Abstract Background Low immunity and sleep disorders are prevalent suboptimal health conditions in contemporary populations, which render them susceptible to the infiltration of pathogenic factors. LJC, which has a long history in traditional Chinese medicine for nourishing the Yin and blood and calming the mind, is obtained by modifying Qiyuan paste. Dendrobium officinale Kimura et Migo has been shown to improve the immune function in sleep-deprived mice. In this study, based on the traditional Chinese medicine theory, LJC was prepared by adding D. officinale Kimura et Migo to Qiyuan paste decoction. Methods Indicators of Yin deficiency syndrome, such as back temperature and grip strength, were measured in each group of mice; furthermore, behavioral tests and pentobarbital sodium-induced sleep tests were performed. An automatic biochemical analyzer, enzyme-linked immunosorbent assay kit, and other methods were used to determine routine blood parameters, serum immunoglobulin (IgG, IgA, and IgM), cont (C3, C4), acid phosphatase (ACP) and lactate dehydrogenase (LDH) levels in the spleen, serum hemolysin, and delayed-type hypersensitivity (DTH) levels. In addition, serum levels of γ-aminobutyric acid (GABA) and glutamate (Glu) were detected using high-performance liquid chromatography (HPLC). Hematoxylin–eosin staining and Nissl staining were used to assess the histological alterations in the hypothalamus tissue. Western blot and immunohistochemistry were used to detect the expressions of the GABA pathway proteins GABRA1, GAD, GAT1, and GABAT1 and those of CD4+ and CD8+ proteins in the thymus and spleen tissues. Results The findings indicated that LJC prolonged the sleep duration, improved the pathological changes in the hippocampus, effectively upregulated the GABA content in the serum of mice, downregulated the Glu content and Glu/GABA ratio, enhanced the expressions of GABRA1, GAT1, and GAD, and decreased the expression of GABAT1 to assuage sleep disorders. Importantly, LJC alleviated the damage to the thymus and spleen tissues in the model mice and enhanced the activities of ACP and LDH in the spleen of the immunocompromised mice. Moreover, serum hemolysin levels and serum IgG, IgA, and IgM levels increased after LJC administration, which manifested as increased CD4+ content, decreased CD8+ content, and enhanced DTH response. In addition, LJC significantly increased the levels of complement C3 and C4, increased the number of white blood cells and lymphocytes, and decreased the percentage of neutrophils in the blood. Conclusions LJC can lead to improvements in immunocompromised mice models with insufficient sleep. The underlying mechanism may involve regulation of the GABA/Glu content and the expression levels of GABA metabolism pathway-related proteins in the brain of mice, enhancing their specific and nonspecific immune functions. Abstracts: Objectives: To elucidate the therapeutic effects and mechanisms of Atractylodes macrocephala extract crystallize (BZEP) and BZEP self-microemulsion (BZEPWR) on metabolic dysfunction-associated fatty liver disease (MAFLD) induced by “high sugar, high fat, and excessive alcohol consumption” based on the gut–liver axis HDL/LPS signaling pathway. Methods: In this study, BZEP and BZEPWR were obtained via isolation, purification, and microemulsification. Furthermore, an anthropomorphic MAFLD rat model of “high sugar, high fat, and excessive alcohol consumption” was established. The therapeutic effects of BZEPWR and BZEP on the model rats were evaluated in terms of liver function, lipid metabolism (especially HDL-C), serum antioxidant indexes, and liver and intestinal pathophysiology. To determine the lipoproteins in the serum sample, the amplitudes of a plurality of NMR spectra were derived via deconvolution of the composite methyl signal envelope to yield HDL-C subclass concentrations. The changes in intestinal flora were detected via 16 S rRNA gene sequencing. In addition, the gut–liver axis HDL/LPS signaling pathway was validated using immunohistochemistry, immunofluorescence, and western blot. Results: The findings established that BZEPWR and BZEP improved animal signs, serum levels of liver enzymes (ALT and AST), lipid metabolism (TC, TG, HDL-C, and LDL-C), and antioxidant indexes (GSH, SOD, and ROS). In addition, pathological damage to the liver, colon, and ileum was ameliorated, and the intestinal barrier function of the model rats was restored. At the genus level, BZEPWR and BZEP exerted positive effects on beneficial bacteria, such as Lactobacillus and norank_f__Muribaculaceae, and inhibitory effects on harmful bacteria, such as unclassified_f__Lachnospiraceae and Blautia. Twenty HDL-C subspecies were detected, and their levels were differentially increased in both BZEPWR and BZEP groups, with BZEPWR exhibiting a stronger elevating effect on specific HDL-C subspecies. Also, the gut–liver axis HDL/LPS signaling pathway was studied, which indicated that BZEPWR and BZEP significantly increased the expressions of ABCA1, LXR, occludin, and claudin-1 proteins in the gut and serum levels of HDL-C. Concomitantly, the levels of LPS in the serum and TLR4, Myd88, and NF-κB proteins in the liver were decreased. Conclusion: BZEPWR and BZEP exert restorative and reversal effects on the pathophysiological damage to the gut–liver axis in MAFLD rats, and the therapeutic mechanism may be related to the regulation of the intestinal flora and the HDL/LPS signaling pathway.

Abstract Ionic memristors can emulate brain‐like functions of biological synapses for neuromorphic technologies. Apart from the widely studied excitatory‐excitatory and excitatory‐inhibitory synapses, reports on memristors with the inhibitory‐inhibitory synaptic behaviors remain a challenge. Here, the first biaxially inhibited artificial synapse is demonstrated, consisting of a solid electrolyte and conjugated microporous polymers bilayer as neurotransmitter, with the former serving as an ion reservoir and the latter acting as a confined transport. Due to the migration, trapping, and de‐trapping of ions within the nanoslits, the device poses inhibitory synaptic plasticity under both positive and negative stimuli. Remarkably, the artificial synapse is able to maintain a low level of stable nonvolatile memory over a long period of time (≈60 min) after multiple stimuli, with feature‐inferencing/‐training capabilities of neural node in neuromorphic computing. This work paves a reliable strategy for constructing nanochannel ionic memristive materials toward fully inhibitory synaptic devices.

BackgroundPassive safety system reliability is generally evaluated through best estimation plus uncertainty (BEPU) analysis. An important step in the evaluation process is sensitivity analysis of parameters, which is used to identify key system parameters to reduce the complexity of the model. However, local sensitivity analysis methods based on linear or monotonic assumptions may yield incorrect sensitivity results for complex nuclear power systems. Meanwhile, applying the global sensitivity method is difficult in practical engineering because of its high calculation cost.PurposeThis study aims to develop an efficient and low-cost global sensitivity analysis method for passive systems under ocean conditions.MethodsFirstly, the low-rank approximation (LRA) method was employed to improve the Sobol method based on Monte Carlo simulation. The number of unknown coefficients was significantly reduced by using the multivariate-based tensor product. Then, the LRA coefficients were used to calculate the sensitivity index, and the validity of the proposed method was verified by addressing several sensitivity analysis benchmark questions. Finally, taking an integrated pressurized water reactor including the passive residual heat removal system as the object, a simulation program for thermal-hydraulics analysis under ocean conditions was developed. And its sensitivity analysis was conducted using the proposed method.ResultsThe results show that the proposed method can accurately identify system key parameters after only 200 simulation calculations taking about 55 min, and the sensitivity ranking results are consistent with those obtained by Sobol method after 1.0×105 simulation calculations taking about 19 d.ConclusionsThe efficient global sensitivity analysis method established in this study can provide effective guidance for reliability analysis and design optimization of passive systems.

There is an increasing global trend toward unhealthy lifestyles and dietary decisions, like the overconsumption of alcohol and high-sugar and high-fat diets (ACHSFDs). Poor eating habits can cause an imbalance of the Treg/Th17 immune axis, thereby leading to an imbalance of anti-inflammatory and pro-inflammatory factors, which damages target organs and the vascular endothelium causing MH. Hawthorn has been shown to improve gastrointestinal function, lower blood pressure, and regulate metabolic abnormalities. However, whether it acts on MH by regulating the Treg/Th17 immune axis remains unclear. In this study, the hawthorn extract reduced blood pressure[systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean blood pressure (MBP)] and improved dyslipidemia in rats with ACHSFD-induced MH. In addition, the extract reduced serum AST and ALT levels and liver coefficient and improved liver fat deposition and fibrosis. On the other hand, the hawthorn extract downregulated RORγt expression, upregulated Foxp3 expression, increased IL-10 levels in the aorta, and reduced IL-17, IL-6, and TNF-α content. Moreover, the extract upregulated eNOS expression in the aorta and increased NO content in the serum. These findings indicate that hawthorn extract can inhibit aortic inflammatory injury and improve vascular endothelial function by regulating the Treg/Th17 immune imbalance.

Nardosinone, a predominant bioactive product from Nardostachys jatamansi DC, is well-known for its promising therapeutic applications, such as being used as a drug on anti-inflammatory, antidepressant, cardioprotective, anti-neuroinflammatory, anti-arrhythmic, anti-periodontitis, etc. However, its stability under varying environmental conditions and its degradation products remain unclear. In this study, four main degradation products, including two previously undescribed compounds [2–deoxokanshone M (64.23%) and 2–deoxokanshone L (1.10%)] and two known compounds [desoxo-narchinol A (2.17%) and isonardosinone (3.44%)], were firstly afforded from the refluxed products of nardosinone in boiling water; their structures were identified using an analysis of the extensive NMR and X–ray diffraction data and the simulation and comparison of electronic circular dichroism spectra. Compared with nardosinone, 2–deoxokanshone M exhibited potent vasodilatory activity without any of the significant anti-neuroinflammatory activity that nardosinone contains. Secondly, UPLC–PDA and UHPLC–DAD/Q–TOF MS analyses on the degradation patterns of nardosinone revealed that nardosinone degraded more easily under high temperatures and in simulated gastric fluid compared with the simulated intestinal fluid. A plausible degradation pathway of nardosinone was finally proposed using nardosinonediol as the initial intermediate and involved multiple chemical reactions, including peroxy ring-opening, keto–enol tautomerization, oxidation, isopropyl cleavage, and pinacol rearrangement. Our findings may supply certain guidance and scientific evidence for the quality control and reasonable application of nardosinone-related products.

D-Allulose is an ultra-low calorie sweetener with broad market prospects. As an alternative to Izumoring, phosphorylation-dephosphorylation is a promising method for D-allulose synthesis due to its high conversion of substrate, which has been preliminarily attempted in enzymatic systems. However, in vitro phosphorylation-dephosphorylation requires polyphosphate as a phosphate donor and cannot completely deplete the substrate, which may limit its application in industry. Here, we designed and constructed a metabolic pathway in Escherichia coli for producing D-allulose from D-fructose via in vivo phosphorylation-dephosphorylation. PtsG-F and Mak were used to replace the fructose phosphotransferase systems (PTS) for uptake and phosphorylation of D-fructose to fructose-6-phosphate, which was then converted to D-allulose by AlsE and A6PP. The D-allulose titer reached 0.35 g/L and the yield was 0.16 g/g. Further block of the carbon flux into the Embden-Meyerhof-Parnas (EMP) pathway and introduction of an ATP regeneration system obviously improved fermentation performance, increasing the titer and yield of D-allulose to 1.23 g/L and 0.68 g/g, respectively. The E. coli cell factory cultured in M9 medium with glycerol as a carbon source achieved a D-allulose titer of ≈1.59 g/L and a yield of ≈0.72 g/g on D-fructose.

Heavy metals are widely distributed in the environment due to the natural processes and anthropogenic human activities. Their migration into no contaminated areas contributing towards pollution of the ecosystems e.g. soils, plants, water and air. It is recognized that heavy metals due to their toxicity, long persistence in nature can accumulate in the trophic chain and cause organism dysfunction. Although the popularity of herbal medicine is rapidly increasing all over the world heavy metal toxicity has a great impact and importance on herbal plants and consequently affects the quality of herbal raw materials, herbal extracts, the safety and marketability of drugs. Effective control of heavy metal content in herbal plants using in pharmaceutical and food industries has become indispensable. Therefore, this review describes various important factors such as ecological and environmental pollution, cultivation and harvest of herbal plants and manufacturing processes which effects on the quality of herbal plants and then on Chinese herbal medicines which influence human health. This review also proposes possible management strategies to recover environmental sustainability and medication safety. About 276 published studies (1988–2021) are reviewed in this paper.

Abstract Background The Mindray BC-6800 haematology analyzer (BC-6800) provides a dedicated flag ‘Infected RBC’ (InR) and the number of InR (InR#)/the permillage of InR (InR‰) in routine blood testing as a screening tool for malaria in endemic areas. This study sought to evaluate the effectiveness of the BC-6800 flag parameter for aiding the diagnosis of malaria. Methods A total of 181 samples were tested using the Mindray BC-6800 haematology analyzer, including 117 malaria-infected samples collected from Yunnan, China, and 64 samples from healthy controls. Microscopy examination was conducted as reference when stained thick blood film revealed the presence of malaria parasites identified as Plasmodium vivax and Plasmodium falciparum. The receiver operating characteristic (ROC) curve analysis was developed using Analyse-it v4.92.3. The Kappa value was determined to evaluate the agreement between BC-6800 and light microscopy. Results The sensitivity of InR‰ generated by BC-6800 for P. vivax and P. falciparum was 88.3 and 24.1%, respectively; specificity of InR‰ for malaria parasites was 84.3 and 84.3%, respectively; positive predictive value and negative predictive value was 89.4 and 82.7% for P. vivax, and 52.8 and 60.3% for P. falciparum. There was a strong correlation between ΔWBC and InR‰ (R2 = 0.9731 for P. vivax and R2 = 0.9757 for P. falciparum). There was also a significant correlation between parasitaemia and InR# in P. vivax-infected samples (R2 = 0.734). InR# was evaluated using ROC curve analysis, the area under the ROC curve is 0.95 with a 95% confidence interval of 0.926 to 0.974, and the cut-off value is 0.01 × 109/L for P. vivax. However, the ring stage and the early trophozoite stage of Plasmodium cannot be detected easily on BC-6800, possibly because of the small size and low nucleic acid content of these stages. Conclusions The findings suggest that the flag ‘InR’ and the parameters ‘InR#/InR‰’ provided by the BC-6800 haematology analyzer could be used to screen for malaria in a clinical setting.

The electrochemical behavior of the chalcocite oxidation process in the presence and absence of microorganisms was investigated using electrochemical techniques, including cyclic voltammetry, anodic polarization curves, Tafel curves, and X-ray photoelectron spectroscopy (XPS) analysis. The research results prove the stepwise dissolution mechanism of chalcocite in the presence and absence of microorganisms. The initial stage of oxidation is initiated at low redox potentials. During the initial stage, the intermediate products of CuxS (1 ≤ x via XPS. The XPS analysis results show that the components of the passivation layer on the electrode surface are complex, including CuS, polysulfide (Sn2-), elemental sulfur (S0) and intermediate oxidation products that contain sulfate (SO42-) and that CuS is the main passivating component; therefore, the oxidation of chalcocite follows the multisulfur method.

Background: Coxsackievirus B3 (CVB3) is the primary cause of infectious myocarditis. Aggressive immunological activation and apoptosis of myocytes contributes to progressive dysfunction of cardiac contraction and poor prognosis. MG-132, a proteasome inhibitor, regulates mitochondrial-mediated intrinsic myocardial apoptosis and downregulates NF-κB-mediated inflammation. Here, we determined whether AMPK pathway participates in MG-132-mediated myocardial protection in viral-induced myocarditis. Methods and results: Acute viral myocarditis models were established by intraperitoneal inoculation of CVB3 in male BALB/c mice. Myocarditis and age-matched control mice were administered MG-132 and/or BML-275 dihydrochloride (BML) (AMPK antagonist) intraperitoneally daily from the day following CVB3 inoculation. MG-132 improved hemodynamics and inhibited the structural remodeling of the ventricle in mice with myocarditis, while BML largely blunted these effects. TUNEL staining and immunochemistry suggested that MG-132 exerts anti-apoptotic and anti-inflammatory effects against CVB3-induced myocardial injuries. BML attenuated the effects of MG-132 on anti-apoptosis and anti-inflammation. Conclusion: MG-132 modulated apoptosis and inflammation, improved hemodynamics, and inhibited the structural remodeling of ventricles in a myocarditis mouse model via regulation of the AMPK signal pathway.

A bottleneck occurs in the application of bioleaching technology to fluoride-containing ore. The reason for this is that fluorine has a strong inhibitory effect on leaching bacteria with the dissolution of fluorine-containing gangue minerals. In this study, we use the chemical properties of fluorine to convert F ions by adding substances that can form stable complexes with F-, which enables the leaching bacteria to tolerate high fluoride environments. In this research, we studied the inhibition mechanism of fluorine on bacteria, and identified its true toxic form (HF). We found that fluoride exhibited a transmembrane inhibitory effect on bacteria. Under fluoride stress conditions, the concentration of intracellular fluoride was significantly higher than that of a non-fluorinated control group, which was about 18% dry cell. We selected common Fe3+ ions in the bioleaching system, and studied the competitive complex detoxification of Fe3+ to F-. Our thermodynamic analysis results show that Fe3+ can compete with HF in first-order competitive complexation reactions whereby the HF complex structure is converted to FeFn3-n. In the presence of ferric ions, we found that the bacteria could tolerate F-concentrations up to 1.0 g·L-1. Our analysis of the Fe and F complex species indicates that bacteria could grow normally when the concentration of Fe3+ ions was five times greater than that of F- ions. Correspondingly, the proportion of FeF2+ components in the solution was ≥ 45%, and the concentration of free fluoride was 2.87×10-5 mol·L-1. The complexation mechanism shows that as the ratio of F--Fe3+ decreases, the concentration of the ligand is relatively lower. Based on the coordination chemistry, the complex of fluoride and iron moves in a lower coordination direction, and the Fe and F complex species can be controlled by adjusting the concentration ratio of F- and Fe3+ in the medium, therefore making it possible for bacteria to grow in a high-fluorine environment.

The preparation of high-quality iron concentrate from pyrite cinder by means of desulphurization has potential industrial applications. On one hand, the comprehensive utilization of pyrite cinder can be achieved, while on the other hand, the environmental problems that it causes can be solved effectively. In this study, the desulfurization technology and its process flowsheet, as well as recent advances, were summarized in detail. There are mainly three desulfurization methods, namely, chemical leaching, combined process, and bioleaching method. Chemical leaching mainly involves the acid leaching and alkali leaching processes. The combined process contains the alkali-acid leaching process, flotation-magnetic separation, gravity separation-flotation, magnetic roasting-magnetic separation, etc. The technical features of these methods were introduced, and it is concluded that bioleaching has a potential application value. The research areas of bioleaching method in a future will be included as:the breeding of effective desulfurization bacteria, cyclic utilization of desulfurization water, synergistic recovery of valuable metals, and basic investigation and engineering technology research in the bioleaching desulfurization process.

Agglomeration technology is one of the most effective measures to enhance the poor heap permeability of ores caused by the presence of fine particles. Selecting the proper acid resistant binder is key to the successful heap leaching of copper ore. The agglomeration of oxide-sulfide mixed copper ore was studied in this paper, and results show that Biometek-WLAG001 is an ideal acid resistant binder. The effects of binder amount, water spray amount, rotational speed, curing time, aging acid addition amount, and aging time on the compressive and wet strengths of agglomerated pellets were investigated in single-factor experiments. The effects of binder amount, curing time, aging acid addition amount, and aging time on the wet strength of agglomerated pellet were further studied via a response surface methodology, and results were optimized and analyzed by Design-Expert 8.0 software. The interaction mechanism between the binder and the ore was investigated by Fourier transform infrared spectroscopy analysis and Zeta potential measurements. Results reveal that the binder and ore undergo chemical adsorption but not electrostatic attraction.
