Diese Arbeit hatte es zum Ziel, eine neue und effiziente Affinitätsplattform für die Aufreinigung und Isolierung von Proteinen basierend auf nicht‐porösen und stabilen Korund‐Partikeln zu entwickeln. Das Rohmaterial wurde kovalent mit Proteinbindern modifiziert, um so die Isolierung spezieller Target‐Proteine aus komplexen biologischen Proben zu realisieren. Für die erste Modifizierungsschicht auf der Korundoberfläche wurden verschiedene Phosphonsäuren und Silane untersucht. Anschließend wurde der etablierte bifunktionale Crosslinker Glutaraldehyd mit dem biokompatibleren verzweigten Polyglycerol (PG) verglichen. Es konnte gezeigt werden, dass Polyglycerol eine hervorragende Alternative zu Glutaraldehyd darstellen kann. Für die angestrebte Anwendung als neues Tool für die Antikörperreinigung wurde humanes IgG mit Protein‐A‐funktionalisierten Korundpartikeln erfolgreich aus Humanplasma isoliert. Um das Anwendungsspektrum der neuen Korundmethode auszuweiten, wurde im zweiten Teil dieser Arbeit eine Affinitätsplattform für die Isolierung Polyhistidin‐getaggten Proteinen basierend auf Metallionen‐funktionalisiertem Korund entwickelt und angewendet. Hauptkriterien dieser Methodenentwicklung waren die schnelle, effiziente und ökonomische Aufreinigung rekombinanter Proteine aus bakteriellen Lysaten in einem säulenfreien Format. Als Modellsystem wurde Polyhistidin‐getaggtes Protein A/G (His6‐PAG) mit Rinderserumalbumin versetzt, was die Entwicklung eines optimierten Protokolls für die Isolierung rekombinanter Proteine ermöglichte. Im direkten Vergleich mit kommerziellen Ni‐NTA‐Agarose‐Beads zeigten die Korundpartikel höhere Proteinreinheiten. Abschließend konnte gezeigt werden, dass Zink eine ideale Alternative als Metallion darstellt, um etwaige Nachteile des Nickels in der Anwendung von Life‐Science‐Methoden zu umgehen und eine zukunftsträchtige Methode mit weiterem Potenzial zu realisieren., This work aimed to develop and establish a novel efficient affinity platform for protein purification based on nonporous, stable, and easily available corundum powder. The material was functionalized covalently with protein binders to isolate and enrich specific proteins from complex biological matrices. Phosphonic acids and silanes were tested as the first modification layer. In the next step, the well‐known bifunctional crosslinker glutaraldehyde was compared with a more biocompatible, hyperbranched polyglycerol (PG). It could have been shown that oxidized polyglycerol is an excellent alternative to glutaraldehyde. Human IgG was purified with protein A functionalized corundum from crude human plasma for the initial purpose of antibody isolation. To broaden the application of functionalized corundum, a novel purification platform for the isolation of poly His‐tagged proteins based on metal‐ion‐functionalized corundum was established.The main goal of this approach was the efficient, economical, and fast purification of recombinant proteins in a column‐free format that can also easily be performed in moderately equipped laboratories. As a model system for method optimization, His‐tagged protein A/G (His6‐PAG), mixed with bovine serum albumin (BSA), was established leading to a purification protocol with minimal nonspecific binding by the variation of the imidazole content in the used binding and washing buffers. Corundum in direct comparison with standard Ni‐NTA agarose beads generated higher purities of isolated proteins. Finally, it was shown that zinc‐functionalized corundum is another efficient approach to circumvent potential negative effects of nickel use in life science applications.

Induktiv gekoppelte Plasma-Massenspektrometrie mit Laserablation (LA-ICP-MS) wird zunehmend für die Einzelzellanalyse eingesetzt, jedoch wird eine weitere verbreitete Verbreitung durch den geringen Durchsatz behindert. Daher wurde in dieser Arbeit der Durchsatz von Einzelzellen-LA-ICP-MS untersucht und verbessert. Zunächst werden die beiden möglichen Ablationsmodi, Bildgebung und Einzelpunktanalyse (SSA), hinsichtlich ihrer analytischen Gütezahlen (Signal-Rausch-Verhältnis, Präzision, Genauigkeit, Durchsatz) verglichen. Hierfür wurden adhärente 3T3-Fibroblastenzellen mit zwei Metallfarbstoffen angefärbt und mit beiden Methoden mehrere Dutzend Zellen vermessen. SSA zeigte überlegene Eigenschaften hinsichtlich Durchsatz und Nachweisgrenzen. Darüber hinaus wurde gezeigt, dass >400 Zellen analysiert werden müssen, um zufriedenstellende Statistiken für einen quantitativen Vergleich der Ergebnisse zu erhalten, was als zu mühsam befunden wurde. Daher wurde ein Einzelzellen-Arraying-Schritt integriert, um eine automatisierte LA-ICP-MS-Analyse zu ermöglichen. Hierfür wurden zwei Arrayingverfahren getestet: Zunächst wurde das mikrofluidische Arraying von Zellen getestet, jedoch verhinderte das Einklemmen von weichen PDMS-Chips eine erfolgreiche Anwendung, und eine Neugestaltung des Chips wäre erforderlich. Daraufhin wurde eine neuartige Technologie getestet, die auf dem Arraying von Tröpfchen in Verbindung mit der Bilderkennung von Zellen beruht, wobei ein Anordnungsdurchsatz von 550 Zellen pro Stunde und eine beispiellose Einzelzellengenauigkeit (> 99%) gefunden wurde. In einem Proof-of-Principle-Experiment wurde ein Zellarray von THP-1-Suspensionszellen mittels LA-ICP-TOF-MS analysiert und erstmals gleichzeitig endogene und exogene Isotope einzelner Zellen als Isotopen-Fingerabdrücke von Zellen mit Nachweisgrenzen von lediglich wenigen hundert attogramm. Schließlich wurden diese Ergebnisse mit der derzeit gebräuchlichsten Analysemethode Single-Cell (sc)-ICP-MS verglichen., Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is increasingly used for single-cell analysis. However, a more widespread use of LA-ICP-MS in single cell analysis is hampered by its low throughput. Hence, in this work the throughput of single cell LA-ICP-MS was studied and improved. First, the two possible ablation modes, imaging and single spot analysis (SSA) of single cells using a large laser spot, are compared regarding their analytical figures of merit (signal to noise, precision, accuracy, throughput), as well as regarding ease of operation and data evaluation. For that, adherent 3T3 fibroblast cells were stained with two metal dyes and several dozen cells were measured using both modes. SSA showed superior characteristics regarding throughput and detection limits. Moreover, it was shown that >400 cells must be analyzed to reach satisfactory statistics for a quantitative comparison of results, which would have been too laborious. Thus, a single cell arraying step was integrated to enable automated LA-ICP-MS analysis. Two different arraying methods were evaluated: First, arraying via hydrodynamic front trapping of cells using a microfluidic device was tested, but clamping of soft PDMS-chips prevented successful arraying and it was concluded that a major redesign of the chip is necessary. Secondly, and a novel technology relying on a microdroplet arrayer in conjunction with image recognition of cells was tested and a moderate arraying throughput (550 cells per hour) and an unprecedented single-cell accuracy (>99%) was found. In a proof of principle experiment, a cell array of THP-1 suspension cells was analyzed using LA-ICP-TOF-MS and endogenic and exogenic isotopes of individual cells were detected for the first time simultaneously as isotopic fingerprints of cells with detection limits as low as hundred attogram. Finally, these results were compared to the currently more commonly used analysis method single-cell (sc)-ICP-MS.

Nephrotoxicity is a major limitation to cisplatin antitumor therapies. Cilastatin, an inhibitor of renal dehydropeptidase-I, was recently proposed as a promising nephroprotector against cisplatin toxicity, preventing apoptotic cell death. In this work, cilastatin nephroprotection was further investigated in a rat model, with a focus on its effect on 76 renal lipids altered by cisplatin, including 13 new cisplatin-altered mitochondrial cardiolipin species. Lipid imaging was performed with MALDI mass spectrometry imaging (MALDI-MSI) in kidney sections from treated rats. Cilastatin was proved to significantly diminish the lipid distribution alterations caused by cisplatin, lipid levels being almost completely recovered to those of control samples. The extent of recovery of cisplatin-altered lipids by cilastatin turned out to be relevant for discriminating direct or secondary lipid alterations driven by cisplatin. Lipid peroxidation induced by cisplatin was also shown to be reduced when cilastatin was administered. Importantly, significant groups separation was achieved during multivariate analysis of cortex and outer-medullary lipids, indicating that damaged kidney can be discerned from the nephroprotected and healthy groups and classified according to lipid distribution. The quest for internal standards useful in MALDI imaging studies goes on to get not only lateral distribution but also reliable relative quantitative information. We developed a method based on application of matrix and dual internal standards to allow intra- and intersample normalization of lipids intensities in kidney sections of control and cisplatin-treated Wistar rats. An inkjet printer was used to deposit a custom-prepared ink with DHB as MALDI matrix, a primary lipids-based internal standard, and a spiked lanthanide as a secondary internal standard. We applied different laser energy and varied the amounts of matrix-internal standards mixture to evaluate the normalization potential of the internal standards. Successful correction of intensity artifacts caused by instrumental drifts was possible, but not those resulting from uneven matrix application. ICP-MS absolute quantification of the lanthanide in the printed layer ensured the reproducibility of the matrix and internal standards application with RSD of 10−15%. Internal standard-normalized data allowed intrasample modification of the MALDI image to make it compatible with the optical image. Normalization to internal standards corrected a 2-fold difference in lipids intensity, which allowed a meaningful comparison of tissue lipids in control and cisplatin-treated kidneys. Gadoliumbasierte Kontrastmittel werden in der Magnetresonanztomographie verwendet und gelangen nach der Exkretion durch behandelte Patienten in Oberflächengewässer, da sie von Klärwerken nur unzureichend entfernt werden. Das Verhalten der Kontrastmittel in der Umwelt und deren Wechselwirkung mit Organismen wurde in dieser Arbeit aufgrund der Toxizität des freien Gd-Ions und der damit verbundenen möglichen Gefahren, wie der Anreicherung in der menschlichen Nahrungskette, näher untersucht. Die bildgebenden elementanalytischen Methoden Laserablation mit induktiv gekoppelter Plasmamassenspektrometrie (LA-ICP-MS) und Röntgenfluoreszenzanalyse mit Synchrotronstrahlung (SRXRF) wurden genutzt, um verschiedene biologische Systeme hinsichtlich ihrer Aufnahme, Verteilung und Abgabe von Gd-basierten Kontrastmitteln zu untersuchen. Zuvor wurden beide Methoden für diese Anwendung analytisch charakterisiert und verglichen. Die Nachweisgrenzen von Gadolinium wurden mittels LA-ICP-MS (0.78 pg) und SRXRF (89 pg) unter jeweils optimierten Bedingungen durch die Kalibrierung über Rückstände eingetrockneter Elementstandardtropfen bestimmt. Anhand von Fadenalgen konnte die Aufnahme und Abgabe der Gd-basierten Kontrastmittel gezeigt werden. Die Abhängigkeit von der Konzentration des Kontrastmittels in der Expositionslösung und die Unabhängigkeit der zeitlichen Aufnahme innerhalb eines Zeitraums von 1-7 Tagen wurden an Wasserlinsen studiert. Gadolinium konnte in einem Kresseblatt quantifiziert werden. Die Verifizierung der LA-ICP-MS-Ergebnisse erfolgte mit der SRXRF und der ICP-MS nach Aufschluss. Auch die Aufnahme und Abgabe von Gd in Wasserflöhen wurde beobachtet. Die Lokalisierung des Gds wurde mittels 3D µ-Computertomographie durch den Vergleich mit einem unbelasteten Wasserfloh aufgeklärt. In allen Untersuchungen wurde Gadolinium in den exponierten Modellorganismen nachgewiesen, woraus geschlussfolgert werden kann, dass die Kontrastmittel aus der Umgebung aufgenommen werden., Gadolinium based contrast agents are used for magnetic resonance imaging. After their excretion by medicated patients they reach surface water passing waste water treat-ment plants where they are not removed sufficiently. The behavior of the contrast agents in the environment and the interaction with organisms was investigated in this work due to the toxicity of the free Gd3+ ion and the associated risks, such as accumulation in the human food chain. In this work, the two elemental analytical imaging methods laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and synchrotron radiation X-ray fluorescence analysis (SRXRF) have been used to investigate the uptake, distribution, and excretion of Gd-based contrast agents by various biological systems. Both methods were analytically characterized and compared for this application. The detection limits of gadolinium were determined under optimized conditions by LA-ICP-MS and SRXRF. With calibration by remains of dried elemental standard droplets detection limits of 0.78 pg absolute amount of gadolinium (LA-ICP-MS), respectively 89 pg (SRXRF) were reached. Based on filamentous algae as water plants the uptake and the excretion of Gd-based contrast agents were revealed. The dependence on concentration of the contrast agent in the exposition solution and the independence of temporal uptake within one to seven days were studied for duckweed. By LA-ICP-MS gadolinium was quantified in a leaf of cress plant. The verification of the results was performed by SRXRF and ICP-MS after digestion. Furthermore, the uptake and distribution of Gd-based contrast agents in higher organisms (water flea) were observed. The exact location of gadolinium was resolved by three-dimensional μ-computed tomography by the comparison of an exposed with a Gd-free water flea. In all studies, gadolinium was detected in the investigated exposed model organisms. It can be concluded that the contrast agents were taken from the environment.

In dieser Arbeit konnte ein humaner IgG1-Antikörper mittels verschiedener massenspektrometrischer Methoden erfolgreich untersucht und charakterisiert werden. Neben der Bestimmung der molekularen Massen des betrachteten Anti-Interleukin-8-Antikörpers und dessen Fragmenten konnte auch die Bildung von Oligomeren untersucht werden. Es wurden verschiedene Oligomere, bis hin zum Pentamer, massenspektrometrisch gemessen und es konnten auch Erkenntnisse gewonnen werden, dass Konzentrationseffekte maßgeblich zur Oligomerbildung bei diesem Antikörper beitragen. Die Antikörperglykosylierung, eine sehr prominente und hoch diverse Modifikation, wurde bei diesem Antikörper ebenfalls gefunden und neben dem intakten Antikörper auch in dessen Fragmenten und im Dimer gemessen. Neben dieser natürlichen Modifikation wurden auch Derivatisierungen des Antikörpers analysiert, dabei wurden unterschiedliche Metallmarkierungen analysiert. Es konnten verschiedene Markierungsgrade bestimmt werden, auch wenn diese nebeneinander als Mischung vorlagen. Der Nachweis, dass der Antikörper auch weiterhin intakt und bindungsfähig ist, konnte erbracht werden. Die Bildung von Komplexen des Antikörpers mit verschiedenen Proteinen konnte ebenfalls mittels Massenspektrometrie gezeigt werden. Weiterhin konnte gezeigt werden, dass es möglich war, eine Quantifizierungsstrategie für Antikörper zu entwickeln. Mit dieser können absolute Quantifizierungen durchgeführt werden, während diese gleichzeitig unabhängig von Art und Struktur des einzusetzenden Standards sind. Damit ist es möglich, die Probleme des Bradford-Assays zu umgehen. Des Weiteren sind nur minimale Kenntnisse über den zu analysierenden Antikörper notwendig, sodass dieses Verfahren auch hervorragend auf bisher nicht charakterisierte Antikörper angewendet werden kann., The aim of this work is the characterization of an antibody. The used human IgG1-antibody was successfully investigated with various mass spectrometric methods. In addition to the determination of the molecular masses of the analyzed Anti-Interleukin-8 antibody itself and its fragments the formation of oligomers was examined. Different oligomers up to pentamers could be mass spectromatrically measured. Resulting from this, it was possible to find that concentration effects are the main factors for the oligomer formation of this particular antibody. The glycosylation of antibodies, a prominent and high diverse modification, was also found in this antibody and was measured in the intact molecule, in its fragments and also the dimer. Next to this native modification, also a derivatization of the antibody was investigated. Different metal containing modifications were analyzed. Thereby it was possible to determine various tagging degrees, even in a mixture of these metal containing modifications. The important question, whether the tagged antibody is still active and capable for binding, can be answered positively. Furthermore the formation of antibody-protein complexes could be shown via mass spectrometry. It was also successful to develop a quantification strategy for this antibody. With this an absolute quantification was possible, which was independent from the type and the structure of the used standard. With this method it is possible to overcome the problems of the Bradford assay. In addition to this only very little information about the antibody is needed for the quantification, which allows to use this method for non characterized antibodies.
