A total of 482 bats representing 32 species and two families were captured in the Amazon forests of the Amapá state in northern Brazil. Nineteen Artibeus planirostris bats (3.9 %) were infested with 160 ticks, all identified as Ornithodoros hasei. Three pools of larvae were screened for rickettsial DNA via polymerase chain reaction (PCR) targeting three rickettsial genes: gltA, ompA and htrA. Only one of them yielded an amplicons of the expected size for all three molecular assays. Comparisons of the obtained sequences including a phylogenetic analysis confirmed the occurrence of "Candidatus Rickettsia wissemanii" in Brazil.

The present study reports on patterns of parasitism by Amblyomma humerale on yellow-footed tortoise, Chelonoidis denticulatus, in the Atlantic Forest of southeastern Brazil. The tortoises were sampled occasionally, from July 2005 to January 2020. We collected 81 adult ticks from seven tortoises (mean = 11.57 ± 6.90 ticks/host), of which 72 were males (88.9%) and nine females (11.1%), resulting in an 8:1 sex ratio (males:females). Males were found in clusters, attached to the carapace. Females occurred mainly isolated, attached to coriaceous integumental areas (n = 7; 77.8%), especially the engorged females (100%). We suggest that the difference between attachment sites is a strategy used by fertilized females to avoid mechanical removal from their host through friction with elements of the environment. Because they undergo a considerable increase of volume when engorged, they move to more sheltered places on their host's surface. The parasitism patterns by A. humerale on C. denticulatus were compatible with those observed in the Amazon region and in the Cerrado-Amazonia transition. The present work provides information about the life history and host-parasite interface of the two species and contributes to understanding the parasitism patterns by A. humerale on C. denticulatus in the Atlantic Forest of southeastern Brazil.

Simple Summary Sarcocystidae is a family of apicomplexan protozoa highly prevalent in vertebrates. The definitive hosts of sarcocystids eliminate oocysts or sporocysts that infect intermediate hosts. After infection, mature tissue cysts (sarcocysts) develop in intermediate hosts, mostly in muscle and neurological tissues. Sarcocysts are infectious for definitive hosts, which acquire them through carnivorous or scavenging habits. Intermediate hosts and definitive hosts are the natural hosts of sarcocystids in which infections are usually mildly or not symptomatic. In 2017, muscular and neurological tissues of 22 birds from Magdalena Islands, southern coast of Chile, were screened for the presence of DNA of sarcocystids. DNA of organisms of the genus Sarcocystis was identified in two Chilean skuas (Stercorarius chilensis). The genetic makeup of the parasite detected in skuas was unprecedented and probably represent a new species in the genus. It is well known that Sarcocystis may cause severe infections in aberrant hosts, which are susceptible animals that do not behave as natural hosts for the parasite and have low resistance to the infection, thus more studies are needed to characterize this parasitosis in skuas and other hosts to understand the epidemiology of the infection and its impact on the health of marine fauna. Abstract Evidence of sarcocystid infection was investigated in samples of 16 penguins (Spheniscus. magellanicus), four Dominican gulls (Larus dominicanus) and two Chilean skuas (Stercorarius chilensis) found in Madalenas Islands, Chile, in 2017. Samples of skeletal muscle, cardiac muscle and brain from all birds were screened by a pan-sarcocystid nested-PCR targeting a short fragment of the gene encoding the small ribosomal unit (nPCR-18Sa). The only two positive samples by nPCR-18Sa, both from skuas, were tested by a nested-PCR directed to the internal transcribed spacer 1 (nPCR-ITS1), also a pan-sarcocystidae nested-PCR, and to a nested-PCR directed to the B1 gene (nPCR-B1), for the exclusive detection of Toxoplasma gondii. The two nPCR-18Sa-positive samples were nPCR-ITS1-positive and nPCR-B1-negative. The nPCR-ITS1 nucleotide sequences from the two skuas, which were identical to each other, were revealed closely related to homologous sequences of Sarcocystis halieti, species found in seabirds of northern hemisphere. Larger fragments of genes encoding 18S and partial sequences of genes coding for cytochrome oxidase subunit 1 were also analyzed, corroborating ITS1 data. The haplotypes found in the skuas are unprecedent and closely related to species that use birds as the definitive host. Further studies need to be carried out to detect, identify and isolate this parasite to understand the epidemiology of the infection and its impact on the health of marine fauna.

Birds are recognized hosts of ticks, especially for the immature stages which may harbor various species and strains of Rickettsia. To explore landscapes inhabited by birds and their ticks would expand the knowledge on host-parasite relationships and the rickettsiae. The aim of this paper was to record the diversity of ticks collected on wild birds and assess the phylogenetic position of a novel Rickettsia strain detected in immature ticks. Birds were captured in the Ibitipoca State Park, located in the Minas Gerais state, southeastern Brazil, as part of a long-term research project on the ecology of ticks, birds and Rickettsia. We found three tick species parasitizing birds: Amblyomma aureolatum (63 larvae, 10 nymphs), Haemaphysalis leporispalustris (28 larvae, seven nymphs) and Amblyomma romarioi (27 larvae). Among these, A. aureolatum was the most abundant species including 54% (73/135) of the collected ticks. New tick-host records were: A. romarioi on Turdus amaurochalinus and H. leporispalustris on Thamnophilus caerulescens, Saltator similis and Zonotrichia capensis. Of the 82 ticks tested for Rickettsia spp. by PCR, two larvae (2.5%) of A. romarioi were infected with 'Candidatus Rickettsia paranaensis', a novel putative Rickettsia species closely related to Rickettsia africae, Rickettsia sibirica and Rickettsia parkeri, as corroborated by our phylogenetic analysis. Finally, we present a list of all records of immature stages of H. leporispalustris on passerine birds in Brazil.

Ticks, along with fleas, are considered the main ectoparasites affecting dogs in Brazil. The literature is rich in reports on the occurrence of ticks, which are vectors of diseases in both dogs and humans. The study of the epidemiology of these arthropods provides important data that can help control the infestation in residential areas where dogs are present. Thus, the study of their occurrence in urban areas and veterinary services is very important. The aim of the present study was to determine the occurrence of ticks in dogs in a veterinary hospital population in the state of Espírito Santo, Brazil. Over a one-year period (July 2012 to July 2013), 1483 dogs brought to veterinary services in south-eastern Brazil were examined. Among the dogs examined, 8.97% presented tick infestation, of which 100% were identified as Rhipicephalus sanguineus sensu lato. Given that this species of tick acts as a vector for diseases, it is important that the official health services monitor the occurrence of infestation in dogs and humans, in order to identify diseases transmitted by these ticks in this region. RESUMO: Os carrapatos, juntamente com as pulgas, são considerados, no Brasil, como os principais ectoparasitas do cão. A literatura é rica em relatos considerando a ocorrência de carrapatos, que são vetores de doenças para cães e humanos. O estudo da epidemiologia de artrópodes é um dado importante que pode ajudar a controlar a infestação em áreas domiciliadas com a presença de cães. Nesse sentido, o estudo de sua ocorrência em áreas urbanas e ou mesmo em serviços médicos veterinários é importante. O objetivo do presente estudo foi verificar a ocorrência de carrapatos em cães em uma população hospitalar do estado de Espírito Santo, Brasil. Durante um ano (julho de 2012 a julho de 2013), foram examinados 1483 cães atendidos em serviços hospitalares no sudeste do Brasil. Entre os cães examinados, 8,97% apresentaram infestação de carrapatos, sendo 100% deles identificados como Rhipicephalus sanguineus sensu lato. Uma vez que se trata de uma espécie de carrapato que atua como vetor de doenças, é importante que os serviços oficiais de saúde mantenham um monitoramento da ocorrência de infestação tanto em cães como em humanos, a fim de identificar doenças transmitidas por esses carrapatos nesta região.

Background Brazilian spotted fever (BSF), caused by the bacterium Rickettsia rickettsii, has been associated with the transmission by the tick Amblyomma sculptum, and one of its main hosts, the capybara (Hydrochoerus hydrochaeris). Methods During 2015–2019, we captured capybaras and ticks in seven highly anthropic areas of São Paulo state (three endemic and four nonendemic for BSF) and in two natural areas of the Pantanal biome, all with established populations of capybaras. Results The BSF-endemic areas were characterized by much higher tick burdens on both capybaras and in the environment, when compared to the BSF-nonendemic areas. Only two tick species (A. sculptum and Amblyomma dubitatum) were found in the anthropic areas; however, with a great predominance of A. sculptum (≈90% of all ticks) in the endemic areas, in contrast to a slight predominance of A. dubitatum (≈60%) in the nonendemic areas. Tick species richness was higher in the natural areas, where six species were found, albeit with a predominance of A. sculptum (≈95% of all ticks) and environmental tick burdens much lower than in the anthropic areas. The BSF-endemic areas were characterized by overgrowth populations of A. sculptum that were sustained chiefly by capybaras, and decreased populations of A. dubitatum. In contrast, the BSF-nonendemic areas with landscape similar to the endemic areas differed by having lower tick burdens and a slight predominance of A. dubitatum over A.sculptum, both sustained chiefly by capybaras. While multiple medium- to large-sized mammals have been incriminated as important hosts for A. sculptum in the natural areas, the capybara was the only important host for this tick in the anthropic areas. Conclusions The uneven distribution of R. rickettsii infection among A. sculptum populations in highly anthropic areas of São Paulo state could be related to the tick population size and its proportion to sympatric A. dubitatum populations., Author summary Brazilian spotted fever (BSF), caused by the bacterium Rickettsia rickettsii, is the deadliest tick-borne disease of the New World. Since 2010, a new rickettsiosis caused by Rickettsia parkeri, a species included in the spotted fever group (SFG) and transmitted by the tick Amblyomma ovale, has been described in Brazil. Considering that A. ovale does occur in the Espirito Santo state (ES), the present study aimed at preliminarily describing the epidemiology of R. parkeri in the Atlantic forest of this state. Between June 2016 and September 2018, 33 villages from nine municipalities of the ES were included in a study for the molecular detection and isolation of SFG rickettsiae from adult Amblyomma ticks collected on dogs with free access to the forest. Serologic screenings against SFG rickettsiae in these animals and their owners (humans) were performed through immunofluorescence assay (IFA) using antigens of Rickettsia rickettsii and R. parkeri. Additionally, local health secretariats were informed on clinical manifestations of R. parkeri infection and told to communicate any suspected case. A total of 280 adult ticks were collected and taxonomically classified as A. ovale (n = 152), Amblyomma aureolatum (n = 127) and Amblyomma sculptum (n = 1). Overall, Rickettsia DNA was detected in 12/266 ticks. The sequencing of PCR products revealed that 0.7% (1/144) and 0.8% (1/121) of the analyzed A. ovale and A. aureolatum ticks were infected by R. parkeri strain Atlantic rainforest, respectively, and 8.3% (10/121) of the A. aureolatum ticks infected by Rickettsia bellii. Among the above PCR-positive ticks, only one isolate from one A. aureolatum tick was successfully established in the laboratory. DNA extracted from the third passage of this isolate was designated as strain M9A and molecularly characterized using primers targeting the Rickettsia gltA gene, whose sequence matched 100% the corresponding sequences of R. bellii. Seroprevalence against SFG rickettsiae in sampled dogs (n = 83) was 41% or 57%, depending on the rickettsial antigen (R. rickettsii strain Taiacu or R. parkeri strain Atlantic rainforest, respectively). A total of 37 (45%) canine sera showed titers to R. parkeri at least fourfold higher than to R. rickettsii antigen. Among humans, 10% (4/41) of the samples reacted to at least one rickettsial antigen, with the highest endpoint titer varying from 64 to 128 for R. rickettsii and R. parkeri; no human serum showed ≥4-fold difference between the highest endpoint titers. Finally, during the study period, suspicions on cases of R. parkeri-rickettsiosis were not informed by the health secretariats. Our results confirm the presence and exposure to R. parkeri strain Atlantic rainforest, associated with two anthropophilic tick species (A. ovale and A. aureolatum) parasitizing domestic dogs with unrestrained access to forest areas. Consequently, the occurrence of R. parkeri infection in humans inhabiting the Atlantic forests of ES should not be discarded.

This study evaluated ticks and rickettsial exposure in 220 free-ranging lowland tapirs, Tapirus terrestris, from 2006 to 2018 in selected areas of three major biomes of Brazil - Atlantic Forest, Pantanal, and Cerrado. Overall, a total of 5970 tick specimens representing the following nine species were collected from tapirs: Amblyomma brasiliense, Amblyomma coelebs, Amblyomma dubitatum, Amblyomma ovale, Amblyomma parvum, Amblyomma sculptum, Amblyomma triste, Haemaphysalis juxtakochi, and Rhipicephalus microplus. Amblyomma sculptum was the most prevalent and abundant tick species in all three biomes; however, mean intensity values for A. sculptum were significantly lower in Atlantic Forest than in the Pantanal or Cerrado, and at the same time, statistically similar among tapirs from Pantanal and Cerrado. Contrastingly, mean intensity values for A. coelebs were significantly higher in the Atlantic Forest than in the other biomes. The remaining tick species were collected in lower numbers, or were exclusive for one biome, e.g., A. brasiliense and H. juxtakochi only in the Atlantic Forest. A total of 177 blood sera (123 individuals plus 54 recaptures) were collected from tapirs and tested for the presence of reactive antibodies to six Rickettsia species by immunofluorescence assay. Overall, 69% (9/13), 49% (62/126), and 66% (25/38) tapir sera from Atlantic Forest, Pantanal and Cerrado, respectively, were seroreactive to at least one Rickettsia species, with no significant difference between the three areas. Although many tapir sera reacted simultaneously to ≥2 Rickettsia species, Rickettsia parkeri elicited highest % seroprevalence and endpoint titers, and was incriminated as the possible agent involved in a homologous reaction in tapirs from the three biomes, where A. ovale was previously found infected by R. parkeri. In fact, seroconversion to R. parkeri was demonstrated in five tapirs that were captured at least twice during the study. This study demonstrated that tapirs were found to be constantly infested by several tick species in the Atlantic Forest, Pantanal and Cerrado biomes; however, the richness of tick infestations was concordant to the tick species known to be established in each biome. Under natural conditions, lowland tapirs were shown to be exposed to tick-borne spotted fever group rickettsiae.

Sheep are intermediate hosts of the protozoon Toxoplasma gondii, a zoonotic parasite that that may cause abortion and reproductive losses in this species. Previous studies have shown that its prevalence is high in sheep herds in Brazil, but no information from the state of Espirito Santo was available. The aim of this study was to improve the epidemiological data in different regions of this state by analyzing occurrences of anti-T. gondii antibodies in young female sheep, before reproductive age, using IFAT (cutoff ≥64). In addition, variables relating to these animals and farm management were investigated for significance of associations. A total of 312 sheep, aged 3 to 12 months, on five farms in five municipalities, were sampled. Among these 312 animals, 135 (43.26%) were reactive, and 70 positive sheep (51.8%) presented high titers (more than three times the cutoff).Eight-month-old sheep presented the lowest occurrence and 7 and 11-month-old sheep, the highest. At least one animal was positive in each municipality, thus confirming the endemic status of toxoplasmosis in sheep in this region. None of the variables analyzed, relating to animals and herds, presented any association with T. gondii occurrence (p > 0.05). The number of females that were close to sexual maturity (12 months) and which were reactive to T. gondii was less than 50%. These ewes would probably be more susceptible to reproductive problems.

The aim of this study was to determine the presence of antibodies to Neospora caninum and Toxoplasma gondii in serum samples of 187 dogs from 30 rural properties surrounding Córrego do Veado Biological Reserve, Espírito Santo State, Brazil. The Reserve is one of the last lowland Atlantic Forest remnants of the region, surrounded by agriculture farms and cattle pastures. The presence of IgG antibodies was determined by the Indirect Fluorescent Antibody Test for T. gondii (cut-off 1:16) and N. caninum (cut-off 1:50). Positive samples were diluted 2-fold until the last positive dilution. Antibodies to T. gondii were found in 77 (47.05%) dogs and antibodies to N. caninum in 22 dogs (11.76%) and one only dog was positive for both infections. No association between T. gondii and N. caninum infection and sex was observed (p>0.05). Control measures to prevent those infections in dogs that living surrounding the reserve and that had contact with wild animals are important to avoid the introduction of N. caninum in wild animals. This was the first study of frequency of occurrence of T. gondii and N. caninum in dogs from Espírito Santo, Brazil. Resumo O objetivo deste estudo foi determinar a presença de anticorpos contra Neospora caninum e Toxoplasma gondii em amostras de soro de 187 cães de 30 propriedades rurais do entorno da Reserva Biológica Córrego do Veado, Espírito Santo, Brasil. A Reserva é um dos últimos remanescentes de Mata Atlântica da região, cercada por fazendas com agricultura e pastagens. A presença de anticorpos IgG foi determinada pela reação de imunofluorescência indireta para T. gondii (ponto de corte de 1:16) e para N. caninum (ponto de corte de 1:50). As amostras positivas foram diluídas na base dois e tituladas. Anticorpos para T. gondii foram encontrados em 77 (47,05%) cães e para N. caninum em 22 (11,76%). Um cão foi positivo para ambas as infecções. Não houve associação entre a infecção pelo T. gondii e N. caninum e o sexo (p>0,05). Medidas de controle, para prevenir essas infecções em cães que vivem no entorno da reserva e com possibilidade de contato com animais selvagens, são importantes para evitar a introdução de N. caninum nos animais selvagens. Este foi o primeiro relato da frequência de anticorpos contra estes coccídios em cães do Espírito Santo, Brasil.

Among the 69 tick species currently recognized in Brazil, only 13 have been reported in the state of Espírito Santo, southeastern Brazil. This study reports new tick records from Espírito Santo, and investigated the infection by rickettsial bacteria in these ticks. During 2012-2016, ticks were occasionally collected from different sources in Espírito Santo. In addition, we revised tick unpublished records from Espírito Santo in a tick collection. A total of 1263 tick specimens comprising 19 tick species (17 Ixodidae, 2 Argasidae) were collected. The following seven tick species are reported for the first time in Espírito Santo: Amblyomma aureolatum, Amblyomma coelebs, Amblyomma ovale, Amblyomma rotundatum, Amblyomma varium, Haemaphysalis juxtakochi, and Ornithodoros hasei. The only tick species previously reported for Espírito Santo, but not found in the present study, is A. longirostre. A total of 194 ticks of 7 species were tested for rickettsial infection. Three Rickettsia species were detected: Rickettsia bellii in A. aureolatum, Rickettsia rhipicephali in H. juxtakochi, and Rickettsia amblyommii in A. humerale. The present study increases the tick fauna of Espírito Santo to 20 species (29% of the Brazilian tick fauna). Additionally, 3 Rickettsia species are reported for the first time in this region of Brazil.

We evaluated infection by Rickettsia spp. and Ehrlichia spp in small mammals and their ticks from two Atlantic forest conservation areas in the state of Rio Grande do Norte, northeastern Brazil. A total of 39 small mammals were captured during 2012–2013, encompassing 33 marsupials (29 Didelphis albiventris, four Monodelphis domestica), three Cricetidae rodents (two Necromys lasiurus, one Rattus rattus), one Caviomorpha rodent (Thrichomys apereoides) and two armadillos (Euphractus sexcinctus). The ticks Amblyomma auricularium, Ixodes loricatus, and Ornithodoros mimon were collected from D. albiventris, whereas only A. auricularium was collected from armadillos. Through immunofluorescence assay with Rickettsia spp. antigens, 6/28 (21%) D. albiventris and the single R. rattus specimen reacted to at least one rickettsial antigen, with highest seroprevalence and endpoint titers to Rickettsia amblyommatis. A total of 150 ticks (126 A. auricularium, nine I. loricatus, 15 O. mimon) was tested for rickettsial infection by PCR, which detected only R. amblyommatis in most of the A. auricularium ticks. Lung and spleen samples were collected from small mammals (two N. lasiurus, six D. albiventris, three M. domestica, one T. apereoides, one R. rattus) and were tested by PCR for Anaplasmataceae agents. The spleen from one D. albiventris contained a new ehrlichial agent, here named as Ehrlichia sp. strain Natal. Phylogenetic analysis inferred from the dsb gene of Ehrlichia spp. indicates that this novel agent is potentially a new species. Future studies should monitor the possible role of rickettsial and/or ehrlichial microorganisms as agents of emerging diseases in these degraded areas of Atlantic forest, just as has occurred with other agents in degraded areas of this biome in southeastern Brazil., Graphical abstract Image 1, Highlights • Amblyomma auricularium, Ixodes loricatus and Ornithodoros mimon ticks were found. • Infection by Rickettsia amblyommatis was detected in A. auricularium ticks. • Wild small mammals presented serological evidence of R. amblyommatis infection. • A potentially novel Ehrlichia species infected the opossum Didelphis albiventris.

Espírito Santo state (southeastern Brazil) is considered an endemic area for spotted fever group rickettsioses. In February 2017, we received in our laboratory seven unfed Amblyomma ovale adult ticks collected by a farmer from his clothes and body (not attached) during a working day in the rural area of Ibiraçu municipality, Espírito Santo state. By polymerase chain reaction (PCR) analyses, targeting gltA and ompA rickettsial genes, the DNA of Rickettsia was detected in 6/7 (85.7%) A. ovale. In all cases, DNA sequencing of PCR products revealed that consensus sequences of both genes were 100% identical to gltA and ompA corresponding sequences of Rickettsia sp. strain Atlantic rainforest retrieved from GenBank. This study reports the first molecular detection of Rickettsia sp. strain Atlantic rainforest in A. ovale ticks from Espírito Santo state. Our findings indicate a new Brazilian state in the southeast region at risk of human infection with this tick-borne emerging rickettsial agent.

Toxoplasmosis has been reported in many avian species, but little information is available from wild penguin populations. Leptospira can infects domestic and wild animals. Spheniscus magellanicus belong to the order Sphenisciformes, family Spheniscidae, and are colonial birds. These seabirds live in temperate waters along the Atlantic shores of South America, and their total population has been estimated to be 1,300,000 breeding pairs. Magdalena Island (Chile) hosts an important breeding colony but, over recent decades, a marked decline in the number of birds has been seen. The objective of this study was to determine occurrences of antibodies against T. gondii and Leptospira spp. in penguins (Spheniscus magellanicus) on Magdalena Island, from where no previous data on these agents were available. Serum samples were collected from 132 penguins on Magdalena Island. Antibodies against Toxoplasma gondii were detected using the modified agglutination test (Titer ≥20), and anti-Leptospira spp. antibodies were detected using the microscopic agglutination test (Titer ≥100). T. gondii antibodies were detected in 57 (43.18%) of the 132 serum samples, with titers that ranged from 20 to 320. None of the penguins in this study was reactive to anti-Leptospira spp. antibodies. This is the first report of T. gondii seropositivity in free-living Magellanic penguins in Chile.

This study evaluated rickettsial infection in ticks collected on toads from the northern Brazilian Amazon (Amapa state), where to our knowledge there are neither records of ticks from amphibians nor rickettsial infections in ticks. During 2016–2017, a total of 22 free-living toads were captured and identified as Rhinella marina. Overall, 12 (54.5%) toads were parasitized by a total of 97 ticks (6 males, 39 females, 31 nymphs, 21 larvae), giving a mean intensity of 8.1 ticks per infested toad. Two tick species were morphologically identified: Amblyomma rotundatum Koch (31 females, 14 nymphs), and Amblyomma dissimile Koch (6 males, 8 females, 17 nymphs). The 21 larvae were morphologically denoted as Amblyomma sp. Five toads were co-infested by A. rotundatum and A. dissimile. Morphological identifications were confirmed by nucleotide sequencing of fragments of the mitochondrial genes 16S rDNA, 12S rDNA and/or COX1. A total of 54 ticks were analyzed for the presence of rickettsial DNA. Eleven (9 females and 2 nymphs) out of 14 A. rotundatum ticks contained Rickettsia bellii. None of the 25 specimens of A. dissimile (6 males, 6 females, 13 nymphs) contained amplifiable rickettsial DNA. From 15 Amblyomma sp. larvae, a pool of 10 individuals contained Rickettsia sp. strain Colombianensi. Sequencing of the 16S rDNA amplicon derived from the positive pool yielded a sequence of A. dissimile. We detected Rickettsia sp. strain Colombianensi for the first time in Brazil. Prior records of this agent were restricted to Colombia and Honduras. In addition, we report the presence of A. rotundatum for the first time in the state of Amapa, where the only other record of A. dissimile was registered over 20 years ago.

The main motivation for this study was to determine the occurrence of Toxoplasma gondii, a cosmopolitan widespread zoonotic parasite distribution that can infect a wide variety of mammals and birds, in Magellanic penguins (Spheniscus magellanicus) in Brazil. In recent decades there has been a significant increase in the number of penguins originating from Argentinian and Chilean Patagonia, where these birds are born, that arrive on the Brazilian coast, where many of them are stranded and rescued. Tissue samples were collected from 330 individuals surveyed from 2012–2015 at the Institute for Marine Animal Research and Rehabilitation (IPRAM) located in Cariacica, state of Espirito Santo, Brazil. Serum were collected from 145 animals surveyed in 2015 for the detection of anti-T. gondii antibodies using the Modified Agglutination Test (MAT ≥20) and 18 birds were positive, with titers of 20 (7 birds), 40 (9 birds) and 80 (2 birds). Mouse bioassay for the isolation of T. gondii was performed using tissues from 54 penguins that were also surveyed in 2015, but no isolates were obtained. DNA from tissue samples of 330 individuals was PCR amplified and sequenced to detect tissue cyst forming coccidians by using pan sarcocystids-directed primers (based on 18S rDNA). These samples were from animals surveyed in 2015 and from frozen stocked tissues from animals surveyed in the years 2012 and 2013. The positives were PCR amplified and sequenced with genus Sarcocystis-specific primers (based on internal transcribed spacer 1, RNA polymerase beta subunit coding gene, and cytochrome B coding gene) and with Sarcocystis falcatula/Sarcocystis neurona- specific primers (based on surface antigens SAG2, SAG3 and SAG4). Sixteen (3.0%) of pectoral muscle samples were positive by all the seven molecular markers and all the samples were identical to each other. Organisms close related to Sarcocystis falcatula were confirmed in all cases. This is the first report on molecular detection of infection by S. falcatula-related organisms and the first report of seropositivity for T. gondii in free-living Magellanic penguins in Brazil. Felids and didephid opossums are definitive hosts of T. gondii and S. falcatula, respectively. Where the penguins acquire the infective forms of the parasites shed by the terrestrial mammals remains to be elucidated.

This work aims to identify and quantify the percentage of Rickettsia spp., Ehrlichia spp., Babesia spp. and Hepatozoon spp. positive pet dogs, and to identify ticks collected on these animals in the state of Espírito Santo, in the Southeast region of Brazil. The study included 378 dogs, 226 females and 152 males, of various breeds and ages (mean age of 4.1 years). All animals were examined for ticks, and whole blood was collected and processed by conventional PCR protocols for Babesia spp., Anaplasmataceae, Hepatozoon spp. and by real-time PCR for Ehrlichia canis. Of the 378 dogs examined, 157 (41.53%) had ticks at the time of the study, which were identified as R. sanguineus s.l. in 154 animals (98.10%), Amblyomma ovale in one animal (0.63%), and Amblyomma sculptum in 2 animals (1.27%). In the PCR for Babesia spp., 5 animals (1.32%) were positive, producing DNA sequences 100% identical to Babesia vogeli. For Anaplasmataceae, 34 animals (9%) were positive, 10 of which generated DNA sequences 100% similar to Ehrlichia canis. The other 24 samples generated fragments 100% identical to Anaplasma platys. In the PCR for Hepatozoon spp, 39 animals (10.31%) were positive, producing sequences 100% identical to Hepatozoon canis. Finally, in the real-time PCR specific for E. canis, 28 animals (7.40%) were positive. Coinfection with 2 or 3 agents was observed in 20 animals (5.29%). Of the 378 dogs sampled, 312 were analyzed by indirect immunofluorescence assay (IFA) for E. canis and five species of Rickettsia (R. rickettsii, R. parkeri, R. amblyommatis, R. rhipicephali and R. bellii). Among them, 71 dogs (22.75%) had a positive reaction for E. canis and 16 dogs (5.13%) had antibody titers higher than 64 to at least one Rickettsia species, 5 of them (1.60%) to R. rickettsii. Samples of blood were collected from 10 equines in the regions where dogs were found with positive serology for any one of the Rickettsia sp. tested. In the municipality of Vila Velha, two equines were positive for R. bellii with inverse titers of 64 and 128. These results indicate that tick-transmitted agents, such as E. canis, A. platys, B. vogeli, H. canis, and several spotted fever group Rickettsia are circulating in the canine population of the Brazilian state of Espirito Santo.

Magellanic penguins (Spheniscus magellanicus) breed on the Atlantic and Pacific coasts of the southernmost parts of South America and migrate northward as far as Peru and Brazil. Serum samples (n = 100) from Magellanic penguins from three zoos and two rehabilitation centers (RCs) in Brazil were assayed for the presence of antibodies to Toxoplasma gondii by means of the modified agglutination test (MAT, cut-off ≥ 20). The penguins were categorized as young (≤4 yr old) or adults (≥4 yr old) and sexed (male, female, or not identified), and data were analyzed using the chi-square test (P ≤ 0.05). Toxoplasma gondii antibodies were found in 28% of penguins: 25.8% males, 27.8% females, 30.3% unknown sex, 25.4% young, and 31.1% adults. Statistical analyses did not find any difference (P > 0.05) with respect to age, sex, or source of birds. This is the first report of T. gondii antibodies in S. magellanicus.

Although Espirito Santo state is considered an endemic area for Brazilian spotted fever (BSF) with related lethal cases, it also constitutes the only state of southeastern Brazil that currently lacks a specific confirmation of the specific rickettsial agent. In an attempt to a species level confirmation of the etiological agent of fatal rickettsiosis cases in Espirito Santo state, in this study we tested human sera obtained between 2015 to 2017 by means of qPCR and subsequent conventional PCR protocols targeting gltA (citrate synthase) and ompA (190-kDA outer membrane protein) rickettsial genes. All samples were found to contain rickettsial DNA through the citrate synthase qPCR protocol. By conventional PCR, rickettsial gltA and ompA specific DNA fragments were detected in 25% (one sample) and 50% (2 samples) of the screened sera, respectively. Obtained consensuses for each gene partial sequences were 100% identical to Rickettsia rickettsii gltA and ompA genes. The present study confirms for the first time R. rickettsii as the etiological agent of a lethal spotted fever group rickettsiosis in human patients from Espirito Santo state.

O gênero Sarcocystis é constituído por várias espécies que se diferenciam pelas características morfológicas, biológicas e moleculares. Foram relatadas mais de 196 espécies encontradas em mamíferos, aves e répteis e somente 26 dessas espécies possuem o ciclo completo conhecido. Toxoplasma gondii é um parasito intracelular obrigatório, com distribuição geográfica cosmopolita, capaz de infectar uma ampla variedade de mamíferos e aves, inclusive o homem, caracterizando seu potencial zoonótico. Nas últimas décadas, a quantidade de pinguins vindos da Patagônia argentina e chilena, região de nascimento dessas aves, para o litoral brasileiro, onde muitos encalham e são resgatados, tem aumentado significativamente. Pouco se sabe sobre as doenças causadas por protozoários nessas aves. O presente estudo teve como objetivo conhecer aspectos epidemiológicos da infecção por coccídios da família Sarcocystidae em pinguins-de-magalhães (Spheniscus magellanicus), através de análises moleculares e sorológicas. Foram realizadas duas campanhas, uma em 2014 e outra em 2015, com a finalidade de obter amostras de sangue e tecidos dos pinguins que vieram a óbito durante reabilitação no Instituto de Pesquisa e Reabilitação de Animais Marinhos (IPRAM) localizada em Cariacica, Espírito Santo. Foram colhidas 514 amostras de tecidos (músculo=342, coração=86, cérebro=86) de 310 indivíduos. Dos tecidos de 54 pinguins foi realizado o bioensaio em camundongos para o isolamento de T. gondii, mas nenhum isolado foi obtido. Amostras de 310 indivíduos tiveram o DNA extraído para a pesquisa de coccídios da família Sarcocystidae utilizando-se os marcadores 18S rDNA, espaçador interno transcrito 1 (ITS1), codificador de proteínas de superfície (SAG)2, SAG3 e SAG4, subunidade beta da RNA polimerase (RPOB) e citocromo B (CytB). Destas, 16 (3.0%) amostras de músculo peitoral foram positivas para o gênero Sarcocystis spp., quando analisadas pelo marcador 18S, e todas com resultados idênticos. Com o ITS1, RPOB e Ctv. foram confirmadas as espécies de Sarcocystis em 12 amostras, todas idênticas a S. falcatula-like. Com os marcadores SAGs foi possível observar que as sequências não tinham variabilidade genética. Das 145 amostras de soro avaliadas para a presença de anticorpos anti-T. gondii, pelo Teste de Aglutinação Modificado (MAT ≥20), 18 aves foram positivas com títulos de: 20 (7 aves), 40 (9 aves) e 80 (2 aves). Este é o primeiro relato de S. falcatula-like e de anticorpos anti - T. gondii em pinguins-de-magalhães de vida livre. The genus Sarcocystis is composed of several species that are differentiated by the morphological, biological and molecular characteristics. More than 196 species found in mammals, birds and reptiles have been reported, and only 26 of these species have the complete known cycle. Toxoplasma gondii is an obligate intracellular parasite with cosmopolitan geographic distribution, capable of infecting a wide variety of mammals and birds, including man, characterizing its zoonotic potential. In recent decades, the number of penguins that have come from Argentine and Chilean Patagonia, the region of birth of these birds, to the Brazilian coast, where many of them are stranded and rescued, has increased significantly. Little is known about the diseases caused by protozoa in these birds. The present study had as objective to study epidemiological aspects of coccidia infection of the family Sarcocystidae in Magellanic Penguins (Spheniscus magellanicus), through molecular and serological analyzes. Two campaigns were carried out, one in 2014 and another in 2015, in order to obtain blood and tissue samples from penguins who died during rehabilitation at the Institute of Research and Rehabilitation of Marine Animals (IPRAM) in the municipality of Cariacica, Espírito Santo. Tissue samples (total = 514: muscle = 342, heart = 86, brain = 86) were collected from 310 birds. From the tissues of 54 penguins the mouse bioassay was performed for the isolation of T. gondii, but no isolates were obtained. Samples of 310 individuals had DNA extracted for coccidia from the Sarcocystidae family using the 18S rDNA, Transcribed internal spacer 1(ITS1), surface protein encoder (SAG)2, SAG3, SAG4, beta subunit of RNA polymerase (RPOB) and citocrome B (CytB) markers. Of these, 16 (3.0%) samples, of pectoral muscle, were positive and all were identical to Sarcocystis spp. when analyzed by the 18S marker. With ITS1, RPOB and CytB the Sarcocystis species were confirmed in 12 samples, all identical to S. falcatula-like. With the SAGs it was possible to observe that the sequences had no genetic variability. Of the 145 serum samples evaluated for the presence of anti-T. gondii antibodies by Modified Agglutination Test (MAT ≥20), 18 were positive with titers of: 20 (7 birds), 40 (9 birds) and 80 (2 birds). This is the first report of S. falcatula-like and the presence of antibodies to T. gondii in free-living magellanic penguins.

In this study, we present a morphological description of immature and adult specimens of Ornithodoros saraivai n. sp., a tick associated with the frog Cycloramphus boraceiensis (Cycloramphidae) at Sao Sebastiao island, located in the Sao Paulo state seaboard, Brazil. While larvae of O. saraivai are ecologically related to Ornithodoros faccinii, another soft tick associated with cycloramphids, the combination of 7 sternal pairs, 16 dorsal pairs, a pyriform dorsal plate and a partially toothed hypostome constitute unique characters of the O. saraivai larvae. One undetermined nymphal instar and adults of O. saraivai are similar to mature specimens of the Ornithodoros talaje species group; however, the O. saraivai specimens can be recognized by the presence of a robust bean-shaped spiracle with a large spiracular plate and more than two long seta in palpal article I. Identical partial sequences of the mitochondrial 16S rDNA gene confirmed the identity for all collected stages and for two cohorts of laboratory-reared larvae of O. saraivai. A Bayesian and Maximum Parsimony inferred phylogenetic trees support the position of O. saraivai in a clade with O. faccinii, suggesting the existence of an Ornithodoros lineage that evolved in association with amphibians.

Human parasitism by the brown dog tick Rhipicephalus sanguineus sensu lato (s. l.), an important parasite in medical and veterinary sciences, is only rarely reported in the American continent. The present investigation reports a R. sanguineus s. l. male tick parasitizing a human in the city of Campo Grande, State of Mato Grosso do Sul, west-central Brazil. This observation is of public health relevance, since R. sanguineus s. l. ticks are known as vectors of spotted fever group rickettsiae to dogs and humans. O parasitismo humano pelo carrapato marrom do cão, Rhipicephalus sanguineus sensu lato (s. l.), um importante parasita para a saúde pública e veterinária, é raramente relatado no continente americano. Este trabalho relata o registro de um macho de R. sanguineus s. l. parasitando um humano na cidade de Campo Grande, estado do Mato Grosso do Sul, no Centro-Oeste do Brasil. Essa observação é relevante para a saúde pública, uma vez que os carrapatos desse complexo são conhecidos como vetores de riquétsias do grupo da febre maculosa para cães e humanos.

Ehrlichia spp. are obligatory intracellular microorganisms that infect hematopoietic, endothelial or blood cells of mammals. Ticks are the only vectors of these agents in nature. To date, the role of birds and their associated ticks as reservoirs of ehrlichiae remains almost unexplored. In this study, we performed a molecular screening for bacteria of Anaplasmataceae family in samples of spleen (n = 72) and lung (n = 17), recovered from 72 carcasses of Magellanic penguins (Spheniscus magellanicus) in Brazil and Chile. One apparently unengorged tick (Ixodes uriae) was also collected while wandering upon one of the carcasses and submitted to molecular analyses as well. Through conventional and nested PCR protocols three genes (16S rRNA, dsb and groEL) of a new Ehrlichia sp. were partially characterized upon organs of three penguins and in the tick coming from Magdalena Island (Chile). First matches after BLASTn comparisons showed that our sequences share 99.4% (16S rRNA), 94.6% (groEL) and 79.3% (dsb) of identity with “Candidatus Ehrlichia ornithorhynchi”, Ehrlichia sp. NS101 and Ehrlichia canis CCZ, respectively. Matrixes of genetic distance including other representatives of the Ehrlichia genus point a 99.4%, 94.0%, and 80.0% of identity with 16S rRNA, groEL and dsb genes from Ehrlichia sp. It25, Ehrlichia sp. NS101, and Ehrlichia chaffeensis San Louis, respectively. A Bayesian phylogenetic analysis of Anaplasmataceae 16S rRNA gene places the detected Ehrlichia sp. into a group with Ehrlichia sp. BAT and Ehrlichia sp. Natal. Although depicting different topologies, Bayesian unrooted phylogenetic trees constructed for groEL and dsb genes position this Ehrlichia sp. into well-supported branches, which reinforces the finding of a new taxon. For the moment, any pathogenic effect of this new Ehrlichia sp. on penguins is still unknown. However, this fact becomes important to assess from a conservation point of view since populations of Magellanic penguins are currently threatened and in an ongoing decrease.

As espécies do gênero Trypanosoma parasitam vertebrados de todas as classes (peixes, anfíbios, répteis, aves e mamíferos) e as espécies do gênero Leishmania parasitam mamíferos no Velho e Novo Mundo. Possuem ciclos de vida com alternância entre vertebrados e invertebrados. A maioria das espécies se desenvolve em artrópodes hematófagos que podem pertencer a diversas ordens e famílias. Os tripanossomas circulam no ambiente silvestre como enzootias, associados com os hospedeiros e seus respectivos ecótopos. A Leishmaniose visceral é uma importante zoonose e possui canídeos silvestres e domésticos como importantes reservatórios conhecidos. Estudos realizados com algumas espécies de tripanossomas apontam uma grande complexidade do ciclo silvestre em biomas brasileiros, incluindo a Mata Atlântica e a diversidade genética de L. (L.) infantum chagasi no Brasil ainda não é conhecida. Ressalta-se o fato que existem poucos trabalhos realizados no Estado do Espírito Santo. Até o momento, não foram avaliados os pequenos mamíferos terrestres, grandes mamíferos (Tapirus terrestris) e morcegos como reservatórios silvestres destes parasitas neste Estado. O presente estudo tem por objetivo principal, o conhecimento da diversidade de parasitas do gênero Leishmania e Trypanosoma em animais silvestres da Reserva Biológica Córrego do Veado e domésticos do seu entorno na cidade de Pinheiros no estado do Espírito Santo através do isolamento, caracterização molecular e estudos filogenéticos com marcadores tradicionais utilizados para o grupo. Foram realizadas duas campanhas de captura nos meses de junho e novembro de 2012 totalizando um esforço de 28800 armadilhas/dia para os pequenos mamíferos terrestres e 12600 h/m2 com um total de 157 indivíduos, pertencentes a cinco ordens distintas, 23 gêneros e 27 espécies. Do total de animais capturados 18 (11.46%) apresentaram hemocultura positivas e 11 (7.01%) culturas foram estabelecidas e criopreservadas com morfologia compatível a parasitas do gênero Trypanosoma. Os isolados foram identificados e posicionados através de sequências da região V7V8 SSU rDNA e um isolados de T. cruzi marinkellei e oito de T. dionisii foram detectados em hospedeiros quirópteros e dois isolados de Tapirus terrestris apresentaram morfologia distinta das já descritas para o gênero Trypanosoma e a caracterização parcial através do gene 18S rDNA completo e gGAPDH apontam para uma nova espécie deste gênero e tratam-se dos primeiros isolados obtidos nestes hospedeiros. Nos cães, foram obtidas cultura de linfonodo poplíteo bilateral e todos animais foram negativos. Além disso, a análise sorológica também foi negativa para todos os animais domésticos amostrados no entorno. The species of the genus Trypanosoma parasites of all vertebrate classes (fish, amphibians, reptiles, birds and mammals) and the genus Leishmania parasites mammals in the New World and have life cycles with alternating between vertebrates and invertebrates. Most species develops in arthropod vectors, which may belong to different orders and families. Trypanosomes circulating in sylvatic enzootic diseases as associated with the hosts and their ecotopes. Most species are not pathogenic, Visceral Leishmaniasis is an important zoonosis and has wild and domestic canids as important reservoirs known. Studies in some species of trypanosomes show a great complexity of the sylvatic cycle in biomes, including the Atlantic and genetic diversity of L. (L.) infantum chagasi in Brazil is not known. We emphasize the fact that there are few studies in the State of Espírito Santo. So far, have not been evaluated small terrestrial mammals, large mammals (Tapirus terrestris) and wild bats as reservoirs of these parasites in this state. This study\'s main objective, knowledge of the diversity of parasites of the genus Leishmania and Trypanosoma in wild Biological Reserve Stream Deer and domestic of your surroundings in the Pinheiros city in the state of Espírito Santo through isolation, characterization and molecular studies with traditional phylogenetic markers used for the group. There were two campaigns capture in June and November 2012 totaling an effort of 28,800 trap / day for small terrestrial mammals and h.m2 12600 with a total of 157 individuals belonging to five distinct orders, 23 genera and 27 species. Of the total of 18 animals captured (11:46%) had positive blood cultures and 11 (7.1%) cultures were established and cryopreserved with morphology the parasites of the genus Trypanosoma. Isolates were identified and positioned through V7V8 region sequences of SSU rDNA and one isolate of T. cruzi marinkellei and eight T. dionisii hosts were detected in bats and two isolates showed a distinct morphology Tapirus terrestris those already described for the genus Trypanosoma and partial characterization by 18S rDNA gene gGAPDH complete and point to a new species of this genus and these are the first isolates from these hosts. In domestic animals, dogs were obtained culture of popliteal lymph node of both hind legs and all animals were negative. Furthermore, serological analysis was also negative for all dogs sampled in the vicinity of the Biological Reserve of Deer Creek.

O Zoologico Municipal de Guarulhos, localizado no estado de Sao Paulo, recebe e recupera animais silvestres do proprio municipio e de alguns municipios limitrofes. Entre os anos de 1997 e 2016 foram coletados carrapatos de animais silvestres (Cariama cristata, Buteo brachyurus, Alouatta guariba, Bradypus variegatus, Tamandua tetradactyla, Puma concolor, Cerdocyon thous, Nasua nasua, Mazama gouazoubira, Didelphis aurita, Sphiggurus villosus, Myocastor coypus e Hydrochoerus hydrochaeris) atendidos pelo Zoologico. Foram identificadas as seguintes especies de carrapatos: Amblyomma aureolatum (Pallas, 1772), Amblyomma calcaratum Neumann, 1899, Amblyomma dubitatum Neumann, 1899, Amblyomma longirostre (Koch, 1844), Amblyomma nodosum Neumann, 1899, Amblyomma parkeri Fonseca e Aragao, 1952, Amblyomma sculptum Berlese, 1888, Amblyomma varium Koch, 1844, Haemaphysalis juxtakochi Cooley, 1946, Ixodes loricatus Neumann, 1899 e Rhipicephalus sanguineus sensu lato (Latreille, 1806). Apesar de todas as associacoes carrapato-hospedeiro ja terem sido previamente relatadas no territorio brasileiro, o presente estudo registra pela primeira vez um ginandromorfo de A. dubitatum.

A toxoplasmose é uma doença de importância zoonótica causada pelo protozoário Toxoplasma gondii. As duas principais vias de transmissão de T. gondii para o homem são a ingestão de oocistos esporulados e a ingestão de carnes cruas ou malcozidas que contenham cistos teciduais viáveis do agente. O processo de maturação úmida poder ser um meio de controle da toxoplasmose adquirida por ingestão de carnes. O objetivo deste estudo foi o refinamento do processo de maturação úmida na inviabilização de cistos de T. gondii em cortes de lombos (m. longissimus,) de suínos experimentalmente infectados. Seis suínos com aproximadamente 90 dias de idade e soronegativos para T. gondii (título < 64 na Reação de Imunofluorescência Indireta RIFI) foram infectados, via oral, com 3×103 oocistos do isolado TgCkBr57 de T. gondii Os oocistos foram obtidos após reativação do isolado em camundongos e infecção, via oral, de um gato com idade aproximada de cinco meses e soronegativo para T. gondii (título < 16 na RIFI). Um suíno soronegativo foi mantido como controle durante todo o experimento. Os sete suínos foram acompanhados, semanalmente, para anticorpos anti-T. gondii por meio da RIFI durante oito semanas. No abate (suínos com 150 dias e 80kg, aproximadamente), as carcaças dos seis suínos foram divididas em seis pares. Após a separação dos cortes, os lombos esquerdos foram embalados a vácuo e submetidos ao processo de maturação úmida por 16, 18 ou 21 dias (dois lombos por período) em câmara refrigerada (-1 a 0°C). Todos os lombos, maturados e não-maturados, foram submetidos ao processo de digestão péptica de tecidos e bioensaio em camundongos (300 camundongos para lombos maturados e 300 para lombos não-maturados), em seus respectivos períodos, para a observação da infecção por T. gondii. Diafragma e língua de todos os suínos também foram colhidos no abate para o bioensaio em camundongos (10 camundongos por suíno), como controle da infecção. Os camundongos eram acompanhados diariamente para observação de sinais de toxoplasmose, quando eram eutanasiados e examinados para a presença de taquizoítas e/ou cistos de T. gondii nos seus tecidos. Animais sobreviventes por seis semanas pós-inoculação eram eutanasiados e examinados para a presença de T. gondii. A soroconversão, com a presença de anticorpos IgM e IgG anti-T. gondii, foi observada nos seis suínos infectados. O número de camundongos infectados com os lombos maturados por 16 dias (69/100) foi próximo aos respectivos lombos não-maturados (80/100); este número foi menor nos grupos inoculados com lombos maturados por 18 dias (28/100), em relação ao respectivo grupo não-maturado (73/100), sugerindo uma diminuição do número de cistos com 18 dias de maturação. Não houve infecção dos camundongos inoculados com lombos maturados por 21 dias (0/100), enquanto no grupo não-maturado respectivo foi 68/100 de infectados. O presente estudo não permitiu o refinamento do processo de maturação úmida para lombos suínos usando os períodos de 16 e 18 dias, mas confirmou a inviabilização de cistos de T. gondii no período de 21 dias de maturação úmida nas condições experimentais apresentadas. Toxoplasmosis is a disease of zoonotic importance caused by the protozoan Toxoplasma gondii. The two main routes of transmission of T. gondii to humans are the ingestion of sporulated oocysts and the ingestion of raw or undercooked meat that contain viable tissue cysts of the agent. The dry ageing process can be a means of controlling toxoplasmosis acquired through meat ingestion. The objective of this study was the refinement of the dry ageing process in the non-viability of T. gondiicysts in cuts of loin (m. longissimus) of experimentally infected pigs. Six pigs with approximately 90 days of age and seronegative for T. gondii (titer < 64 by Indirect Fluorescent Antibody TEST IFAT) were orally infected with 3×103 oocysts of the T. gondii isolate TgCkBr57. Oocysts were obtained after reactivation of the isolate in mice and oral infection of a cat aged approximately five months and seronegative for T. gondii (titer < 16 by IFAT). One seronegative swine was maintained as a control throughout the experiment. The seven pigs were followed up weekly for anti-T. gondii antibodies through RIFI for eight weeks. At slaughter (swine aged 150 days and weighing approximately 80 kg), the carcasses of the six pigs were divided into six pairs. After separation of the cuts, the left loins were vacuum packed and submitted to the dry ageing process for 16, 18 or 21 days (two loins per period) in a refrigerated chamber (-1 to 0ºC). All loins, aged and non-aged, were submitted to the process of peptic digestion of tissues and bioassay in mice (300 mice for aged loins and 300 for non-aged loins), in their respective periods, to observe the infection by T. gondii. Diaphragm and tongue of all pigs were also collected at slaughter for the mouse bioassay (10 mice per pig) as an infection control. The mice were monitored daily for signs of toxoplasmosis, when they were euthanized and examined for the presence of tachyzoites and/or T. gondii cysts in their tissues. Animals surviving for six weeks post- inoculation were euthanized and examined for the presence of T. gondii. Seroconversion was observed in the six infected pigs, with the presence of IgM and IgG anti-T. gondii antibodies. The number of mice infected with loins aged for 16 days (69/100) was close to the respective non-aged loins (80/100); this number was lower in the groups inoculated with loins aged for 18 days (28/100), in relation to the respective non-aged group (73/100), suggesting a decrease in the number of cysts with 18 days of dry ageing. There was no infection in mice inoculated with loins aged for 21 days (0/100), while in the respective non-aged group there was 68/100 infected. The present study did not allow the refinement of the dry ageing process for swine loins using the periods of 16 and 18 days but confirmed the non-viability of T. gondii cysts with 21 days of dry ageing vacuum-packed pork under the experimental conditions presented.
