Abstract

ABSTRACT This analysis of real‐world data examines the efficacy, safety, and long‐term outcomes of renal denervation in hypertensive patients for up to 10 years. Sixty‐five consecutive patients underwent renal denervation (RDN) (single operator) for uncontrolled resistant hypertension. Efficacy was defined as the interindividual change of office (OBPM) and ambulatory blood pressure monitoring (ABPM) at 1, 6, 12 months, 2–4 and 5–10 years after RDN. Medication changes, renal function, and device generation disparities were analyzed. Of these patients, 42 received RDN with a first‐generation device, while 23 underwent the procedure with a second‐generation device. Baseline demographics included a predominantly male cohort (57.6%) with an average age of 60.3 years. The mean number of medications at baseline was 4.3. OBPM at baseline was 169.0/87.4 mmHg, and ABPM at baseline was 153.4/88.4 mmHg. Abstract

In this work, pectin aerogels loaded with zinc oxide (ZnO) were produced for a possible application in food packaging. Using a green synthesis procedure, ZnO nanoparticles were obtained via beetroot extract as a reducing, capping, and stabilizing agent; the obtained ZnO particles, with an average size of 300 nm, were loaded into pectin/alginate-based aerogels. Supercritical drying was used to produce the biopolymers-based aerogels, which were initially formulated with different polymer concentrations (2 wt% and 4 wt%) and various ratios between pectin and alginate (9:1, 7:3, 1:1) to identify the optimal configuration that would guarantee a balance between strength and morphology, resulting in better properties in the final structure. The aerogels enriched with different ZnO percentages (5 %, 10 %, and 15 %) were later produced. The aerogels showed a porous structure with uniform pore size distribution, influenced by the ZnO loading. The liquids’ absorption test of the samples assessed that higher concentrations of ZnO could reduce the available surface area, limiting the aerogels’ absorption capacity. The potent antimicrobial activity of ZnO and pectin/alginate aerogels against the non-pathogenic Escherichia coli and the pathogenic Staphylococcus aureus strains was demonstrated using a minimum lethal dose, highlighting the possibility of employing such materials in food packaging.

Abstract

This study aims to explore an advanced protocol for characterising dietary fibre (DF) fractions to meet the growing demand for accurate and reliable data. Although current enzymatic-gravimetric approaches, e.g., AOAC and Van Soest analysis, provide information about soluble and insoluble DF quantification, they present limitations related to the lack of fractions characterisation. To overcome these limitations, the proposed protocol integrates the official AOAC 991.43 method with the sequential fibre fractionation by exploiting the different resistance of the fibre fractions to acid hydrolysis treatments (TFA and H2SO4), utilising hazelnut shells as a case-study. Each hydrolysed fraction was quantified and characterised through GC–MS analysis of monosaccharides. The data obtained for hemicellulose, cellulose, and lignin fractions were then discussed and compared with the Van Soest method. This approach yields a comprehensive procedure applicable to different food and nutraceutical products, emphasising the importance of DF characterisation for a deeper understanding of their bio-functional properties.

Abstract

In most human tumors, the MAPK pathway is constitutively activated. Since p90RSK is downstream of MAPK, it is often hyperactive and capable of phosphorylating oncogenic substrates. We have previously shown that p90RSK phosphorylates MDM2 at S166, promoting p53 degradation in follicular thyroid carcinomas. Thus, the inhibition of p90RSK restores p53 expression, which in turn inhibits cell proliferation and promotes apoptosis. In the present study, we demonstrated that the p90RSK/MDM2/p53 pathway proved to be an excellent target in the therapy of tumors with MAPK hyperactivation. For this purpose, we selected p53wt melanoma, lung and medullary thyroid carcinoma cell lines with high activation of p90RSK. In these cell lines, we demonstrated that the p90RSK/MDM2/p53 pathway is implicated in the regulation of the cell cycle and apoptosis through p53-dependent transcriptional control of p21 and Bcl-2. Furthermore, with an immunohistochemical evaluation of primary melanomas and lung tumors, which exhibit highly activated p90RSK compared to corresponding normal tissue, we demonstrated that MDM2 stabilization was associated with p90RSK phosphorylation. The results indicate that p90RSK is able to control the proliferative rate and induction of apoptosis through the regulation of p53wt levels by stabilizing MDM2 in selected tumors with constitutively activated MAPKs, making p90RSK a new attractive target for anticancer therapy.
