The South American Fur Seal Arctocephalus australis is one of the most widely distributed South American otariid species. In Brazil, during austral winter months specimens of A. australis are regularly found along the Rio Grande do Sul and Santa Catarina coasts. Occasionally, vagrants have been recorded along the southeastern coast but rarely moves north of Rio de Janeiro, at 23°S. On 01 May 2015, fishermen noticed in the surf zone an unidentified large animal in Ilhéus, Bahia, northeastern Brazil. Severely wounded, it was still alive when first spotted. Numerous shark bites were observed in the rostrum and flippers, the peduncle was severely lacerated and the intestines were protruding. The specimen was identified as an adult of South American Fur Seal based on the combination of the characteristic coloration of the pelage, head shape, body size, the relatively large eye size, and mainly due to the pronounced three cusped teeth in post canines. Fishermen decided to butcher the carcass and share the meat among their companions. The present record, however, is of special concern as human consumption South American Fur Seal seems to be unusual in Brazil. Pieces of meat were shared among fishermen. In addition, small pieces of blubber were used as bait in the local shark long-line fishery. Consumption of an injured and presumably weak marine mammal carcass points to an increasing risk of contact between potentially harmful viruses, bacteria and fungi and humans. This note represents an alert to public health specialists and wildlife authorities in Brazil. It calls the urgency of an outreach campaign advertising to the potential risks of ingesting bushmeat either from land or aquatic sources.

Objetivou-se com este trabalho realizar um levantamento do uso popular de plantas medicinais por mulheres da Comunidade Quilombola de Furadinho em Vitória da Conquista-Bahia. O trabalho foi realizado na comunidade Quilombola de Furadinho, visando identificar as principais plantas utilizadas com fins medicinais, as formas de obtenção, sua indicação medicinal, a parte da planta utilizada, as formas de preparo, bem como as formas de obtenção e transmissão deste conhecimento. Para a realização dessa pesquisa foram entrevistadas 14mulheres com idades variadas, moradoras dessa comunidade, através de questionários semi estruturados, e análise descritiva. Constatou-se um vasto conhecimento relacionado ao uso de plantas medicinais entre as entrevistadas, o qual foi adquirido de forma oral através de pais e avós. As plantas medicinais utilizadas são em sua maioria cultivadas em quintais, sendo as folhas a parte mais utilizada nas preparações medicamentosas, e a principal forma de preparo são os chás. Foram identificadas 30 plantas com fins medicinais e terapêuticos, sendo a Erva cidreira, o Capim santo, o Hortelã grosso, a Arruda e a Erva doce as mais utilizadas. Popularized use of medicinal plants by women of Quilombo of Furadinho community in Vitoria da Conquista, Bahia, Brazil Abstract: This work aimed to carry out a survey of the popular use of medicinal plants by women of Furadinho Quilombo Community in Vitória da Conquista, Bahia. The work was conducted in the Furadinho Quilombo Community, aiming at identifying the main plants used for medical purposes, the ways of obtaining, its indication medicinal, the part of the plant used, the forms of preparation, as well as ways of obtaining and transmission of this knowledge. For the realization of this research were interviewed 14 women with ages varied, dwell in this community, through semi-structured questionnaires, and descriptive analysis. It was found a vast knowledge related to the use of medicinal plants among the interviewees, which was purchased in oral form by parents and grandparents. Medicinal plants used are mostly grown in quintals, being the sheets the most used in medicinal preparations, and the main form of preparation are the teas. Were identified 30 plants with medicinal and therapeutic purposes, being the lemon grass, the Grass holy, the Mint grosso, Arruda and Erva Doce the most used.

Abstract Background Spinal ventral root avulsion results in massive motoneuron degeneration with poor prognosis and high costs. In this study, we compared different isoforms of basic fibroblast growth factor 2 (FGF2), overexpressed in stably transfected Human embryonic stem cells (hESCs), following motor root avulsion and repair with a heterologous fibrin biopolymer (HFB). Methods In the present work, hESCs bioengineered to overexpress 18, 23, and 31 kD isoforms of FGF2, were used in combination with reimplantation of the avulsed roots using HFB. Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease that progressively affects motoneurons, causing muscle atrophy and evolving to death. Astrocytes inhibit the expression of MHC-I by neurons, contributing to a degenerative outcome. The present study verified the influence of interferon β (IFN β) treatment, a proinflammatory cytokine that upregulates MHC-I expression, in SOD1G93A transgenic mice. For that, 17 days old presymptomatic female mice were subjected to subcutaneous application of IFN β (250, 1,000, and 10,000 IU) every other day for 20 days. Rotarod motor test, clinical score, and body weight assessment were conducted every third day throughout the treatment period. No significant intergroup variations were observed in such parameters during the pre-symptomatic phase. All mice were then euthanized, and the spinal cords collected for comparative analysis of motoneuron survival, reactive gliosis, synapse coverage, microglia morphology classification, cytokine analysis by flow cytometry, and RT–qPCR quantification of gene transcripts. Additionally, mice underwent Rotarod motor assessment, weight monitoring, and neurological scoring. The results show that IFN β treatment led to an increase in the expression of MHC-I, which, even at the lowest dose (250 IU), resulted in a significant increase in neuronal survival in the ALS presymptomatic period which lasted until the onset of the disease. The treatment also influenced synaptic preservation by decreasing excitatory inputs and upregulating the expression of AMPA receptors by astrocytes. Microglial reactivity quantified by the integrated density of pixels did not decrease with treatment but showed a less activated morphology, coupled with polarization to an M1 profile. Disease progression upregulated gene transcripts for pro- and anti-inflammatory cytokines, and IFN β treatment significantly decreased mRNA expression for IL4. Overall, the present results demonstrate that a low dosage of IFN β shows therapeutic potential by increasing MHC-I expression, resulting in neuroprotection and immunomodulation.

Abstract Understanding morphological and physiological changes under different light conditions in native fruit species in juveniles’ stage is important, as it indicate the appropriate environment to achieve vigorous saplings. We aimed to verify growth and morphophysiological changes under shade gradient in feijoa (Acca sellowiana (O. Berg) Burret) to achieve good quality saplings adequate to improve cultivation in orchards. The saplings were grown for twenty-one-month under four shading treatments (0%, 30%, 50%, and 80%). Growth, photosynthetic pigments, gas exchanges, chlorophyll fluorescence, and leaf anatomy parameters were evaluated. Saplings under full sun and 30% shade had higher height and diameter growth and dry mass accumulation due to higher photosynthesis rate. As main acclimatization mechanisms in feijoa saplings under 80% shade were developed larger leaf area, reduced leaf blade thickness, and enhanced quantum yield of photosystem II. Even so, the net CO2 assimilation and the electron transport rate was lower and, consequently, there was a restriction on the growth and dry mass in saplings under deep shade. Therefore, to obtain higher quality feijoa saplings, we recommend that it be carried out in full sun or up to 30% shade, to maximize the sapling vigor in nurseries and, later, this light environment can also be used in orchards for favor growth and fruit production.

Abstract Background Treatment of spinal cord injury is dependent on neuronal survival, appropriate synaptic circuit preservation, and inflammatory environment management. In this sense, mesenchymal stem cell (MSC) therapy is a promising tool that can reduce glial reaction and provide trophic factors to lesioned neurons. Methods Lewis adult female rats were submitted to a unilateral ventral funiculus cut at the spinal levels L4, L5, and L6. The animals were divided into the following groups: IA (intramedullary axotomy), IA + DMEM (Dulbecco’s modified Eagle’s medium), IA + FS (fibrin sealant), IA + MSC (106 cells), and IA + FS + MSC (106 cells). Seven days after injury, qPCR (n = 5) was performed to assess gene expression of VEGF, BDNF, iNOS2, arginase-1, TNF-α, IL-1β, IL-6, IL-10, IL-4, IL-13, and TGF-β. The cellular infiltrate at the lesion site was analyzed by hematoxylin-eosin (HE) staining and immunohistochemistry (IH) for Iba1 (microglia and macrophage marker) and arginase-1. Fourteen days after injury, spinal alpha motor neurons (MNs), evidenced by Nissl staining (n = 5), were counted. For the analysis of astrogliosis in spinal lamina IX and synaptic detachment around lesioned motor neurons (GAP-43-positive cells), anti-GFAP and anti-synaptophysin immunohistochemistry (n = 5) was performed, respectively. Twenty-eight days after IA, the gait of the animals was evaluated by the walking track test (CatWalk; n = 7). Results The site of injury displayed strong monocyte infiltration, containing arginase-1-expressing macrophages. The FS-treated group showed upregulation of iNOS2, arginase-1, proinflammatory cytokine (TNF-α and IL-1β), and antiinflammatory cytokine (IL-10, IL-4, and IL-13) expression. Thus, FS enhanced early macrophage recruitment and proinflammatory cytokine expression, which accelerated inflammation. Rats treated with MSCs displayed high BDNF-positive immunolabeling, suggesting local delivery of this neurotrophin to lesioned motoneurons. This BDNF expression may have contributed to the increased neuronal survival and synapse preservation and decreased astrogliosis observed 14 days after injury. At 28 days after lesion, gait recovery was significantly improved in MSC-treated animals compared to that in the other groups. Conclusions Overall, the present data demonstrate that MSC therapy is neuroprotective and, when associated with a FS, shifts the immune response to a proinflammatory profile.

ABSTRACT The control of the temperature of the storage air have the ability to improve a better preservation of some types of agricultural products. However, the most effective temperature for storage as well as the duration of storage varies between products. Therefore, special attention should be concentrated on the storage temperature and its effects on the integrity and longevity of the produce. The objective of the present study was to evaluate the effects of storage temperature and storage period on the quality of castor beans. Castor seeds with a water content of approximately 6.1% (w.b.) were stored for 180 days at temperatures of 15, 25 and 35 °C. The quality of the seeds was evaluated every 45 days throughout the study period by measuring dry matter loss, electrical conductivity, color, and the free fatty acid and peroxide content of the crude oil extracted. Our results indicated that: a) higher storage temperature negatively affect the quality of the seeds and the extracted oil; b) the negative effect of temperature increase with longer storage period; c) the storage temperature of 15 °C least affect the quality of castor bean and the extracted oil.

We recently proposed a new surgical approach to treat ventral root avulsion, resulting in motoneuron protection. The present work combined such a surgical approach with bone marrow mononuclear cells (MC) therapy. Therefore, MC were added to the site of reimplantation. Female Lewis rats (seven weeks old) were subjected to unilateral ventral root avulsion (VRA) at L4, L5 and L6 levels and divided into the following groups (n = 5 for each group): Avulsion, sealant reimplanted roots and sealant reimplanted roots plus MC. After four weeks and 12 weeks post-surgery, the lumbar intumescences were processed by transmission electron microscopy, to analyze synaptic inputs to the repaired α motoneurons. Also, the ipsi and contralateral sciatic nerves were processed for axon counting and morphometry. The ultrastructural results indicated a significant preservation of inhibitory pre-synaptic boutons in the groups repaired with sealant alone and associated with MC therapy. Moreover, the average number of axons was higher in treated groups when compared to avulsion only. Complementary to the fiber counting, the morphometric analysis of axonal diameter and “g” ratio demonstrated that root reimplantation improved the motor component recovery. In conclusion, the data herein demonstrate that root reimplantation at the lesion site may be considered a therapeutic approach, following proximal lesions in the interface of central nervous system (CNS) and peripheral nervous system (PNS), and that MC therapy does not further improve the regenerative recovery, up to 12 weeks post lesion.
