Background: As vitiligo progresses, autophagy becomes more and more important. Objectives: To validate potential genes associated with autophagy in vitiligo through bioinformatics analysis and experimental testing. Materials and Methods: Dataset GSE75819 of mRNA expression profiles was obtained from GEO. After data normalisation, gene set enrichment analyse enrichment analysis and abundance analysis of infiltrating immune cells were performed. A list of autophagy-related differentially expressed genes (ARDEGs) associated with vitiligo was generated using R software. Protein–protein interaction (PPI) analysis, correlation analysis, and enrichment analysis on gene ontology (GO) and Kyoto encyclopaedia of genes and genome (KEGG) pathways were conducted on the ARDEG data. The microRNAs associated with hub genes were predicted using the TargetScan database. Finally, RNA expression of 10 hub genes and Western blotting (WB) of autophagy pathway factors were further verified. Results: From the lesions of 15 vitiligo patients, 44 ARDEGs were identified. PPI analysis demonstrated that these ARDEGs interacted with each other. GO and KEGG analyses of ARDEGs revealed that several enriched terms were associated with macroautophagy (biological process), vacuolar membranes (cellular components), cysteine-type peptidase activity (molecular function), and autophagy in animals, neurodegeneration-multiple disease pathways, and apoptosis. In vitiligo lesions, qRT-PCR and sequencing validation analyses showed expression levels of CCL2, RB1CC1, TP53, and ATG9A that were consistent with bioinformatic analysis of the microarray. WB results also showed that autophagy-related proteins were differentially expressed. Conclusions: Forty-four potential ARDEGs were identified in vitiligo by bioinformatic analysis. Vitiligo may be affected by autophagy regulation through CCL2, RB1CC1, TP53, and ATG9A.

Abstract Apple moldy core is a fungus-infested disease that is extremely insidious, usually occurring inside the fruit, making it very difficult to distinguish from the exterior with the naked eye. Using VIS/NIR transmission spectroscopy, this study successfully detected moldy core apples. By combining four wavelength selection algorithms (CARS, CARS-SPA, MC-UVE, and MC-UVE-SPA) with four classifiers (SVM, ELM, KNN, and LDA-KNN), discrimination models were established for two-class and three-class classifications. MC-UVE-SPA-LDA-KNN achieved an AUC of 0.99 and an accuracy of 98.82% for two-class classification, while MC-UVE-SPA achieved an AUC of 0.99 and an accuracy of 97.64% for three-class classification. This confirms MC-UVE-SPA as an effective tool for selecting wavelengths specific to moldy core apples, facilitating precise identification and differentiation of apple states. This study advances dynamic online detection of early-stage moldy core conditions in apples, reducing post-harvest disease occurrence and preserving fruit quality effectively. Graphical Abstract

Zinc ash is a serious defect of hot-dip galvanized process and has great impact on products with high surface level requirements. During galvanizing process, the zinc ash formed by the oxidation of zinc vapor in a furnace snout was easy to adhere to the surface of strip steel, influencing the surface quality of the coating. At present, the effective control measures were to control the dew point by humidifying the furnace snout. If the dew point of the furnace snout was too high or too low, it would produce defects. In this paper, the control methods of selecting the appropriate dew point by controlling the process parameters such as pressure and flow were expounded according to the existing working conditions.

Abstract Background Ureteral fibro-epithelial polyp (UFP) is a rare benign ureteral tumor, and surgical removal of the polyps is still the preferred solution. Although many cases have reported polyps extending to the bladder, our case was the first to report a huge UFP that underwent endoscopic laser resection to highlight the urethra and cause severe end hematuria permanently. Case presentation In 2019, a 37-year-old woman came to the hospital because of hematuria and a dark red extraurethral mass. CTU inspection showed: filling defect between the right ureter and the bladder at the entrance of the bladder. After ureteroscopy, it was found that the ureteral mass came out of the urethral orifice. Then, under the direct view of the ureteroscope, a Ho:YAG laser was used to remove the tumor by cutting off along the its base, and the patient was discharged 3 days after the operation. Conclusion Urethral polyps from the ureter should be considered in the differential diagnosis of urethral neoplasms. Ho:YAG laser resection under ureteroscopy is an effective option for treating UFP, but be careful of ureteral stricture after surgery.

Brain decoding is to predict the external stimulus information from the collected brain response activities, and visual information is one of the most important sources of external stimulus information. Decoding functional magnetic resonance imaging (fMRI) based on visual stimulation is helpful in understanding the working mechanism of the brain visual function regions. Traditional brain decoding algorithms cannot accurately extract stimuli features from fMRI. To address these shortcomings, this paper proposed a brain decoding algorithm based on a graph convolution network (GCN). Firstly, 11 regions of interest (ROI) were selected according to the human brain visual function regions, which can avoid the noise interference of the non-visual regions of the human brain; then, a deep three-dimensional convolution neural network was specially designed to extract the features of these 11 regions; next, the GCN was used to extract the functional correlation features between the different human brain visual regions. Furthermore, to avoid the problem of gradient disappearance when there were too many layers of graph convolutional neural network, the residual connections were adopted in our algorithm, which helped to integrate different levels of features in order to improve the accuracy of the proposed GCN. The proposed algorithm was tested on the public dataset, and the recognition accuracy reached 98.67%. Compared with the other state-of-the-art algorithms, the proposed algorithm performed the best.

One normal outburst and three mini-outbursts have been detected by Rossi X-ray Timing Explorer satellite after 2000 in the well-known black hole X-ray binary XTE J1550-564. In this work, we explore the hysteresis effect of the four outbursts, which is a phenomenon that a similar spectral state transition occurs at different luminosity in an outburst of black hole X-ray binary. A q-like track was found in the hardness-intensity diagram of the normal outburst in 2000 but not in the three mini-outbursts that only occur in the Low/Hard state. The results demonstrate that the hysteresis effect is not apparent in the three mini-outbursts and the X-ray spectra are harder than that of the normal outburst at the same photon count rate. Furthermore, the results of the correlation analysis show that the Γ − F2−10keV correlation of mini-outburst maintain negative in the Low/Hard state with the harder spectra than that of the normal outburst at the same X-ray flux. The X-ray spectral evolution can be well-explained by the state-transition model from the Shakura–Sunyaev disk to the advection-dominated accretion flow, which implies that the three mini-outbursts of XTE J1550-564 might originate from a smaller discrete accretion event.

The influence of microcrack on the ignition of polymer-bonded explosives is missing a comprehensive description of the cracking microstructure characteristics. In this article, the mechanical–thermal–chemical response of octahydro-1,3,5,7-tetranitro-tetrazocine (HMX)-based polymer-bonded explosives (PBXs) under low-velocity impact is investigated by a dynamic damage viscoelastic model with tension–compression asymmetrical microcrack evolution and a friction-based hotspot formation mechanism. The microcrack evolution fully considers cracking nucleation and growth, where the nucleation depends on tension or compression condition and the growth follows the law of the energy-release rate. The simulation concerns the mechanical and ignition response of HMX-based PBX in the Steven test. The cracking formation from tension or compression is captured, and the damage morphology is well simulated. The simulation on the ignition response reveals the effect of the cracking microstructure characteristics, including the initial microcrack density number and the tension–compression asymmetrical nucleation rate. In addition, heterogeneous microcrack density number is constructed by unimodal normal distribution and bimodal normal distribution, corresponding to pristine and damaged scenarios, respectively. The uncertainties in the microcrack density number are propagated and quantities in the mechanical–thermal–chemical-coupled model and further, the effect on the ignition, are obtained. A fluorescence assay for the detection of 4-nitrophenol (4-NP), α-glucosidase (α-Glu) activity and α-Glu inhibitors (AGIs) is developed based on the inner filter effect (IFE), a flexible and simple signal transfer strategy. In this assay, silicon nanoparticles (Si NPs) synthesized under mild and easily accessible conditions are employed as fluorescent indicators. 4-NP efficaciously quenches the fluorescence of Si NPs through the IFE at a very rapid rate, thus achieving 4-NP detection in a mix-to-read manner, which is suitable for on-site detection. The quenching mechanism has been comprehensively studied and confirmed. More significantly, based on the fact that 4-NP can be generated through α-Glu-catalyzed hydrolysis of 4-nitrophenyl-α- D -glucopyranoside (NPG), the fluorescence detection of α-Glu activity is legitimately achieved by employing NPG as the substrate. The linear ranges for 4-NP and α-Glu activity detection are 0.5–60 μM and 0.5–60 mU mL−1 with low detection limits of 0.074 μM and 0.094 mU mL−1, respectively. This method not only can preciously assay targets in real samples, but is also capable of screening AGIs as drugs as well as assessing their inhibition efficiency. [ABSTRACT FROM AUTHOR]
