Rainfall infiltration is the main inducing factor for the instability of unsaturated soil slopes, and root water uptake and reinforcement play an important role in preventing shallow landslide. In order to explore the influence of vegetation root on the soil hydraulic and mechanical properties under rainfall, a self-designed soil permeability coefficient measuring device considering the effects of vegetation was used to study the soil water characteristic curve (SWCC) and permeability coefficient of Festuca Arundinacea, Ophiopogon Japonicus, Ligustrum Vicaryi and bare soil under two different rainfall conditions (3.0mm/h and 5.0mm/h) were studied. Then, the direct shear tests of root-soil composite with different water contents and root contents were carried out. Finally, the slope stability under different rainfall and vegetation was simulated by GeoStudio. Results show that: root water uptake can effectively reduce soil water content and increase soil suction, and its influence range is about 2-3 times the length of the root system. Root water uptake can also significantly improve the soil water retention capacity. The air entry value of vegetation soil is larger than that of bare soil, and the permeability coefficient of vegetation soil is about one order of magnitude lower than that of bare soil. Among the three different types of vegetation, the effect of Festuca Arundinacea and Ophiopogon Japonicus on soil water content and suction is more significant than Ligustrum Vicaryi. Root reinforcement mainly increases the soil shear strength by improving the cohesion of the root-soil composite, but has little effect on the internal friction angle. The cohesion of the root-soil composite is affected by soil water content, root content and root distribution, which increases with the increase of root content, and decreases with the increase of water content. When the roots are vertically distributed, the cohesion of the root-soil composite is greater than when the roots are placed horizontally and inclined. Vegetation can effectively improve the stability of the shallow slope under various rainfall conditions, but has little effect on the stability of a deep slope. The safety factor of all three types of vegetated slopes is higher than that of bare soil slopes.

This paper explores the micro characteristics of unsaturated sand in triaxial shearing by using X-ray computed tomography (X-ray CT). To obtain higher resolution, a mini-triaxial set-up is designed in which the sample is miniaturised to 1 cm in diameter and 2 cm long, allowing scans with a pixel size of 9 μm. Samples are sheared in the mini-triaxial set-up at different constant suction levels (therefore, different degree of saturation). In the meanwhile, the samples are scanned by X-ray CT at various deformation stages, about 0, 2, 5, 10 and 15% axial strain. The three-dimensional (3D) reconstructed image is trinarised based on a region growing technique, which gives access to the microstructure of the solid, liquid and air phases. Then, the 3D image is subdivided into representative volume elements, with length ≈3·8D50, which gives local information of degree of saturation and porosity. It is observed that the sample dilates and water drains out during triaxial test under constant suction condition. The local study shows that the porosity increase and water desaturation are more significant in the middle part of the sample, especially for a higher suction value. The glazed tile is an important building material used throughout the history of traditional Chinese architecture. Architectural glazed tiles used to decorate the screen walls of ancient China are studied scientifically for the first time. More than 30 glazed tile samples from the screen walls of the 15th to 18th century AD of the Hancheng Confucian Temple and Town God’s Temple in Shaanxi Province were carefully investigated using SEM–EDS and XRD. Microstructure and chemistry indicated the raw materials, the recipes and the technological choices used to produce the paste and glaze of the glazed tile samples studied. The causes for the key degradation processes of these glazed tiles used as building materials in the screen walls have also been discussed. This work has clear implications for the restoration and conservation treatments on these kinds of ancient Chinese building materials.

A triple-source CT system is proposed for micro-scale testing of geological materials. This study aims at reducing the projection acquisition time by two-thirds compared to a conventional single-source CT system. The proposed system with different positioning errors in the source-to-object distance (SOD) was simulated and tested using the Shepp-Logan phantom model, as well as slices of sand, glass beads, and concrete samples. Furthermore, the imaging quality of a single-source and the triple-source CT system with different dead detector pixels was compared. The results showed that within the maximum allowable positioning error, the pixel differences between the simulated and the original images are close to zero, and the structural similarities are greater than 0.96. In the presence of dead detector pixels, the quality of the simulated images in the triple-source CT system is superior to that of a single-source CT system. The presented triple-source CT system performs well in high-quality image reconstruction.

Determining the safety factor by limit equilibrium methods is based on different assumed slip surfaces and equilibrium equations for forces and moments to be satisfied. The safety factor can be predicted using empirical correlations instead of going through lengthy calculations. In this paper, empirical equations between the factor of safety (FS), unit weight (γ), cohesion (c), internal friction angle (φ), slope angle (α), and height of the slope (H) have been proposed for Spencer’s, Morgenstern-Price, Janbu’s simplified, Bishop’s simplified, and Lowe-Karafiath methods. Data of 50 slopes having homogeneous soil strata with non-identical shearing strength parameters, containing different geometries and the water table below the base of the slope, have been selected to determine FS using Slope-W and for the development of correlations using SPSS software. Unit weight, cohesion, friction angle, slope angle, and height of the slope vary from 11.5 to 21 (kN/m3), 0 to 137 (kPa), 0 to 39 (°), 11 to 78 (°), and 3.5 to 22 (m), respectively. Multiple regression analyses were performed on Slope-W outcomes, and equations for the prediction of FS were formulated. Validation of the proposed FS equations was done both internally and externally as well. The validation graphs are drawn between Slope-W FS versus developed equations for FS of each method. The correlations are helpful for the calculation of FS to have a quick prediction of failure of a slope, and ultimately preventive measures can be taken. The regression constant (R2) values for Spencer’s, Morgenstern-Price, Janbu’s simplified, Bishop’s simplified, and Lowe-Karafiath methods are 0.940, 0.940, 0.922, 0.932, and 0.935, respectively.

β-Glucosidase belongs to the glycoside hydrolase I family, which is widely present in multiple species and responds to various biotic and abiotic stresses. In rice, whether β-glucosidase is involved in the interaction between plants and microorganisms is not clear. In this study, we found that the expression of several genes encoding β-glucosidases, including OsBGLU19 and OsBGLU23, were upregulated after inoculation with Xanthomonas oryzae pv. oryzicola (Xoc) and downregulated after inoculation with X. oryzae pv. oryzae (Xoo). The respective insertion mutants of OsBGLU19 and OsBGLU23, bglu19 and bglu23, were more susceptible to Xoc infection. The expression of OsAOS2, a key gene in the jasmonic acid signal pathway, was dramatically downregulated after inoculation with Xoc in the bglu19 and bglu23 mutants. Simultaneously, the expression of downstream disease resistance-related genes, such as OsPR1a, OsPR5 and a key transcription factors OsWRKY72 were obviously downregulated. The resistance mediated by OsBGLU19 and OsBGLU23 to bacterial leaf streak is related to disease resistance-related genes above mentioned.

The critical state is an important concept for saturated and partially saturated granular materials as the strength and volume become constant and unique under continuous shear. By incorporating the water bridge effect, the mechanical behaviours of wet granular matters can be studied by the discrete element method (DEM). A series of DEM simulations are performed following the conventional triaxial loading path for dry and wet granular materials, and different suction values are applied at various confining stress levels. Unique critical state behaviours have been observed in both macroscopic and microscopic scales. It shows that the confining stress level plays an important role in the critical state behaviour of wet granular materials. The critical stress ratio for a wet material is not a constant value at different stress levels, and it is found that both the critical stress ratio and void ratio in wet granular matters are also much higher with a low confining stress. A framework is proposed by considering both the contact stress and the capillary stress effects to model the critical state lines. At large strain, the coordination number, the mean inter-particle force and fabric anisotropies evolve to constant critical state values for both dry and wet materials. The macro-parameters formulating the critical state stress ratio are found to be associated with the critical state anisotropies in solid skeleton and water phase fabrics, respectively.

Rapidly and randomly drifted reference frames will shorten the link distance and decrease the secure key rate of realistic quantum key distribution (QKD) systems. However, an actively or inappropriately implemented calibration scheme will increase complexity of the systems and may open security loopholes. In this article, we present a free-running reference-frame-independent (RFI) QKD scheme, where measurement events are classified into multiple slices with the same misalignment variation of reference frames and each slice performs the post-processing procedure individually. We perform the free-running RFI QKD experiment with a fiber link of 100km and the misalignment of the reference frame between Alice and Bob is varied more than 29 periods in a 50.7-hour experiment test. The average secure key rate is about 734 bps with a total loss of 31.5 dB, which achieves the state-of-art performance of the long-distance RFI QKD implementations. Our free-running RFI scheme can be efficiently adapted into the satellite-to-ground and drone based mobile communication scenarios, as it can be performed with rapidly varying reference frame and a loss more than 40 dB, where no secure key can be obtained by the original RFI scheme., Comment: 5 pages, 5 figures

The long-debated effective stress definition of unsaturated soils is believed to be correlated to suction, the degree of saturation, and the less mentioned interfacial areas. The tensile strength of unsaturated soils is directly associated with the effective stress definition in theory, and it is also crucial in engineering practices. For unsaturated soils, the relationship between suction and degree of saturation can be described as the water retention curve (WRC), which is related to the pore-size distribution. In the meanwhile, the air-water interfacial area is also regarded as a function of the degree of saturation, and parameters of the function are determined by the soil's pore structures. For granular soils, the pore-size distribution is usually in a unimodal shape and, therefore, the strength properties can also be related to the particle gradation parameters. In this study, a preliminary estimation method is proposed for the tensile strength of unsaturated sandy soils based on basic particle gradation parameters. In this method, with basic physical features considered, WRC is estimated from a characteristic grain diameter (d60) and the coefficient of uniformity (Cu). The function to determine the air-water interfacial area is also formulated by soil gradation parameters of the mean grain-size (d50), the coefficient of uniformity (Cu), and the void ratio. The proposed tensile strength estimation is compared with experimental measurements on sandy soils, which shows fair agreement, especially for the conventional split plate method. Increases in seepage force reduce the effective stress of particles and result in the erosion of particles, producing heave failure and piping. Sheet piles/cutoff walls are often employed in dams to control the seepage. In this study, a computational fluid dynamics solver involving two fluid phases was developed and coupled with discrete element method software to simulate the piping process around a sheet pile/cutoff wall. Binary-sized particles were selected to study the impact of fine particles on the mechanisms of seepage. The seepage phenomenon mainly appeared among fine particles located in the downstream side, with the peak magnitudes of drag force and displacement occurring around the retaining wall. Based on the particle-scale observations, the impact of seepage produced a looser condition for the region concentrated around the retaining wall and resulted in an anisotropic condition in the soil skeleton. The results indicate that heave behavior occurs when the drag force located adjacent to the boundary on the downstream side is larger than the corresponding weight of the bulk soil.

Introduction Non-small cell lung cancer (NSCLC) is the most common cause of cancer-related deaths in the world. MALAT1 and SOX9 have important roles in tumour formation and development in several types of cancers. However, little is known about the function and co-relationship of these 2 factors in NSCLC in vivo. Objectives To explore the role of MALAT1 and SOX9 expression relationship, their clinical pathological characteristics and OS on NSCLC patients. Methods Paired of primary lung cancer tissues and the matched tumour adjacent tissues were collected in 121 NSCLC patients. MALAT1 and SOX9 mRNA expression was measured by SYBR green q RT-PCR assay. SOX-9 protein expression was measured by streptavidin-peroxidase (SP) staining method. Results MALAT1and SOX9 expression was higher in NSCLC tissues than the adjacent tissues, and they have positive correlation. Moreover, SOX9 protein expression was higher in NSCLC tissues, especially in MALAT1 mRNA higher expressed NSCLC tissues. MALAT1 and SOX9 mRNA expression were associated with age (x2 =11.474, P = .009), tumour size (x2 =26.839, P = .000), TNM stage (x2 =8.010, P = .046) and LEL. (x2 =53.908, P = .000). NSCLC patients with higher MALAT1 and SOX9 mRNA expression had poorer OS rates. Conclusions MALAT1 and SOX9 could be used as prognostic co-biomarker in NSCLC.

Wet granular materials are three-dimensionally simulated by the discrete element method with water bridges incorporated between particles. The water bridges are simplified as toroidal shapes, and the matric suction is constantly maintained in the material. A comparison with experimental tests in the literature indicates that the toroidal shape approximation may be one of the best choices with high practicability and decent accuracy. Mechanical behaviours of wet granular materials are studied by triaxial tests. Effects of particle size distributions and void ratios are investigated systematically in this study. The hydraulic limit of the pendular state is also discussed. It gives the capillary cohesion function which is not only determined by the degree of saturation but also positively correlated to relative density and particle size polydispersity and inversely proportional to mean particle size. Furthermore, the capillary strengthening effect is also analysed microscopically in aid of the Stress–Force–Fabric relationship, mainly in fabric anisotropy, coordination number and stress transmission pattern, which revealed the micro-mechanisms of the additional effective stress induced by capillary effect.

Recent evidence highlights the crucial regulatory roles of long non-coding RNAs (lncRNA) in tumor biology. Metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) has important roles in regulating the proliferation, invasion and migration of lung cancer cell. High MALAT1 expression in lung cancer was related to poorer clinicopathologic features in this study. MALAT1 knockdown alone was sufficient to amplify DDP-induced repression of cell viability. MALAT1 knockdown could also sensitized DDP-resistant lung cancer cells (A549/DDP and H1299/DDP) to DDP. Further assays indicated that MALAT1 acted as a competing endogenous RNA to upregulate SOX9 expression by sponging miR-101 in DDP-resistant cancer cells, through Wnt signaling pathway. Moreover, SOX9 could bind to the promoter of MALAT1 to activate its transcription. Taken together, MALAT1, miR-101 and SOX9 form a feedback loop to enhance the chemo-resistance of lung cancer cell to DDP; this MALAT1-miR-101-SOX9 feedback loop plays an important role in the chemo-resistance of lung cancer cell to DDP and may serve as a potential target for cancer treatment.

AT treatment leads to apoptosis in various cell types, including breast cancer cells, by regulating different signaling pathways. Iron oxide (Fe3O4) magnetic nanoparticles (MNPs) are nanomaterials with biocompatible activity and low toxicity. In the present study, the possible benefits of AT in combination with MNPs on non-small-cell lung cancer (NSCLC) were explored in in vitro and in vivo studies. AT-MNP treatment contributed to apoptosis in NSCLC cells, as evidenced by activation of the caspase3-signaling pathway, which was accompanied by downregulation of the antiapoptotic proteins Bcl2 and BclXL, and upregulation of the proapoptotic signals Bax and Bad. The death receptors of TRAIL were also elevated following AT-MNP treatment in a p53-dependent manner. Furthermore, a mouse xenograft model in vivo revealed that AT-MNP treatment exhibited no toxicity and suppressed NSCLC growth compared to either AT or MNP monotherapies. In conclusion, this study suggests a novel therapy to induce apoptosis in suppressing NSCLC growth in a p53-dependent manner by combining AT with Fe3O4 MNPs. Estimating hydraulic conductivity from particle size distribution (PSD) is an important issue for various engineering problems. Classical models such as Hazen model, Beyer model, and Kozeny-Carman model usually regard the grain diameter at 10% passing ( d10) as an effective grain size and the effects of particle size uniformity (in Beyer model) or porosity (in Kozeny-Carman model) are sometimes embedded. This technical note applies the dimensional analysis (Buckingham's ∏ theorem) to analyze the relationship between hydraulic conductivity and particle size distribution (PSD). The porosity is regarded as a dependent variable on the grain size distribution in unconsolidated conditions. It indicates that the coefficient of grain size uniformity and a dimensionless group representing the gravity effect, which is proportional to the mean grain volume, are the main two determinative parameters for estimating hydraulic conductivity. Regression analysis is then carried out on a database comprising 431 samples collected from different depositional environments and new equations are developed for hydraulic conductivity estimation. The new equation, validated in specimens beyond the database, shows an improved prediction comparing to using the classic models.

This study proposed two pedotransfer functions (PTFs) to estimate sandy soil water retention curves. It is based on the van Genuchten's water retention model and from a semi-physical and semi-statistical approach. Basic gradation parameters of d60 as particle size at 60% passing and the coefficient of uniformity Cu are employed in the PTFs with two idealized conditions, the monosized scenario and the extremely polydisperse condition, satisfied. Water retention tests are carried out on 8 granular materials with narrow particle size distributions as supplementary data of the UNSODA database. The air entry value is expressed as inversely proportional to d60 and the parameter n, which is related to slope of water retention curve, is a function of Cu. The proposed PTFs, although have fewer parameters, have better fitness than previous PTFs for sandy soils. Furthermore, by incorporating with the suction stress definition, the proposed pedotransfer functions are imbedded in shear strength equations which provide a way to estimate capillary induced tensile strength or cohesion at a certain suction or degree of saturation from basic soil gradation parameters. The estimation shows quantitative agreement with experimental data in literature, and it also explains that the capillary induced cohesion is generally higher for materials with finer mean particle size or higher polydispersity.

Pervious concrete piles (PCP) have high permeability and strength. During installation by vibrating-sinking tube construction, liquified soil migrates into PCP, leading to clogging. To study PCP clogging, model tests and X-ray computed tomography analyses were carried out. The clogging depth, porosity, and excess pore water pressure dissipation were characterized. The effect of geotextile on PCP anti-clogging was evaluated. The results show an increase in target porosity and sampling depth significantly increased the PCP clogging depth. A preliminary model was proposed to evaluate the clogging of PCP. The results show that clogging can be solved by surrounding PCP with geotextile.

Sclerotinia sclerotiorum is one of the most devastating necrotrophic phytopathogens. Virulence of the hyphae of this fungus at different ages varies significantly. Molecular mechanisms underlying this functional distinction are largely unknown. In this study, we confirmed the effect of mycelial culture time/age on virulence in two host plants and elucidated its molecular and morphological basis. The virulence of the S. sclerotiorum mycelia in plants dramatically decreases along with the increase of the mycelial age. Three-day-old mycelia lost the virulence in plants. Comparative proteomics analyses revealed that metabolism pathways were comprehensively reprogrammed to suppress the oxalic acid (OA) accumulation in old mycelia. The oxaloacetate acetylhydrolase (OAH), which catalyzes OA biosynthesis, was identified in the S. sclerotiorum genome. Both gene expression and protein accumulation of OAH in old mycelia were strongly repressed. Moreover, in planta OA accumulation was strikingly reduced in old mycelia-inoculated plants compared with young vegetative mycelia-inoculated plants. Furthermore, supply with 10 mmol L−1 OA enabled the old mycelia infect the host plants, demonstrating that loss of virulence of old mycelia is mainly caused by being unable to accumulate OA. Additionally, aerial mycelia started to develop from 0.5-day-old vegetative mycelia and dominated over 1-day-old mycelia grown on potato dextrose agar plates. They were much smaller in hypha diameter and grew significantly slower than young vegetative mycelia when subcultured, which did not maintain to progenies. Collectively, our results reveal that S. sclerotiorum aerial hyphae-dominant old mycelia fail to accumulate OA and thereby lose the virulence in host plants.

A regenerable electrochemiluminescence aptasensor for thrombin (TB) has been developed using dual co-reactants of poly(ethylenimine) (PEI) and thiosemicarbazide (TSC) for signal amplification. Herein, Au nanoparticle (AuNPs) were in-situ electrodeposited on glassy carbon electrode (GCE) in advance, and then the colloidal gold nanoparticle (nano-Au) and TSC were alternately self-assembled on AuNPs/GCE to obtain nano-Au/TSC/AuNPs multi-layers for 5’-thiolated partly complementary strand (PCS) immobilization. Subsequently, the multifunctional binding aptamer (MBA), consisted of an adenosine triphosphate (ATP) binding aptamer (ABA) and a thrombin binding aptamer (TBA), was hybridized with PCS to generate double-strands DNA (PCS/MBA). Meanwhile, the signal probe was fabricated by immobilizing detection thrombin aptamer (TBA 2) on the surface of PEI enhanced Ru(bpy)32+ doped nanospheres. On the basis of sandwich-type tactics, TB was measured quantitatively in the range from 1 × 10−5 nM to 0.1 nM. More importantly, the aptasensor could regenerate by treating with ATP to take off MBA based on ATP-induced conformation change, leaving only PCS onto the modified electrode to re-hybridize with MBA. The proposed aptasensor exhibited good selectivity, acceptable stability and satisfactory regenerability.

Herein, a novel "on-off" electrochemiluminescence (ECL) aptasensor for highly sensitive determination of thrombin has been constructed based on the triple quenching of the effect of hemin/G-quadruplex DNAzymes upon the Ru(II) complex-based ECL system. First, a strong initial ECL signal was achieved by the dual amplification strategies of (i) intramolecular coreaction of a self-enhanced Ru(II)-based molecule (PTCA-PEI-Ru(II)) and (ii) intermolecular coreaction between PTCA-PEI-Ru(II) and nicotinamide adenine dinucleotide (NADH), which was named the signal-on state. Then, a novel triple quenching of the effect of multifunctional hemin/G-quadruplex DNAzymes upon the Ru(II) complex-based ECL system was designed to realize the desirable signal-off state, which was outlined as follows: (i) the hemin/G-quadruplex DNAzymes mimicked NADH oxidase to oxidize NADH and in situ generate the H2O2, consuming the coreactant of NADH; (ii) its active center of hemin could oxidize the excited state PTCA-PEI-Ru(II)* to PTCA-PEI-Ru(III), making the energy and electron transfer quench; (iii) it also acted as horseradish peroxidase (HRP) to catalyze the H2O2 for in situ producing the quencher of O2. Based on triple quenching of the effect of hemin/G-quadruplex DNAzymes, the highly sensitive "on-off" thrombin aptasensor was developed with a wide linear detection range of 1.0 × 10(-14) M to 1.0 × 10(-10) M and a detection limit down to the femtomolar level.

Kruppel-like factor 2 (KLF2) expression is diminished in many malignancies. However, its expression and role in nonsmall-cell lung cancer (NSCLC) remain unknown. In this study, we found that KLF2 levels were decreased in NSCLC tissues compared with adjacent normal tissues. Its expression level was significantly correlated with TNM stages, tumor size, and lymph node metastasis. Moreover, patients with low levels of KLF2 expression had a relatively poor prognosis. Furthermore, knockdown of KLF2 expression by siRNA could promote cell proliferation, while ectopic expression of KLF2 inhibited cell proliferation and promoted apoptosis in NSCLC cells partly via regulating CDKN1A/p21 and CDKN2B/p15 protein expression. Our findings present that decreased KLF2 could be identified as a poor prognostic biomarker in NSCLC and regulate cell proliferation and apoptosis.

The new broken-explosive warhead for back deep underground target and airport runway hard target has a unique advantage. Through analyzing hard target characteristics of the concrete , process using ANSYS/LS-DYNA software to break-explosive warhead of a warhead forming process and penetration into concrete target is studied by numerical simulation. Capillarity in wet granular materials induces cohesion and increases the material strength due to the attractive forces acting on capillary bridges. In the funicular state, water bridges may be not only formed between two grains but also binding three or more particles, which breaks the axial symmetry of the liquid bridge. This work presents a fundamental study on capillary forces and rupture behaviours of funicular water bridges between three spherical bodies at equilibrium (or static) configurations. Funicular water clusters are numerically solved by an energy minimization approach. Experimental comparisons are made by measuring capillary forces and these confirm the validity of the numerical solutions. Evolutions of capillary forces and rupture distances are investigated systematically by moving two spheres away from the centre. The fixed water volume condition and the constant mean curvature condition are studied respectively. Comparisons are made between the un-coalesced pendular liquid rings and the coalesced funicular bridge. For a same fixed total water volume, the capillary force is weakened by water bridge coalescence to a funicular bridge when the spheres are packed together, but the situation may vary for different contact angles and inter-particle distances. For the constant mean curvature condition, water bridge coalescence does not alter capillary force significantly when particles are packed closely, but the discrepancy is larger by increasing the gap. Funicular water bridge rupture criteria are also proposed based on the studied configurations. It is observed that in general the transmission from pendular to funicular state extends the rupture distance when it has a relatively high water volume or low air-water pressure difference.

To investigate the gene expression, localization and potential functions of Schistosoma japonicum chymotrypsin-like protease (SjCTRL) in the host, and evaluate its potential immune-protection efficacy against S. japonicum infection in mice.The physicochemical properties of SjCTRL and its phylogenetic relationship with homologous genes from other species were analyzed with bioinformatic software. The distribution of SjCTRL transcripts in 26-day-old worms was investigated using whole-mount in situ hybridization. The transcriptional levels of SjCTRL in male and female worms at four development stages (14, 18, 22, and 26 days after infection) were measured with quantitative real-time PCR. The SjCTRL-dsRNA was prepared and used to induce RNA interference (RNAi) in 26-day-old worms via soaking in vitro, and confocal microscopy was used to observe the morphological changes of worms after RNAi. Primers were designed to amplify the encoding sequence (excluding the transmembrane region) from the S. japonicum cDNA. The truncated gene was subcloned into the pET-28a plasmid, transformed into E. coli BL21 (DE3) for expression. Mice were immunized with the purified recombinant protein and challenged with cercariae. The worms and mouse liver were collected on day 35 after the challenge, and the worm-reduction rate and egg-reduction rate were calculated.The in situ hybridization results showed that SjCTRL mRNA was located in the posterior segment of intestinal tract of female worms, and having abundence only in 26-day-old female worms. After RNAi with SjCTRL-dsRNA, the mRNA expression was reduced to 25.7% (P0.05), without significant morphological changes. Using the recombinant plasmid pET-28a/SjCTRL, expression of insoluble SjCTRL protein was induced. Mice immunized with this protein gained a worm-reduction rate of 25.4% and an egg-reduction rate of 80.5% in liver after being challenged with cercariae.This study proves a high transcriptional level of SjCTRL in the posterior segment of intestinal tract in 26-day-old female worms, which can be reduced by RNAi treatment in vitro. Immunization with the SjCTRL protein can reduce adult worms and liver eggs.

Calcium and calmodulin-dependent protein kinase (CCaMK) is a member of calcium/calmodulin-dependent protein kinase superfamily and is essential to microbe- plant symbiosis. To date, the distribution of CCaMK gene in plants has not yet been completely understood, and its function in plant disease resistance remains unclear. In this study, we systemically identified the CCaMK genes in genomes of 44 plant species in Phytozome and analyzed the function of tomato CCaMK (SlCCaMK) in resistance to various pathogens. CCaMKs in 18 additional plant species were identified, yet the absence of CCaMK gene in green algae and cruciferous species was confirmed. Sequence analysis of full-length CCaMK proteins from 44 plant species demonstrated that plant CCaMKs are highly conserved across all domains. Most of the important regulatory amino acids are conserved throughout all sequences, with the only notable exception being observed in N-terminal autophosphorylation site corresponding to Ser 9 in the Medicago truncatula CCaMK. CCaMK gene structures are similar, mostly containing six introns with a phase profile of 200200 and the exception was only noticed at the first exons. Phylogenetic analysis demonstrated that CCaMK lineage is likely to have diverged early from a calcium-dependent protein kinase (CDPK) gene in the ancestor of all nonvascular plant species. The SlCCaMK gene was widely and differently responsive to diverse pathogenic stimuli. Furthermore, knock-down of SlCCaMK reduced tomato resistance to Sclerotinia sclerotiorum and Pseudomonas syringae pv. tomato (Pst) DC3000 and decreased H2O2 accumulation in response to Pst DC3000 inoculation. Our results reveal that SlCCaMK positively regulates disease resistance in tomato via promoting H2O2 accumulation. SlCCaMK is the first CCaMK gene proved to function in plant disease resistance.

Herein, a new electrochemiluminescence (ECL) strategy for enzyme-free microRNA-21 (miR-21) amplified detection was designed based on target-catalyzed hairpin assembly by combining the signal-amplification capability of both intramolecular and intermolecular ECL co-reaction. In this strategy, two hairpin DNA probes of H1 and H2 were designed as capture probes and detection probes, respectively. To be specific, the capture probes of H1 were immobilized on the multilayer interface of AuNPs and thiosemicarbazide (TSC) assembly on the single-walled carbon nanohorns decorated electrode, while the detection probes of H2 was anchored on the nanocarriers of gold nanoparticals functionalized reduced graphene oxide (Au-rGO) which were tagged with the self-enhanced ruthenium complex (PEI-Ru(ΙΙ)) in advance. Based on the target-catalyzed hairpin assembly, target miR-21 could trigger the hybridization of H1 and H2 to further be released for initiating the next hybridization process to capture a large number of H2 bioconjugates on the sensing surface. Herein, the TSC was used not only as a coupling reagent to attach the AuNPs via Au-S and Au-N bonds but also as a novel intermolecular coreactant to enhance the ECL intensity, and the PEI-Ru(ΙΙ) as emitters exhibited enhanced ECL efficiency. Therefore, a strong ECL signal was achieved by the dual amplification strategies of target recycle and the intramolecular/intermolecular co-reaction of PEI-Ru(ΙΙ) and TSC. The designed protocol provided an ultrasensitive ECL detection of miR-21 down to the sub-femtomolar level with a linear response about 6 orders of magnitude (from 1.0 × 10(-16)M to 1.0 × 10(-11)M) with a relatively low detection limit of 0.03 fM (S/N=3).

The cyclic nucleotide gated channel (CNGC) is suggested to be one of the important calcium conducting channels. Nevertheless, genome-wide identification and systemic functional analysis of CNGC gene family in crop plant species have not yet been conducted. In this study, we performed genome-wide identification of CNGC gene family in the economically important crop tomato (Solanum lycopersicum L.) and analyzed function of the group IVb SlCNGC genes in disease resistance. Eighteen CNGC genes were identified in tomato genome, and four CNGC loci that were misannotated at database were corrected by cloning and sequencing. Detailed bioinformatics analyses on gene structure, domain composition and phylogenetic relationship of the SlCNGC gene family were conducted and the group-specific feature was revealed. Comprehensive expression analyses demonstrated that SlCNGC genes were highly and widely responsive to diverse stimuli and the expression profile was gene-dependent. Pharmacological assays showed that the putative CNGC activators cGMP and cAMP enhanced resistance against Sclerotinia sclerotiorum. Silencing of group IVb SlCNGC genes significantly enhanced resistance to fungal pathogens Pythium aphanidermatum and S. sclerotiorum, strongly reduced resistance to viral pathogen Tobacco rattle virus, while attenuated PAMP- and DAMP-triggered immunity as shown by obvious decrease of the flg22- and AtPep1-elicited hydrogen peroxide accumulation in SlCNGC-silenced plants. Additionally, silencing of these SlCNGC genes significantly altered expression of a set of Ca2+ signaling genes including SlCaMs, SlCDPKs and SlCAMTA3. Collectively, our results reveal that group IV SlCNGC genes regulate a wide range of resistance in tomato probably by affecting Ca2+ signaling.

Calcium-dependent protein kinases (CDPKs) and CDPK-related kinases (CRKs) play multiple roles in plant. Nevertheless, genome-wide identification of these two families is limited to several plant species, and role of CRKs in disease resistance remains unclear. In this study, we identified the CDPK and CRK gene families in genome of the economically important crop tomato (Solanum lycopersicum L.) and analyzed their function in resistance to various pathogens. Twenty-nine CDPK and six CRK genes were identified in tomato genome. Both SlCDPK and SlCRK proteins harbored an STKc_CAMK type protein kinase domain, while only SlCDPKs contained EF-hand type Ca(2+) binding domain(s). Phylogenetic analysis revealed that plant CRK family diverged early from CDPKs, and shared a common ancestor gene with subgroup IV CDPKs. Subgroup IV SlCDPK proteins were basic and their genes contained 11 introns, which were distinguished from other subgroups but similar to CRKs. Subgroup I SlCDPKs generally did not carry an N-terminal myristoylation motif while those of the remaining subgroups and SlCRKs universally did. SlCDPK and SlCRK genes were differently responsive to pathogenic stimuli. Furthermore, silencing analyses demonstrated that SlCDPK18 and SlCDPK10 positively regulated nonhost resistance to Xanthomonas oryzae pv. oryzae and host resistance to Pseudomonas syringae pv. tomato (Pst) DC3000, respectively, while SlCRK6 positively regulated resistance to both Pst DC3000 and Sclerotinia sclerotiorum in tomato. In conclusion, CRKs apparently evolved from CDPK lineage, SlCDPK and SlCRK genes regulate a wide range of resistance and SlCRK6 is the first CRK gene proved to function in plant disease resistance.
