Abstract

Today researchers look for a possibility to keep cells for a long time without losing their viability. For that cryopreservation is often used. In this process it is necessary that the cells are not destroyed so cryoprotective agents (CPA) are needed. At the moment 5 to 10 % dimethylsulfoxide (DMSO) is mostly used, but this chemical is cytotoxic to the cells. So an alternative is needed. In this work experiments are made with an icebindingprotein (IBP) of Hypogastrura harveyi, as an alternative to DMSO. It was shown in previous studies that this protein isn’t cytotoxic for the cells, with crude extract and purified inclusion bodies it even seems that the mixtures have a positive effect on growth and proliferation. As a first step the protein was produced heterologous in E. coli and then the crude extract and the purified inclusion bodies were used for experiments on the influence of the IBP on the cryopreservation of mesenchymal stem cells from the common marmoset monkey Callithrix jacchus. In the process it was found that the protein could not replace DMSO completely. But it was possible to show that the DMSO-concentration can be reduced by adding the IBP.

Abstract

When intense lightwaves accelerate electrons through a solid, the emerging high-order harmonic (HH) radiation offers key insights into the material.sup.1-11. Sub-optical-cycle dynamics--such as dynamical Bloch oscillations.sup.2-5, quasiparticle collisions.sup.6,12, valley pseudospin switching.sup.13 and heating of Dirac gases.sup.10--leave fingerprints in the HH spectra of conventional solids. Topologically non-trivial matter.sup.14,15 with invariants that are robust against imperfections has been predicted to support unconventional HH generation.sup.16-20. Here we experimentally demonstrate HH generation in a three-dimensional topological insulator--bismuth telluride. The frequency of the terahertz driving field sharply discriminates between HH generation from the bulk and from the topological surface, where the unique combination of long scattering times owing to spin-momentum locking.sup.17 and the quasi-relativistic dispersion enables unusually efficient HH generation. Intriguingly, all observed orders can be continuously shifted to arbitrary non-integer multiples of the driving frequency by varying the carrier-envelope phase of the driving field--in line with quantum theory. The anomalous Berry curvature warranted by the non-trivial topology enforces meandering ballistic trajectories of the Dirac fermions, causing a hallmark polarization pattern of the HH emission. Our study provides a platform to explore topology and relativistic quantum physics in strong-field control, and could lead to non-dissipative topological electronics at infrared frequencies. Abstract

Ultrafast charge separation after photoexcitation is a common phenomenon in various van-der-Waals (vdW) heterostructures with great relevance for future applications in light harvesting and detection. Theoretical understanding of this phenomenon converges towards a coherent mechanism through charge transfer states accompanied by energy dissipation into strongly coupled phonons. The detailed microscopic pathways are material specific as they sensitively depend on the band structures of the individual layers, the relative band alignment in the heterostructure, the twist angle between the layers, and interlayer interactions resulting in hybridization. We used time- and angle-resolved photoemission spectroscopy combined with tight binding and density functional theory electronic structure calculations to investigate ultrafast charge separation and recombination in WS2-graphene vdW heterostructures. We identify several avoided crossings in the band structure and discuss their relevance for ultrafast charge transfer. We relate our own observations to existing theoretical models and propose a unified picture for ultrafast charge transfer in vdW heterostructures where band alignment and twist angle emerge as the most important control parameters.

Abstract

Mixing is a crucial parameter for a wide range of processes in bio-/chemical engineering. In multiphase flows, the aim is a high and uniform mixing effect throughout the reaction volume without negative effects on the discrete phase. These are usually caused by the hydromechanical stress exerted by the mixing element. The present study introduces a novel reactor concept to overcome the use of fast rotating, locally acting mixing elements common in stirred tank reactors. Therefore, the reactor is designed of a flat, horizontally mounted cylinder, which enables mixing of the enclosed volume by its rotation around the central axis. To evaluate the mixing performance, a colorimetric method for mixing time determination is presented and the spatial evolution of mixing kinetics visualized. Abstract

Malaria risk is highly heterogeneous. Understanding village and household-level spatial heterogeneity of malaria risk can support a transition to spatially targeted interventions for malaria elimination. This analysis uses data from cross-sectional prevalence surveys conducted in 2014 and 2016 in two villages (Megiar and Mirap) in Papua New Guinea. Generalised additive modelling was used to characterise spatial heterogeneity of malaria risk and investigate the contribution of individual, household and environmental-level risk factors. Following a period of declining malaria prevalence, the prevalence of P. falciparum increased from 11.4 to 19.1% in Megiar and 12.3 to 28.3% in Mirap between 2014 and 2016, with focal hotspots observed in these villages in 2014 and expanding in 2016. Prevalence of P. vivax was similar in both years (20.6% and 18.3% in Megiar, 22.1% and 23.4% in Mirap) and spatial risk heterogeneity was less apparent compared to P. falciparum. Within-village hotspots varied by Plasmodium species across time and between villages. In Megiar, the adjusted odds ratio (AOR) of infection could be partially explained by household factors that increase risk of vector exposure, such as collecting outdoor surface water as a main source of water. In Mirap, increased AOR overlapped with proximity to densely vegetated areas of the village. The identification of household and environmental factors associated with increased spatial risk may serve as useful indicators of transmission hotspots and inform the development of tailored approaches for malaria control.

One of the research questions of the joint research project “Efficient Storage and Processing Techniques for Wood Chips” was, if storing technically dried wood chips outdoors with a fleece cover leads to the same results concerning fuel quality and dry matter losses as storage in a storehouse. Therefore, an outdoor experiment was conducted from June to November 2018 at a wood chip storage site near Munich. Two 80 lcm - piles of technically dried wood chips from spruce energy roundwood were built up, equipped with temperature sensors and 36 balance bags with wood chip samples. One pile was put up outside and covered with a vapor-permeable fleece (PolyTex, 200 g/m², Zill GmbH & Co. KG). As a reference, a second pile was put inside a storehouse nearby. The wood chips were technically dried to a moisture content of approx. 14 w-% by a biomass service contractor prior to storage. After five months the balance bags were removed and analyzed. There was hardly any change in moisture content and no dry matter loss. In addition to the main trial described above, two side trials were executed. A large-scale laboratory experiment was carried out from July to September 2018 to determine the average precipitation retention capacity (PRC) of the fleece. The weighted mean of the PRC was 84.7 %, considering the local precipitation distribution. For light rainfall it can be even more than 90 %. Furthermore, a laboratory experiment was conducted to estimate the influence of aging and usage of the fleece on its PRC.
