The dinoflagellate genus Scrippsiella is a common member of phytoplankton and their cysts are also frequently reported in coastal sediments worldwide. However, the diversity of Scrippsiella in Korean waters has not been fully investigated. Here, several isolates of Scrippsiella precaria collected from Korean waters and germinated from resting cysts were examined using light and scanning electron microscopy. The resting cysts were characterized by pointed calcareous spines and one or two red accumulation bodies, and the archeopyle was mesoepicystal, representing the loss of 2–4′ and 1–3a paraplates. Rounded resting cysts were found in culture, and an increase in spine length was observed until 8 days of development. Korean isolates of S. precaria had the plate formula of Po, X, 4′, 3a, 7″, 6C, 4S, 5‴, 2⁗. There were differences in the cell size and location of the red body between Korean isolates and previously described cells of S. precaria. In addition, the Korean isolates of S. precaria had two types of the 5″ plate that either contacted the 2a plate or not. Molecular phylogeny based on internal transcribed spacer (ITS) and large subunit (LSU) rDNA sequences revealed that the Korean isolates were nested within the subclade of PRE (S. precaria and related species) in the clade of Scrippsiella sensu lato, and that the PRE subclade had two ribotypes: ribotype 1 consisting of the isolates from Korea, China, and Australia, and ribotype 2 consisting of the isolates from Italy and Greece. Lineages between isolates of ribotype 1 were likely to be related to the dispersal by ocean currents and ballast waters from international shipping, and the two types of spine shapes and locations of the 5″ plates may be a distinct feature for ribotype 1.

The present study was performed to investigate the anti-inflammatory activity of Tetragonia tetragonoides hydro- sols (TTH) and its underlying mechanism in lipopolysaccharide (LPS)-induced RAW 264.7 cells. Gas chromatog- raphy (GC) coupled with mass spectrometry and retention index calculations showed that TTH were mainly com- posed of tetratetracontane (29.5 %), nonacosane (27.6 %), and oleamide (17.1 %). TTH significantly decreased the production of nitric oxide (NO), prostaglandin E2 (PGE2), interleukin (IL)-6, and IL-1β in LPS-stimulated RAW 264.7 cells. Consistent with these observations, TTH treatment decreased the protein expression levels of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2). The molecular mechanism of its anti-inflamma- tory activity was found to be associated with inhibition of nuclear factor-kappa B (NF-κB) phosphorylation and nuclear translocation of NF-κB 65. Furthermore, TTH markedly suppressed the LPS-induced phosphorylation of mitogen-activated protein kinases (MAPKs). The anti-inflammatory properties of the supercritical fluid extract of Ishige okamurae (SFEIO) on lipopolysaccharide (LPS)-stimulated murine RAW 264.7 macrophages. The lipid profile of the SFEIO, reviled the presence of palmitic acid (220.2 mg/g), linoleic acid (168.0 mg/g), and oleic acid (123.0 mg/g). SFEIO was found to exert it’s anti-inflammatory effects through inhibiting nitric oxide (NO), prostaglandin E2 (PGE2), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 production in LPS-stimulated RAW 264.7 cells, without inducing cytotoxicity. SFEIO did not effect on the LPS-induced p38 kinase phosphorylation, whereas it attenuated the extracellular-related signaling kinase (ERK) and c-Jun N-terminal kinase (JNK) phosphorylation. Furthermore, SFEIO inhibited the LPS-induced IκB-α degradation and p50 NF-κB activation. In this study, the antioxidant properties of 80 % ethanol extracts of 16 species of plants from Jeju Island in Korea were evaluated using various antioxidant assays, including the DPPH (1,1-Diphenyl-2-pricrylhydrazyl) radical scavenging, superoxide scavenging, xanthine oxidase inhibition and hydrogen peroxide scavenging activities. Among the 16 plant extracts tested, CN-13 showed strong antioxidant properties in the DPPH radical scavenging and hydrogen peroxide scavenging tests. The CN-13 ethanol extract was thus selected to be used for further experiments, and was separated into various fractions using four different organic solvents (n-hexane, methylene chloride, ethyl acetate and butanol). The ethyl acetate fraction of CN-13 extract evidenced strong DPPH radical scavenging properties as compared to the other fractions. The ethyl acetate fraction also strongly inhibited DNA-damage induced by hydrogen peroxide-oxidative damage in a mouse lymphoma (L5178Y-R) cell line. The anticancer effects of trans-1,3-diphenyl-2,3-epoxypropan-1-one (DPEP), a chalcone derivative, were investigated in human leukemia HL-60 cells. Treatment of HL-60 cells with various concentration of DPEP resulted in a sequence of events characteristic of apoptosis, including loss of cell viability, morphological changes, and increased sub-G1 DNA content. We demonstrated that DPEP elevates reactive oxygen species (ROS) levels in HL-60 cells, and that the ROS scavenger N-acetylcysteine (NAC) could block DPEP-induced ROS generation and apoptosis. Western blot analysis revealed that DPEP inhibits Bcl-xL expression, leading to caspase-3 activation and poly-ADP-ribose polymerase (PARP) cleavage, thereby inducing apoptosis. However, NAC pre-treatment significantly inhibited the activation of caspase-3 and PARP cleavage and reduced Bcl-xL levels. These findings provide the first evidence that DPEP may inhibit the growth of HL-60 cells and induce apoptosis through a ROS-mediated Bcl-xL pathway., EXCLI Journal; 14:Doc900; ISSN 1611-2156
