Abstract

Artificial Intelligence (AI) based image analysis has an immense potential to support diagnostic histopathology, including cancer diagnostics. However, developing supervised AI methods requires large-scale annotated datasets. A potentially powerful solution is to augment training data with synthetic data. Latent diffusion models, which can generate high-quality, diverse synthetic images, are promising. However, the most common implementations rely on detailed textual descriptions, which are not generally available in this domain. This work proposes a method that constructs structured textual prompts from automatically extracted image features. We experiment with the PCam dataset, composed of tissue patches only loosely annotated as healthy or cancerous. We show that including image-derived features in the prompt, as opposed to only healthy and cancerous labels, improves the Fr\'echet Inception Distance (FID) from 178.8 to 90.2. We also show that pathologists find it challenging to detect synthetic images, with a median sensitivity/specificity of 0.55/0.55. Finally, we show that synthetic data effectively trains AI models.

Abstract

Aging is associated with an increased risk of cardiovascular disease and death. Here we show that oral supplementation of the natural polyamine spermidine extends the lifespan of mice and exerts cardioprotective effects, reducing cardiac hypertrophy and preserving diastolic function in old mice. Spermidine feeding enhanced cardiac autophagy, mitophagy and mitochondrial respiration, and it also improved the mechano-elastical properties of cardiomyocytes in vivo, coinciding with increased titin phosphorylation and suppressed subclinical inflammation. Spermidine feeding failed to provide cardioprotection in mice that lack the autophagy-related protein Atg5 in cardiomyocytes. In Dahl salt-sensitive rats that were fed a high-salt diet, a model for hypertension-induced congestive heart failure, spermidine feeding reduced systemic blood pressure, increased titin phosphorylation and prevented cardiac hypertrophy and a decline in diastolic function, thus delaying the progression to heart failure. In humans, high levels of dietary spermidine, as assessed from food questionnaires, correlated with reduced blood pressure and a lower incidence of cardiovascular disease. Abstract

The age-induced deterioration of the organism results in detrimental and ultimately lethal pathologies. The process of aging itself involves a plethora of different mechanisms that should be subverted concurrently to delay and/or prevent age- related maladies. We have identified a natural compound, 4,4ʹ-dimethoxychalcone (DMC), which promotes longevity in yeast, worms and flies, and protects mice from heart injury and liver toxicity. Interestingly, both the DMC-mediated lifespan extension and the cardioprotection depend on macroautophagy/autophagy whereas hepatoprotection does not. DMC induces autophagy by inhibiting specific GATA transcription factors (TFs), independently of the TORC1 kinase pathway. The autophagy-independent beneficial effects of DMC might involve its antioxidative properties. DMC treatment results in a phylogenetically conserved, systemic impact on the metabolome, which is most prominently characterized by changes in cellular amino acid composition. Altogether, DMC exerts multiple, geroprotective effects by igniting distinct pathways, and thus represents a potential pharmacological agent that delays aging through multipronged effects.

Accumulating evidence suggests that GATA TFs also play a role in lifespan regulation. In a recent study, we identified a natural polyphenol, 4,4’-dimethoxychalcone (DMC), that extends lifespan depending on reduced activity of distinct GATA TFs. Prolonged lifespan by DMC treatment depends on autophagy, a protective cellular self-cleansing mechanism. In yeast, DMC reduces the activity of the GATA TF Gln3 and, genetic deletion of Gln3 is sufficient to increase autophagy levels during cellular aging. In addition, we observed similar changes in the abundance of several amino acids in the metabolome of DMC-treated and GATA/Gln3 depleted cells. Here, we examine current data on the involvement of GATA TFs in the regulation of autophagy and longevity in different organisms and explore if GATA TFs might be suitable targets for anti-aging interventions.

Altogether, spermidine represents a cardio- and vascular-protective autophagy inducer that can be readily integrated in common diets.

Abstract

Case summary: A 10-year-old male neutered British Shorthair cat was presented with a 6 month history of lethargy, weight loss and alopecia. Clinical examination revealed widespread alopecia of the ventral abdomen and hindlimbs. The skin in these areas was smooth and shiny and hairs could be easily epilated. Spontaneous pruritus was observed. Cytological examination of superficial impression smears showed a severe Malassezia species dermatitis and pyoderma. Ectoparasites could not be detected and no sign of dermatophytosis was visible in trichograms and Wood s lamp analysis. Abdominal ultrasound found a focally thickened wall of the large intestine and multiple nodules in the liver. Fine-needle aspirates from lymph nodes, liver and altered colonic wall were consistent with an undifferentiated malignant neoplasia. The cat was euthanased at the owners request, owing to potential neoplasia with metastatic spread. At necropsy a metastasising carcinoma of the colonic wall was found, as well as a paraneoplastic alopecia.Relevance and novel information: Feline paraneoplastic alopecia has been reported in association with pancreatic carcinoma, bile duct carcinoma and hepatocellular carcinoma, as well as with neuroendocrine pancreatic carcinoma and hepatosplenic plasma cell tumour. This is the first reported case of feline paraneoplastic alopecia associated with a colon carcinoma.

Abstract

This dissertation is a portfolio of 7 selected pieces completed between 2009-2016. Together they trace a transitional arc into the current branch of my work. Earlier pieces (Fox Bride, Slip Trains, and Bird in the Well) form something of a bridge into this transition, embodying many through-lines of my current language in an earlier form. I feel it's fitting that these pieces retain peculiarities of scoring and notational practice that have since evolved, but that accurately depict my working methods at the time. In every sense, they express this unfolding of a project. The pieces in the latter half of this portfolio begin to incorporate performance systems that use transducers and custom-built mechanisms to extend and amplify resonances of acoustic instruments. Starting from pieces for solo performer with 'live mechanics' and developing more recently into expanding configurations of ensembles, the resulting pieces build around malleable qualities of fibrillation and pulsation, using these as a kind of live connective tissue within a body of instruments. There is something tectonic that transducers can achieve, creating an underbelly of sound that is both amorphic and uniquely visceral. When worked into an actively layered field, they can yield an enveloping quality that is as much about body-feel as it is about audible sound. Pulsation feels inherently intimate – from the semi-regularity of blood-flow to the tremors of cells and nervous systems. There is something richly primal in this deeper morse code. Navigating this spectra largely through acoustic means is my way of digging into these very physical aspects of sound that have a permeating intensity in live performance. This project is ongoing and includes pieces spanning this recent period that are still in progress.
