The Lower Karamay Formation in the Xia 77 well block of the Xiazijie Oilfield in the Junggar Basin features a complex oil and water relationship in its ultra-low porosity and ultra-low permeability reservoirs. These reservoirs are characterized by low production, high water content, low oil saturation, poor correlation between porosity and permeability, unclear relationship between reservoir parameters and logging responses and difficult identification of oil and water layers. Conventional methods for evaluating and predicting reservoir parameters are poorly suited for this block. Through the analysis of lithology, physical properties and oil-bearing characteristics, it was determined that the reservoir lithology of the Lower Karamay Formation is dominated by glutenite and gravelly sandstones, with mixed-layers of illite and smectite as the dominant clay mineral. The reservoirs are characterized by low porosity and ultra-low permeability with primary intergranular and residual intergranular pores as the main storage space. By establishing an oil saturation interpretation model, it was confirmed that the reservoirs in this area are low oil saturation reservoirs, with oil saturation generally ranging between 36%-55%. The physical properties and oil content of glutenite reservoirs are superior to those of medium to fine sandstones, with reservoir physical properties controlling oil content and exhibiting low saturation characteristics. Electrical properties are influenced by both oil content and lithology. Through studying the formation mechanism of low oil saturation oil reservoirs, it was found that the microscopic pore structure of the reservoirs is the main cause of low oil saturation. By selecting sensitive parameters and utilizing data from natural gamma, resistivity, and acoustic time difference logging, BP neural network technology based on machine learning was introduced to calculate and predict porosity, permeability, and water saturation for the Lower Karamay Formation in Xia 77 well block. The prediction accuracy of reservoir parameters exceeded 80%. The conclusions and methods derived from this study can provide a basis and reference for the prediction of physical parameters in low oil saturation tight sandstone reservoirs.

The Coptidis Rhizoma and Bovis Calculus herb pair possesses clearing heat and detoxifying effects. The aim of this study was to reveal the effects and mechanisms of the herb pair in the treatment of NASH by network pharmacology and experimental verification. A network pharmacology-based approach was employed to predict the putative mechanism of the herb pair against NASH. The high-fat diet (HFD) and methionine/choline deficient (MCD) diet induced NASH models were used to evaluate efficacy and mechanism of the herb pair. Network pharmacological analysis showed that the herb pair modulated NOD-like receptor pathway. In the HFD mice, herb pair reduced body weight, blood sugar, serum ALT, AST, TBA, TC, TG and LDL-C contents, also improved the general morphology and pathological manifestations. Hepatic transcriptomics study showed that herb pair attenuated NASH by regulating NOD-like receptor signaling pathway. Western blotting showed that herb pair reduced the protein expression levels of NLRP3, cleaved Caspase-1 and cleaved IL-1β. In the MCD mice, herb pair also reduced serum ALT, ALT and TBA levels, improved liver pathological manifestations, inhibited the protein expression levels of NLRP3, cleaved Caspase-1 and cleaved IL-1β. Our findings proved that the Coptidis Rhizoma and Bovis Calculus herb pair attenuates NASH through suppression of NLRP3 inflammasome activation. This will demonstrate effective pharmacological evidence for the clinical application of herb pair.

In recent years, China has experienced frequent chemical production accidents. This study collates 1900 briefings of such accidents from 2012 to 2023, sourced from a variety of repositories. By employing association rule mining, we analyzed the connections between causative factors and patterns of these accidents. The analysis revealed significant association rules characterized by high lift values, severe consequences, and patterns not previously recognized. A network model was constructed utilizing Gephi® software to represent the causative factors of these accidents. Through a centrality analysis of the network nodes, key factors contributing to these incidents were identified. Moreover, a SARIMAX model was developed and validated using time series data to predict future accident trends in chemical production. The forecasts generated by this model provide valuable insights for chemical production sectors, highlighting periods with an increased likelihood of accidents. Conclusively, this integration of data mining and predictive modeling could provide a critical method for improving safety protocols and enhancing risk management in chemical industry.

ABSTRACTPeriodontitis has recently been defined as a dysbiotic disease caused by an imbalanced oral microbiota. The transition from commensal microbial communities to periodontitis-associated ones requires colonization by specific pathogens, including Porphyromonas gingivalis. We previously reported an antagonistic relationship between Streptococcus cristatus and P. gingivalis. To determine the role of S. cristatus in altering the interactions of P. gingivalis with other oral bacteria in a complex context, we collected dental plaque samples from patients with periodontitis and assigned them to two groups based on the ratios of S. cristatus and P. gingivalis. We then characterized the microbial profiles of the dental plaque samples using shotgun metagenomic sequencing and compared the oral microbial composition and functional capabilities of the group with high S. cristatus-P. gingivalis ratios with the low ratio group. Taxonomic annotation revealed significant differences in the microbial composition at both the genus and species levels between the low and high S. cristatus-P. gingivalis ratio groups. Notably, a higher microbial diversity was observed in the samples with low S. cristatus-P. gingivalis ratios. Furthermore, the antibiotic resistance gene profiles of the two groups were also distinct, with a significantly increased abundance of the genes in the dental plaque samples with low S. cristatus-P. gingivalis ratios. It, therefore, indicates that the S. cristatus-P. gingivalis ratios influenced the virulence potential of the oral microbiome. Our work shows that enhancing the S. cristatus-P. gingivalis ratio in oral microbial communities can be an attractive approach for revising the dysbiotic oral microbiome.IMPORTANCEPeriodontitis, one of the most common chronic diseases, is linked to several systemic diseases, such as cardiovascular disease and diabetes. Although Porphyromonas gingivalis is a keystone pathogen that causes periodontitis, its levels, interactions with accessory bacteria and pathobionts in the oral microbiome, and its association with the pathogenic potential of the microbial communities are still not well understood. In this study, we revealed the role of Streptococcus cristatus and the ratios of S. cristatus and P. gingivalis in modulating the oral microbiome to facilitate a deeper understanding of periodontitis and its progression. The study has important clinical implications as it laid a foundation for developing novel non-antibiotic therapies against P. gingivalis and improving the efficiency of periodontal treatments.

In order to delay the browning and prolong the shelf life of fresh-cut eggplant during storage, the optimal concentrations of furfuryl thioacetate (FT) on overall sensory quality, browning degree, the activities of enzymes related to browning, contents of total phenol and quinone, and total antioxidant capacity of fresh-cut eggplant during storage at 4 ℃ were investigated. The results showed that 0.64 mmol/L FT treatment significantly inhibited the browning, maintained its overall sensory quality, and prolonged the shelf life of fresh-cut eggplant during storage at 4 ℃ up to day 5. Further studies indicated that 0.64 mmol/L FT treatment suppressed the activities of polyphenol oxidase (PPO) and peroxidase (POD), increased phenylalanine ammonialyase (PAL) activity and DPPH free radical scavenging rate compared with control. In addition, 0.64 mmol/L FT treatment also decreased the contents of total phenol and quinone. Therefore, FT, as a novel anti-browning inhibitor, can delay the browning of fresh-cut eggplant and prolong its shelf life.

The aging behavior of microplastics (MPs) in the environment has garnered significant attention, yet the exact aging process undergone by microplastics during advanced oxidation still remains unclear. This study emphasized elucidating the oxidation and dechlorination mechanisms of polyvinyl chloride microplastics (PVC MPs) within the UV-activated sodium percarbonate system (UV/SPC). After 35 h of UV/SPC (0.05 M) treatment, it exhibited effective dechlorination with a Cl− release of 100.62 ± 13.17 mg/L. And this treatment resulted in the fragmentation and release of fragments from the PVC MPs, thereby contributing to their further aging and dechlorination. In addition, an increase in oxygen-containing functional groups was identified. Quenching experiments confirm that ·OH and ·CO3− are the primary active species present in the system. In summary, the aging of PVC MPs results in a process of fragmentation, releasing fragments that are more fragile and susceptible to mineralization compared to the original PVC MPs. The release of chloride ions originates from the mineralization of these fragments released from PVC MPs by ·OH and ·CO3−. This study has provided valuable insights into the aging mechanisms of MPs in practical wastewater treatment.

Introduction Sepsis is a life-threatening disease that damages multiple organs and induced by the host’s dysregulated response to infection with high morbidity and mortality. Heart remains one of the most vulnerable targets of sepsis-induced organ damage, and sepsis-induced cardiomyopathy (SIC) is an important factor that exacerbates the death of patients. However, the underlying genetic mechanism of SIC disease needs further research. Methods The transcriptomic dataset, GSE171564, was downloaded from NCBI for further analysis. Gene expression matrices for the sample group were obtained by quartile standardization and log2 logarithm conversion prior to analysis. The time series, protein-protein interaction (PPI) network, and functional enrichment analysis via Gene Ontology and KEGG Pathway Databases were used to identify key gene clusters and their potential interactions. Predicted miRNA-mRNA relationships from multiple databases facilitated the construction of a TF-miRNA-mRNA regulatory network. In vivo experiments, along with qPCR and western blot assays, provided experimental validation. Results The transcriptome data analysis between SIC and healthy samples revealed 221 down-regulated, and 342 up-regulated expressed genes across two distinct clusters. Among these, Tpt1, Mmp9 and Fth1 were of particular significance. Functional analysis revealed their role in several biological processes and pathways, subsequently, in vivo experiments confirmed their overexpression in SIC samples. Notably, we found TPT1 play a pivotal role in the progression of SIC, and silencing TPT1 showed a protective effect against LPS-induced SIC. Conclusion In our study, we demonstrated that Tpt1, Mmp9 and Fth1 have great potential to be biomarker of SIC. These findings will facilitated to understand the occurrence and development mechanism of SIC.

Since the proposal of the neurovascular unit (NVU) theory, it has become almost mandatory for neuroprotective medicines against ischaemic stroke (IS) to focus on this unit. Refined Qingkailing (RQKL) is a compound composed of hyodeoxycholic acid, geniposide, baicalin and cholic acid, which has shown great potential in the treatment of IS, but its effect on NVU has not been fully studied. The purpose of this study was to investigate the potential biological pathways that underlie the protective effects of RQKL against NVU damage induced by oxygen–glucose deprivation and re-oxygenation (OGD/R). Using in vitro OGD/R models, we looked into whether RQKL protects the NVU. In order to create an in vitro NVU that resembles IS, we created an OGD/R injury model using primary cultures of brain microvascular endothelial cells, neurons, and astrocytes. Based on our results, we present evidence, for the first time, that RQKL treatment of the injury caused by OGD/R significantly (1) kept the blood brain barrier (BBB) functioning and maintained the architecture of the neurons, (2) mitigated the oxidative stress damage, inflammatory cytokine release, and neuronal death, and (3) upregulated the expression of neurotrophic factors generated from glial cells and the brain in the in vitro model. Therefore, RQKL has a variety of preventive effects against NVU damage caused by OGD/R. RQKL may be a suitable medication for treating IS in a clinical setting.

The dramatic changes in global climate on human health have been extremely severe. The immune disorder caused by low temperature and high humidity (LTHH) have become a severe public health issue. Clinically, Jingfang granule (JF) has the effect of dispelling cold and eliminating dampness, and is widely used in the treatment of cold caused by wind and cold, autoimmune diseases, and COVID-19 with cold-dampness stagnating in the lung pattern. Our study aims to elucidate the effect of JF on LTHH-induced immune disorders in mice as well as the underlying mechanisms. In this study, JF increased the spleen index, improved fecal character, repaired the intestinal barrier and alleviated intestinal inflammatory responses. Most importantly, JF ameliorated immune disorder in LTHH mice, which was manifested primarily by the significant increase in gdT, CD8+ Tcm, and CD8+ Tem cells, as well as the decrease in TH1, TH17, CD4+ Tem1, CD4+ Tem2, immature NK, mature NK cells, and M1-like macrophages. Interestingly, the JF treatment not only regulated the gut microbiota by decreasing the abundance of harmful bacteria, as well as up-regulating the abundance of beneficial bacteria, but also ameliorated the metabolic disorders by reversing the levels of fecal metabolites to normality. The results of the correlation analysis demonstrated a significant association among gut microbiota, fecal metabolites and immune cells. In addition, JF inhibited the TLR4/NF-κB/NLRP3 pathway in LTHH mice. In conclusion, our results suggested that JF alleviated inflammation and immune disorders in LTHH mice by restoring gut microbiota and fecal metabolism.

Objective: To explore the therapeutic capacity of the Liangxue Xiaoban (LXXB) decoction and its disassembled prescriptions in the modulation of T cell subsets and recurrence-related indexes of psoriasis using a psoriasis-like mouse model. Methods: The psoriasis model was generated by the treatment of BALB/c mice (n = 48) with imiquimod. Mice were divided into six groups: control, psoriasis model, tripterygium glycosides, LXXB decoction, Liangxue decoction, and Qingqi decoction. After the intervention period, the interleukin (IL)-17A, IL-22, and interferon-γ levels in mice were examined and hematoxylin and eosin staining was conducted to determine pathological changes in the skin tissues. T cell subset changes in the skin-draining lymph nodes were analyzed using flow cytometry, and the expression levels of the associated transcription factors and recurrence-related indexes in the skin tissues were determined using a polymerase chain reaction. Results: LXXB decoction attenuated the levels of CD8+ T, Th17, and Th1 cells and induced an increase in the Th2 and Treg cell levels. The disassembled prescriptions promoted or inhibited specific subsets of T cells to improve the symptoms of psoriasis. Notably, the LXXB and Liangxue decoctions suppressed the expression of IL-22 at both the gene and protein levels and restored the CD103 and IL-15 expressions in the skin tissue to the normal range. Conclusion: LXXB decoction exerted significant immunoregulatory effects on T cell subsets and improved the recurrence-related indexes. Interestingly, the Liangxue prescription appeared to have a therapeutic advantage in terms of Th17 modulation and psoriasis recurrence, while the Qingqi prescription performed better in Treg immunoregulation.

Background and aimsNon-alcoholic fatty liver disease(NAFLD) is common worldwide and has previously been reported to be associated with sleep traits. However, it is not clear whether NAFLD changes sleep traits or whether the changes in sleep traits lead to the onset of NAFLD. The purpose of this study was to investigate the causal relationship between NAFLD and changes in sleep traits using Mendelian randomization.MethodsWe proposed a bidirectional Mendelian randomization (MR) analysis and performed validation analyses to dissect the association between NAFLD and sleep traits. Genetic instruments were used as proxies for NAFLD and sleep. Data of genome-wide association study(GWAS) were obtained from the center for neurogenomics and cognitive research database, Open GWAS database and GWAS catalog. Three MR methods were performed, including inverse variance weighted method(IVW), MR-Egger, weighted median.ResultsIn total,7 traits associated with sleep and 4 traits associated with NAFLD are used in this study. A total of six results showed significant differences. Insomnia was associated with NAFLD (OR(95% CI)= 2.25(1.18,4.27), P = 0.01), Alanine transaminase levels (OR(95% CI)= 2.79(1.70, 4.56), P =4.71×10-5) and percent liver fat(OR(95% CI)= 1.31(1.03,1.69), P = 0.03). Snoring was associated with percent liver fat (1.15(1.05,1.26), P =2×10-3), alanine transaminase levels (OR(95% CI)= 1.27(1.08,1.50), P =0.04).And dozing was associated with percent liver fat(1.14(1.02,1.26), P =0.02).For the remaining 50 outcomes, no significant or definitive association was yielded in MR analysis.ConclusionGenetic evidence suggests putative causal relationships between NAFLD and a set of sleep traits, indicating that sleep traits deserves high priority in clinical practice. Not only the confirmed sleep apnea syndrome, but also the sleep duration and sleep state (such as insomnia) deserve clinical attention. Our study proves that the causal relationship between sleep characteristics and NAFLD is the cause of the change of sleep characteristics, while the onset of non-NAFLD is the cause of the change of sleep characteristics, and the causal relationship is one-way.

Objective: The Qinzhi Zhudan formula (QZZD) exhibits a prominent therapeutic effect in the treatment of vascular dementia (VaD). This study combined a network pharmacology approach and experimental validation to identify the underlying biological mechanism of QZZD against VaD. Methods: Male Wistar rats received bilateral common carotid artery occlusion (BCCAO) surgery, and after 4 weeks of intragastric administration of QZZD, the therapeutic effect was assessed using the Morris water maze test and cerebral blood flow (CBF) assessment. Hematoxylin and eosin staining, Nissl staining, and electron microscopy were used to measure the histopathological changes in the neurons of rats. The effect of QZZD treatment on hippocampal neurotransmitters was assessed by high-performance liquid chromatography with electrochemical detection and liquid chromatography mass spectrometry. Immunofluorescence was used to observe VaD-induced microglia activation. The inflammatory cytokine levels were assessed by enzyme linked immunosorbent assay. Western blot was used to examine the TNFR1-mediated TNF pathway, which was screened out by network pharmacology analysis. Results: QZZD treatment alleviated pathological changes and neuronal damage in VaD rats and attenuated their cognitive impairment. In addition, QZZD increased CBF and the expression of acetylcholine and 5-hydroxytryptamine in the hippocampal region. Notably, QZZD inhibited microglial activation and the expression of IL-6 and TNF-α. Network pharmacology and western blot indicated that QZZD inhibited the levels of TNFR1, NF-κBp65, p-ERK, TNF-α, and IL-6, which are related to the TNFR1-mediated TNF signaling pathway. Conclusion: QZZD clearly improved learning and memory function, reduced brain pathological damage, elevated CBF and hippocampal neurotransmitter levels, and alleviated neuroinflammation of VaD rats partly by inhibiting the TNFR1-mediated TNF pathway, indicating its potential value in the clinical therapy of VaD.

Plant height is one of the most important agronomic traits that affects yield in wheat, owing to that the utilization of dwarf or semi-dwarf genes is closely associated with lodging resistance. In this study, we identified a semi-dwarf mutant, jg0030, induced by γ-ray mutagenesis of the wheat variety ‘Jing411’ (wild type). Compared with the ‘Jing411’, plant height of the jg0030 mutant was reduced by 7%-18% in two years’ field experiments, and the plants showed no changes in yield-related traits. Treatment with gibberellic acid (GA) suggested that jg0030 is a GA-sensitive mutant. Analysis of the frequency distribution of plant height in 297 F3 families derived from crossing jg0030 with the ‘Jing411’ indicated that the semi-dwarf phenotype is controlled by a major gene. Using the wheat 660K SNP array-based Bulked Segregant Analysis (BSA) and the exome capture sequencing-BSA assay, the dwarf gene was mapped on the long arm of chromosome 2B. We developed a set of KASP markers and mapped the dwarf gene to a region between marker PH1 and PH7. This region encompassed a genetic distance of 55.21 cM, corresponding to a physical distance of 98.3 Mb. The results of our study provide a new genetic resource and linked markers for wheat improvement in molecular breeding programs.

Gegen Qinlian Decoction is one of the commonly used classical prescriptions, which consists of four herbs: Gegen (Puerariae Lobatae Radix), Huangqin (Scutellariae Radix), Huanglian (Coptidis Rhizoma), and Zhigancao Glycyrrhizae Radix et Rhizoma Praeparata cum Melle. The clinical application experience of Gegen Qinlian Decoction is as follows: first, according to Zhongjing Zhang's original text, heat distressing the large intestine with unresolved exterior syndrome is the original meaning of Gegen Qinlian Decoction syndrome. In clinical practice, bacillary dysentery, gastrointestinal cold, etc., belong to simultaneous exterior and interior disease, which are completely consistent with the original meaning of this prescription; second, the clinical digestive system diseases with exuberant heat of the large intestine as the core manifestation cannot be treated merely based on the exterior syndrome; third, due to the interior–exterior relationship between the lung and the large intestine and between the meridians, the use of Gegen (Puerariae Lobatae Radix), Huangqin (Scutellariae Radix), and Huanglian (Coptidis Rhizoma) can make the source of body clear, and the waste qi can be released and the pore can be easily opened, so this prescription can also be used for exogenous fever; fourth, with multidimensional comprehensive understanding of the syndrome, pathogenesis, symptoms and pharmacology, Gegen Qinlian Decoction has also been further applied to the treatment of hypertension, diabetes, etc.

Broccoli (Brassica oleracea L. var. Italic) is rich in nutrition. However, it is susceptible to yellowing after harvest, leading to nutritional and economic losses. In this study, diacetyl, a natural food additive compound, was selected to inhibit the yellowing of broccoli florets and maintain the nutrient quality during storage time. It was found that 20 μl L–1 diacetyl treatment for 12 h could significantly delay the yellowing and decrease the weight loss and lignin content of broccoli florets. Meanwhile, diacetyl could maintain higher contents of chlorophyll, vitamin C and flavonoids and suppress the transcript levels of chlorophyll degradation–related genes in broccoli florets. Moreover, accumulations of reactive oxygen species (ROS) were inhibited by diacetyl treatment. Under diacetyl treatment, the generation of ethylene was prevented by inhibiting the activities and related-gene expressions of 1-aminocyclopropane-1-carboxylic acid (ACC) synthase and ACC oxidase. Based on our findings, exogenous diacetyl could be employed as a novel bioactive molecule for retarding the yellowing and maintaining the quality of postharvest broccoli.

There is yet no effective drug for Alzheimer’s disease (AD) which is one of the world’s most common neurodegenerative diseases. The Qin-Zhi-Zhu-Dan Formula (QZZD) is derived from a widely used Chinese patent drug–Qing-Kai-Ling Injection. It consists of Radix Scutellariae, Fructus Gardeniae, and Pulvis Fellis Suis. Recent study showed that QZZD and its effective components played important roles in anti-inflammation, antioxidative stress and preventing brain injury. It was noted that QZZD had protective effects on the brain, but the mechanism remained unclear. This study aims to investigate the mechanism of QZZD in the treatment of AD combining network pharmacology approach with experimental validation. In the network pharmacology analysis, a total of 15 active compounds of QZZD and 135 putative targets against AD were first obtained. Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were then applied to clarify the biological mechanism. The anti-inflammatory mechanism of QZZD was proved, and a synthetic pathway–TNFR1-ERK1/2-NF-κBp65 signaling pathway was obtained. On the basis of the above discoveries, we further validated the protective effects QZZD on neurons with an APP/PS1 double transgenic mouse model. Weight change of the mice was monitored to assess QZZD’s influence on the digestive system; water maze experiment was used for evaluating the effects on spatial learning and memory; Western blotting and immunohistochemistry analysis were used to detect the predicted key proteins in network pharmacology analysis, including Aβ, IL-6, NF-κBp65, TNFR1, p-ERK1/2, and ERK1/2. We proved that QZZD could improve neuroinflammation and attenuate neuronal death without influencing the digestive system in APP/PS1 double transgenic mice with dementia. Combining animal pharmacodynamic experiments with network pharmacology analysis, we confirmed the importance of inflammation in pathogenesis of AD, clarified the pharmacodynamic characteristics of QZZD in treating AD, and proved its neuroprotective effects through the regulation of TNFR1-ERK1/2-NF-κBp65 signaling pathway, which might provide reference for studies on treatment of AD in the future.

The non-natural mineralization of CaCO3 with special structures or morphologies is generated during the migration of crude oil and is the main form of scale in alkaline/surfactant/polymer (ASP) flooding in oilfields, adversely affecting oil recovery and causing environmental pollution. To date, the mineralization of aragonite superstructures and the role of heavy alkyl-benzene sulfonate (HABS) in mineralization are still unclear. In this work, aragonite-based superstructures of CaCO3 crystals were obtained in an O/W emulsion with HABS to help deepen the understanding of the diversified growth of CaCO3 scaling in oilfields. As a result, rosette-like, bouquet-like, and dumbbell-shaped CaCO3 crystals with vaterite–aragonite, aragonite, and calcite–aragonite phases were formed with 200 mg/L HABS concentration at 45 °C for 60 min and spherical vaterite phase stabilized at a high HABS concentration (800 mg/L and 1000 mg/L). Rhombohedral calcite content experienced a fluctuation of about 40% as the HABS concentration varied. Needle-like and bundle-like aragonite precipitates were generated with increasing temperatures from 65 °C to 85 °C. Thus, HABS affects the nucleation and growth of the precipitated CaCO3 solid, leading to modifications in the structure and morphology of the crystals. The synergistic effect between HABS and temperature can regulate ion pairs with the calcium ions and block sites that are essential to the incorporation of new solutes into the crystal lattice, which leads to the heterogeneous nucleation of vaterite and aragonite on calcite, forming aragonite-based superstructures in kerosene emulsion. This work may enrich the understanding of CaCO3 mineralization in oilfields, and also provide a novel strategy for manufacturing organic–inorganic composites.

Abstract Background PTI1 (Pto-interacting 1) protein kinase belongs to the receptor-like cytoplasmic kinase (RLCK) group of receptor-like protein kinases (RLK), but lack extracellular and transmembrane domains. PTI1 was first identified in tomato (Solanum lycopersicum) and named SlPTI1, which has been reported to interact with bacterial effector Pto, a serine/threonine protein kinase involved in plant resistance to bacterial disease. Briefly, the host PTI1 specifically recognizes and interacts with the bacterial effector AvrPto, which triggers hypersensitive cell death to inhibit the pathogen growth in the local infection site. Previous studies have demonstrated that PTI1 is associated with oxidative stress and hypersensitivity. Results We identified 12 putative PTI1 genes from the genome of foxtail millet (Setaria italica) in this study. Gene replication analysis indicated that both segmental replication events played an important role in the expansion of PTI1 gene family in foxtail millet. The PTI1 family members of model plants, i.e. S. italica, Arabidopsis (Arabidopsis thaliana), rice (Oryza sativa), maize (Zea mays), S. lycopersicum, and soybean (Glycine max), were classified into six major categories according to the phylogenetic analysis, among which the PTI1 family members in foxtail millet showed higher degree of homology with those of rice and maize. The analysis of a complete set of SiPTI1 genes/proteins including classification, chromosomal location, orthologous relationships and duplication. The tissue expression characteristics revealed that SiPTI1 genes are mainly expressed in stems and leaves. Experimental qRT-PCR results demonstrated that 12 SiPTI1 genes were induced by multiple stresses. Subcellular localization visualized that all of foxtail millet SiPTI1s were localized to the plasma membrane. Additionally, heterologous expression of SiPTI1–5 in yeast and E. coli enhanced their tolerance to salt stress. Conclusions Our results contribute to a more comprehensive understanding of the roles of PTI1 protein kinases and will be useful in prioritizing particular PTI1 for future functional validation studies in foxtail millet.

Background: Qinzhi Zhudan Formula (QZZD), optimized from Angong Niuhuang Wan, consists of Radix Scutellariae, Fructus Gardeniae and Pulvis Fellis Suis. We had investigated the neuroprotective effects of QZZD and its active components, and demonstrated that it could treat cerebral ischemia and dementia through multiple pathways and mechanisms. Nevertheless, toxicological data on this formula still remains limited. In the study, we sought to examine the toxicological effects of QZZD during the treatment and recovery periods.Methods: We investigated potential toxicities of QZZD in Sprague-Dawley (SD) rats via 28-day gavage administration. SD rats were randomly divided into control group and treatment groups of A (0.5 g/kg/d QZZD), B (1.5 g/kg/d QZZD), and C (5.0 g/kg/d QZZD). The 56-day course includes treatment period (administration with water or QZZD once a day for 28 consecutive days) and recovery period (28 days). The rats received daily monitoring of general signs of toxicity and mortality, as well as weekly determination of body weight and food consumption. Moreover, the complete blood cell count, biochemistry, coagulation, and urine indicators, organ weights, and histopathological report were analyzed respectively at the end of the treatment and recovery periods.Results: There was no death related to the active pharmaceutical ingredients of QZZD during the treatment period. The maximum no observed adverse effect level (NOAEL) was 0.5 g/kg/d, which is approximately 16.7 times of the equivalent dose of clinical dose in rats. In group TB (1.5 g/kg/d QZZD) and TC (5.0 g/kg/d QZZD), there were adverse effects of blue coloring of tail skin, weight loss, a significant increase of total bilirubin (TBIL), blackening of liver and kidney in gross examination, hyperplasia of bile duct and karyomegaly of hepatocytes in histopathological examination. Besides, in females rats, the food consumption was reduced, while in male rats, there was decrease in triglycerides (TG) and slight increase in white blood cell (WBC) count and neutrophils. In group TC (5.0 g/kg/d QZZD), the indicators of red blood cell (RBC) count, hemoglobin (HGB) and hematocrit (HCT) were decreased slightly, while the platelet count (PLT) was increased. However, these changes were not considered to be toxicologically significant because they resolved during the recovery period.Conclusion: Overall, QZZD exhibited a good safety profile. The maximum no observed adverse effect level was 0.5 g/kg/d, and no target organs toxicity were identified. The present findings might confirm the safety of QZZD in clinical practices.

Calculus bovis (CB), a famous traditional Chinese medicine, is commonly used to treat various inflammatory diseases, including oral ulcers. However, the potential mechanisms of CB in the treatment of oral ulcers are still limited. The present study aimed to use a network pharmacology-based approach to understand the multi-compound, multi-target, multi-pathway effect of CB against oral ulcers. The network pharmacology included the following main parts: first, we screened bioactive compounds and predicted corresponding targets via multiple databases; second, we used R software to analyze related Gene Ontology (GO) and KEGG terms; third, we calculated the network proximity on the protein–protein interaction background and identified potential modules. Finally, a rat oral ulcer model was used to validate the therapeutic effect of CB against oral ulcers and test the expression of important targets. Through network pharmacology analysis, 12 bioactive compounds were initially screened out. The network proximity Z score was -4.37, indicating the potential therapeutic efficacy of CB against oral ulcers. Six potential modules were identified, while module 1 was related to the NOD-like receptor signaling pathway and module 6 was related to apoptosis. Experiments confirmed the therapeutic efficacy of CB against oral ulcers. Western blotting indicated that CB upregulated the anti-apoptosis protein Bcl-xl and downregulated the pro-apoptosis protein Bax and the nlrp3 signal. In conclusion, CB exerted a curative effect on oral ulcers by inhibiting apoptosis and nlrp3 activation.

Asian cultivated rice consists of two main subspecies, xian/indica (XI) and geng/japonica (GJ), and GJ accessions have significantly lower nitrogen-use efficiency (NUE) than XI accessions. In order to facilitate genetic improvement of NUE in GJ accessions, we conducted haplotype analysis of 14 cloned NUE genes using 36 rice germplasm accessions with high-quality reference genome and developed 18 intragenic markers for elite haplotypes, which were then used to evaluate NUE genes in another 41 genetically diverse germplasm accessions from 12 countries and 71 approved GJ cultivars from northern provinces of China. Our results show that elite haplotypes of 12 NUE genes are mainly existed in XI accessions, but few is distributed in GJ accessions. The number of elite haplotypes carried by an XI accession can reach 10, while that carried by a GJ accession is less than 3. Surprisingly, the elite haplotype of gene DEP1 is nearly fixed in approved GJ cultivars, and elite haplotypes of gene MYB61 and NGR5 have been introduced into some approved GJ cultivars. The developed intragenic markers for NUE genes and evaluated 77 genetically diverse rice accessions could be of great use in the improvement of NUE in GJ cultivars.

Nasogastric feeding tube plays an important role in administering enteral feeding and drug delivery for poststroke patients with consciousness disorders or poststroke dysphagia. Nevertheless, placement of nasogastric tubes is not without any risk of potential harm. Inadvertent malposition into the trachea or the distal tracheobronchial tree could induce severe pulmonary complications. As for poststroke patients with long-term dysphoria, such tubes have to be replaced periodically to prevent the overdue service of the tubes. Therefore, the risk of feeding tube misplacement into pulmonary system for these patients is increased. Here, we present a case of a 79-year-old poststroke patient with hydropneumothorax induced by malposition of nasogastric tube into the right pleura after routine replacement, accompanied by acute anterior wall myocardial infarction.

Abstract Background Asthma, a common respiratory disease, is harmful biological effect to our health. As a traditional Chinese medicine for asthma, Majie cataplasm could alleviate the symptoms of asthma and its compositions have immunomodulatory effects. Previous experiments showed that Majie cataplasm was an effective approach to mitigate asthma airway remodeling and had the potential to regulate Th2 cytokines of IL-5 and IL-13. Therefore, our further research focuses on the explanation about the regulatory effect of Majie cataplasm on reshaping Th1/Th2 through their related transcription factors. Methods In this experiment, the launch of asthma model was made by inducing with Ovalbumin (OVA) in C57 mice (n = 40), including 4 groups: the untreated control group (n = 10), the asthma model group (n = 10), the dexamethasone group (n = 10) and the Majie cataplasm group (n = 10). After the intervention, all groups of animals got detected for serum IgE levels, and HE staining of lung tissues was to observe and examine pathological changes. Meanwhile, we analyzed the secretion of IL-4+ T cells and IFN-γ+ T cells in spleen by flow cytometry. The expressions of transcription factor STAT6 mRNA, GATA-3 mRNA and T-bet mRNA in lung tissues was tested by PCR, and western blot had been used to detect levels of JAK2 and STAT3. Background: Xiaoqinglong decoction (XQLD) is a classic Chinese medicinal formula that is widely used to treat allergic asthma. Recently, the use of XQLD to treat allergic asthma has inspired research to determine its mechanism of action. Because dendritic cells (DCs) and the T helper 1 (Th1)/Th2 cytokine balance play important roles in allergic asthma, the present work aimed to assess how these immune system components are affected by XQLD. Methods: Thirty-six female BALB/C mice were randomly divided into three groups: an ovalbumin-based allergic asthma model group, a XQLD treatment group, and a control group. Histology was performed with haematoxylin and eosin staining and immunohistochemical staining. Bronchoalveolar lavage fluid and blood were collected from the animals and used to analyze the composition of inflammatory cells and expression levels of the cytokines interleukin (IL)-5 and IL-13. This paper is concerned with detection of dispersed defects in the resin eyeglass. At present, manual detection is always used in industry, which inevitably causes impairing eyes and a high rate of false-negative detection. Statistics show that its average accuracy and the average detection time are about 85% and 10s, respectively. We for the first time propose an automatic approach to detection of dispersed defects in resin eyeglass, based on the machine vision technology. It is observed that the refractivity of the normal and the defective regions of an eyeglass are different, and thus the reflection image is also collected in our system in addition to the normal transmission image. Such an optical imaging system is modelled and its analysis shows the gray-scale gradient difference between the normal and defective regions in the acquired image is dramatically enhanced under the illumination of a point light source with our designed system. An image processing algorithm is then developed to reveal the above difference and detect dispersed defects in resin eyeglasses. Our simulation study verifies the proposed approach. Further, its experimental evaluation was carried out and the result was consistent with the simulation one, showing that its detection accuracy and the average detection time were 97.50% and 0.636s, respectively, which meet the requirements for online detection of dispersed defects.

Enzymatic browning is the main quality issue of fresh-cut stem lettuce (Lactuca sativa L. var. angustana Irish). In this research, the effect of diacetyl on the browning and browning-related mechanisms of fresh-cut stem lettuce was explored. The data showed that diacetyl treatment with 10 μL L−1 inhibited the browning of fresh-cut stem lettuce and extended the shelf life by over 8 d at 4 °C compared with the control. Diacetyl treatment repressed gene expression and decreased the activities of PAL (phenylalanine ammonia-lyase), C4H (cinnamate-4-hydroxylase) and 4CL (4-coumarate-CoA ligase), which thus reduced the accumulation of individual and total phenolic compounds. Moreover, diacetyl enhanced the antioxidant ability and reduced ROS accumulation, improving the anti-browning ability and indirectly suppressing the biosynthesis of phenolic compounds. These results indicated that diacetyl treatment repressed the browning of fresh-cut stem lettuce by regulating the phenylpropanoid metabolism pathway and antioxidant ability. This study is the first to report that diacetyl has an effective anti-browning role for fresh-cut stem lettuce.

Nitric oxide (NO) is a gaseous free radical that has been become a potential tool to maintain the quality of postharvest horticultural produce. It plays important roles in delaying ripening, alleviating chilling injury, preventing browning, and enhancing disease resistance. The regulatory function of NO is achieved through the post-transcriptional modification of proteins, such as tyrosine nitration, S-nitrosylation, and nitroalkylation. Secondly, NO can also induce the expression of stress-related genes by synergistically interacting with other signaling substances, such as Ca2+, ethylene (ETH), salicylic acid (SA), and jasmonic acid (JA). Here, research progress on the role of NO and its donors in regulating the quality of postharvest fruits and vegetables under storage is reviewed. The function of NO crosstalk with other phytohormones is summarized. Future research directions for NO commercial application and the endogenous NO regulatory mechanism are also discussed.

Excess soluble salts in saline soils are harmful to most plants. Understanding the biochemical responses to salts in plants and studying the salt tolerance-associated genetic resources in nature will contribute to the improvement of salt tolerance in crops. As an emerging model crop, foxtail millet (Setaria italica L.) has been regarded as a novel species for stress resistance investigation. Here, the dynamic proteomic and phosphoproteomic profiling of two foxtail millet varieties of An04 and Yugu2 with contrasting salt tolerance characteristics were investigated under salt stress. In total, 10,366 sites representing to 2,862 proteins were detected and quantified. There were 759 and 990 sites corresponding to 484 and 633 proteins identified under salinity in An04 and Yugu2, respectively, and 1,264 and 1,131 phosphorylation sites corresponding to 789 and 731 proteins were identified between these two varieties before and after salt stress, respectively. The differentially-regulated phosphoproteins (DRPPs) were mainly involved in signal transduction, regulation of gene expression, translation, ion transport, and metabolism processes. Yugu2 possessed signal perception and transduction capabilities more rapidly and had a more intense response compared with An04 upon salinity. The sucrose metabolism pathway, in particularly, might play a vital role in salt response in foxtail millet, which not only provides UDP-glucose for the cellulose synthesis and energy production, but also promotes flavonoid related synthesis to enhance the salt tolerance ability. Over-expressing the phospho-mimic sucrose synthase (SuS) (SuSS10D) in soybean roots enhanced salt tolerance compared with over-expressing SuS lines. The knowledge of this research will shed light on elucidating the mechanisms of salt response, and pave the way for crop varieties innovation and cultivation under salinity and stresses.

Non-alcoholic fatty liver disease (NAFLD) has become the most prevalent liver disease in China. Sinisan (SNS) is a traditional Chinese medicine formula that has been widely used in treating chronic liver diseases, including NAFLD. However, its underlying biological mechanisms are still unclear. In this study, we employed a network pharmacology approach consisting of overlapped terms- (genes or pathway terms-) based analysis, protein-protein interaction (PPI) network-based analysis, and PPI clusters identification. Unlike the previous network pharmacology study, we used the shortest path length-based network proximity algorithm to evaluate the efficacy of SNS against NAFLD. And we also used random walk with restart (RWR) algorithm and Community Cluster (Glay) algorithm to identify important targets and clusters. The screening results showed that the mean shortest path length between genes of SNS and NAFLD was significantly smaller than degree-matched random ones. Six PPI clusters were identified and ten hub targets were obtained, including STAT3, CTNNB1, MAPK1, MAPK3, AGT, NQO1, TOP2A, FDFT1, ALDH4A1, and KCNH2. The experimental study indicated that SNS reduced hyperlipidemia, liver steatosis, and inflammation. Most importantly, JAK2/STAT3 signal was inhibited by SNS treatment and was recognized as the most important signal considering the network pharmacology part. This study provides a systems perspective to study the relationship between Chinese medicines and diseases and helps to discover potential mechanisms by which SNS ameliorates NAFLD.

Background: Baicalin (BCL), a candidate drug for ischemic stroke, has been indicated to protect neurons by promoting brain-derived neurotrophic factor (BDNF). However, the cellular source of BDNF release promoted by baicalin and its detailed protective mechanism after ischemia/reperfusion remains to be studied. The aim of this study was to investigate the neuroprotective mechanisms of baicalin against oxygen–glucose deprivation/reoxygenation (OGD/R) in a neuron–astrocyte coculture system and to explore whether the BDNF-TrkB pathway is involved.Methods and Results: A neuron–astrocyte coculture system was established to elucidate the role of astrocytes in neurons under OGD/R conditions. The results demonstrated that astrocytes became reactive astrocytes and released more BDNF in the coculture system to attenuate neuronal apoptosis and injury after OGD/R. BCL maintained the characteristics of reactive astrocytes and obviously increased the expression of cyclic AMP response element-binding protein (CREB) and the levels of BDNF in the coculture system after OGD/R. To further verify whether BDNF binding to its receptor tyrosine kinase receptor B (TrkB) was required for the neuroprotective effect of baicalin, we examined the effect of ANA-12, an antagonist of TrkB, on NA system injury, including oxidative stress, inflammation, and apoptosis induced by OGD/R. The results showed that treatment of NA systems with ANA-12 significantly attenuated the neuroprotection of BCL. The phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) and mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK) pathways are two important downstream cascades of signaling pathways activated by BDNF binding to TrkB. We investigated the expressions of TrkB, PI3K, Akt, MAPK, and ERK. The results demonstrated that baicalin significantly increased the expressions of TrkB, PI3K/AKT, and MAPK/ERK.Conclusion: The neuroprotective effects of baicalin against oxidative stress, inflammation, and apoptosis were improved by astrocytes, mainly mediated by increasing the release of BDNF and its associated receptor TrkB and downstream signaling regulators PI3K/Akt and MAPK/ERK1/2.

Abstract Background Cerebral ischemia is the second-leading cause of death and the main cause of permanent adult disabilities worldwide. Qingkailing (QKL) injection, a patented Chinese medicine approved by the China Food and Drug Administration, has been widely used in clinical practice to treat cerebral ischemia in China. The NOD-like receptor pyrin 3 (NLRP3) inflammasome is activated in cerebral ischemia and thus, is an effective therapeutic target. AMP-activated protein kinase (AMPK) is an important regulator inhibiting NLRP3 inflammasome activation. Methods We investigated the potential of QKL injection to provide neuroprotection after cerebral ischemia in a rat model of middle cerebral artery occlusion (MCAO). Adult male Sprague-Dawley rats (210–230 g) were randomly divided into three groups which consist of sham, MCAO and 3 ml/kg QKL. Rats in the QKL group received intraperitoneal injections of 3 ml/kg QKL, while rats in other groups were given saline in the same volumes. After 90 min ischemia and 24 h reperfusion, neurological function, laser speckle imaging, brain infarction, brain water content and brain blood barrier permeability were examined and cell apoptosis at prefrontal cortex were evaluated 24 h after MCAO, and western blot and real-time quantitative polymerase chain reaction was also researched, respectively. Results Intraperitoneal administration of QKL alleviated neurological deficiencies, cerebral infarction, blood-brain barrier permeability, brain oedema and brain cell apoptosis after MCAO induction. QKL decreased pro-inflammatory cytokines, TNF-α, IL-6 and IL-1β, and increased anti-inflammatory cytokines, IL-4 and IL-10. Furthermore, QKL activated phosphorylated AMPK, decreased oxidative stress and decreased NLRP3 inflammasome activation. Conclusions QKL relieved cerebral ischemia reperfusion injury and suppressed the inflammatory response by inhibiting AMPK-mediated activation of the NLRP3 inflammasome. These results suggest that QKL might have potential in treating brain inflammatory response and attenuating the cerebral ischemia-reperfusion injury.

Objective: As a traditional herbal formula, Mahuang Xixin Fuzi decoction (MXFD) has been used to treat allergic rhinitis (AR). It regulates the transcription factor GATA3 in T cells, blocks Th2 skewing and rebalances the Th1/Th2 immunity. Type 2 innate lymphoid cells (ILC2s) are closely associated with GATA3. However, it remains unknown whether ILC2s could be one mechanism through which MXFD acts. We sought to elucidate the efficacy and mechanism of action of this decoction in AR treatment. Methods: Forty C57BL/6J female mice were equally divided into control, model, loratadine- and MXFD-treated groups in random. AR was induced by ovalbumin (OVA), and then treatment with loratadine or MXFD was administered. AR scores were evaluated and compared before and after treatment. Pathological changes in the nasal mucosa and lung were observed using hematoxylin-eosin staining of tissue samples. Activation of ILC2 in nasal mucosa was assessed by immunofluorescence and quantitative polymerase chain reaction analysis. ILC2 cell count in lungs was measured by flow cytometry and levels of interleukin- (IL) 4, IL-5 and IL-13 in serum were detected by ELISA. Results: The decoction alleviated the symptoms of AR in mice, improved vascular congestion and expansion, glandular hyperplasia and inflammatory cell infiltration in mouse nasal mucosa and slowly improved the interstitial pneumonia and lesions. Meanwhile, MXFD reduced the number and percentage of ILC2s in the nasal mucosa and lungs, downregulated the expression of GATA3 mRNA and RORα mRNA, and reduced the related inflammatory cytokine levels, including IL-4, IL-5 and IL-13. Conclusion: These data suggest that inhibition of ILC2s by MXFD may be an important means by which to treat AR. Objective: To study the effects of social stress (CS) and social stress combined with high-fat diet on fat mobilization as a candidate mechanism for the induction or aggravation of non-alcoholic fatty liver disease (NAFLD). Methods: Thirty-two male Wistar rats (250 ± 10 g) were randomly allocated to a blank control group (BC), a high-fat diet group (HFD), a CS group, and a combined CS and high-fat diet group (CS + HFD). Rats were sacrificed and tissues were collected after 8 weeks. Liver and body mass were measured and used to calculate the liver index. Serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and free fatty acids (FFAs) were measured. Liver sections were examined microscopically after oil red O and hematoxylin and eosin staining. The relative mRNA expression of acetyl-CoA carboxylase (ACCase), hydroxymethylglutaryl coenzyme A (HMG-CoA) reductase in liver, and hormone-sensitive lipase (HSL), and adipose triglyceride lipase (ATGL) in subcutaneous adipose tissue, were measured by real-time PCR. The liver concentrations of triglyceride, reactive oxygen species, and ACCase were measured by ELISA and HSL activity was determined using turbidimetry. Results: NAFLD developed in the CS, HFD, and CS + HFD groups, with the most severe NAFLD being in the CS + HFD group. Serum AST, ALT, and FFA, liver index, and hepatic triglyceride, FFA, and tumor necrosis factor-α were significantly higher in both the CS and CS + HFD groups. However, food intake and ACCase mRNA expression were lower. The mRNA expression of HSL and ATGL in adipose tissue was much higher, and HSL activity was higher in the CS group than in the BC group, and in the CS + HFD group than in the HFD group. Conclusion: We have successfully established two models of stress-induced NAFLD, suggesting stress can induce and aggravate NAFLD by promoting fat mobilization through upregulation of HSL and ATGL. Objective: To investigate the diversity of experiences of sadness emotion and physiological responses among female college students with high neuroticism (HN) and low neuroticism (LN) during the late luteal phase (LLP) and early follicular phase (EFP). Methods: Forty-two participants were classified as the HN group and 41 as the LN group based on their neuroticism score (Eysenck Personality Questionnaire Short Scale for Chinese). Participants watched sadness-inducing experimental films during the LLP and EFP. Results: The HN group showed similar sadness intensities in both phases. The LN group showed higher sadness intensity in the LLP than the EFP. The LN group's sadness scale assessments were lower than the HN group in both phases. The blood volume pulse in the HN group fell in both the LLP and EFP (both P .05). Between-group comparisons showed heart rate was significantly lower in the HN group than the LN group during EFP (P = .009), but there were no statistically significant differences in subjective experience and physiological data during LLP. The HN group had stronger sadness feelings and greater sadness arousal than the LN group during EFP. There were significant associations between menstrual cycle-related changes in sadness ratings and changes in physiological outcomes. Conclusion: Women with HN have less fluctuation in sadness than those with LN during the menstrual cycle, but experience strong sadness feelings in the LLP. Women with HN show stronger physiological responses in the EFP than those with LN, and have the highest responses in the LLP. Keywords: Neuroticism, Menstruation, Sadness, Emotional responses

In this paper, the issue of stabilization for uncertain rectangular descriptor fractional order systems (FOS) with $0< \alpha < 1$ is considered by designing dynamic compensators. Firstly, the uncertain rectangular descriptor FOS is reconstructed into an augmented uncertain square descriptor FOS. Due to introducing the augmented plant, dynamic compensator can equivalently be transformed into static output feedback. Secondly, two methods for the static output feedback controller design are provided. All results are expressed as a series of linear matrix inequalities (LMIs). Finally, three practical numerical examples are given to verify the effectiveness of the results proposed in this paper.

Asthma poses a threat to human health, and its pathogenesis is closely related to the neuroimmune system. Majie cataplasm can not only regulate the immune system but also the nervous system in asthma patients for its components. We speculate that Majie cataplasm may relieve asthmatic patients with sensitivity to hormone or not by regulating the body's neuroimmune system. Methods: In this experiment, a mouse model of asthma was well established by ovalbumin. The lung function of animals was examined and pathological changes in the lung tissue were assessed by hematoxylin-eosin staining. Serum immunoglobulin E (IgE), calcitonin gene-related peptide (CGRP) and neurokinin A (NKA) were measured by ELISA. The location of CGRP, CD3 and neutrophil in lung tissue and their expressions were detected by immunofluorescence staining. In addition, contents of CGRP mRNA, Substance P (SP) mRNA, interleukin (IL)-17 mRNA and interleukin(IL)-13 mRNA were detected by quantitative polymerase chain reaction. Results: Compared with the asthma model group, Majie cataplasm and dexamethasone can not only equivalently relieve airway hyperresponsiveness, but also make the content of serum IgE reduced. In addition, they can lower the content of serum CGRP and NKA after OVA stimulation, and this effect was more obvious for Majie cataplasm. Our results also showed that Majie Cataplasm and dexamethasone could inhibit the secretion of CGRP and the infiltration of T lymphocytes together with neutrophils in lung tissue and reduce expressions of CGRP mRNA, SP mRNA, IL-17 mRNA and IL-13 mRNA in lung tissue. Conclusion: Majie cataplasm effectively relieves expressions of neuropeptides such as CGRP, reduces the infiltration of immune cells in lung tissue, regulates the body's neuroimmune system, and has a therapeutic potential for both Th2 asthma and neutrophilic asthma.

Stroke is the second leading cause of death after heart disease globally and cerebral ischemic stroke accounts for approximately 70% of all incident stroke cases. We selected four main compounds from a patent Chinese medicine, Qingkailing (QKL) injection, including baicalin from Scutellaria baicalensis Georgi (Huang Qin), geniposide from Gardenia jasminoides J. Ellis (Zhizi), and cholic acid and hyodeoxycholic acid from Bovis Calculus (Niuhuang) with a ratio of 4.4:0.4:3:2.6 m/m, to develop a more efficacious and safer modern Chinese medicine injection against ischemic stroke, refined QKL (RQKL). In this study, we investigated multiple targets, levels, and pathways of RQKL by using an integrative pharm\acology combining experimental validation approach. In silica study showed that RQKL may regulate PI3K-Akt, estrogen, neurotrophin, HIF-1, MAPK, Hippo, FoxO, TGF-beta, NOD-like receptor, apoptosis, NF-kappa B, Wnt, chemokine, TNF, Toll-like receptor signaling pathways against ischemic stroke. The experimental results showed that RQKL improved neurological function and prevented infract volume and blood-brain-barrier damage. RQKL inhibited microgliosis and astrogliosis, and protected neurons from ischemic/reperfusion injury. RQKL also inhibited cell apoptosis and affecting the ratio of the anti-apoptosis protein B-cell lymphoma-2 (Bcl2) and pro-apoptosis protein Bcl2-associated X protein (Bax). Western blot analysis showed that RQKL activated AKT/PI3K signaling pathway and antibody array showed RQKL inhibited inflammatory response and decreased proinflammatory factor Tnf, Il6, and Il1b, and chemokines Ccl2, Cxcl2, and Cxcl3, and increased anti-inflammatory cytokine Il10. In conclusion, RQKL protected tissue against ischemic stroke through multiple-target, multiple signals, and modulating multiple cell-types in brain. This study not only promoted our understanding of the role of RQKL against ischemic stroke, but also provided a pattern for the study of Chinese medicine combining pharmaceutical Informatics and system biology methods.

Abstract Background Foxtail millet (Setaria italica L. P. Beauv) has been considered as a tractable model crop in recent years due to its short growing cycle, lower amount of repetitive DNA, inbreeding nature, small diploid genome, and outstanding abiotic stress-tolerance characteristics. With modern agriculture facing various adversities, it’s urgent to dissect the mechanisms of how foxtail millet responds and adapts to drought and stress on the proteomic-level. Results In this research, a total of 2474 differentially expressed proteins were identified by quantitative proteomic analysis after subjecting foxtail millet seedlings to drought conditions. 321 of these 2474 proteins exhibited significant expression changes, including 252 up-regulated proteins and 69 down-regulated proteins. The resulting proteins could then be divided into different categories, such as stress and defense responses, photosynthesis, carbon metabolism, ROS scavenging, protein synthesis, etc., according to Gene Ontology annotation. Proteins implicated in fatty acid and amino acid metabolism, polyamine biosynthesis, hormone metabolism, and cell wall modifications were also identified. These obtained differential proteins and their possible biological functions under drought stress all suggested that various physiological and metabolic processes might function cooperatively to configure a new dynamic homeostasis in organisms. The expression patterns of five drought-responsive proteins were further validated using western blot analysis. The qRT-PCR was also carried out to analyze the transcription levels of 21 differentially expressed proteins. The results showed large inconsistency in the variation between proteins and the corresponding mRNAs, which showed once again that post-transcriptional modification performs crucial roles in regulating gene expression. Conclusion The results offered a valuable inventory of proteins that may be involved in drought response and adaption, and provided a regulatory network of different metabolic pathways under stress stimulation. This study will illuminate the stress tolerance mechanisms of foxtail millet, and shed some light on crop germplasm breeding and innovation.

Objective: Research on the biological mechanisms by which the liver controls dispersion and regulates emotion usually focuses on psychological stress. This study investigated the biological mechanisms underlying the liver's regulation of emotion using the Trier Social Stress Test (TSST). Differences in subjective stress, subjective emotion, and physiological responses were examined between healthy women (Group A) and depressed women with liver qi stagnation syndrome (Group B). Method: Subjects were 72 women who met the inclusion criteria after physical examination. The EPQ-RSC, BAI, and BDI-21 were used to assess personality and emotion. Subjects completed a 5-point scale to measure subjective stress. Heart rate and heart rate variability were measured. To measure changes in activity of the locus coeruleus–norepinephrine (LC-NE)/autonomic nervous system and the HPA axis, saliva samples were collected at the end of the rest and preparation periods, and 5 time pionts after the TSST task. The PAD Emotion Scale was completed at 4 time points before and after the TSST task. Saliva cortisol was tested. These indexes were compared between the two groups. Objective: Ephedra asarum aconite decoction is used in the clinical treatment of allergic rhinitis. We investigated the effects of ephedra asarum aconite decoction on Th1/Th2 balance in two animal models of allergic rhinitis and in activated T cells in vitro. Methods: Animal models of simple and compound kidney yang deficiency types of allergic rhinitis were treated with ephedra asarum aconite decoction to observe the efficacy of this traditional Chinese medicine formula on allergic symptoms, serum cytokines, and tissue RNA levels. We also cultured and activated T cells from the mouse. After treatment with ephedra asarum aconite decoction, we tested the proliferation and differentiation of T cells. Results: Ephedra asarum aconite decoction relieved allergy symptoms in both types of animal models, and was more effective in the compound kidney yang deficiency model. In addition to its anti-allergic efficacy, ephedra asarum aconite decoction significantly improved the health status of animals in the compound kidney yang deficiency groups, significantly made their bodyweight rising up, and significantly increased the T-bet mRNA/GATA-3 mRNA ratio. Moreover, ephedra asarum aconite decoction promoted the proliferation of activated T cells and inhibited the differentiation to Th2 cells in vitro. Conclusions: Ephedra asarum aconite decoction regulates the balance of Th1/Th2 to ameliorate allergic conditions.

Objective: To assess the efficacy of Qingre Lishi Tuihuang therapy (QLTT) for acute icteric hepatitis B infection. Methods: Eight electronic databases were searched from inception to December 2016 with no language restrictions for reports of randomized controlled trials evaluating the effect of QLTT treating acute icteric hepatitis B. Two researchers independently extracted detailed data and assessed methodological quality. Review Manager 5.3.0 software was used to analyze the data. Results: A total of 13 randomized controlled trials involving 2238 participants were included in this review. The methodological quality was generally poor. The results indicated that supplemented Yinchenhao decoction combined with non-specific treatments was more effective in improving the cure rate (risk ratio = 1.80; 95% CI 1.21–2.68) and reducing the serum levels of total bilirubin (mean difference = −29.74; 95% CI −31.91 to −27.57) and aspartate aminotransferase. Other self-made prescriptions conforming to QLTT plus non-specific treatments had beneficial effect for acute icteric hepatitis B in curing this disease (risk ratio = 1.48; 95% CI 1.27–1.73), as well as for negative seroconversion of HBeAg (risk ratio = 1.39; 95% CI 1.11–1.74). Supplemented Yinchenhao decoction plus non-specific treatments was more effective than other self-made prescriptions conforming to QLTT in reducing serum total bilirubin level. Conclusion: Qingre Lishi Tuihuang therapy appears to improve effect based on non-specific treatments for the treatment of acute icteric hepatitis B. However, it is premature to draw confirmative conclusions, owing to the poor methodological quality and high clinical heterogeneity of the included trials. Further well-designed clinical randomized controlled trials with large sample sizes should be undertaken.

Abstract Background Nonalcoholic fatty liver disease (NAFLD) represents one of the most common forms of liver disease worldwide, and it is always regarded as a consequence of a sedentary, food-abundant lifestyle, sitting for an extended time, and a low physical activity level, which often coincide with chronic and long-lasting psychological stress. A Chinese medicine Sinisan (SNS) may be a potential formula for treating this kind of disease. Methods In this study, a long-term chronic restraint stress protocol was used to investigate the mechanism underlying stress-induced NALFD. Objective: To investigate the molecular mechanism underlying the development of diarrhea-predominant irritable bowel syndrome (IBS-D) with spleen and kidney yang deficiency (SKYD) using a proteomics approach. Methods: Male Sprague–Dawley rats (n = 22) were divided into IBS-D (n = 12) and normal control (n = 10) groups. SKYD was then modeled in IBS-D rats by a combination of acetic acid enema, bondage, rectal dilation, tail stimulation, and Senna gavage. Colon tissue samples were subsequently collected and examined by Q Exactive mass spectrometry to identify differentially expressed proteins between the two groups. Results: The occurrence of SKYD/IBS-D was associated with ribosomal protein S23 (Rps23), protein phosphatase 2 catalytic subunit alpha (Pp2a), and growth factor receptor-bound protein 2 (Grb2), which are involved in the ribosome, neurotrophin signaling, and Janus kinase–signal transducer and activator of transcription (JAK–STAT) signaling pathways. Conclusion: These data suggest that SKYD/IBS-D pathophysiology likely involves inflammation, cell growth, apoptosis, stress granule formation, immune activation, loss of epithelial cell integrity, and visceral hypersensitivity.

Dy ^3+ -doped YCa _4 O(BO _3 ) _3 (Dy:YCOB) crystal was successfully grown by the Czochralski technique. The absorption cross section at 453 nm was 0.28 × 10 ^–21 cm ^2 , that is related to the ^6 H _15/2 → ^4 I _15/2 transition. The Judd-Ofelt parameters Ω _t (t = 2, 4, 6) were 1.62 × 10 ^–20 , 0.10 × 10 ^–20 , 0.41 × 10 ^–20 cm ^2 , respectively. The emission cross section assigned to the transition ^4 F _9/2 → ^6 H _13/2 was 1.02 × 10 ^–21 cm ^2 . The ^4 F _9/2 energy level’s fluorescence lifetime was 900 μ s.

Graphene nanoplatelets (GNPs) and carbon nanotubes (CNTs) are two kinds of significant carbon fillers of conductive switching composites with excellent nonlinear I-V property for protecting overvoltage damages of electronic equipment. In this research, GNPs-CNTs hybrid were fabricated and mixed with epoxy resin (ER) by the way of solution blending. Due to the better morphological features and conductivity of GNPs-CNTs hybrid than pure GNPs or CNTs, the GNPs-CNTs/ER composite could exhibit regulatable I-V behaviors with diverse weight ratios of graphene oxide (GO) to multiwalled carbon nanotubes (MWCNTs) and filler concentrations. Especially, the samples (A-0.7, C-0.7 and C-0.8) with proper filler concentrations and weight ratios of GO to MWCNTs could not only possess good nonlinear conductive characteristic, but exhibit stable reversibility throughout multiple measurements as well, which indicates the GNPs-CNTs/ER composite is more usable and practicable for actual overvoltage protection than previous carbon composite. Furthermore, the mechanisms of regulatable I-V behavior of GNPs-CNTs/ER composite were discussed.

Vanadium dioxide is a typical representative of a strong electronic correlation system. Triggered by stimuli such as heat and electric fields, VO _2 can undergo a reversible transition from insulating to metallic phase and has therefore attracted attention of many researchers. In this work, a hydrothermal method was used to prepare nano-VO _2 particles with phase transition. According to the phenomenology, finite element simulation and theoretical calculation models of the electro-induced phase transition (E-MIT) in the VO _2 -filled Polyethylene glycol were established. Electrode-gap effect on the phase-transition voltage of E-MIT was discussed. Furthermore, the phase transition and heat distribution processes were analyzed. The results revealed that high-temperature conductive channels are formed in the VO _2 structure during the electro-induced phase transition. Moreover, position of these channels is determined by the high-voltage edge effect of the electrode and the heat dissipation effect of the material. The ideal adiabatic E-MIT process was also evaluated. The phase change voltage curve of the theoretical model was well matched with the simulation model. The results reveal that the temperature in the channel is significantly lower than the typical phase change temperature when a phase transition occurs. In addition, owing to an applied voltage, the resistance of the VO _2 composite material can drop suddenly even if the temperature rise is small. The analysis results verified that Joule heating is an important factor in the electro-induced phase transition of VO _2 -filled polymers. The experimental results are helpful in understanding the E-MIT mechanism of VO _2 composite materials and the subsequent application of these materials.

Objective: To evaluate the efficacy and safety of Chinese herbal medicine (CHM) for the treatment of functional dyspepsia (FD). Methods: Web of Science, PubMed, EMBASE, Cochrane Library, and four other Chinese electronic databases, including China National Knowledge Infrastructure (CNKI), Chinese Biological Medical Database (CBM), Chinese Scientific Journals Database (VIP), and WanFang Database were used to search (up to Feb, 2016) for randomized, double-blind, placebo-controlled trials recruiting adults with FD treated with CHM. Study selection, data extraction, quality assessment, and data analyses were conducted based on Cochrane standards using Review Manager software. Results: Fourteen publications (1424 patients) were included. Evidence revealed that CHM was more efficacious than the placebo in improving global dyspepsia symptoms (RR, 1.45; 95% CI, 1.31–1.60), Chinese medicine syndrome (CMS) (RR, 1.36; 95% CI, 1.23–1.50), and quality of life (SMD, 0.30; 95% CI, 0.15–0.45) in FD patients. Furthermore, the difference in the incidence of adverse events between CHM and placebo groups had no statistical significance (RR, 1.06; 95% CI, 0.66–1.70). Conclusion: This meta-analysis demonstrates that CHM has a therapeutic potential in treating FD with a certain safety. However, due to the restricted number of trials included, well-planned, long-term studies are necessary to provide credible evidence.
