Abstract

BackgroundThe extent to which vaccinated persons diagnosed with coronavirus disease 2019 (COVID-19) can transmit to other vaccinated and unvaccinated persons is unclear.MethodsUsing data from the San Francisco Department of Public Health, this report describes outcomes of household contact tracing during 29 January-2 July 2021, where fully vaccinated patients with COVID-19 were the index case in the household.ResultsAmong 248 fully vaccinated patients with breakthrough infections, 203 (82%) were symptomatic and 105 were identified as the index patient within their household. Among 179 named household contacts, 71 (40%) contacts tested, over half (56%) were fully vaccinated and the secondary attack rate was 28%. Overall transmission from a symptomatic fully vaccinated patient with breakthrough infection to household contacts was suspected in 14 of 105 (13%) of households. Viral genomic sequencing of samples from 44% of fully vaccinated patients showed that 82% of those sequenced were infected by a variant of concern or interest and 77% by a variant carrying mutation(s) associated with resistance to neutralizing antibodies.ConclusionsTransmission from fully vaccinated symptomatic index patients to vaccinated and unvaccinated household contacts can occur. Indoor face masking and timely testing of all household contacts should be considered when a household member receives a positive test result in order to identify and interrupt transmission chains.

Abstract

Recent advances in nucleation and crystal growth have revolutionized our understanding and control of crystallization processes. This paper highlights key developments in this field and the processes and technologies involved in its continuous growth. Advanced computational models have allowed for precise prediction of nucleation rates and crystal morphologies, facilitating the rational design of materials with desired properties. Innovative strategies have also emerged, enabling enhanced control over crystal growth kinetics and crystallographic orientations. Process intensification strategies, including microreactors and membrane crystallization, enhance nucleation rates and crystal growth. Advances in the potential-driven growth of metal crystals from ionic liquids, including protic ionic liquids (PILs) and solvate ionic liquids (SILs), are discussed. Lastly, current research gaps and future prospects in the field of nucleation and crystal formation are highlighted. The integration of cutting-edge experimental techniques, computational modeling, and novel strategies will drive the understanding of nucleation and crystal growth processes, allowing for the development of materials with tailored properties and enhanced functionality across multiple disciplines. Abstract

Mycobacterium tuberculosisdrug resistance is emerging and new drug targets are needed. Tryptophan biosynthesis is necessary for M. tuberculosisreplication and virulence. Indole-3-glycerol phosphate synthase (IGPS) catalyzes a step in M. tuberculosistryptophan biosynthesis and has been suggested as a potential anti-infective target, but our understanding of this enzyme is limited. To aid in inhibitor design and gain a greater mechanistic picture of this enzyme, there is a need to understand the roles of active site amino acids in ligand binding and catalysis. In this work, we explored the roles of conserved active site amino acids Glu57, Lys59, Lys119, Glu168, and Glu219. Mutation of each to Ala results in loss of all detectable activity. The Glu57Gln, Lys59Arg, Lys119Arg, Glu168Gln, and Glu219Asp mutations result in large activity losses, while Glu219Gln has enhanced activity. Analysis of the enzymatic data yields the following main conclusions: (A) Lys119 is the likely catalytic acid in the CdRP ring closure step. (B) Glu168 stabilizes a charged reaction intermediate and may also be the catalytic base. (C) Glu57, Glu219, and Lys119 form a closely arranged triad in which Glu57 and Glu219 modulate the pKaof Lys119, and thus overall activity. This increased understanding of inter- and intramolecular interactions and demonstration of the highly coordinated nature of the M. tuberculosisIGPS active site provide new mechanistic information and guidance for future work with this potential new drug target.

Abstract

Objective: Despite successful endovascular therapy, a proportion of stroke patients exhibit long‐term functional decline, regardless of the cortical reperfusion. Our objective was to evaluate the early activation of the adaptive immune response and its impact on neurological recovery in patients with large vessel occlusion (LVO). Methods: Nineteen (13 females, 6 males) patients with acute LVO were enrolled in a single‐arm prospective cohort study. During endovascular therapy (EVT), blood samples were collected from pre and post‐occlusion, distal femoral artery, and median cubital vein (controls). Cytokines, chemokines, cellular and functional profiles were evaluated with immediate and follow‐up clinical and radiographic parameters, including cognitive performance and functional recovery. Results: In the hyperacute phase (within hours), adaptive immune activation was observed in the post‐occlusion intra‐arterial environment (post). Ischemic vascular tissue had a significant increase in T‐cell‐related cytokines, including IFN‐γ and MMP‐9, while GM‐CSF, IL‐17, TNF‐α, IL‐6, MIP‐1a, and MIP‐1b were decreased. Cellularity analysis revealed an increase in inflammatory IL‐17+ and GM‐CSF+ helper T‐cells, while natural killer (NK), monocytes and B‐cells were decreased. A correlation was observed between hypoperfused tissue, infarct volume, inflammatory helper, and cytotoxic T‐cells. Moreover, helper and cytotoxic T‐cells were also significantly increased in patients with improved motor function at 3 months. Interpretation: We provide evidence of the activation of the inflammatory adaptive immune response during the hyperacute phase and the association of pro‐inflammatory cytokines with greater ischemic tissue and worsening recovery after successful reperfusion. Further characterization of these immune pathways is warranted to test selective immunomodulators during the early stages of stroke rehabilitation. [ABSTRACT FROM AUTHOR]

Abstract

Understanding how species can coexistence in light of competition has long been a focus of ecological study. One of the many mechanisms that have been proposed to explain how species with overlapping niches can coexist is that they differ in their behavioral patterns. In our study we investigated how rodents in the genera Dipodomys and Peromyscus partition their habitat spatially and how their behaviors differ. To test for behavioral differences and spatial habitat partitioning in these species we monitored animals using traps and wildlife cameras and mapped their habitat use in space. We found that Peromyscus displayed risk averse strategies in their foraging and habitat use whereas Dipodomys displayed more variability in their movements and were more likely to take risks. These interspecific patterns help us to better understand community ecology as a whole, and can have broad implications for the structure of communities in sensitive desert habitats., CEC Research Volume 1, Issue 2

Abstract

Larvae of the Arctiinae use an impressive arsenal of defenses to protect themselves against natural enemies. Several species within this subfamily have been studied for their defensive capabilities, specifically the use of plant secondary metabolites. Life history and sequestration patterns of secondary metabolites have been well documented for Grammia incorrupta; however, little information exists for other species of Grammia. Furthermore, information on sequestration patterns during the immature stages of several Grammia spp. is largely unknown. To address these gaps in knowledge, I am conducting a comparative study of the chemical ecology and life history traits for the virgin tiger moth (Grammia virgo), the little virgin tiger moth (Grammia virguncula) and the figured tiger moth (Grammia figurata) throughout larval development. I divided larvae into four treatment groups: one on white clover (Trifolium repens), one on broadleaf plantain (Plantago major), one on narrowleaf plantain (Plantago lanceolata), and one on a wheat-germ based artificial diet. I collected data on development time for each instar, larval weight at each instar and overall survivorship. A subset of larvae was also harvested at each instar; these samples were prepared for chemical analysis to determine presence or absence of aucubin and catalpol at different instars. Aucubin and catalpol are found in one or both of the Plantago species used in this study. Here I will present results of the feeding trials and the chemical analyses of the Grammia tiger moths and discuss implications of these results on the understanding the chemical ecology and development of these species.

En las últimas décadas el Burnout ha sido una patología de impulso investigativo por su amplia presencia y foco de manifestación en el área laboral, con la llegada del Covid-19 a los profesionales de la salud se les atribuye una serie de estresores dentro de las cuales se puede mencionar como, la asignación de tareas administrativas y técnicas distintas de las habituales, falta de adecuada capacitación, dilemas éticos y morales asociados a la restricción o falta de recursos, aumento de las expectativas de la población respecto de la capacidad sanitaria, discriminación social por temor al contagio, presión de los medios de comunicación, los cuales son precipitantes para generar sintomatologías de depresión ansiedad y afección emocional contribuyendo a la aparición del síndrome de Burnout. De tal forma, en la presente investigación se pueden analizar sesgos en las repuestas entre los criterios a analizar del síndrome como lo es la despersonalización, cansancio emocional, realización personal, ya que estos datos se presentan tanto dispersos como agrupados, lo cual permite ofrecer información base para próximas investigaciones académicas visualizando la presencia del agotamiento mental, emocional y físico en personal de enfermería expuestos en el periodo de pandemia en la ciudad de Ibagué Tolima. In recent decades the Burnout has been a pathology of research impulse for its wide presence and focus of manifestation in the area of work, exposing a series of symptomatologies that by their holistic origin within the activities of the individual open debates about their genesis and their precipitating factors and/or environments. However, burnout has now been recognized by WHO as an occupational disease. In this way, in the present research we can analyze biases in the responses between the criteria to analyze the syndrome such as depersonalization, emotional fatigue, personal fulfillment, since these data are presented both dispersed and grouped, providing background information for further academic research by visualizing the presence of mental, emotional and physical exhaustion in nursing personnel exposed during the pandemic period in the city of Ibague Tolima.
