Abstract

RNA terminal phosphorylase B (RTCB) has been shown to play a significant role in multiple physiological processes. However, the specific role of RTCB in the mouse colon remains unclear. In this study, we employ a conditional knockout mouse model to investigate the effects of RTCB depletion on the colon and the potential molecular mechanisms. We assess the efficiency and phenotype of Rtcb knockout using PCR, western blot analysis, histological staining, and immunohistochemistry. Compared with the control mice, the Rtcb-knockout mice exhibit compromised colonic barrier integrity and prominent inflammatory cell infiltration. In the colonic tissues of Rtcb-knockout mice, the protein levels of TNF-α, IL-8, and p-p65 are increased, whereas the levels of IKKβ and IκBα are decreased. Moreover, the level of GSK3β is increased, whereas the levels of Wnt3a, β-catenin, and LGR5 are decreased. Collectively, our findings unveil a close association between RTCB and colonic tissue homeostasis and demonstrate that RTCB deficiency can lead to dysregulation of both the NF-κB and Wnt/β-catenin signaling pathways in colonic cells.

15. How to Identify Boundary Conditions with Contrasty Metric?

Abstract

The boundary conditions (BCs) have shown great potential in requirements engineering because a BC captures the particular combination of circumstances, i.e., divergence, in which the goals of the requirement cannot be satisfied as a whole. Existing researches have attempted to automatically identify lots of BCs. Unfortunately, a large number of identified BCs make assessing and resolving divergences expensive. Existing methods adopt a coarse-grained metric, generality, to filter out less general BCs. However, the results still retain a large number of redundant BCs since a general BC potentially captures redundant circumstances that do not lead to a divergence. Furthermore, the likelihood of BC can be misled by redundant BCs resulting in costly repeatedly assessing and resolving divergences. In this paper, we present a fine-grained metric to filter out the redundant BCs. We first introduce the concept of contrasty of BC. Intuitively, if two BCs are contrastive, they capture different divergences. We argue that a set of contrastive BCs should be recommended to engineers, rather than a set of general BCs that potentially only indicates the same divergence. Then we design a post-processing framework (PPAc) to produce a set of contrastive BCs after identifying BCs. Experimental results show that the contrasty metric dramatically reduces the number of BCs recommended to engineers. Results also demonstrate that lots of BCs identified by the state-of-the-art method are redundant in most cases. Besides, to improve efficiency, we propose a joint framework (JAc) to interleave assessing based on the contrasty metric with identifying BCs. The primary intuition behind JAc is that it considers the search bias toward contrastive BCs during identifying BCs, thereby pruning the BCs capturing the same divergence. 24. Exploring the driving factors of bryophyte assemblage distribution patterns in Tibet.

Abstract

Plant communities are complex systems shaped by a combination of deterministic and stochastic ecological processes. Bryophytes are an essential component of plant diversity in natural ecosystems, yet our understanding of their community ecology needs to catch up to that of other organisms. The unique geological history, alpine climatic conditions, and high habitat heterogeneity of Tibet provide suitable areas for bryophytes to survive in the alpine regions. Therefore, field surveys were conducted across 184 plots in forest, thicket, and herbaceous vegetation of Tibet to investigate the role of deterministic processes such as biological interactions and abiotic effects, along with stochastic processes, in shaping the distribution of bryophyte assemblages. We employed various analytical methods, including mixed effects models, partial least squares path modeling, null model analysis, and neutral community models. The study showed that bryophyte richness was highest in forests. Bryophyte assemblages showed greater segregation in forest and thicket environments compared to herbaceous vegetation. As the influence of stochastic processes increased, that of deterministic processes decreased from forests through thickets to herbaceous vegetation. Deterministic processes were the main driving forces for the bryophyte assemblage pattern. Soil properties and climatic factors, particularly pH played a key role in determining bryophyte patterns in Tibet. This study has deepened our comprehension of how deterministic and stochastic ecological processes interplay and shape bryophyte distribution patterns in Tibet. [ABSTRACT FROM AUTHOR]

Abstract

Investigating intraspecific trait variability is crucial for understanding plant adaptation to various environments, yet research on lithophytic mosses in extreme environments remains scarce. This study focuses on Indusiella thianschanica Broth. Hal., a unique lithophytic moss species in the extreme environments of the Tibetan Plateau, aiming to uncover its adaptation and response mechanisms to environmental changes. Specimens were collected from 26 sites across elevations ranging from 3642 m to 5528 m, and the relationships between 23 morphological traits and 15 environmental factors were analyzed. Results indicated that coefficients of variation (CV) ranged from 5.91% to 36.11%, with gametophyte height (GH) and basal cell transverse wall thickness (STW) showing the highest and lowest variations, respectively. Temperature, elevation, and potential evapo-transpiration (PET) emerged as primary environmental drivers. Leaf traits, especially those of the leaf sheath, exhibited a more pronounced response to the environment. The traits exhibited apparent covariation in response to environmental challenges and indicated flexible adaptive strategies. This study revealed the adaptation and response patterns of different morphological traits of I. thianschanica to environmental changes on the Tibetan Plateau, emphasizing the significant effect of temperature on trait variation.
