- Abstract
Pyrimidine-based compounds, a group of heterocyclic compounds, exhibit a range of biological and pharmacological activities. They can be synthesized through various methods, including condensation, cyclization, and cross-coupling reactions. Notably, Suzuki and Sonogashira reactions are commonly employed in cross-coupling to incorporate aryl and alkynyl groups into the pyrimidine core. These reactions, catalyzed by palladium and copper complexes, are known for their high efficiency, selectivity, and mild reaction conditions. In this review article, we will discuss the recent advancements in pyrimidine-based compound derivative synthesis using Sonogashira and Suzuki coupling reactions. We will focus on the following aspects: 1. the general principles and mechanisms of Sonogashira and Suzuki coupling reactions; 2. the synthesis of different classes of pyrimidine-based compound derivatives using Sonogashira and Suzuki coupling reactions; 3. the biological activities and pharmacological properties of pyrimidine-based compound derivatives synthesized by Sonogashira and Suzuki coupling reactions.
