Here, we investigated the myeloprotective effects of trilaciclib among patients at increased risk of CIM.Patients and Methods: Data were pooled from three randomized, double-blind, placebo-controlled, phase 2 clinical studies of trilaciclib administered prior to chemotherapy in patients with extensive-stage small cell lung cancer (ES-SCLC). Myeloprotective outcomes were evaluated in patient subgroups based on age (< 65 or ≥ 65 years), risk of chemotherapy-induced febrile neutropenia (FN), and risk of anemia or red blood cell (RBC) transfusions. For the FN and anemia analyses, risk factors were identified from published literature and used to classify patients into FN and anemia risk categories. Subgroup analysis based on age was also performed on patient reported outcome (PRO) measures.Results: In total, 123 patients received trilaciclib and 119 patients received placebo. Myeloprotective benefits of trilaciclib were observed regardless of age, with greater effects observed among patients aged ≥ 65 years. Across FN risk factors and categories, trilaciclib had beneficial effects on neutrophil-related endpoints vs placebo, with greater effects observed in patients at higher risk of FN. Effects on RBC-related endpoints favored trilaciclib vs placebo, regardless of anemia risk factors and categories. Improvements in PROs with trilaciclib were observed irrespective of age group, but with greater improvements and less deterioration from baseline observed in older patients.Conclusion: By both decreasing the incidence of CIM and improving quality of life, trilaciclib has the potential to allow patients receiving chemotherapy for ES-SCLC, including patients who are older or more vulnerable to CIM, to receive chemotherapy on schedule and at standard-of-care doses, and to improve the experience for patients receiving chemotherapy to treat ES-SCLC.Clinical Trial Numbers: NCT02499770; NCT03041311; NCT02514447.Keywords: trilaciclib, myelosuppression, myeloprotection, myelopreservation, chemotherapy, small cell lung cancer

20. Testing and Validation of an Image-Based, Pose and Shape Reconstruction Algorithm for Didymos Mission

Abstract

The recent and continuously growing scientific interest for asteroids composition and its relation to solar system’s origins led the space industry to put high effort into asteroid missions during the last years. Nowadays, several space companies are investing resources into Hera mission (which has been proposed by the European Space Agency). In addition to visiting the Didymos asteroid binary system to acquire important scientific discoveries, Hera mission will also be the first mission to operate autonomously around an asteroid. The spacecraft will fuse data from its Asteroid Framing Camera (AFC), star-tracker, laser altimeter, thermal infrared camera and inertial sensors to build up a coherent model of its surroundings and be able to behave autonomously. Despite that, the AFC is considered to be the most crucial data source because of its dual science and navigation functionality. In fact, although images acquired by the camera are going to be used for scientific purposes, during the mission their main usage will be devoted to navigating autonomously around the main asteroid, Didymain. Because of this reason, several hardware-in-the-loop tests are being made in laboratories to analyze the accuracy of specific feature detection, extraction and tracking algorithms. Along with the computer vision part, navigation filters are being tested on the captured images to prove the feasibility of shape reconstruction and relative pose estimation (i.e., relative position, velocity, attitude and angular velocity). This paper presents an algorithm which has been built to use images acquired by a camera to reconstruct Didymain’s relative pose and shape and validates it with both computer simulations and experimental tests. First, the algorithm is going to be explained in all its parts: the computer vision block makes use of feature detection and extraction methods (e.g., KAZE, SIFT, etc.) and tracking algorithms (e.g., Kanade–Lucas–Tomasi Feature Tracker) to extract meaningful information from the captured images; the navigation filter, developed following an Unscented Kalman-like scheme, is going to use those data to reconstruct the asteroid’s pose and shape. Second, a simulator built in Matlab environment, which uses a 3D CAD of the asteroid to generate synthetic images in closed loop, is being used to prove the feasibility of this concept: several simulation scenario with different initial conditions will be analyzed to prove the robustness of this approach. Finally, the algorithm is going to be tested and validated through experimental tests performed at platform-art©, GMV’s Advanced Robotic Testbed in Madrid, where real images of a Didymain asteroid’s mock-up are going to be framed and used in the loop. The results obtained by the software testing and hardware-in-the-loop validation steps show that the proposed technique attains a good estimation accuracy as long as the relative configuration between spacecraft and asteroid is not singular (thus unobservable), meeting mission requirements and thus being suitable for an on-board implementation.

Abstract

Sur une initiative toulousaine, nous avons organisé, pour la quatrième année, une journée événementielle en centre-ville de Bayonne, afin de sensibiliser le grand public à la notion de santé respiratoire. Le 8 mai 2024, nous avons installé un village composé de différentes animations: stand d’information anti-tabac, stand pollution de l’air intérieur et extérieur, jeux éducatifs pour les enfants, initiation à l’activité physique et enfin la possibilité de réaliser une spirométrie. Nous rapportons ici les résultats des 223 spirométries effectuées chez les passants venant tester leur souffle spontanément.

Abstract

Functional loss of E-cadherin is frequent during tumor progression and occurs through a variety of mechanisms, including proteolytic cleavage. E-cadherin downregulation leads to the conversion of a more malignant phenotype promoting Epithelial to Mesenchymal Transition (EMT). The UBC9/SUMO pathway has been also shown to be involved in the regulation of EMT in different cancers. Here we found an increased expression of UBC9 in the progression of Head and Neck Cancer (HNC) and uncovered a role for UBC9/SUMO in hampering the HPV-mediated E-cadherin cleavage in HNC.

Abstract

We present the set-up and performances of the TOP-IMPLART accelerator as a proton source for radiation testing of space components. The TOP-IMPLART proton accelerator is a pulsed fully linear machine aimed at active intensity modulated proton therapy with a final energy of 150 MeV under development at ENEA Frascati Research Centre. Presently the machine offers a beam extraction point on the horizontal line at 35 MeV, with a current up to 50 µA in a 3 µs long pulse and maximum repetition frequency of 50 Hz. An in-air irradiation set-up 1.8 m downstream the accelerator exit window is available with an effective beam energy of about 30 MeV, ±5% transverse homogeneity on a 40 mm diameter beam spot, and intensity repeatability within ±5%. While the proton beams main characteristics (energy, intensity, average fluence) are the same of conventional cyclotron-based cancer treatment facilities, already used for space components qualification, its time structure is significantly different, with high instantaneous dose rate. This research aims at demonstrating the possibility to use an accelerator with these characteristics for radiation hardness assurance testing. A commercial 6-axis integrated MEMS inertial navigation system (accelerometer, gyroscope) was selected as device under test. These devices could be used for example as sensors for attitude determination in University Cubesat missions. Irradiation with proton beams allows simultaneous exploration of total dose, displacement damage and some single-event effect of such components. The results of experimental tests aimed to define an operational procedure are reported: the irradiation set-up is described in detail and the characterization of radiation effect on the component is reported highlighting the consequence of the device performance degradation in terms of the overall navigation system accuracy. These results are considered a first step towards the use of the TOP-IMPLART linear accelerator as a meaningful alternative to traditional cyclotron for proper space qualification of electronic components.
