Teaching robotics with the robot operating system (ROS) is valuable for instating good programming practices but requires significant setup steps from the learner. Providing a ready-made ROS learning environment over the web can make robotics more accessible; however, most of the previous remote labs have abstracted the authentic ROS developer environment either for didactical or technological reasons, or do not give the possibility to program physical robots. In this article, we present a remote web lab that employs virtual network computing and Docker to serve in-browser desktop workstations, where learning tasks can be completed on both the physical and simulated robots. The learners can reserve access to the remote lab through a learning management interface, which also includes tools for administering the remote lab. The system allows anyone to experiment with ROS without configuring any software locally and was successfully trialed in an online ROS course.

The aim of this paper was to provide an overview of the productivity of the stump lifting device Pallari KH-160, the distribution of working time, and to assess the dependence of the time and productivity of lifting operations on stump diameter in four different sample areas. Viru and Orguse sample areas were located in the Hepatica site type, Elva sample area in the Oxalis site type, and Rõuge test area in the Myrtillus site type. Viru sample area had smaller stumps compared to the others and had more stumps affected by decay. The highest productivity in terms of dry mass or volume was observed in stump lifting in Rõuge sample area, where 9.8 t of stumps were lifted per hour based on dry mass. The productivity of Viru and Orguse sample areas was lower, but these sample areas are located in the Hepatica site type, characterized by limestone substrates and leached soils. In contrast, Elva and Rõuge test areas are in the Oxalis and Myrtillus site types, respectively, with soils that are predominantly sandy. As the diameter of the stumps increased, the time required per stump increased, but the lifting speed per unit mass also increased. In other words, the productivity of lifting in stands with larger stumps is higher than in stands with smaller stumps. For example, the average productivity of lifting 10 cm diameter stumps is 0.8 t/h, for 20 cm stumps it is 2.2 t/h, and for 60 cm stumps it is 11.8 t/h.

Educational robots offer a platform for training aspiring engineers and building trust in technology that is envisioned to shape how we work and live. In education, accessibility and modularity are significant in the choice of such a technological platform. In order to foster continuous development of the robots as well as to improve student engagement in the design and fabrication process, safe production methods with low accessibility barriers should be chosen. In this paper, we present Robotont 3, an open-source mobile robot that leverages Fused Deposition Modeling (FDM) 3D-printing for manufacturing the chassis and a single dedicated system board that can be ordered from online printed circuit board (PCB) assembly services. To promote accessibility, the project follows open hardware practices, such as design transparency, permissive licensing, accessibility in manufacturing methods, and comprehensive documentation. Semantic Versioning was incorporated to improve maintainability in development. Compared to the earlier versions, Robotont 3 maintains all the technical capabilities, while featuring an improved hardware setup to enhance the ease of fabrication and assembly, and modularity. The improvements increase the accessibility, scalability and flexibility of the platform in an educational setting.

The paper presents a study of the effect of laser irradiation of crystalline chalcogenide films of lead selenide (PbSe) on their electrical characteristics caused by irreversible modification of the structure due to valence reconfiguration of lead as a result of its oxidation. The study of the modification features of the electrical properties of the films was carried out because of laser exposure to nanosecond pulses with a wavelength of 1064 nm. Measurements of the electrical characteristics of PbSe films were carried out using the four-probe method. It was shown that when the current was directed parallel to the laser tracks recorded in the darkening mode, the resistance of the modified film decreased by 44 % compared to the original sample, and with the perpendicular direction of the current, the resistance increased by 153 %. The resistance of the film increased more than 27 times after laser irradiation in the bleaching mode, regardless of the direction of the current relative to the laser tracks. The experimentally measured temperature and its gradient along the laser spot on the film in the darkening and bleaching modes turned out to be in good agreement with the proposed mathematical model of the thermal effect of laser pulses. It has been shown that the processes of laser modification of films occur at lower temperatures than during standard heat treatment in a furnace. The obtained results can be applied in the development of photodetectors in the middle IR range of the spectrum based on PbSe film.

Online platforms that allow non-residents to register Arms have emerged to boost economic development goals in jurisdictions ranging from Wyoming (United States) to Estonia. They create novel governance challenges that fall between governance frameworks. The global tax governance agenda needs to address the role of such platforms, which often involve conflicts between economic policy aspirations and other goals. Our Estonian case study demonstrates the inability of authorities to perform background checks of numerous non-resident entrepreneurs, as national administrative capacities get strained. Building on the nascent tax spillover approach, we analyse administrative spillover effects caused by online incorporation platforms in international taxation. Mapping de facto administrative capacities requires analysing conflicts between governmental priorities and the obstacles of sharing information between administrative and criminal procedures. When the non-resident community grows compared with the size of the domestic economy, supervisory systems tailored for domestic entrepreneurs become strained. We show that resolving this policy conflict assumes targeted investments into administrative capabilities from skilled personnel to data exchange and interorganizational coordination. IntroductionThe ability of species to colonize newly suitable habitats under rapid climate change can be constrained by migration processes, resulting in a shift of the leading edge lagging behind the ameliorating climate, i.e. migration lag. The importance and extent of such migration lags during the forest expansion after the Last Glacial Maximum (LGM) are still debated. Similarly, the relative importance of the main drivers of post-glacial vegetation dynamics (temperature, dispersal limitation, and competition) is still discussed in the literature.MethodsWe used the dynamic global vegetation model LPJ-GM 2.0 to reconstruct the range shifts of 16 competing major European tree species after the LGM (18.5 ka BP) until recent times (0 ka BP). We simulated two dispersal modes by allowing free establishment whenever the climatic conditions suited the species (free dispersal), or by accounting for migration processes in the simulated vegetation dynamics (dispersal limitation). We then calculated thermal and range shift velocities, competition at establishment, thermal and dispersal lags for each species and dispersal mode. Finally, we compared our simulated range shift velocities with pollen-derived migration rates.ResultsThe simulation assuming limited dispersal resulted in more accurate migration rates as compared to pollen-derived migration rates and spreading patterns. We found no marked migration lags in the post-glacial establishment of pioneer species (Pinus sylvestris and Betula pubescens). Under the free dispersal mode, the remaining temperate species expanded rapidly and almost synchronously across central Europe upon climate warming (Bølling-Allerød interstadial). Differently, the northward spread of temperate species simulated under dispersal limitation happened mainly during the Holocene and in successive waves, with late spreaders (e.g. Fraxinus excelsior) experiencing multi-millennial dispersal lags and higher competition.DiscussionOur simulation under dispersal constraints suggests that the post-glacial tree expansion in Europe was mainly driven by species-specific thermal requirements and dispersal capacity, which in turn affected the order of taxa establishment and thus the degree of competition. Namely, taxa with less cold-tolerance and relatively low dispersal ability experienced the highest migration lags, whereas the establishment of pioneer species was mostly in equilibrium with the climate.

The development of novel biologically active nanopharmaceuticals is a topical problem of medicine. Water-soluble fullerene derivatives are of particular interest due to their ability to regulate intracellular metabolism of reactive oxygen species (ROS) by direct oxidation or effects on oxidative and signaling enzymes. Here, we studied the effect of a water-soluble chlorine-containing derivative of C60 fullerene on human embryonic lung fibroblasts. MTT tests, intracellular ROS visualization, detection of the ROS-associated gene and protein expression, repair, cell proliferation and cell cycle regulation, and quantitation of oxidative DNA damage were used. Within the first three hours after exposure, antioxidant effects were revealed. Later, oxidative damage appeared. Thus, the studied compound had an ambiguous effect on ROS metabolism associated with a switch in the regulatory effect, which must be taken into account when assessing its biological activity and toxicity.

Photothermal therapy (PTT) has attracted increasing interest as a complementary method to be used alongside conventional therapies. Despite a great number of studies in this field, only a few have explored how temperatures affect the outcome of the PTT at nanoscale. In this work, we study the necrosis/apoptosis process of cancerous cells that occurs during PTT, using a combination of local laser heating and nanoscale fluorescence thermometry techniques. The temperature distribution within a whole cell was evaluated using fluorescence lifetime imaging microscopy during laser-induced hyperthermia. For this, gold nanorods were utilized as nanoheaters. The local near-infrared laser illumination produces a temperature gradient across the cells, which is precisely measured by nanoscale thermometry. This allows one to optimize the PTT conditions by varying concentration of gold nanorods associated with cells and laser power density. During the PTT procedure, such an approach enables an accurate determination of the percentages of apoptotic and necrotic cells using 2D and 3D models. According to the performed cell experiments, the influence of temperature increase during the PTT on cell death mechanisms has been verified and determined. Our investigations can improve the understanding of the PTT mechanisms and increase its therapeutic efficiency while avoiding any side effects.

BACKGROUND: Schizophrenia and suicidal behavior are associated with shortening in the length of telomeres. The aim of the study was to compare the content (pg/mcg) of telomeric repeat in DNA isolated from peripheral blood cells in three groups of subjects: patients with schizophrenia and a history of suicide attempts, patients with schizophrenia without suicidal tendencies, and healthy control volunteers. METHODS: Relapses according to gender and age were examined in 47 patients with schizophrenia with suicidal behavior, 47 patients without self-destructive conditions, and 47 volunteers with healthy control and maintenance for the content of telomeric and the number of copies of mitochondrial DNA (mtDNA) in peripheral blood leukocytes. RESULTS: Analysis of determining the content of telomeric repeat (TR) in the DNA of massive weight gain in the series: patients with schizophrenia and suicidal attempts patients with schizophrenia without suicidal observations healthy controls (22528.4 (227 [190; 250]) vs. 24321 (245 [228; 260]) vs. 25517.9 (255 [242; 266]), p 0.005. The same trend is observed for the number of mtDNA copies (257101.5 (250 [194; 297])) vs. 262.359.3 (254 [217; 312]) vs. 27279.9 (274 [213; 304]); p=0.012), but no significant differences were recorded. CONCLUSIONS: For the first time, the phenomenon of telomere shortening was discovered in schizophrenics with suicidal risk. The length of the telomere corresponds to the parameter of a biological marker an objectively measured indicator of normal or pathological processes, but gaining an idea of its reliability is still necessary for verification with an assessment of its sensitivity, specificity, and positive and negative predictive value. The telomere may be considered a putative predictive indicator of suicidal risk.

The fragment of satellite III (f-SatIII) is located in pericentromeric heterochromatin of chromosome 1. Cell with an enlarged f-SatIII block does not respond to various stimuli and are highly stress-susceptible. The fraction of f-SatIII in the cells of schizophrenia patients changed during antipsychotic therapy. Therefore, antipsychotics might reduce the f-SatIII content in the cells. We studied the action of haloperidol, risperidone and olanzapine (3 h, 24 h, 96 h) on human skin fibroblast lines (n = 10). The f-SatIII contents in DNA were measured using nonradioactive quantitative hybridization. RNASATIII were quantified using RT-qPCR. The levels of DNA damage markers (8-oxodG, γ-H2AX) and proteins that regulate apoptosis and autophagy were determined by flow cytometry. The antipsychotics reduced the f-SatIII content in DNA and RNASATIII content in RNA from HSFs. After an exposure to the antipsychotics, the autophagy marker LC3 significantly increased, while the apoptosis markers decreased. The f-SatIII content in DNA positively correlated with RNASATIII content in RNA and with DNA oxidation marker 8-oxodG, while negatively correlated with LC3 content. The antipsychotics arrest the process of f-SatIII repeat augmentation in cultured skin fibroblasts via the transcription suppression and/or through upregulated elimination of cells with enlarged f-SatIII blocks with the help of autophagy.

In order to achieve visionary concepts such as Society 5.0 and Industry 5.0, there is a growing need for people who are able to create innovative robotic technologies. Training students to become such skilled professionals requires transitioning from often toy-like educational platforms with significant hardware limitations to costly research robots with full ROS (Robot Operating System) support. To aid in this transition, we propose Robotont – an open-source omnidirectional mobile robot platform with both physical hardware and a digital twin. Robotont enables robotics education with professional tools as well as provides researchers with a capable mobility platform for validating and demonstrating scientific results. Robotont has successfully been used for university teaching, professional education, and online courses about ROS and robotics.

Introduction: Mild cognitive impairments (MCI) accompanying aging are associated with oxidative stress. The ability of cells to respond to stress is determined by the protein synthesis level, which depends on the ribosomes number. Ribosomal deficit was documented in MCI. The number of ribosomes depends, together with other factors, on the number of ribosomal genes copies. We hypothesized that MCI is associated with low rDNA CN in the elderly person genome.Materials and Methods: rDNA CN and the telomere repeat (TR) content were determined in the DNA of peripheral blood leukocytes of 93 elderly people (61–91 years old) with MCI and 365 healthy volunteers (16–91 years old). The method of non-radioactive quantitative hybridization of DNA with biotinylated DNA probes was used for the analysis.Results: In the MCI group, rDNA CN (mean 329 ± 60; median 314 copies, n = 93) was significantly reduced (p < 10–15) compared to controls of the same age with preserved cognitive functions (mean 412 ± 79; median 401 copies, n = 168) and younger (16–60 years) control group (mean 426 ± 109; median 416 copies, n = 197). MCI is also associated with a decrease in TR DNA content. There is no correlation between the content of rDNA and TR in DNA, however, in the group of DNA samples with rDNA CN > 540, TR content range was significantly narrowed compared to the rest of the sample.Conclusion: Mild cognitive impairment is associated with low ribosomal genes copies in the elderly people genomes. A low level of rDNA CN may be one of the causes of ribosomal deficit that was documented in MCI. The potential possibilities of using the rDNA CN indicator as a prognostic marker characterizing human life expectancy are discussed.

The development of systems capable of efficiently eliminating organic dyes from wastewater is crucial for the environment. In this context, TiO2-based photocatalytic systems are of particular interest. However, the development of eco-friendly and single-step methods for the fabrication of such systems remains a challenge to their practical use. Here, we demonstrate a fabrication approach for a catalytic system composed of TiO2 nanocoating created on the surface of a bulk titanium plate by exposure to a nanosecond laser. The exposure conditions affect the relief and phase composition of the nanocoating, as revealed by a combination of scanning electron microscopy (SEM), contact profilometry, Raman, High-resolution X-ray photoelectron spectroscopy (HR XPS), and grazing incidence X-ray diffraction (GIXRD) methods. Using the GIXRD method, we establish that nanocoatings consist of a mixture of anatase and rutile grains with sizes of 22.7–24.4 and 29.8–32.2 nm, respectively. HR XPS analysis demonstrates that changes in the number of pulses per spot modify the anatase surface content from 40.6 to 70.4%. The samples with a higher anatase content exhibit up to 6.3 times higher catalytic activity, which is confirmed by methylene blue degradation when exposed to UV light and can be related to the synergistic effect of the anatase–rutile heterojunction. The reusability of the samples is supported by repeated degradation experiments. The photocatalytic activity of the obtained TiO2 nanocoatings is improved by Au nanoparticles deposited on their surface by magnetron sputtering. The proposed approach lays the foundation for the application of nanosecond laser-assisted technologies in the eco-friendly fabrication of catalytic systems. [ABSTRACT FROM AUTHOR]
