The oceanic sink for anthropogenic CO2 (Cant) that humans have emitted into the atmosphere has been pivotal for limiting global warming. The transport of Cant from the surface into the ocean interior, where most of it is accumulating, is the rate limiting step for this uptake. Yet multi-decadal trends in the ocean interior storage of Cant have not been assessed at global scale. We determined such trends by applying the eMLR(C*) regression method to ocean interior observations collected between 1989 and 2020, and found that the global ocean storage of Cant grew by 29 ± 3 Pg C dec-1 and 27 ± 3 Pg C dec-1 (±1σ) from 1994 to 2004 and 2004 to 2014, respectively. Although the two growth rates are not significantly different, they imply a reduction of the oceanic uptake fraction of the anthropogenic emissions from 36 ± 4 % to 27 ± 3 % during the respective decades. We attribute this reduction to a decrease of the ocean buffer capacity and changes in ocean circulation. In the Atlantic Ocean, the maximum storage rate shifted from the Northern to the Southern Hemisphere, plausibly caused by a weaker formation rate of North Atlantic Deep Waters and an intensified ventilation of mode and intermediate waters in the Southern Hemisphere. Between 1994 and 2004, the oceanic Cant accumulation exceeded the net air-sea flux by 8 ± 4 Pg C dec-1, suggesting a loss of natural carbon from the ocean during this decade. Our results reveal a substantial vulnerability of the ocean carbon sink.

The aim of this study was to compare the neuropsychological functioning and symptoms of female and male high school athletes following a single concussion during the school year. The baseline test scores of the Immediate Post-Concussion Assessment and Cognitive Testing (ImPACT) of 309 concussed athletes (169 females, 221 males) in varied sports were obtained. After a sport-related concussion, the athletes were re-administered the ImPACT, on the average, 7.25 days after the injury. MANCOVA compared the ImPACT baseline scores, post-concussion scores, and baseline-to-post-concussion changes of the female and male athletes, with p set at 0.01. The only significant sex comparison found females performing better on baseline visual motor speed (p < 0.001). Otherwise, no sex differences were found in baseline test scores, post-concussion scores, and baseline-to-post-concussion changes. The findings tentatively suggest that there are insignificant sex differences in the consequences of sport-related concussion after a week post-injury. Future studies on sex comparisons after a concussion are needed focusing on the acute and sub-acute phases of the recovery period.

Introduction Exercise is an important therapeutic strategy for heart failure (HF). However, the myocardial effects of resistance exercise during HF are not completely understood. In this study, we investigated the influence of resistance exercise on cardiac remodeling and molecular myocardial changes of rats with myocardial infarction (MI)-induced HF. Methods Three months after MI induction or simulated surgery, Wistar rats were divided into three groups: Sham (n=14); MI (n=9); and MI subjected to resistance exercise (MI-R, n=13). Exercised rats trained 3 times a week during 12 weeks performing four climbs in a ladder with progressive loads. Cardiac structure and left ventricular function were assessed by echocardiogram. Myocyte diameters were measured in histological sections. Energy metabolism, lipid hydroperoxide, antioxidant enzymes activity, malondialdehyde, and protein carbonylation were evaluated by spectrophotometry. NADPH oxidase subunits (Nox2, Nox4, p22phox and p47phox) gene expression was assessed by RT-PCR. Statistical analysis: ANOVA and Tukey test or Dunn's test. Results Mortality did not differ between MI-R and MI groups. MI-R and MI presented dilated left atrium and left ventricle with systolic and diastolic dysfunction. Exercise improved maximum carrying load with no changes in cardiac structure or left ventricle function. Myocyte diameter was lower in MI than Sham and MI-R. Lactate dehydrogenase and creatine kinase activities were lower in MI than Sham. Activity of citrate synthase and catalase was lower in MI and MI-R than Sham. Lipid hydroperoxide concentration was lower in MI-R than MI. Nox2 and p22phox gene expression was higher in MI-R than Sham. Gene expression of Nox4 was higher in both infarcted groups and gene expression of p47phox was lower in MI than Sham. Conclusion Resistance exercise is safe and well tolerated by infarcted rats. Exercise increases maximum carrying load and reduces myocardial oxidative stress with no changes in cardiac structure or left ventricle function of infarcted rats. Background Sodium-glucose cotransporter 2 (SGLT2) inhibitors have beneficial effects on the cardiovascular system in diabetes mellitus (DM) patients. However, as most clinical trials were performed in type 2 DM, the effects of SGLT2 inhibitors in patients with type 1 DM still need further clarification. In this study, we evaluated the effects of long-term treatment with the SGLT2 inhibitor dapagliflozin on cardiac remodeling, myocardial function, and energy metabolism in rats with type 1 DM. Methods Male Wistar rats were divided into three groups: control (C, n=15); DM (n=15); and DM treated with dapagliflozin (DM+DAPA, n=15) for 30 weeks. DM was induced by streptozotocin; DAPA was added to the rat chow (5 mg/kg/day). Cardiac performance was evaluated by echocardiogram and myocardial function in isolated left ventricular (LV) papillary muscle preparations. Myocardial energy metabolism enzyme activities were evaluated by spectrophotometry. Statistical analyzes: ANOVA and Tukey or Kruskal-Wallis and Dunn. Results DM+DAPA had lower glycemia than DM [C 112 (108–116); DM 531 (522–535)*; DM+DAPA 267 (179–339) mg/dL; p Conclusion Long-term dapagliflozin treatment attenuates cardiac remodeling and myocardial dysfunction and preserves hexokinase, phosphofructokinase and pyruvate kinase activity in rats with type 1 diabetes mellitus. Introduction Skeletal muscle energy metabolism is commonly altered in heart failure patients, with a metabolic shift from oxidative to glycolytic muscle fiber. These changes contribute to reduced functional capacity. Sodium glucose co-transporter type 2 (SGLT2) inhibitors improve cardiovascular outcomes in both diabetic and non-diabetic patients, as well as those with and without heart failure. However, the effects of SGLT2 inhibitors on skeletal muscle during heart failure have not been established. The aim of this study was to assess the metabolic effect of empagliflozin (EMPA) on skeletal muscle of rats with myocardial infarction (MI)-induced heart failure. Methods One week after MI induction or simulated surgery, male Wistar rats were divided into four groups: Sham (n=10), Sham+Empa (n=12), MI (n=10), and MI+Empa (n=09). EMPA was added to rat chow (5 mg/kg/day). Rats were supplied with ad libitum water and chow for 12 weeks. Infarct size was measured by histological analysis. Metabolic enzyme activity in the soleus muscle was assessed by spectrophotometry. Statistical analysis: ANOVA and Tukey, and Student's t tests. Results Only rats with infarction size greater than 35% of total left ventricle area were included in this study. Infarction size did not differ between infarcted groups (MI 41.8±4.2; MI+Empa 40.7±5.7 of total left ventricle area). In the MI soleus muscle, metabolic enzyme activity of glucose-6-phosphate-dehydrogenase, citrate synthase and beta-hydroxy-acyl-dehydrogenase was higher than the Sham group. These changes were not observed in the MI+Empa group. MI+Empa had lower hexokinase, phosfructokinase, and pyruvate kinase activity (glycolytic metabolism enzymes), and lower citrate synthase and glucose-6-phosphate-dehydrogenase activity than MI. Conclusion Chronic treatment with SGLT2 inhibitor empagliflozin prevents metabolic abnormalities in skeletal muscle in infarcted rats. The poultry sector demands alternative additives to antibiotics that can be used as performance enhancers. Therefore, this experiment was conducted to evaluate the probiotics effects on performance, intestinal health, and redox status of 720 broilers exposed to heat stress from 15 days of age. Eight dietary treatments were evaluated: basal diet (BD) without antibiotic and probiotic (T1); BD supplemented with antibiotic zinc bacitracin (T2), BD supplemented with commercial probiotic of Bacillus subtilis DSM 17,299 (T3), BD supplemented with non-commercial probiotic of Lactococcus lactis NCDO 2118, Lactobacillus delbrueckii CNRZ 327, Escherichia coli CEC15, or Saccharomyces boulardii (T4 to T7), and BD simultaneously supplemented with the four non-commercial probiotics (T8). Feed intake, weight gain, and feed conversion were determined in the period from 1 to 42 days of age. Carcass and cuts yield, abdominal fat deposition, cloacal temperature, weight and length of intestine, activity of myeloperoxidase and eosinophilic peroxidase enzymes in the jejunum, jejunal histomorphometry, relative gene expression in the jejunum (occludin, zonulin, interleukin-8, cholecystokinin, ghrelin, and heat shock protein-70), and liver (heat shock protein-70), in addition to malondialdehyde level and superoxide dismutase activity in the intestine, liver, and blood, were measured in broilers at 42 days old. As main results, broilers fed T1 diet exhibited lower weight gain (3.222 kg) and worse feed conversion (1.70 kg/kg). However, diets containing non-commercial probiotics resulted in up to 3.584 kg of weight gain and improved feed conversion by up to 10%, similar to that observed for broilers of the T2 and T3 groups.

Nonalcoholic fatty liver disease (NAFLD), often associated with obesity, is becoming one of the most common liver diseases worldwide. It is estimated to affect one billion individuals and may be present in approximately 25% of the population globally. NAFLD is viewed as a hepatic manifestation of metabolic syndrome, with humans and animal models presenting dyslipidemia, hypertension, and diabetes. The gut-liver axis has been considered the main pathogenesis branch for NAFLD development. Considering that foods or beverages could modulate the gastrointestinal tract, immune system, energy homeostasis regulation, and even the gut-liver axis, we conducted an exploratory study to analyze the effects of kombucha probiotic on hepatic steatosis, glucose tolerance, and hepatic enzymes involved in carbohydrate and fat metabolism using a pre-clinical model. The diet-induced obese mice presented glucose intolerance, hyperinsulinemia, hepatic steatosis, increased collagen fiber deposition in liver vascular spaces, and upregulated TNF-alpha and SREBP-1 gene expression. Mice receiving the kombucha supplement displayed improved glucose tolerance, reduced hyperinsulinemia, decreased citrate synthase and phosphofructokinase-1 enzyme activities, downregulated G-protein-coupled bile acid receptor, also known as TGR5, and farnesol X receptor gene expression, and attenuated steatosis and hepatic collagen fiber deposition. The improvement in glucose tolerance was accompanied by the recovery of acute insulin-induced liver AKT serine phosphorylation. Thus, it is possible to conclude that this probiotic drink has a beneficial effect in reducing the metabolic alterations associated with diet-induced obesity. This probiotic beverage deserves an extension of studies to confirm or refute its potentially beneficial effects.

Background: Current research on concussion incidence in youth athletes (age Hypothesis: Concussion injuries commonly occur in high school sports programs. The likelihood of concussion among student-athletes (aged 13-18 years) depends on the sport they are participating in as well as the sex of the athlete. Study Design: Descriptive epidemiology study. Level of Evidence: Level 4. Methods: A retrospective analysis of all Hawaii high school athletes aged 13 to 18 years participating in 14 sports from 2011 through 2017 was performed as part of a statewide standardized concussion assessment and management program. Results: A total of 5993 concussions were identified among 92,966 athletes. The overall concussion rate was 0.96 (95% CI, 0.94-0.99). Girls’ judo had the highest concussion rate (1.92; 95% CI, 1.68-2.17) followed by football (1.60; 95% CI, 1.53-1.66). The concussion rate for boys (1.0; 95% CI, 0.97-1.03) was higher than that for girls (0.91; 95% CI, 0.87-0.95); however, in 4 of the 5 sports in which both girls and boys participated, girls had a higher rate of concussion injury. Conclusion: The likelihood of concussion among student-athletes aged 13 to 18 years may be higher than previously thought and varies depending on sport and sex. Clinical Relevance: Epidemiologic data on concussion injury in children and adolescents are useful in accurately determining the relative risks of high school sports participation and may be valuable in determining the appropriate allocation of health care and scholastic resources for student-athletes, as well as the impact of rule and training modifications designed to improve participant safety.

Context It is unclear whether vitamin D supplementation reduces risk of falls, and results from randomized controlled trials (RCTs) are conflicting. Objective The objective of this work is to determine whether 2000 IU/day of supplemental vitamin D3 decreases fall risk. Design VITamin D and OmegA-3 TriaL (VITAL) is a double-blind, placebo-controlled RCT including 25 871 adults, randomly assigned November 2011 to March 2014 and treated for 5.3 years (median). Setting This is a nationwide study. Participants Men 50 years or older and women 55 years or older (mean age, 67.1 years) without cancer or cardiovascular disease at baseline participated in this study. Interventions Interventions included vitamin D3 (cholecalciferol; 2000 IU/day) and/or omega-3 fatty acids (1 g/day) or respective placebos in a 2 × 2 factorial design. Main Outcome Measures Main outcome measures include 2 or more falls and falls resulting in a doctor or hospital visit. Results Baseline serum total 25-hydroxyvitamin D (25[OH]D) level was 77 nmol/L; characteristics were well-balanced between groups. Numbers of participants with 2 or more falls were similar between active and placebo groups (9.8% vs 9.4%). Over 5 years, there were no differences in the proportion having 2 or more falls (odds ratio [OR] = 0.97; 95% CI, 0.90-1.05, P = .50), falls resulting in a doctor visit (OR = 1.03; 95% CI, 0.94-1.13, P = .46), or resulting in a hospital visit (OR = 1.04; 95% CI, 0.90-1.19, P = .61) between groups. Results did not differ between those with baseline 25(OH)D less than 50 vs 50 nmol/L or greater or other cut points. Conclusion Daily supplemental vitamin D3 vs placebo did not decrease fall risk in generally healthy adults not selected for vitamin D insufficiency. This large RCT does not indicate that supplemental vitamin D should be used for primary prevention of falls in the US population.

This study aims to create controlled fine space by electrospinning, and to develop the electrode materials for high-performance energy devices. With the popularization of mobile devices, household appliances, hybrid vehicles, electric vehicles, and the like, the use of power storage devices is expanding, and further performance improvements are required. In this study, a novel electrode material was developed by compositing Si with carbon nanofibers derived from polyacrylonitrile (PAN) by electrospinning and heat treatment. The texture and structure of the nanofibers were observed and analyzed by scanning electron microscopy (SEM), energy dispersive X-ray spectrometry (EDX) and transmission electron microscopy (TEM) combined with image processing. Nano spaces were created in the CNFs and Si particles were able to be contained in the CNFs. In the second and subsequent cycles of the charge/discharge experiments of lithium ion battery (LIB) electrode made from the materials, the capacity was more than twice the theoretical capacity using graphite, and good cycle performance was obtained.

Huanglongbing (HLB), caused by ‘Candidatus Liberibacter asiaticus’ (CLas), is a devastating disease of citrus. After initial infection, CLas quickly colonizes the root system before canopy symptoms develop. There is limited understanding of CLas movement from roots to canopy and local and systemic effects on root dynamics. Using split-root rhizoboxes and late summer below-the-split bud inoculation, effects of local infection on systemic disease development were studied. Upward bacterial movement from roots is linked to seasonal flushes and CLas population in roots. CLas stayed isolated to one side of the roots for at least 8 months, until the spring flush. HLB caused differential root responses depending on tree age at infection. Systemic effects, independent of CLas movement, occur very early after infection. Stimulation of root growth occurred on noninfected roots prior to CLas detection in 1.5-year-old trees but decreased in 2.5-year-old trees. Independent of tree age, root growth was stimulated during spring root flushes after CLas population stabilized. Root dieback began simultaneously with detection of CLas in roots (6 weeks postinoculation). Infection and tree age altered root lifespan. In total, 1.5-year-old CLas-infected roots from summer and fall flushes had 3 and 6 weeks reduced lifespan. In contrast, 2.5-year-old CLas-infected plants lifespan was unaffected. Season affected root lifespan with late summer root flush lifespan was three times shorter than fall or spring root flushes. Split-root inoculation allowed study of local and systemic effects of CLas infection in roots, information crucial to prolonging the productivity of HLB-affected trees. O consumo de produtos oriundos da piscicultura tem crescido consideravelmente, e tende a aumentar nas próximas décadas, tanto pela demanda do consumidor, como pelas inovações tecnológicas que a mesma vem passando, principalmente nas técnicas de processamento para desenvolvimento de produtos práticos e convenientes. Com isso, o objetivo do presente estudo foi realizar uma revisão bibliográfica acerca das tecnologias emergentes para o processamento de pescado. As novas tecnologias de processamento de alimentos, como aquecimento por infravermelho, aquecimento ôhmico, alta pressão e campos elétricos pulsados, oferecem alimentos seguros e de alta qualidade, no entanto, uma série de questões foram identificadas que atrasam a implementação mais ampla dessas técnicas, incluindo as questões do enorme custo de investimento e a falta de regulamentações adequadas no país. O Brasil apresenta grande potencial de expansão das atividades aquícolas, mas ainda há a necessidade de desenvolvimento de projetos de apoio técnico na área, bem como de maior divulgação das pesquisas para os produtores. O estudo demonstrou que, dentre todos os processamentos emergentes, o processamento em alta pressão, ainda segue sendo o tratamento térmico mais bem sucedido.

A aquicultura é a única forma de garantir pescados suficientes para o mundo, haja visto que os estoques mundiais de peixes selvagens estão se esgotando em uma velocidade cada vez maior. A aquicultura precisa de tecnologias inovadoras para aumentar a produção de peixes. Tecnologias inovadoras, incluindo edição de genoma, inteligência artificial, sistemas de recirculação, proteínas e óleos alternativos para substituir refeições e óleos de peixe, vacinação oral, pode fornecer soluções para uma aquicultura sustentável e lucrativa. Esta revisão tem como objetivo abordar a tecnologia da informação digital, bem como integrar essas tecnologias à aquicultura visando uma melhorar produtividade, sustentabilidade e lucratividade.

O aumento da demanda mundial por carne de peixe implica em melhoramento de novas espécies para atender ao crescente mercado. Brasil possui um grande potencial para o desenvolvimento do setor aquícola, por apresentar condições favoráveis, tais como clima favorável e água em grande quantidade, favorecendo assim, o crescimento da aquicultura no país. Entretanto, poucos animais explorados comercialmente são geneticamente melhorados. Para contribuir com este crescimento da demanda por peixes, os programas de melhoramento genético de peixes tornam-se uma alternativa para melhorar a produtividade. Programas de melhoramento eficientes podem contribuir consideravelmente para o desenvolvimento da piscicultura por reduzir os custos de produção, melhorar a resistência dos organismos cultivados a doenças, melhorar o aproveitamento alimentar e a qualidade dos produtos. Neste contexto, o presente trabalho tem como objetivo realizar uma revisão de programas de melhoramento genético de tilápia e espécies nativas no Brasil.

Na aquicultura, os procedimentos de manejo dos peixes são muitas vezes traumáticos, podendo causar graves reações fisiológicas e bioquímicas. Com o aumento da produção aquícola, percebe-se a necessidade de conhecer a influência do manejo sobre a qualidade da carne. Assim, o objetivo desta revisão bibliográfica foi avaliar como o estresse pode afetar a qualidade da carne em peixes. Através da análise de trabalhos disponíveis, pode-se perceber que o estresse afeta a qualidade da carne do pescado, já que a natação vigorosa leva ao uso intenso do músculo branco. Animais estressados antes do abate entram em rigor mortis mais rapidamente, diminuindo o pH da carne, sua vida útil e qualidade. Além disso, o manuseio e processamento do peixe durante o rigor mortis causam menor rendimento do filé e perda da qualidade da carne. O estresse agudo antes do abate pode desencadear carnes com textura menos firme, ao passo de que estresse de longa duração acarreta em aumento da firmeza da carne. Além disso, o estresse pode provocar aumento na incidência de gaping e diminuição da capacidade de retenção de água. A cor da carne varia muito entre espécies de peixes, podendo ou não estar associada ao estresse ante mortem.

Significance Extrachromosomal circular DNA (eccDNA) plays a role in human diseases such as cancer, but little is known about the impact of eccDNA in healthy human biology. Since eccDNA is a tiny fraction of nuclear DNA, artificial amplification has been employed to increase eccDNA amounts, resulting in the loss of native compositions. We developed an approach to enrich eccDNA populations at the native state (naïve small circular DNA, nscDNA) and investigated their origins in the human genome. We found that, in human sperm, the vast majority of nscDNA came from high-copy genomic regions, including the most variable regions between individuals. Because eccDNA can be incorporated back into chromosomes, eccDNA may promote human genetic variation., Extrachromosomal circular DNA (eccDNA) originates from linear chromosomal DNA in various human tissues under physiological and disease conditions. The genomic origins of eccDNA have largely been investigated using in vitro–amplified DNA. However, in vitro amplification obscures quantitative information by skewing the total population stoichiometry. In addition, the analyses have focused on eccDNA stemming from single-copy genomic regions, leaving eccDNA from multicopy regions unexamined. To address these issues, we isolated eccDNA without in vitro amplification (naïve small circular DNA, nscDNA) and assessed the populations quantitatively by integrated genomic, molecular, and cytogenetic approaches. nscDNA of up to tens of kilobases were successfully enriched by our approach and were predominantly derived from multicopy genomic regions including segmental duplications (SDs). SDs, which account for 5% of the human genome and are hotspots for copy number variations, were significantly overrepresented in sperm nscDNA, with three times more sequencing reads derived from SDs than from the entire single-copy regions. SDs were also overrepresented in mouse sperm nscDNA, which we estimated to comprise 0.2% of nuclear DNA. Considering that eccDNA can be integrated into chromosomes, germline-derived nscDNA may be a mediator of genome diversity.
