Abstract

Artificial intelligence (AI) has gained significant attention in healthcare consultation due to its potential to improve clinical workflow and enhance medical communication. However, owing to the complex nature of medical information, large language models (LLM) trained with general world knowledge might not possess the capability to tackle medical-related tasks at an expert level. Here, we introduce EyeGPT, a specialized LLM designed specifically for ophthalmology, using three optimization strategies including role-playing, finetuning, and retrieval-augmented generation. In particular, we proposed a comprehensive evaluation framework that encompasses a diverse dataset, covering various subspecialties of ophthalmology, different users, and diverse inquiry intents. Moreover, we considered multiple evaluation metrics, including accuracy, understandability, trustworthiness, empathy, and the proportion of hallucinations. By assessing the performance of different EyeGPT variants, we identify the most effective one, which exhibits comparable levels of understandability, trustworthiness, and empathy to human ophthalmologists (all Ps>0.05). Overall, ur study provides valuable insights for future research, facilitating comprehensive comparisons and evaluations of different strategies for developing specialized LLMs in ophthalmology. 6. Integrating Bioinspired Natural Adhesion Mechanisms into Modified Polyacrylate Latex Pressure-Sensitive Adhesives.

Abstract

For polyacrylate latex pressure-sensitive adhesives (PSAs), high peel strength is of crucial significance. It is not only a key factor for ensuring the long-lasting and effective adhesive force of polyacrylate latex PSAs but also can significantly expand their application scope in many vital fields, such as packaging, electronics, and medical high-performance composite materials. High peel strength can guarantee that the products maintain stable and reliable adhesive performance under complex and variable environmental conditions. However, at present, the peel strength capacity of polyacrylate latex PSAs is conspicuously insufficient, making it difficult to fully meet the urgent market demand for high peel strength, and severely restricting their application in many cutting-edge fields. Therefore, based on previous experimental studies, and deeply inspired by the adhesion mechanism of natural marine mussels, in this study, a traditional polyacrylate latex PSA was ingeniously graft-modified with 3,4-dihydroxybenzaldehyde (DHBA) through the method of monomer-starved seeded semi-continuous emulsion polymerization, successfully synthesizing novel high-peel-strength polyacrylate latex pressure-sensitive adhesives (HPSAs) with outstanding strong adhesion properties, and the influence of DHBA content on the properties of the HPSAs was comprehensively studied. The research results indicated that the properties of the modified HPSAs were comprehensively enhanced. Regarding the water resistance of the adhesive film, the minimum water absorption rate was 4.33%. In terms of the heat resistance of the adhesive tape, it could withstand heat at 90 °C for 1 h without leaving residue upon tape peeling. Notably, the adhesive properties were significantly improved, and when the DHBA content reached 4.0%, the loop tack and 180° peel strength of HPSA4 significantly increased to 5.75 N and 825.4 gf/25 mm, respectively, which were 2.5 times and 2 times those of the unmodified PSA, respectively. Such superior adhesive performance of HPSAs, on the one hand, should be attributed to the introduction of the bonding functional monomer DHBA with a rich polyphenol structure; on the other hand, the acetal structure formed by the grafting reaction of DHBA with the PSA effectively enhanced the spatial network and crosslink density of the HPSAs. In summary, in this study, the natural biological adhesion phenomenon was ingeniously utilized to increase the peel strength of pressure-sensitive adhesives, providing a highly forward-looking and feasible direct strategy for the development of environmentally friendly polyacrylate latex pressure-sensitive adhesives. Abstract

A polymer micelle-based drug delivery system has faced many challenges due to the lack of stability especially after being diluted in blood, resulting in a premature release. Herein, we developed camptothecin (CPT)-conjugated prodrug (CPTP) micelles in which CPT was grafted to the poly(ethylene glycol)-poly(glutamic acid) block copolymer via a disulfide bond linker for a redox-triggered drug release. Then, the cisplatin (CDDP)-crosslinked CPT-prodrug micelles (CPTP/CDDP) with a hybrid complex as a stable structure were successfully established via the CDDP (Pt)-carboxyl (COOH) chelate interaction. The resulting dual CPTP/CDDP had an average hydrodynamic radius of about 50 nm with a narrow distribution, which was conducive to the promotion of solid tumor accumulation. Importantly, CPT chemical bonding to the polymer backbone obviously stabilizes the CPT-prodrug micelles and prolongs their circulation time. Moreover, both CPT and CDDP are clinically used antitumor drugs; CDDP not only behaves as an ancillary anticarcinogen but also serves as a crosslinker to restrain the untimely burst release of CPT and to achieve synergistic antitumor efficacy. In addition, the CPTP/CDDP also exhibited a sustained reduction responsive release of CPT accompanied by the dissociation of the CDDP-COOH complex. This design ingeniously solved the contradiction between the stability and release of polymer micelle-based nanomedicines. Both in vitro and in vivo tests demonstrated an amazing antineoplastic efficacy compared with free drugs (CPT or CDDP) and just their physical mixing, indicating great promise for cancer treatment.

Temozolomide (TMZ) is the only clinical first-line chemotherapy drug for malignant glioma up to present. Due to poor aqueous solubility and toxic effects, TMZ is still inefficient and limited for clinical glioma treatment.Methods: UiO-66-NH2 nanoparticle is a zirconium-based framework, constructed by Zr and 2-amino-1,4-benzenedicarboxylic acid (BDC-NH2) with octahedral microporous structure, which can be decomposed by the body into an ionic form to discharge. We prepared the nanoscale metal-organic framework (MOF) of UiO-66-NH2 to load TMZ for therapy of malignant glioma, TMZ is released from UiO-66-NH2 through a porous structure. The ultrasound accelerates its porous percolation and promotes the rapid dissolution of TMZ through low-frequency oscillations and cavitation effect. The biological safety and antitumor efficacy were evaluated both in vitro and in vivo.Results: The prepared TMZ@MOF exhibited excellent biocompatibility and biosafety due to minimal drug leakage without ultrasound intervention. We further used the flank model of glioblastoma to verify the in vivo therapeutic effect. Abstract

Poly(amino acid)-type macromolecules (PAAs), poly(ethylene glycol)-b-poly(aspartic acid) (PEG-PAAs), and poly(ethylene glycol)-b-poly(glutamic acid) (PEG-PGAs) were synthesized, and their states in aqueous solutions were studied. PEG-PAAs and PEG-PGAs were first used as novel polycarboxylate superplasticizers, and their dispersion and adsorption behaviors were investigated in cement slurries in the absence and presence of montmorillonite (MMT) clay. The results demonstrated that both PEG-PAAs and PEG-PGAs not only showed excellent water reducing and fluidity retention capabilities but also exhibited remarkable MMT clay tolerance. In contrast to the stretched molecular conformation of traditional polycarboxylate superplasticizers (PCEs), the hydrophobic main chain and the side methylene and ethylene linkers connected to the carboxylic groups of PAA and PGA segments resulted in self-assembly of the PEG-PAAs and PEG-PGAs in aqueous solutions, and the self-assembled nanosized aggregates exhibited relatively uniform spherical morphologies with average sizes of 30–100 nm. Carboxylic groups located on the periphery of the nanoaggregates acted as anchors and adsorbed on the surfaces of the cement particles. The nanoaggregates and the stretched hydrophilic PEG side chains, which jointly produced and enhanced the steric hindrance to prevent cement particle aggregation and hydration, resulted in notable dispersion capabilities. Total organic carbon (TOC), X-ray diffraction (XRD), and thermogravimetry–differential scanning calorimetry (TG-DSC) analyses revealed that the adsorption between the MMT clay and PAA nanoaggregates was surface adsorption (rather than interlayer adsorption), and this significant finding could be ascribed to the enlarged steric hindrance of the PAA nanoaggregates, which prevented the interlamination adsorption consumption of MMT clay to PAAs and revealed the outstanding clay insensitivity. This study provides a new perspective and strategy to design novel poly(amino acid)-type superplasticizers (PAAs) with excellent dispersion performances and improved clay tolerance.

Abstract

Polyethylene terephthalate (PET) and CO2, two chemical wastes that urgently need to be transformed in the environment, are converted simultaneously in a one‐pot catalytic process through the synergistic coupling of three reactions: CO2 hydrogenation, PET methanolysis and dimethyl terephthalate (DMT) hydrogenation. More interestingly, the chemical equilibria of both reactions were shifted forward due to a revealed dual‐promotion effect, leading to significantly enhanced PET depolymerization. The overall methanol yield from CO2 hydrogenation exceeded the original thermodynamic equilibrium limit since the methanol was in situ consumed in the PET methanolysis. The degradation of PET by a stoichiometric ratio of methanol was significantly enhanced because the primary product, DMT was hydrogenated to dimethyl cyclohexanedicarboxylate (DMCD) or p‐xylene (PX). This synergistic catalytic process provides an effective way to simultaneously recycle two wastes, polyesters and CO2, for producing high‐value chemicals. [ABSTRACT FROM AUTHOR]
