies involving N-aryl-3-trifluoromethyl pyrido[1,2- a]benzimidazoles (PBI) identified several compounds possessing potent in vitro activities against the asexual blood, liver, and gametocyte stages of the Plasmodium parasite with no cross-resistance to chloroquine. Frontrunner lead compounds with good in vitro absorption, distribution, metabolism, and excretion (ADME) profiles were subjected to in vivo proof-of-concept studies in NMRI mice harboring the rodent P. berghei infection. This led to the identification of compounds 10 and 49, effecting 98% and 99.93% reduction in parasitemia with mean survival days of 12 and 14, respectively, at an oral dose of 4 × 50 mg/kg. In vivo pharmacokinetics studies on 10 revealed slow absorption, low volume of distribution, and low clearance profiles. Furthermore, this series displayed a low propensity to inhibit the human ether-a-go-go-related gene (hERG) potassium ion channel whose inhibition is associated with cardiotoxicity.

Further structure-activity relationship (SAR) studies on the recently identified pyrido[1,2-a]benzimidazole (PBI) antimalarials have led to the identification of potent, metabolically stable compounds with improved in vivo oral efficacy in the P. berghei mouse model and additional activity against parasite liver and gametocyte stages, making them potential candidates for preclinical development. Inhibition of hemozoin formation possibly contributes to the mechanism of action.

There is an urgent need for the development of new antimalarial compounds. As a result of a phenotypic screen, several compounds with potent activity against the parasite Plasmodium falciparum were identified. Characterization of these compounds is discussed, along with approaches to optimise the physicochemical properties. The in vitro antimalarial activity of these compounds against P. falciparum K1 had EC50 values in the range of 0.09-29 μM, and generally good selectivity (typically >100-fold) compared to a mammalian cell line (L6). The quantity and diversity of the microbial community in soil make it possibly the most difficult of all the natural ecosystems. It is thought to be challenging to culture up to 99% of the microorganisms in a given environment. The intricacy of microbial variety is impacted by numerous interconnected factors, including as soil structure, water content, biotic activity, pH, and fluctuations in climate. Environmental DNA isolation and purification are often the first steps in the soil metagenomic analysis process. Creating genomic DNA libraries and then using them for high-throughput sequencing or library screening are the main steps in the application of metagenomics. These genomic sequences are currently being used to advance our knowledge of the ecology and physiology of these bacteria as well as for new biotechnological and medicinal applications. To completely comprehend the intricacies involved in the operation of microbial communities and the interactions between different microorganisms within specific niches, metagenomic sequences are employed. This study focuses on the latest advancements in biotechnological approaches and procedures for identifying novel genes from uncultured microorganisms and intricate microbial habitats. This paper uses uniquely detailed large-scale commercial real estate investment data to examine how financial markets perceived the attractiveness of investing in UK regions during the last two decades. Our analysis demonstrates that prior to the 2008 Global Financial Crisis all regions of the UK were perceived similarly in terms of risks, whereas the crisis engendered a flight to safety of capital into London. Other UK regions shifted rapidly into junk bond territory, and have remained there ever since. These asymmetric capital shocks led up to a profound and adverse shift in the subsequent productivity growth of the UK regional economies. [ABSTRACT FROM AUTHOR]

With the amendment to the German Climate Change Act in 2021, the Federal Government of Germany has set the target to become greenhouse gas neutral by 2045. Reaching this ambitious target requires multisectoral efforts, which in turn calls for interdisciplinary collaboration: the Net-Zero-2050 project of the Helmholtz Climate Initiative serves as an example of successful, interdisciplinary collaboration with the aim of producing valuable recommendations for action to achieve net-zero CO2 emissions in Germany. To this end, we applied an interdisciplinary approach to combining comprehensive research results from ten German national research centers in the context of carbon neutrality in Germany. In this paper, we present our approach and the method behind the interdisciplinary storylines development, which enabled us to create a common framework between different carbon dioxide removal and avoidance methods and the bigger carbon neutrality context. Thus, the research findings are aggregated into narratives: the two complementary storylines focus on technologies for net-zero CO2 emissions and on different framing conditions for implementing net-zero CO2 measures. Moreover, we outline the Net-Zero-2050 results emerging from the two storylines by presenting the resulting narratives in the context of carbon neutrality in Germany. Aiming at creating insights into how complementary and related expertise can be combined in teams across disciplines, we conclude with the project's lessons learned. This paper sheds light on how to facilitate cooperation between different science disciplines with the purpose of preparing joint research results that can be communicated to a specific audience. Additionally, it provides further evidence that interdisciplinary and diverse research teams are an essential factor for defining solution spaces for complex, interdisciplinary problems. The climate mitigation potential of terrestrial carbon dioxide removal (tCDR) methods depends critically on the timing and magnitude of their implementation. In our study, we introduce different measures of efficiency to evaluate the carbon removal potential of afforestation and reforestation (AR) and bioenergy with carbon capture and storage (BECCS) under the low-emission scenario SSP1-2.6 and in the same area. We define efficiency as the potential to sequester carbon in the biosphere in a specific area or store carbon in geological reservoirs or woody products within a certain time. In addition to carbon capture and storage (CCS), we consider the effects of fossil fuel substitution (FFS) through the usage of bioenergy for energy production, which increases the efficiency through avoided CO2 emissions. These efficiency measures reflect perspectives regarding climate mitigation, carbon sequestration, land availability, spatiotemporal dynamics, and the technological progress in FFS and CCS. We use the land component JSBACH3.2 of the Max Planck Institute Earth System Model (MPI-ESM) to calculate the carbon sequestration potential in the biosphere using an updated representation of second-generation bioenergy plants such as Miscanthus. Our spatially explicit modeling results reveal that, depending on FFS and CCS levels, BECCS sequesters 24–158 GtC by 2100, whereas AR methods sequester around 53 GtC on a global scale, with BECCS having an advantage in the long term. For our specific setup, BECCS has a higher potential in the South American grasslands and southeast Africa, whereas AR methods are more suitable in southeast China. Our results reveal that the efficiency of BECCS to sequester carbon compared to "nature-based solutions" like AR will depend critically on the upscaling of CCS facilities, replacing fossil fuels with bioenergy in the future, the time frame, and the location of tCDR deployment. Purpose: In 2020, mining activity by Rio Tinto destroyed rock caves in Western Australia's Juukan Gorge that are considered sacred sites by the First Nation Peoples of that area, the Puutu Kunti Kurrama and Pinikura (PKKP) Peoples. This paper examines the public response to the damage caused at this culturally sensitive site and identifies cultural heritage protection strategies that emerged in the aftermath of this catastrophic event. Design/methodology/approach: This research applies a qualitative case study method and analysis of open-sourced official policy documents, media reports and published institutional statements. Findings: The research identified specific cultural heritage protection strategies, including stakeholder-driven advocacy and shared values approach to business practices to help foster a greater appreciation of the connections between people, objects and lands. Whilst the mining activities were considered lawful, significant gaps in the legislation to protect heritage sites were also exposed. Originality/value: Using a recent case that occurred in 2020, this paper unpacks how the motivations for accessing minerals can override cultural sensibilities and legal/ethical frameworks established to protect cultural heritage. This paper brings to light the liabilities associated with the mining industry when operating in a culturally significant environment where appropriate due diligence to manage cultural heritage is not thoroughly applied. The paper highlights the role the community can play in demanding improved corporate social responsibility which can, in turn, act as a strategy for cultural heritage protection. This study investigates the impact of organic (OF) and conventional farming (CF) on soil fungal communities in banana monoculture plantations on Madeira Island. We hypothesized that OF promotes beneficial fungal groups over harmful ones, sustaining soil health. Soil samples were collected from six plantations (three OF and three CF) for ITS amplicon sequencing to assess fungal diversity. Results showed that OF significantly enhanced fungal alpha-diversity (Shannon–Wiener index) and Evenness. The phylum Ascomycota dominated OF systems, while Basidiomycota prevailed in CF. Mortierella, a beneficial genus, was abundant in OF and is observed in CF but was less evident, being the genus Trechispora the most well represented in CF agrosystems. Additionally, OF was associated with higher soil pH and Mg levels, which correlated positively with beneficial fungal groups. Functional analysis revealed that OF promoted saprotrophic fungi, crucial for the decomposition of organic matter and nutrient cycling. However, both systems exhibited low levels of arbuscular mycorrhizal fungi, likely due to high phosphorus levels. These findings suggest that organic practices can enhance soil fungal diversity and health, although attention to nutrient management is critical to further improving soil–plant–fungi interactions. Agricultural soils are responsible for ecological functions and services that include primary production of food, fiber and fuel, nutrient cycling, carbon cycling and storage, water infiltration and purification, among others. Fungi are important drivers of most of those ecosystem services. Given the importance of fungi in agricultural soils, in this study, we aimed to characterize and analyse the changes of the soil fungal communities of three cropping systems from Madeira Island, where family farming is predominant, and investigate the response of fungi and its functional groups to soil physicochemical properties. To achieve that, we sequenced amplicons targeting the internal transcribed spacer 1 (ITS1) of the rRNA region, to analyse soil samples from 18 agrosystems: 6 vineyards (V), 6 banana plantations (B) and 6 vegetable plantations (H). Our results showed that alpha diversity indices of fungal communities are similar in the three cropping systems, but fungal composition and functional aspects varied among them, with more pronounced differences in B. Ascomycota, Basidiomycota, and Mortierellomycota were the main phyla found in the three cropping systems. Agaricomycetes and Sordariomycetes are the predominant classes in B, representing 23.8 and 22.4%, respectively, while Sordariomycetes (27.9%) followed by Eurotiomycetes (12.3%) were the predominant classes in V and Sordariomycetes (39.2%) followed by Tremellomycetes (8.9%) in the H. Saprotrophs are the fungal group showing higher relative abundance in the three cropping systems, followed by plant pathogens. Regarding symbionts, endophytes were highly observed in B, while mycorrhizal fungi was predominant in V and H. The structure of fungal communities was mainly correlated with soil content of P, K, N, Fe, and Cu. In addition, we identified bioindicators for each cropping system, which means that cultivated crops are also drivers of functional groups and the composition of communities. Overall, the three cropping systems favored diversity and growth of taxa that play important roles in soil, which highlights the importance of conservative management practices to maintain a healthy and resilient agrosystem. The substance principal used to synthesize these materials is fibroin, obtained from the silkworm's cocoon (Bombyx mori) in the form of a filament, but can be converted into nanometric particles. After the synthesis, the obtained samples were characterized by size and Zeta potential (via nanosizer), X-Ray diffraction (XRD), specific area determination by the BET method, Infrared Spectroscopy with Fourier Transform with attenuated diffuse reflectance (FTIR), transmission electron microscopy (TEM). The silk fibroin nanoparticles (SFNp) were immobilized on polylactic acid (PLA)-knitted fabric and functionalized with chitosan for FTIR and XRD analyses, in addition to cytotoxicity analysis with colorimetric test (MTT) and cell adhesion. The DLS, FTIR and MET results confirm the formation of spherical silk fibroin nanoparticles with average sizes between 48 and 156nm, and zeta potential of − 19.95mV which characterizes a good dispersibility. Cell-viability assessment showed that SFNp is a non-cytotoxic material at low concentrations up to 48h, confirming that the nanoparticle synthesis method using the ternary system is an effective and low-cost alternative. This technique has great potential for use in the development of biomaterials, due to the production of knitted tissues with PLA functionalized with chitosan and immobilized with SFNp, exhibiting mechanical strength and biocompatibility. Soil moisture movement reveals the hydrological environment and irrigation characteristics of plants, which is very important and basic hydrological problem. A soil moisture movement algorithm based on cellular automata suitable for greenhouse was proposed to simulate the lateral flow and vertical penetration of soil water in different soil layers. Artemisia annua was used as an experimental plant, and two numerical simulations were set up to determine the minimum water replenishment and the degree of upward soil modification. To verify the performance of the model, corn was used as an experimental plant for application simulation. The numerical simulation results show that the effect of only water for plant cells is superior to soil cells, and the disturbance behavior of the underlying soil layer should be minimized during the cultivation process. The average MAE/RMSE of all soil layers is 0.86/1.06 and in deep layer (50 cm and 60 cm) is 0.53/0.64. It shows the model has a certain prediction and simulation ability, especially in deep soil layers. The proposed algorithm can simply calculate soil flow, set the minimum water replenishment, and evaluate the water replenishment efficiency, which can provide a theoretical reference for the water replenishment and soil replacement scheme. This paper argues that a new 'domestic regime' has taken hold in Anglo-American economies since 2008. The combination of austere fiscal policy and loose monetary policy produced sustained house-price inflation in an otherwise stagnant economy. This combined with broadband-enabled digital platforms, a highly flexible labour market, and a routinely undervalued social reproductive sector to transform how capitalism operates in these economies. Taking the United Kingdom as a central case, our goal in this paper is to articulate what constitutes a 'domestic regime', and to locate this concept within wider sociological and political-economic debates. To adequately grasp the contemporary regime, we suggest that it needs to be considered in all its multifaceted, interlocking dimensions: the financial, the infrastructural, the reproductive and the productive. Designing low-cost network layouts is an essential step in planning linked infrastructure. For the case of capacitated trees, such as oil or gas pipeline networks, the cost is usually a function of both pipeline diameter (i.e. ability to carry flow or transferred capacity) and pipeline length. Even for the case of incompressible, steady flow, minimizing cost becomes particularly difficult as network topology itself dictates local flow material balances, rendering the optimization space non-linear. The combinatorial nature of potential trees requires the use of graph optimization heuristics to achieve good solutions in reasonable time. In this work we perform a comparison of known literature network optimization heuristics and metaheuristics for finding minimum-cost capacitated trees without Steiner nodes, and propose novel algorithms, including a metaheuristic based on transferring edges of high valency nodes. Our metaheuristic achieves performance above similar algorithms studied, especially for larger graphs, usually producing a significantly higher proportion of optimal solutions, while remaining in line with time-complexity of algorithms found in the literature. Data points for graph node positions and capacities are first randomly generated, and secondly obtained from the German emissions trading CO2 source registry. As political will for applications and storage for hard-to-abate industry CO2 emissions is growing, efficient network design methods become relevant for new large-scale CO2 pipeline networks. This study investigates the escape of Mercury's sodium‐group ions (Na+‐group, including ions with m/q from 21 to 30 amu/e) and their dependence on true anomaly angle (TAA), that is, Mercury's orbital phase around the Sun, using measurements from MESSENGER. The measurements are categorized into solar wind, magnetosheath, and magnetosphere, and further divided into four TAA intervals. Na+‐group ions form escape plumes in the solar wind and magnetosheath, with higher fluxes along the solar wind's motional electric field. The total escape rates vary from 0.2 to 1 × 1025 atoms/s with the magnetosheath being the main escaping region. These rates exhibit a TAA dependence, peaking near the perihelion and similar during Mercury's remaining orbit. Despite Mercury's tenuous exosphere, Na+‐group ions escape rate is comparable to other inner planets. This can be attributed to several processes, including that Na+‐group ions may include several ion species, efficient photoionization frequency for elements within Na‐group, etc. Plain Language Summary: Atmospheric escape is defined as the loss of atmospheric particles in the form of neutrals and ions into outer space. Understanding atmospheric escape is a fundamental science question for studying the evolution of planetary atmosphere and habitability, as it can provide insight into how life can form on a planet. While atmospheric escape has been extensively studied in Venus, Earth, and Mars through in situ measurements and simulations, it remains poorly understood at Mercury. In this study, we investigate the escape of the most abundant heavy ions at Mercury, specifically the Na+‐group ions, using MESSENGER's measurements. Our findings show that the escape rates of the Na+‐group ions are dependent on Mercury's orbital phase around the Sun, exhibiting a seasonal effect with rates from 0.2 to 1 × 1025 atoms/s. This rate is comparable to the ion's escape rates at other inner planets, which is surprising given that Mercury only has a tenuous exosphere. Phosphorus (P) is an important and irreplaceable macronutrient. It is central to energy and information storage and exchange in living cells. P is an element with a "broken geochemical cycle" since it lacks abundant volatile compounds capable of closing the P cycle. P fertilizers are critical for global food security, but the reserves of minable P are scarce and non-evenly distributed between countries of the world. Accordingly, the risks of global crisis due to limited access to P reserves are expected to be graver than those entailed by competition for fossil hydrocarbons. Paradoxically, despite the scarcity and value of P reserves, its usage is extremely inefficient: the current waste rate reaches 80% giving rise to a plethora of unwanted consequences such as eutrophication leading to harmful algal blooms. Microalgal biotechnology is a promising solution to tackle this challenge. The proposed review briefly presents the relevant aspects of microalgal P metabolism such as cell P reserve composition and turnover, and the regulation of P uptake kinetics for maximization of P uptake efficiency with a focus on novel knowledge. The multifaceted role of polyPhosphates, the largest cell depot for P, is discussed with emphasis on the P toxicity mediated by short-chain polyPhosphates. Opportunities and hurdles of P bioremoval via P uptake from waste streams with microalgal cultures, either suspended or immobilized, are discussed. Possible avenues of P-rich microalgal biomass such as biofertilizer production or extraction of valuable polyPhosphates and other bioproducts are considered. Background: Social isolation and loneliness can occur in all age groups, and they are linked to increased mortality and poorer health outcomes. There is a growing body of research indicating inconsistent findings on the effectiveness of interventions aiming to alleviate social isolation and loneliness. Hence the need to facilitate the discoverability of research on these interventions. Objectives: To map available evidence on the effects of in‐person interventions aimed at mitigating social isolation and/or loneliness across all age groups and settings. Search Methods: The following databases were searched from inception up to 17 February 2022 with no language restrictions: Ovid MEDLINE, Embase, EBM Reviews—Cochrane Central Register of Controlled Trials, APA PsycInfo via Ovid, CINAHL via EBSCO, EBSCO (all databases except CINAHL), Global Index Medicus, ProQuest (all databases), ProQuest ERIC, Web of Science, Korean Citation Index, Russian Science Citation Index, and SciELO Citation Index via Clarivate, and Elsevier Scopus. Selection Criteria: Titles, abstracts, and full texts of potentially eligible articles identified were screened independently by two reviewers for inclusion following the outlined eligibility criteria. Data Collection and Analysis: We developed and pilot tested a data extraction code set in Eppi‐Reviewer. Data was individually extracted and coded. We used the AMSTAR2 tool to assess the quality of reviews. However, the quality of the primary studies was not assessed. Main Results: A total of 513 articles (421 primary studies and 92 systematic reviews) were included in this evidence and gap map which assessed the effectiveness of in‐person interventions to reduce social isolation and loneliness. Most (68%) of the reviews were classified as critically low quality, while less than 5% were classified as high or moderate quality. Most reviews looked at interpersonal delivery and community‐based delivery interventions, especially interventions for changing cognition led by a health professional and group activities, respectively. Loneliness, wellbeing, and depression/anxiety were the most assessed outcomes. Most research was conducted in high‐income countries, concentrated in the United States, United Kingdom, and Australia, with none from low‐income countries. Major gaps were identified in societal level and community‐based delivery interventions that address policies and community structures, respectively. Less than 5% of included reviews assessed process indicators or implementation outcomes. Similar patterns of evidence and gaps were found in primary studies. All age groups were represented but more reviews and primary studies focused on older adults (≥60 years, 63%) compared to young people (≤24 years, 34%). Two thirds described how at‐risk populations were identified and even fewer assessed differences in effect across equity factors for populations experiencing inequities. Authors' Conclusions: There is growing evidence that social isolation and loneliness are public health concerns. This evidence and gap map shows the available evidence, at the time of the search, on the effectiveness of in‐person interventions at reducing social isolation and loneliness across all ages and settings. Despite a large body of research, with much of it published in more recent years, it is unevenly distributed geographically and across types of interventions and outcomes. Most of the systematic reviews are of critically low quality, indicating the need for high quality reviews. Introduced plants create lasting abiotic and biotic changes to soil that are increasingly implicated as major drivers of invasion success, yet models for investigating patterns in the impacts of these soil legacies are lacking. Here, we used Bohemian knotweed (Reynoutria × bohemica) to present a novel approach for testing whether accounting for diverse evolutionary drivers can clarify how an invader's soil legacy mediates its success. For most tested species, R. × bohemica's soil legacy did not influence seedling performance irrespective of its evolutionary history or the evolutionary novelty of species that share its soil. However, we found the relative importance of legacy effects versus other mechanisms of invader dominance depends on the life stage of the plants growing in soil conditioned by the invader. While the effects of R. × bohemica's soil legacy on seedling performance were context dependent, emergence was, on average, higher in R. × bohemica‐conditioned soils than in unconditioned soils. Thus, contrary to theory, R. × bohemica's soil legacy not only fails to impede potential competitors but may also facilitate them. Our results are particularly powerful: we detected novel insights in the face of biological complexity, even for R. × bohemica, which likely does not depend on heterospecific plant–soil feedbacks for its competitive dominance in its introduced range. Designing rigorous investigations that explore how plant species' evolutionary histories mediate an invader's soil legacy across diverse abiotic environments, as shown here, is key for producing insights in other systems. Encapsulins are protein nanocompartments that regulate cellular metabolism in several bacteria and archaea. Myxococcus xanthus encapsulins protect the bacterial cells against oxidative stress by sequestering cytosolic iron. These encapsulins are formed by the shell protein EncA and three cargo proteins: EncB, EncC, and EncD. EncB and EncC form rotationally symmetric decamers with ferroxidase centers (FOCs) that oxidize Fe+2 to Fe+3 for iron storage in mineral form. However, the structure and function of the third cargo protein, EncD, have yet to be determined. Here, we report the x-ray crystal structure of EncD in complex with flavin mononucleotide. EncD forms an α-helical hairpin arranged as an antiparallel dimer, but unlike other flavin-binding proteins, it has no ß-sheet, showing that EncD and its homologs represent a unique class of bacterial flavin-binding proteins. The cryo-EM structure of EncA-EncD encapsulins confirms that EncD binds to the interior of the EncA shell via its C-terminal targeting peptide. With only 100 amino acids, the EncD α-helical dimer forms the smallest flavin-binding domain observed to date. Unlike EncB and EncC, EncD lacks a FOC, and our biochemical results show that EncD instead is a NAD(P)H-dependent ferric reductase, indicating that the M. xanthus encapsulins act as an integrated system for iron homeostasis. Overall, this work contributes to our understanding of bacterial metabolism and could lead to the development of technologies for iron biomineralization and the production of iron-containing materials for the treatment of various diseases associated with oxidative stress. Protein capsids are a widespread form of compartmentalization in nature. Icosahedral symmetry is ubiquitous in capsids derived from spherical viruses, as this geometry maximizes the internal volume that can be enclosed within. Despite the strong preference for icosahedral symmetry, we show that simple point mutations in a virus-like capsid can drive the assembly of unique symmetry-reduced structures. Starting with the encapsulin from Myxococcusxanthus, a 180-mer bacterial capsid that adopts the well-studied viral HK97 fold, we use mass photometry and native charge detection mass spectrometry to identify a triple histidine point mutant that forms smaller dimorphic assemblies. Using cryoelectron microscopy, we determine the structures of a precedented 60-mer icosahedral assembly and an unexpected 36-mer tetrahedron that features significant geometric rearrangements around a new interaction surface between capsid protomers. We subsequently find that the tetrahedral assembly can be generated by triple-point mutation to various amino acids and that even a single histidine point mutation is sufficient to form tetrahedra. These findings represent a unique example of tetrahedral geometry when surveying all characterized encapsulins, HK97-like capsids, or indeed any virus-derived capsids reported in the Protein Data Bank, revealing the surprising plasticity of capsid self-assembly that can be accessed through minimal changes in the protein sequence. This article describes dynamical simulations of the assembly of an icosahedral protein shell around a bicomponent fluid cargo. Our simulations are motivated by bacterial microcompartments, which are protein shells found in bacteria that assemble around a complex of enzymes and other components involved in certain metabolic processes. The simulations demonstrate that the relative interaction strengths among the different cargo species play a key role in determining the amount of each species that is encapsulated, their spatial organization, and the nature of the shell assembly pathways. However, the shell protein–shell protein and shell protein–cargo component interactions that help drive assembly and encapsulation also influence cargo composition within certain parameter regimes. These behaviors are governed by a combination of thermodynamic and kinetic effects. In addition to elucidating how natural microcompartments encapsulate multiple components involved within reaction cascades, these results have implications for efforts in synthetic biology to colocalize alternative sets of molecules within microcompartments to accelerate specific reactions. More broadly, the results suggest that coupling between self-assembly and multicomponent liquid–liquid phase separation may play a role in the organization of the cellular cytoplasm. Societal Impact Statement: Plants and bacteria interact in complex ways that are crucial to the health and productivity of native vegetation and croplands. While the range of characterised plant‐beneficial bacterial traits continues to grow, key questions remain regarding the distribution and mobility of genes associated with these traits. This work explores the diversity of mobile genetic elements carried by bacteria associated with plant root surfaces, assessing their capacity to help shape plant–bacterial interactions. The significance of this work lies in the potential to contribute to new strategies for enhancing plant health, promoting sustainable agriculture and managing plant diseases in an era when we must respond to environmental change. Summary: Integrons are gene capture and expression systems that contribute to bacterial adaptation. Integron research has mainly focused on the role that these elements play in spreading antimicrobial resistance. However, their contribution to niche adaptation is potentially much broader because integrons can sample the vast repertoire of diverse functions encoded by integron gene cassettes. Integrons and gene cassettes have been identified in many bacterial lineages residing in soil and water across varied ecosystems, but there has been little investigation of integrons in plant‐associated bacteria.Bacteria and plants have complex, dynamic relationships that influence plant health and productivity. To investigate whether integrons contribute to adaptative processes in plant microbiomes, we examined gene cassette and microbial taxonomic profiles in rhizoplanes of four important crop species grown under controlled glasshouse conditions. We identified 38,546 unique gene cassettes, including elements carrying genes associated with antibiotic resistance, type II toxin–antitoxin systems and genes with putative functions associated with plant growth promotion, along with a larger set encoding genes of unknown functions.Rhizoplane microbiomes of different plant species showed more similarity in their community composition profiles than in their gene cassette profiles, with complex and distinct suites of gene cassettes associated with each plant species, suggesting that gene cassettes might have a role in specific plant–bacterial interactions.We show that rhizoplane microbiomes carry diverse integron gene cassettes that could play a role in establishing and maintaining rhizoplane communities. Microbial diversity profoundly influences soil ecosystem functions, making it vital to monitor community dynamics to comprehend its structure. Our study focused on six agrosystems in Madeira Archipelago, analysing bacteria, archaea, fungi and AMF through classical microbiology and molecular techniques. Despite distinct edaphoclimatic conditions and management practices, bacterial structures exhibited similarities, with Alphaproteobacteria at 18%–20%, Bacilli at 11%–18% and Clostridia at 9%–14%. The predominance of copiothrophic groups suggested that soil nutrient content was the driver of these communities. Regarding archaea, the communities changed among sites, and it was evident that agrosystems provided niches for methanogens. The Crenarchaeota varied between 15% and 29%, followed by two classes of Euryarchaeota, Methanomicrobia (17%–25%) and Methanococci (4%–32%). Fungal communities showed consistent composition at the class level but had differing diversity indices due to management practices and soil texture. Sordaryomycetes (21%–28%) and Agaricomycetes (15%–23%) were predominant. Conversely, AMF communities appeared to be also influenced by the agrosystem, with Glomus representing over 50% of the community in all agrosystems. These insights into microbial groups' susceptibilities to environmental conditions are crucial for maintaining healthy soil and predicting climate change effects on agrosystems' productivity, resilience and sustainability. The most common aging-related neurodegenerative disorder is Alzheimer's disease (AD), of which the main symptom is memory disturbance. Though the mechanism of AD pathogenesis is not fully defined, abnormal aggregation of amyloid beta (Aβ) plaques and tau have been considered as key factors and main histological hallmarks of the disease. Astrocyte is responsible for the control of cells and the environment around brain and spinal cord cells. Astrocytes have been implicated with AD. However, the exact function of astrocytes in AD has not been established. In this study, we investigated the regulation of astrocytes in the AD model using primary cultures. We have demonstrated that oligomerized Aβ is toxic to neurons and can induce cell death in primary cultures. In the primary cultures containing neurons and astrocytes, amyloid beta uptake was observed in both neurons and astrocytes. To verify if the uptake of amyloid beta in astrocytes is dependent on neurons, we separated and cultured primary astrocytes with no neurons. Amyloid uptake was still observed in this pure astrocyte culture, suggesting that the uptake of amyloid beta is a neuron-independent function of astrocytes. Astrocyte activation was observed in both pure and mixed cultures. Taken together, our data suggest that astrocyte is activated by oligomerized Aβ and uptakes it, which is independent of neurons. Wearable electronic textiles (e‐textiles) have emerged as a transformative technology revolutionizing healthcare monitoring and communication by seamlessly integrating with the human body. However, their practical application has been limited by the lack of compatible and sustainable power sources. Various energy sources, including solar, thermal, mechanical, and wind, have been explored for harvesting, leading to diverse energy harvesting technologies, such as photovoltaic, thermoelectric, piezoelectric, and triboelectric systems. Notably, 2D materials have gained significant attention as attractive candidates for energy harvesting and storage in e‐textiles due to their unique properties, such as high surface‐to‐volume ratio, mechanical strength, and electrical conductivity. This comprehensive review explores the utilization of 2D materials in textile‐based energy harvesters, covering their preparation, fabrication, and characterization strategies. Foam injection has been suggested and demonstrated as a potential gas‐based solution for enhancing oil recovery from underground reservoirs. While the pore‐scale behavior of foam has been investigated for many years, most of those studies were done at room temperature and pressure. A high‐pressure high‐temperature (HPHT) cell was constructed to relax this contributing simplification and evaluate foam behavior under reservoir conditions. The performance of foam injection into a fractured reservoir was investigated experimentally at vertical and horizontal pathways utilizing a 2D microfluidic device. The results showed foam had a higher lamellae density in the fracture network and could direct fluids to the matrix. This was mainly attributed to the created pressure drop in the fractured media dictated by foam self‐tuning ability, which is a higher viscosity in areas with higher conductivity. Generating a viscous crossflow in horizontal tests leads to enhanced recovery relative to gas injection. But the presence of gravity in the vertical tests causes the drainage of the foam film and the segregation of the gas and surfactant solution due to gravity, which ultimately reduces the stability of the foam. Comparing the performance of gas and foam in the vertical injection scenario reveals that foam injection results in a considerably more enhanced oil recovery. The oleic phase had a detrimental impact on foam strength and reduced fluids diversion from fracture to matrix. The sizes of gas bubbles were also larger in the presence of the oleic phase. Removal of atmospheric carbon dioxide is being considered a suitable option for reducing the recent global rise in atmospheric temperature. The impact of the removal on some climate parameters—near-surface air temperature (TAS), maximum near-surface air temperature (TASMAX), minimum near-surface air temperature (TASMIN) and surface temperature (TS) over West Africa was assessed in this paper. We used CNRM-ESM1-C1 model simulation output consisting of 1%yr−1 CO2 removal from the atmosphere which was compared with CRU observational dataset. Four climatological periods 1990–2019 (reference period), 2040–2069, 2070–2099 and 2100–2129 were considered, and hence the impacts levels in each of the two West African regions, Sahel and Guinea, were estimated in each period with respect to the reference period. The comparison with CRU demonstrated that CNRM-ESM1-C1 model captured temperature variations within major locations in Mauritania, Mali, Niger, Burkina Faso and Senegal with an indication of an underestimation of temperature at locations above 18° N. The value of each parameter was projected to decrease progressively the periods and much impacts were also projected in the last period for the two regions. Time of retreat to 2 °C reduction target is projected a decade before the year 2100 and will occur earlier with greater impact in the Guinea region than in Sahel region. The root mean square deviation of each ensemble member was found at RMSD < 0.5 with respect to the model ensemble mean per parameter, although RMSD > 0.5 was found with GFDL-ESM4 model for TAS and TS. Naturally derived alkaloids belong to a class of quite significant organic compounds. Coptisine, a benzyl tetrahydroisoquinoline alkaloid, is one of the major bioactive constituents in Coptis chinensis Franch., which is a famous traditional Chinese medicine. C. chinensis possesses many kinds of functions, including the ability to eliminate heat, expel dampness, purge fire, and remove noxious substances. In Asian countries, C. chinensis is traditionally employed to treat carbuncle and furuncle, diabetes, jaundice, stomach and intestinal disorders, red eyes, toothache, and skin disorders. Up to now, there has been plenty of research of coptisine with respect to its pharmacology. Nevertheless, a comprehensive review of coptisine-associated research is urgently needed. This paper was designed to summarize in detail the progress in the research of the pharmacology, pharmacokinetics, safety, and formulation of coptisine. The related studies included in this paper were retrieved from the following academic databases: The Web of Science, PubMed, Google scholar, Elsevier, and CNKI. The cutoff date was January 2023. Coptisine manifests various pharmacological actions, including anticancer, antimetabolic disease, anti-inflammatory disease, and antigastrointestinal disease effects, among others. Based on its pharmacokinetics, the primary metabolic site of coptisine is the liver. Coptisine is poorly absorbed in the gastrointestinal system, and most of it is expelled in the form of its prototype through feces. Regarding safety, coptisine displayed potential hepatotoxicity. Some novel formulations, including the β -cyclodextrin-based inclusion complex and nanocarriers, could effectively enhance the bioavailability of coptisine. The traditional use of C. chinensis is closely connected with the pharmacological actions of coptisine. Although there are some disadvantages, including poor solubility, low bioavailability, and possible hepatotoxicity, coptisine is still a prospective naturally derived drug candidate, especially in the treatment of tumors as well as metabolic and inflammatory diseases. The vertical flow constructed wetlands (VFCW) for the treatment of domestic wastewater has become a conventional and cost effective treatment system with one of the major disadvantage of elevated nitrate concentrations. The present study makes an effort in providing a new design of anaerobic denitrification unit termed as anaerobic chamber (AC) which was introduced after two-stage VFCW to remove nitrates from the treated wastewater (WW). The AC provided all the essential conditions of effective denitrification such as anaerobic environment with enough carbon and nitrogen source. To understand the pollutant removal mechanism in AC, microbial diversity and functional annotation was studied by metagenomic analysis of sequences obtained from biofilm formed in AC. The efficiency of AC was measured with respect to physicochemical wastewater quality parameters. The removal efficiencies were 88, 65, 43 and 27% for total nitrogen, nitrate (NO3), ammoniacal-nitrogen (NH4) and ortho-phosphate respectively. The microbial flora was much more diverse and unique pertaining to anaerobic microbes in AC compared to WW with total of 954 and 1191 genuses respectively with minimum abundance of 10 hits. The metagenomes exhibited 188% more Archaea in the AC than WW where Crenarchaeota, Euryarchaeota, Korarchaeota, Nanoarchaeota and Thaumarchaeota were major phyla with 60 genuses. The nitrogen metabolism was reported in terms of assimilatory nitrate reductase. As the class, Proteobacteria, Actinobacteria were prominent in WW, whereas Proteobacteria, Chloroflexi in AC were abundant. Cells have used compartmentalization to implement complex biological processes involving thousands of enzyme cascade reactions. Enzymes are spatially organized into the cellular compartments to carry out specific and efficient reactions in a spatiotemporally controlled manner. These compartments are divided into membrane-bound and membraneless organelles. Mimicking such cellular compartment systems has been a challenge for years. A variety of artificial scaffolds, including liposomes, polymersomes, proteins, nucleic acids, or hybrid materials have been used to construct artificial membrane-bound or membraneless compartments. These artificial compartments may have great potential for applications in biosynthesis, drug delivery, diagnosis and therapeutics, among others. This review first summarizes the typical examples of cellular compartments. In particular, the recent studies on cellular membraneless organelles (biomolecular condensates) are reviewed. We then summarize the recent advances in the construction of artificial compartments using engineered platforms. Finally, we provide our insights into the construction of biomimetic systems and the applications of these systems.
