Buçalışmada, Türkiye için endemik bir bitki olan Origanum acutidens (Hand.-Mazz.) Ietsw. iki farklı çözücü (Su ve %80 etanol) ile ekstrakte edildi. O.acutidens örnekleri Bingöl-Topalan'dan toplandı. (Toplayıcı kayıt numarası:MMH 1157). O. acutidens’in etanol vesu ekstraktlarının antioksidan aktiviteleri, toplam fenolik flavonoidiçeriklerinin yanı sıra element ve fenolik bileşik analizleri yapıldı. Tümanaliz sonuçları 1 mg/mL ekstrakt konsantrasyonuna göre hesaplandı. Analizsonuçlarına göre DPPH aktivitesinde; su ekstraktının % 77,53’ü etanolekstraktının ise % 90,69’u inhibisyon, FRAP aktivitesinde su için 0,34 mg/mL veetanol için 0,96 mg/mL FeSO4 eşdeğerinde aktivite gösterdiği tespitedildi. Toplam fenolik madde tayinindesu ekstraktının 86,48 mg /mL etanol ekstraktının ise 142,78 mg /mL gallik asiteşdeğeri total fenolik madde bulundu. Total flavonoid madde analizinde ise su ekstraktı için 280,58 mg/mL etanolekstraktı için ise 503,82 mg/mL rutin eşdeğeri flavonoid madde içeriği tespitedildi. Ayrıca O. acutidensbitkisinde 15 farklı fenolik bileşik ve 32 farklı element tespit edildi., In thisstudy, Origanum acutidens (Hand.-Mazz.)Ietsw. (zemul), an endemic plant forTurkey, were extracted with two different solvents (Water and 80 % ethanol). O. acutidens samples were collected fromBingöl-Topalan (Collector registration number: MMH 1157). In this study,antioxidant activities, total phenolic and flavonoid content, alsoidentification of element and phenolic compounds of O. acutidens (ethanol and water extracts) were analyzed. Allanalysis results were calculated at concentration of 1 mg/ml extract. Accordingto these results, for DPPH activity; It was found that the water extract showedactivity of 77.53% and ethanol extract 90.69% inhibition, at FRAP activity 0.34mg / mL for water and 0.96 mg / mL FeSO4 equivalent for ethanolextracts. Total phenolic content was calculated as 86.48 mg / mL in waterextract and 142.78 mg / mL gallic acid equivalent total phenolic substance inethanol extract. Total flavonoid analysis revealed 280.58 mg/mL for waterextract and 503.82 mg / mL routine equivalent flavonoid content for ethanolextract. In addition, 15 different phenolic compounds and 32 different elementswere detected in O. acutidens.

In this present study, it was aimed to determine the composition of the essential oil obtained by water distillation from the aerial parts of Origanum acutidens. The oil was analysed by GC-FID and GC/MS. Carvacrol (69.3%), p-cymene (8.3%), y-terpinene (6.3%), β-caryophyllene (2.7%) and borneol (2.2%) were found as main constituents. In addition, headspace-solid phase microextraction using a Polydimethylsiloxane-Divinylbenzene (PDMS/DVB – 65 μm) fiber of O. acutidens was performed and analysed by GC-FID and GC/MS. The main components were identified as: carvacrol (44.0%), p-cymene (25%), ϒ-terpinene (8%), β-caryophyllene (6.1%) and borneol (6.1%), respectively.

Aim: Nowadays, studies on the use of medicinal plants and their essential oils in cancer treatment are increasing rapidly and these studies are very important both scientifically and economically. However, the collection of plants from nature causes extinction. Therefore, it is of great importance to cultivate plants, especially endemic species, to reproduce and consume them. In our study, cytotoxic and membrane damaging effects of essential oils obtained from wild and cultured forms of Origanum acutidens (Hand.-Mazz.) Ietswaart (Lamiaceae) (O. acutidens), which is endemic to Turkey, on non-small-cell lung cancer (NSCLC) cells H1299 and A549 were compared. Material and Method: The level of malondialdehyde, an oxidative stress biomarker, was determined in cell lysates. Assessment of cell viability was made by CellTiter-Blue® Cell Viability Assay and 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay after 10- 250 μg/mL concentrations of wild and cultured forms of O. acutidens essential oil treated to H1299 and A549 cells for 24, 48 and 72 h. Malondialdehyde levels were assayed for determining the membrane damaging effects. Results: Cell viability of H1299 and A549 cells incubated with essential oils obtained from wild and cultured forms of O. acutidens was found to decrease depending on concentration and time. 179 µg/mL, 157 µg/mL, and 132 µg/mL were calculated as IC50 values of wild form of O. acutidens essential oil on H1299 cells for 24, 48, and 72 h, respectively by MTT assay. 150 µg/mL, 131 µg/mL, and 110 µg/mL were calculated as IC50 values of wild form of O. acutidens essential oil on H1299 cells for 24, 48, and 72 h, respectively by resazurin-based assay. 118 µg/mL, 99 µg/mL, and 69 µg/mL were calculated as IC50 values of wild form of O. acutidens essential oil on A549 cells for 24, 48, and 72 h, respectively by MTT assay. 98 µg/mL, 83 µg/mL, and 57 µg/mL were calculated as IC50 values of wild form of O. acutidens essential oil on A549 cells for 24, 48, and 72 h, respectively by resazurin-based assay. Essential oils obtained from wild and cultured forms of O. acutidens increased malondialdehyde level on both H1299 and A549 cells. The greatest membrane damage was observed in A549 cells treated with wild form of O. acutidens essential oil. Conclusion: Essential oils obtained from wild and cultured forms of O. acutidens had cytotoxic effect on lung cancer cells and it has been demonstrated that they showed this effect by causing membrane damage in cells.

The main objective of the current work is to search the antioxidant activities of Origanum acutidens leaves by using DPPH, FRAP and CUPRAC methods severally. Three well known antioxidant compounds (BHA, BHT and ascorbic acid) were used as standards for comparing the extracts. Also, phenolic compounds of Origanum acutidens leaves were identified by UHPLC-ESI-MS/MS. The high concentrations of rosmarinic acid (11158.99 ppb) quinic acid (3200.84 ppb) and naringenin (1238.45 ppb) were detected quantitatively.

Ietswaart and to determine its chemical composition. Materials and Methods: The essential oil was obtained by hydrodistillation and analyzed by gas chromatography/mass spectrometry (GC-MS). The cytotoxic effect of the essential oil on the growth of human colorectal adenocarcinoma (HT-29) and human cervical adenocarcinoma (HeLa) cell lines was investigated by xCELLigence Real Time Cell Analyzer instrument. Moreover, radical scavenging effect of the oil was determined by using DPPH method. Results: The main constituents of the oil were found to be carvacrol (61.69%), p-cymene (17.32%), γ-terpinene (4.05%), and borneol (3.96%). The essential oil of O. acutidens exhibited significant cytotoxic effect against HeLa and HT-29 cell lines at the tested concentrations. The essential oil had moderate DPPH radical scavenging activity compared to butylated hydroxytoluene (BHT). Conclusion: The data in the present study clearly demonstrated inhibitory effect of the oil on two human cancer cell lines. According to these results and other reported studies, this observed high effect may be attributed to the presence of the carvacrol component in the oil.
