27. Experimental study of dispersive clay modified by calcium lignosulfonate

Abstract

Dispersive clay is a type of water-sensitive soil, which has the characteristics of dispersivity and material loss when encountering water. The influential factors and modification mechanism of dispersive clay modified by calcium lignosulfonate (CL) were studied using dispersivity test, mechanical property test, chemical property test, microstructure test and simulated rainfall scouring test. The test results indicated that with the increase in CL content, the dispersivity, slake durability and erosion resistance of the modified soil were gradually improved, and it had a good modification effect when the CL content reached 3.0%. The unconfined compressive strength (UCS) first increased and then decreased, while the coefficient of consolidation first decreased and then increased, and both achieved extreme values at the CL content of 0.5%. With the increase in curing age, the dispersivity and slake durability of the modified soil decreased gradually, the coefficient of consolidation reduced significantly, while the UCS increased gradually. At the curing age of 28 d, the UCS values of the modified soil with the CL contents of 0.5% and 3.0% increased by 50% and 20%, respectively, compared with that of dispersive clay. The engineering properties of dispersive clay were improved by the addition of CL mainly through reduction of thickness of electric double layer, cation bridging, particle cementation and hydrophobic effect of hydrophobic group. However, when the CL content was too high, the CL would preferentially combine with itself and weaken the attraction between soil particles, which would increase the porosity and decrease the mechanical properties of soil. It is concluded that CL has a good modification effect on the dispersive clay, in terms of significant improvement of water sensitivity, water stability and erosion resistance of dispersive clay.

Abstract

Enhancing influenza vaccine cross-protection is imperative to alleviate the significant public health burden of influenza. Heterologous sequential immunization may synergize diverse vaccine formulations and routes to improve vaccine potency and breadth. Here we investigate the effects of immunization strategies on the generation of cross-protective immune responses in female Balb/c mice, utilizing mRNA lipid nanoparticle (LNP) and protein-based PHC nanoparticle vaccines targeting influenza hemagglutinin. Our findings emphasize the crucial role of priming vaccination in shaping Th bias and immunodominance hierarchies. mRNA LNP prime favors Th1-leaning responses, while PHC prime elicits Th2-skewing responses. We demonstrate that cellular and mucosal immune responses are pivotal correlates of cross-protection against influenza. Notably, intranasal PHC immunization outperforms its intramuscular counterpart in inducing mucosal immunity and conferring cross-protection. Sequential mRNA LNP prime and intranasal PHC boost demonstrate optimal cross-protection against antigenically drifted and shifted influenza strains. Our study offers valuable insights into tailoring immunization strategies to optimize influenza vaccine effectiveness. Here the authors show that sequential immunization utilizing mRNA lipid nanoparticle (LNP) and protein-based nanoparticle vaccines can synergize to enhance influenza vaccine potency and breadth. Abstract

Glioblastoma is the most fatal and insidious malignancy, due to the existence of the blood–brain barrier (BBB) and the high invasiveness of tumor cells. Abnormal mitochondrial viscosity has been identified as a key feature of malignancies. Therefore, this study reports on a novel fluorescent probe for mitochondrial viscosity, called ZVGQ, which is based on the twisted intramolecular charge transfer (TICT) effect. The probe uses 3-dicyanomethyl-1,5,5-trimethylcyclohexene as an electron donor moiety and molecular rotor, and triphenylphosphine (TPP) cation as an electron acceptor and mitochondrial targeting group. ZVGQ is highly selective, pH and time stable, and exhibits rapid viscosity responsiveness. In vitro experiments showed that ZVGQ could rapidly recognize to detect the changes in mitochondrial viscosity induced by nystatin and rotenone in U87MG cells and enable long-term imaging for up to 12 h in live U87MG cells. Additionally, in vitro 3D tumor spheres and in vivo orthotopic tumor-bearing models demonstrated that the probe ZVGQ exhibited exceptional tissue penetration depth and the ability to penetrate the BBB. The probe ZVGQ not only successfully visualizes abnormal mitochondrial viscosity changes, but also provides a practical and feasible tool for real-time imaging and clinical diagnosis of glioblastoma. Abstract

The hot compressive experiments of nickel-based alloy 617 were carried out at the different deformation conditions on a Gleeble-3500 thermal-simulator.The rule of the process parameters on the dynamic recrystallization microstructure evolution of alloy 617 under different deformation conditions were analyzed.Microstructure evolution models of dynamic recrystallization were constructed based on computational analyzed on the experiment data including strain-stress curves and metallographic photos.The models were implanted into finite element software Deform-3D in order to predict microstructure evolution during hot forging.The results show that the simulated results are in good agreement with the actual results, prediction control on the microstructure of alloy 617 during hot forging is achieved and the accuracy of models is verified.
