Alterations in photosynthetic performance of lutein-deficient mutant lut2 and wild type (wt) of Arabidopsis thaliana were followed after treatment with low temperature and high light for 6 d. The obtained results indicated lower electrolyte leakage, lower excitation pressure, and higher actual photochemical efficiency of PSII in lut2 plants exposed to combined stress compared to wt plants. This implies that lut2 is less susceptible to the applied stress conditions. The observed lower values of quantum efficiency of nonphotochemical quenching and energy-dependent component of nonphotochemical quenching in lut2 suggest that nonphotochemical quenching mechanism(s) localized within LHCII could not be involved in the acquisition of higher stress tolerance of lut2 and alternatives to nonphotochemical quenching mechanisms are involved for dissipation of excess absorbed light. We suggest that the observed enhanced capacity for cyclic electron flow and the higher oxidation state of P700 (P700+), which suggests PSI-dependent energy quenching in lut2 plants may serve as efficient photoprotective mechanisms, thus explaining the lower susceptibility of lut2 to the combined stress treatments.

Haberlea rhodopensis Friv. is unique with its ability to survive desiccation to an air-dry state during periods of extreme drought and freezing temperatures. To understand its survival strategies, it is important to examine the protective mechanisms not only during desiccation but also during rehydration. We investigated the involvement of alternative cyclic electron pathways during the recovery of photosynthetic functions after freezing-induced desiccation. Using electron transport inhibitors, the role of PGR5-dependent and NDH-dependent PSI-cyclic electron flows and plastid terminal oxidase were assessed during rehydration of desiccated leaves. Recovery of PSII and PSI, the capacity of PSI-driven cyclic electron flow, the redox state of plastoquinone pool, and the intersystem electron pool were analyzed. Data showed that the effect of alternative flows is more pronounced in the first hours of rehydration. In addition, the NDH-dependent cyclic pathway played a more determining role in the recovery of PSI than in the recovery of PSII.

The recovery of photosynthetic activity during rehydration of Haberlea rhodopensis from drought- and freezing-induced desiccation were investigated. The water uptake during the initial 15 h was slow thus preventing cellular damages. The results showed faster recovery of quantum efficiency of PSII in plants rehydrated after freezing stress (RAF) compared to plants rehydrated after drought stress (RAD) and the most significant differences between them were evident after 9-15 h of rehydration. Following rehydration, PSI activity recovered faster compared to PSII and in contrast to PSII, its activity was higher in RAD compared to RAF plants. During the first hours of rehydration, prominent alterations in energy transfer between photosynthetic complexes occurred as revealed by 77 K fluorescence of isolated thylakoids. High proportion of thermal energy dissipation in dry plants and during the first hours of rehydration protects them from photooxidation; the role of PSII reaction center quenching during the recovery was suggested.

Purpose: The effect of the Earth’s ionosphere and interplanetary plasma on the radio astronomy observations, in particular, interferometric ones, at decameter waves has been investigated. The amplitude and phase fluctuations of an incident electromagnetic wave field are observed upon diffraction by a plasma layer with inhomogeneities of electron density. In interferometric observations, the phase fluctuations of an output signal cause a decrease in coherence time and an increase in the interferometer response spectrum width that limits the time of coherent averaging and reduces the instrument sensitivity. The purpose of the study is to assess the degree of ionospheric and interplanetary scintillations influence on the sensitivity of interferometric observations and to determine the possibility of sensitivity increasing that is needed in observations of angular structure of weak compact radio sources. Design/methodology/approach: To determine the contribution of ionospheric scintillations to the broadening of an interferometer output signal spectrum, the radio source with such an angular size, which prevents its scintillations on the solar wind inhomogeneities, but makes it possible to observe the source using the URAN-2 interferometer with the baseline of 153 km, was used. Processing a large amount of observational data made it possible to determine the signal spectrum width at the output of thе interferometer under the condition of amplitude and phase scintillations on ionospheric inhomogeneities. Then, the interferometer was used to observe a compact radio source, whose signal fluctuations are affected by both the ionosphere and the interplanetary medium. The preliminary obtained spectral widths of the ionosphere scintillations made it possible to single out the effect of just interplanetary inhomogeneities on the interferometer response. Findings: The constraints on the time of coherent averaging imposed by the influence of interplanetary plasma have been determined. Some suggestions as for changing the method of observation with the URAN interferometers at decameter wavelengths are made for more correct and efficient study of weak compact objects. The influence of large-scale features of the solar wind structure, related to the form of lines of force of the interplanetary magnetic field and to the form of spatial spectrum of inhomogeneities on the sensitivity of low-frequency interferometric observations was found. An increase in turbulence of the interplanetary medium beyond the Earth’s orbit lasting up to several days, conjugate with manifestations of the sporadic solar activity, was also detected. Conclusions: The study allows to determine the interplanetary medium influence on the limitation of sensitivity in the interferometric observations and find optimal elongations for studying the angular structure of weak radio sources. An account of solar activity events can be used for a short-term forecast when determining the strategy of highly sensitive observations. The proposed method for detecting the solar wind turbulence can be used in studying the process of interaction of the solar events with the interplanetary medium.

Some physiological aspects of interactions between wheat and Zymoseptoria tritici were studied by measuring changes in some stress markers, photosynthetic activity, and the extent of thermal energy dissipation. Four winter wheat genotypes varying in their level of resistance, determined on the basis of necrotic leaf area and presence of pycnidia, were inoculated at a seedling stage and measurements were performed at 0, 2, 17, and 23 d after the inoculation. Physiological activity of wheat genotypes showed that keeping the membrane integrity and the extent of oxidative stress is of significant importance for pathogen resistance. It could be suggested that the changes in electrolyte leakage, malondialdehyde content, and values of quantum yield of PSII electron transport and nonphotochemical fluorescence quenching can be used to screen the degree of tolerance to Septoria tritici blotch in different wheat genotypes.

Porphyridium aerugineum is a unicellular freshwater red microalga that synthesizes and secretes into the culture medium an extracellular polysaccharide (EPS). In this study, algal growth and polysaccharide production, as well as the antioxidant capacity and antitumor effect of Porphyridium aerugineum EPS (PaEPS), were investigated. Cultivation of the microalgae was carried out in a photobioreactor under controlled conditions. Algal growth and the amount of EPS were monitored daily. The accumulated polysaccharide was extracted and lyophilized. At the end of cultivation, the concentration of microalgal biomass and PaEPS reached 3.3 and 1.2 g L−1, respectively. To examine the antioxidant capacity of PaEPS, FRAP and ABTS assays were performed. The cytotoxic activity of PaEPS was evaluated on the tumor cell lines MCF-7 (breast cancer) and HeLa (cervical adenocarcinoma) and on BJ (a non-tumor human skin fibroblast cell line), using MTT assay. The results obtained indicated that P. aerugineum polysaccharide exhibited a high ABTS radical-scavenging activity reaching up to 55%. The cytotoxic effect was best expressed in MCF-7 cells treated for 72 h with 1000 µg/mL PaEPS, where tumor cell proliferation was inhibited by more than 70%. Importantly, the PaEPS treatments did not significantly affect the viability of BJ cells. These findings promote the biotechnological production of P. aerugineum extracellular polysaccharide and reveal its potential as an anticancer and antioxidant agent for future applications.

Introduction. Shellfish poisoning could be caused by ingestion of shellfish contaminated by bacteria, viruses, heavy metals, pesticides, marine biotoxins etc. The objective of the study was to determine the theoretical human exposure to shellfish toxins of Bulgarian consumers. The incidence of registered cases of intoxications due to marine biotoxins ingestion – “Toxic effect: Other fish and shellfish poisoning (T61.2)” in regional hospitals was investigated. Materials and methods. Acute exposure (AE), chronic exposure (CE) and hazard quotient (HQ) are calculated based on experimentally-determined levels of shellfish toxins in mussels harvested from North Bulgarian coast in the period 2012 – 2018. We performed a retrospective analysis of the frequency of the diagnosis T61.2, based on medical documentation for the period 1.05.2012 – 7.12.2018 in hospitals in Varna and Dobrich, Bulgaria. Results. Shellfish toxins that most often occur in mussels are domoic acid (DA), yessotoxin (YTX) and pectenotoxin-2 (PTX2). The calculated maximum AE was 1.00 mg/kg bw for DA, 0.02 mg/kg bw for YTX and 0.0045 mg/kg bw for PTX2, respectively. Maximum CE for DA was 0.01 mg/kg bw/day. All estimated values were much lower than those legislated by European Food Safety Authority thresholds. The exposure to marine biotoxins was estimated based on contamination data of mussels harvested from the same region. The analysis for the whole period showed that its frequency is lower than 1%. Clinicians report that this diagnosis is presumptive, based on recent ingestion of shellfish and presence of symptoms consistent with shellfish poisoning. Conclusions. Estimated theoretical acute and chronic exposures agree upon the incidence of clinical cases with the studied diagnosis. A parallel investigation on a prolonged period is required, to increase the accuracy of these results.

Resurrection plants present an attractive model for studying the mechanisms of desiccation tolerance. In addition to drought, the presence of light during desiccation is extremely dangerous. In the present study, we investigated the effect of light during the desiccation of shade and sun Haberlea rhodopensis from two different habitats by measuring the changes in electrolyte leakage, malondialdehyde and proline content, and photosynthetic and antioxidant activities. Moreover, the plasticity and acclimation ability of plants to changing light intensities were studied by desiccating shade plants under high light and sun plants under low light. The most significant differences between shade and sun plants were observed under moderate dehydration. Regardless of some decline in PSII activity in sun plants, it was much higher compared to shade plants. The lower PSII efficiency in the latter was accompanied by a higher extent of excitation pressure and consequently significant enhancement in non-photochemical quenching, Y(NPQ), and especially in the fraction of energy that is passively dissipated as heat and fluorescence, Y(NO). The activity of antioxidant enzymes remained high during the desiccation of H. rhodopensis , being higher in the sun compared to shade plants in an air-dried state. In addition, shade and sun plants showed high acclimation capacity when desiccated at opposite light intensities.

Diabetic cardiomyopathy is a major etiological factor in heart failure in diabetic patients, characterized by mitochondrial oxidative metabolism dysfunction, myocardial fibrosis, and marked glycogen elevation. The aim of the present study is to evaluate the effect of endurance training and prebiotic xylooligosaccharide (XOS) on the activity of key oxidative enzymes, myocardial collagen, and glycogen distribution as well as some serum biochemical risk markers in streptozotocin-induced type 1 diabetic rats. Male Wistar rats (n = 36) were divided into four diabetic groups (n = 9): sedentary diabetic rats on a normal diet (SDN), trained diabetic rats on a normal diet (TDN), trained diabetic rats on a normal diet with an XOS supplement (TD-XOS), and sedentary diabetic rats with an XOS supplement (SD-XOS). The results show that aerobic training managed to increase the enzyme activity of respiratory Complex I and II and the lactate dehydrogenase in the cardiomyocytes of the diabetic rats. Furthermore, the combination of exercise and XOS significantly decreased the collagen and glycogen content. No significant effects on blood pressure, heart rate or markers of inflammation were detected. These results demonstrate the beneficial effects of exercise, alone or in combination with XOS, on the cardiac mitochondrial enzymology and histopathology of diabetic rats.

The purpose of this study is to study the effects of ostarine alone and in combination with endurance training in sexually mature, male Wistar rats. The rats were divided into a treadmill-trained group and a sedentary group. Half of each group received either ostarine or vehicle for 8 weeks (n = 10 each, in total n = 40). We examined some functional, hormonal, and anthropometric parameters and the myogenic gene expression of myostatin, insulin-like growth factor-1 (IGF-1), and vascular endothelial growth factor-A (VEGF-A) in m. gastrocnemius. Ostarine decreased submaximal endurance and increased myogenic gene expression of myostatin but had no effect on maximal time to exhaustion and grip strength. Training increased submaximal endurance, maximal time to exhaustion, and grip strength. Our results indicate that both exercise and ostarine treatment had no significant effects on serum levels of luteinizing hormone, follicle-stimulating hormone, and testosterone, or on the myogenic gene expression of IGF-1 and VEGF-A. Neither ostarine nor the training had a significant effect on the testis, liver, and heart weights. In conclusion, ostarine had no effect on anthropometric and hormonal parameters but increased the myostatin gene expression in muscle. The SARM treatment decreased submaximal endurance without affecting maximal time to exhaustion, and training increased both metrics., (© 2024. The Author(s).)

Tomato ( Solanum lycopersicum L.) plants, wild type Ailsa Craig , and carotenoid mutant tangerine that accumulates prolycopene instead of all- trans -lycopene were exposed to a combined treatment by low light and low temperature for 5 days. The ability of plants to recover from the stress after development for 3 days at control conditions was followed as well. The suffered oxidative stress was evaluated by the extent of pigment content, lipid peroxidation, membrane stability, and H 2 O 2 generation. The level of MDA content under combined treatment in tangerine implies that the mutant demonstrates lower sensitivity to stress in comparison with Ailsa Craig . The oxidative protective strategy of plants was estimated by following the antioxidant and antiradical activity of phenolic metabolites, including anthocyanins, as well as the activities of antioxidant enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX) and catalase (CAT). Presented results revealed that the oxidative stress was much stronger expressed after exposure of both types of plants to low light combined with low temperature compared to that after treatment with only low light. The most significant antioxidant protection was provided by phenolic substances, including anthocyanins. The lower sensitivity of tangerine plants to low light can be attributed to the higher activity of the antioxidant enzyme CAT.

Prenatal stress (PNS), which alters the hypothalamic-pituitary-adrenal axis function in the offspring, predisposes to insulin resistance (IR) in later life and is associated with numerous disorders, including cognitive and memory impairments. At present, our main goal is to assess the effects of chronic piromelatine (Pir) administration, a melatonin analogue, on PNS-provoked IR in the periphery and the hippocampus in male and female offspring. Pregnant Sprague-Dawley rats were exposed to chronic stress (one short-term stressor on a daily basis and one long-term stressor on a nightly basis) from the first gestation week until birth. Vehicle or Pir 20 mg/kg were administered intraperitoneally for 21 days. Plasma glucose, serum insulin levels, and the homeostasis model assessment of insulin resistance (HOMA-IR) were determined as markers of peripheral IR. For the hippocampal IR assessment, insulin receptors (IRs) and glucose transporter 4 (GLUT4) were examined. Prenatally stressed offspring of both sexes indicated enhanced plasma glucose and serum insulin concentrations, increased HOMA-IR, and decreased hippocampal GLUT4 only in male rats. The PNS-induced changes were corrected by chronic treatment with Pir. The present results suggest that the melatoninergic compound Pir exerts beneficial effects on altered glucose/insulin homeostasis in PNS-exposed offspring.

The prolonged consumption of a high-fat diet (HFD) leads to abnormal growth of the visceral adipose tissue (VAT), increased macrophage infiltration, and altered secretion of biologically active molecules. This is considered as a precondition for the development of obesity, inflammation, and obesity-related disorders. Therefore, we studied HFD-induced changes in the tissue levels of the inflammatory markers C-reactive protein, serum amyloid-A, and interleukin-4 in healthy male Wistar rats. The animals were first divided at random into two groups subjected to either a standard or a high-fat diet. The initial effect of the diet was evaluated after fourteen weeks. In order to study the diet duration effect, the standard diet was given to twelve animals from the HFD group, while the remaining continued with the HFD for an additional four weeks. Our results showed that the HFD barely affected body mass index, conicity, relative fat mass, and Lee indices, whereas it provoked adipocyte hypertrophy and gradually increased the levels of both the pro- and anti-inflammatory markers. The switch from the high-fat to the standard diet resulted in the comparatively fast restoration of the baseline levels of the studied molecules. Although, the prolonged consumption of an HFD causes adipocyte hypertrophy in healthy male animals, the inflammatory process in VAT is well-coordinated, time-dependent, and reversible.

The effects of combination treatments using the selective androgen receptor modulators (SARMs) ostarine (OST) or ligandrol (LIG) with treadmill exercise (TE) were studied in healthy adult rats. Fifteen-week-old male Wistar rats were divided into groups ( n = 10/group). Experiment 1 consisted of (1) Control group: sedentary rats receiving vehicle; (2) OST: sedentary rats receiving OST; (3) TE: training rats receiving vehicle; (4) OST + TE: training rats receiving OST. Experiment 2 consisted of (1) LIG: sedentary group receiving LIG; (2) LIG + TE: training group receiving LIG. The TE regime was as follows: 25 m/min, 5° elevation, 40 min, five times/week, and the sedentary regime was 5 min, three times/week. OST and LIG were administered subcutaneously (0.4 mg/kg body weight/day, five times/week). After eight weeks, bone samples underwent microcomputed tomographical, biomechanical, histological, and ashing analyses. All the treatments had weak effects on the bone structure without affecting bone biomechanics. The OST + TE improved bone structure, while the LIG + TE had unfavorable effects. In serum, OST, OST + TE, and LIG + TE altered cholesterol and lipoprotein levels; TE did not change the serum parameters. The SARM treatments had no clear bone benefit, and the serum effects can be considered as side effects. TE represents a safe treatment. Because SARMs are increasingly applied in gyms along with physical activities, attention should be paid to possible side effects.

The desiccation tolerance of plants relies on defense mechanisms that enable the protection of macromolecules, biological structures, and metabolism. Although the defense of leaf tissues exposed to solar irradiation is challenging, mechanisms that protect the viability of the roots, yet largely unexplored, are equally important for survival. Although the photosynthetic apparatus in leaves contributes to the generation of oxidative stress under drought stress, we hypothesized that oxidative stress and thus antioxidative defense is also predominant in the roots. Thus, we aimed for a comparative analysis of the protective mechanisms in leaves and roots during the desiccation of Haberlea rhodopensis . Consequently, a high content of non-enzymatic antioxidants and high activity of antioxidant enzymes together with the activation of specific isoenzymes were found in both leaves and roots during the final stages of desiccation of H. rhodopensis . Among others, catalase and glutathione reductase activity showed a similar tendency of changes in roots and leaves, whereas, unlike that in the leaves, superoxide dismutase activity was enhanced under severe but not under medium desiccation in roots. Nitric oxide accumulation in the root tips was found to be sensitive to water restriction but suppressed under severe desiccation. In addition to the antioxidative defense, desiccation induced an enhanced abundance of dehydrins, ELIPs, and sHSP 17.7 in leaves, but this was significantly better in roots. In contrast to leaf cells, starch remained in the cells of the central cylinder of desiccated roots. Taken together, protective compounds and antioxidative defense mechanisms are equally important in protecting the roots to survive desiccation. Since drought-induced damage to the root system fundamentally affects the survival of plants, a better understanding of root desiccation tolerance mechanisms is essential to compensate for the challenges of prolonged dry periods.

Since water scarcity is one of the main risks for the future of agriculture, studying the ability of different wheat genotypes to tolerate a water deficit is fundamental. This study examined the responses of two hybrid wheat varieties (Gizda and Fermer) with different drought resistance to moderate (3 days) and severe (7 days) drought stress, as well as their post-stress recovery to understand their underlying defense strategies and adaptive mechanisms in more detail. To this end, the dehydration-induced alterations in the electrolyte leakage, photosynthetic pigment content, membrane fluidity, energy interaction between pigment-protein complexes, primary photosynthetic reactions, photosynthetic and stress-induced proteins, and antioxidant responses were analyzed in order to unravel the different physiological and biochemical strategies of both wheat varieties. The results demonstrated that Gizda plants are more tolerant to severe dehydration compared to Fermer, as evidenced by the lower decrease in leaf water and pigment content, lower inhibition of photosystem II (PSII) photochemistry and dissipation of thermal energy, as well as lower dehydrins' content. Some of defense mechanisms by which Gizda variety can tolerate drought stress involve the maintenance of decreased chlorophyll content in leaves, increased fluidity of the thylakoid membranes causing structural alterations in the photosynthetic apparatus, as well as dehydration-induced accumulation of early light-induced proteins (ELIPs), an increased capacity for PSI cyclic electron transport and enhanced antioxidant enzyme activity (SOD and APX), thus alleviating oxidative damage. Furthermore, the leaf content of total phenols, flavonoids, and lipid-soluble antioxidant metabolites was higher in Gizda than in Fermer.

Resurrection plants have the unique ability to restore normal physiological activity after desiccation to an air-dry state. In addition to their desiccation tolerance, some of them, such as Haberlea rhodopensis and Ramonda myconi , are also freezing-tolerant species, as they survive subzero temperatures during winter. Here, we compared the response of the photosynthetic apparatus of two other Gesneriaceae species, Ramonda serbica and Ramonda nathaliae , together with H. rhodopensis , to cold and freezing temperatures. The role of some protective proteins in freezing tolerance was also investigated. The water content of leaves was not affected during cold acclimation but exposure of plants to -10 °C induced dehydration of plants. Freezing stress strongly reduced the quantum yield of PSII photochemistry (Y(II)) and stomatal conductance ( g s ) on the abaxial leaf side. In addition, the decreased ratio of F v /F m suggested photoinhibition or sustained quenching. Freezing-induced desiccation resulted in the inhibition of PSII activity, which was accompanied by increased thermal energy dissipation. In addition, an increase of dehydrins and ELIPs was detected, but the protein pattern differed between species. During recovery, the protein abundance decreased and plants completely recovered their photosynthetic activity. Thus, our results showed that R. serbica, R. nathaliae, and H. rhodopensis survive freezing stress due to some resurrection-linked traits and confirmed their freezing tolerance.

Haberlea rhodopensis is a unique resurrection plant of high phenotypic plasticity, colonizing both shady habitats and sun-exposed rock clefts. H. rhodopensis also survives freezing winter temperatures in temperate climates. Although survival in conditions of desiccation and survival in conditions of frost share high morphological and physiological similarities, proteomic changes lying behind these mechanisms are hardly studied. Thus, we aimed to reveal ecotype-level and temperature-dependent variations in the protective mechanisms by applying both targeted and untargeted proteomic approaches. Drought-induced desiccation enhanced superoxide dismutase (SOD) activity, but FeSOD and Cu/ZnSOD-III were significantly better triggered in sun plants. Desiccation resulted in the accumulation of enzymes involved in carbohydrate/phenylpropanoid metabolism (enolase, triosephosphate isomerase, UDP-D-apiose/UDP-D-xylose synthase 2, 81E8-like cytochrome P450 monooxygenase) and protective proteins such as vicinal oxygen chelate metalloenzyme superfamily and early light-induced proteins, dehydrins, and small heat shock proteins, the latter two typically being found in the latest phases of dehydration and being more pronounced in sun plants. Although low temperature and drought stress-induced desiccation trigger similar responses, the natural variation of these responses in shade and sun plants calls for attention to the pre-conditioning/priming effects that have high importance both in the desiccation responses and successful stress recovery.

Haberlea rhodopensis belongs to the small group of angiosperms that can survive desiccation to air-dry state and quickly restore their metabolism upon rehydration. In the present study, we investigated the accumulation of sHSPs and the extent of non-photochemical quenching during the downregulation of photosynthesis in H. rhodopensis leaves under desiccation at optimum (23 °C) and high temperature (38 °C). Desiccation of plants at 38 °C caused a stronger reduction in photosynthetic activity and corresponding enhancement in thermal energy dissipation. The accumulation of sHSPs was investigated by Western blot. While no expression of sHPSs was detected in the unstressed control sample, exposure of well-hydrated plants to high temperature induced an accumulation of sHSPs. Only a faint signal was observed at 50% RWC when dehydration was applied at 23 °C. Several cross-reacting polypeptide bands in the range of 16.5-19 kDa were observed in plants desiccated at high temperature. Two-dimensional electrophoresis and immunoblotting revealed the presence of several sHSPs with close molecular masses and pIs in the range of 5-8.0 that differed for each stage of treatment. At the latest stages of desiccation, fourteen different sHSPs could be distinguished, indicating that sHSPs might play a crucial role in H. rhodopensis under dehydration at high temperatures.

Resurrection plants are able to deal with complete dehydration of their leaves and then recover normal metabolic activity after rehydration. Only a few resurrection species are exposed to freezing temperatures in their natural environments, making them interesting models to study the key metabolic adjustments of freezing tolerances. Here, we investigate the effect of cold and freezing temperatures on physiological and biochemical changes in the leaves of Haberlea rhodopensis under natural and controlled environmental conditions. Our data shows that leaf water content affects its thermodynamical properties during vitrification under low temperatures. The changes in membrane lipid composition, accumulation of sugars, and synthesis of stress-induced proteins were significantly activated during the adaptation of H. rhodopensis to both cold and freezing temperatures. In particular, the freezing tolerance of H. rhodopensis relies on a sucrose/hexoses ratio in favor of hexoses during cold acclimation, while there is a shift in favor of sucrose upon exposure to freezing temperatures, especially evident when leaf desiccation is relevant. This pattern was paralleled by an elevated ratio of unsaturated/saturated fatty acids and significant quantitative and compositional changes in stress-induced proteins, namely dehydrins and early light-induced proteins (ELIPs). Taken together, our data indicate that common responses of H. rhodopensis plants to low temperature and desiccation involve the accumulation of sugars and upregulation of dehydrins/ELIP protein expression. Further studies on the molecular mechanisms underlying freezing tolerance (genes and genetic regulatory mechanisms) may help breeders to improve the resistance of crop plants.

Epilepsy is a brain disorder characterized by recurrent epileptic seizures and neurobiological, physiological, mood, and cognitive consequences. In the last decade, the beneficial effects of regular physical exercise have been investigated in patients with neurodegenerative diseases such as epilepsy. However, data on its beneficial effects and underlying mechanisms are still insufficient. The objective of the current study was to investigate the effects of endurance training, applied before and after pilocarpine (Pilo) administration, on status epilepticus (SE) severity, and its relation to epileptogenesis deleterious consequences during the chronic epileptic phase. Long-term aerobic training, applied four weeks before SE and eight weeks after SE, elevated the threshold to induce SE and reduced spontaneous motor seizures. The protective effect of this alternative approach on seizure susceptibility resulted in improved memory responses, and alleviated comorbid depression in epileptic rats. The exercised epileptic rats had improved markers of oxidative stress by decreasing lipid peroxidation and increasing the levels of glutathione and activity of superoxide dismutase in the rat hippocampus. Aerobic training managed to ameliorate the neuroinflammation by decreasing the levels of TNF-α and IL-1β in the hippocampus. Our results suggest that regular physical training predisposes the subjects to crucial plastic changes, leading to increased resistance to SE and the development of epileptogenesis.

Type 1 diabetes mellitus is characterized with decreased microbial diversity. Gut microbiota is essential for the normal physiological functioning of many organs, especially the brain. Prebiotics are selectively fermentable oligosaccharides [xylooligosaccharides (XOS), galactooligosaccharides, etc.] that promote the growth and activity of gut microbes and influence the gut-brain axis. Aerobic exercise is a non-pharmacological approach for the control of diabetes and could improve cognitive functions. The potential beneficial effect of XOS and/or aerobic training on cognition, the lipid profile and oxidative stress markers of experimental rats were evaluated in this study. Male Wistar rats were randomly divided into three streptozotocin-induced diabetic groups and a control group. Some of the rats, either on a XOS treatment or a standard diet, underwent aerobic training. The results showed that the aerobic training independently lowered the total cholesterol levels compared to the sedentary diabetic rats ( p = 0.032), while XOS lowers the malondialdehyde levels in the trained diabetic rats ( p = 0.034). What is more the exercise, independently or in combination with XOS beneficially affected all parameters of the behavioral tests. Haberlea rhodopensis is a unique desiccation-tolerant angiosperm that also survives winter frost. As, upon freezing temperatures, H. rhodopensis desiccates, the taxon is proposed to survive low temperature stress using its desiccation tolerance mechanisms. To reveal the validity of this hypothesis, we analyzed the structural alterations and organization of photosynthetic apparatus during the first hours of recovery after drought- and freezing-induced desiccation. The dynamics of the ultrastructure remodeling in the mesophyll cells and the restoration of the thylakoid membranes shared similarities independent of the reason for desiccation. Among the most obvious changes in thylakoid complexes, the proportion of the PSI-LHCII complex strongly increased around 70% relative water content (RWC), whereas the proportion of Lhc monomers decreased from the beginning of rehydration. We identified enhanced levels of cyt b 6 f complex proteins that contributed to the enhanced electron flow. The high abundance of proteins related to excitation energy dissipation, PsbS, Lhcb5, Lhcb6 and ELIPs, together with the increased content of dehydrins contributed to the preservation of cellular integrity. ELIP expression was maintained at high levels up to 9 h into recovery. Although the recovery processes from drought- and freezing-induced desiccation were found to be similar in progress and time scale, slight variations indicate that they are not identical.

The pineal gland is suggested to be an essential area involved in the programming of fertility, growth, aging, and death of mammals via the released hormone melatonin.The present study aimed to ascertain the effect of melatonin deficit on several physiological and metabolic parameters, closely associated with the aging process, at certain stages of ontogenesis. Sham and rats with pinealectomy, operated at ages 3, 14, and 18-months, respectively, were tested two months later. Sham rats demonstrated an age-related decline of muscle strength, exercise endurance, motor activity, food intake, calorimetric parameters, and impaired lipid profile. Pinealectomy reduced the maximal time to exhaustion and body weight gain while diminished motor activity, food intake, O 2 consumption, CO 2 production, and energy expenditure during the Dark phase in the youngest rat group. In addition, melatonin deficit elevated arterial blood pressure (systolic, diastolic, and mean arterial pressure) and increased serum glucose and triglyceride level in 3-month-old rats while decreased the liver enzyme activity in 14-month-old rats. In conclusion, the present study brought new insights confirming the complex impact of melatonin deficit on important physiological, metabolic and biochemical markers related to aging and demonstrated for the first time that the lack of melatonin hormone is harmful in young adult rats., (Copyright © 2022 Elsevier Inc. In this study, the contribution of nonenzymatic (ascorbate, glutathione) and enzymatic antioxidants (superoxide dismutase, catalase, glutathione reductase, glutathione S-transferase) in the first hours of recovery of the resurrection plant Haberlea rhodopensis from drought- and freezing-induced desiccation was assessed. The initial stage of recovery after desiccation is critical for plants, but less investigated. To better understand the alterations in the activity of antioxidant enzymes, their isoenzyme patterns were determined. Our results showed that ascorbate content remained high during the first 9 h of rehydration of desiccated plants and declined when the leaves' water content significantly increased. The glutathione content remained high at the first hour of rehydration and then strongly decreased. The changes in ascorbate and glutathione content during recovery from drought- and freezing-induced desiccation showed great similarity. At the beginning of rehydration (1-5 h), the activities of antioxidant enzymes were significantly increased or remained as in dry plants. During 7-24 h of rehydration, certain differences in the enzymatic responses between the two plant groups were registered. The maintenance of a high antioxidant activity and upregulation of individual enzyme isoforms indicated their essential role in protecting plants from oxidative damage during the onset of recovery.
