Background: The origins of life on the Earth required chemical entities to interact with their environments in ways that could respond to natural selection. The concept of interpretation, where biotic entities use signs in their environment as proxy for the existence of other items of selective value in their environment, has been proposed on theoretical grounds to be relevant to the origins and early evolution of life. However this concept has not beendemonstrated empirically.Results: Here, we present data that certain catalytic RNA sequences have properties that would enable interpretationof divalent cation levels in their environment. By assaying the responsiveness of two variants of the Tetrahymena ribozyme to the Ca2+ ion as a sign for the more catalytically useful Mg2+ ion, we show an empirical proof-of-principle that interpretation can be an evolvable trait in RNA, often suggested as a model system for early life. In particular we demonstrate that in vitro, the wild-type version of the Tetrahymena ribozyme is not interpretive, in that it cannot use Ca2+ as a sign for Mg2+. Yet a variant of this sequence containing five mutations that alter its ability to utilize the Ca2+ ion engenders a strong interpretive characteristic in this RNA. Conclusions: We have shown that RNA molecules in a test tube can meet the minimum criteria for the evolution of interpretive behaviour in regards to their responses to divalent metal ion concentrations in their environment. Interpretation in RNA molecules provides a property entirely dependent on natural physico-chemical interactions, but capable of shaping the evolutionary trajectory of macromolecules, especially in the earliest stages of life's history. The mitochondrial genome of greenlip abalone,Haliotis laevigata, is reported. MiSeq and HiSeq sequencing of one individual was assembled to yield a single 16,545 bp contig. The sequence shares 92% identity to theH. rubramitochondrial genome (a closely related species that hybridize withH. laevigatain the wild). The failure of synthetic small-diameter vascular grafts has been attributed to a mismatch in the compliance between the graft and native artery, driving mechanisms that promote thrombosis and neointimal hyperplasia. Additionally, the buckling of grafts results in large deformations that can lead to device failure. Although design features can be added to lessen the buckling potential, the addition is detrimental to decreasing compliance (e.g., reinforcing coil). Herein, we developed a novel finite element framework to inform vascular graft design by evaluating compliance and resistance to buckling. A batch-processing scheme iterated across the multi-dimensional design parameter space, which included three parameters: coil thickness, modulus, and spacing. Three types of finite element models were created in FEBio for each unique coil-reinforced graft parameter combination to simulate pressurization, axial buckling, and bent buckling, and results were analyzed to quantify compliance, buckling load, and kink radius, respectively, from each model. Importantly, model validation demonstrated that model predictions agree qualitatively and quantitatively with experimental observations. Subsequently, data for each design parameter combination were integrated into an optimization function for which a minimum value was sought. The optimization values identified various candidate graft designs with promising mechanical properties. Our investigation successfully demonstrated the model-directed framework identified vascular graft designs with optimal mechanical properties, which can potentially improve clinical outcomes by addressing device failure. In addition, the presented computational framework promotes model-directed device design for a broad range of biomaterial and regenerative medicine strategies.
