King coconut (Cocos nucifera var. aurantiaca) is a tropical nut with notable nutritional and medicinal values. King coconut water (KCW) and king coconut kernel (KCK) are sweet and refreshing. They undergo several changes during maturation. The objective of this study was to assess the physico-chemical changes of KCW and KCK with maturity. Three different maturity stages (6, 7, and 8 months) of the variety aurantica, known locally as 'Nawasi Thembili' were studied for several physico-chemical parameters. Whole nut weight (g), nut-circumference (cm), colour (water/kernel) (*L), nut-water volume (mL), kernel weight (g), and kernel thickness (mm) were assessed as physical parameters, while pH, total soluble solids (oBrix), titratable acidity (as % ascorbic acid), total sugars (g/100g; g/100mL), and minerals (mg/L; mg/kg) were analysed as chemical attributes. Sugars were estimated using Agilent 1260 HPLC and ICP-MS was used to measure the mineral profile. Results revealed that all the tested physical parameters of KCW and KCK increased significantly (p < 0.05) with maturity. The pH, TSS, total sugars, and acidity of KCW increased significantly (p < 0.05), while mineral content decreased significantly (p < 0.05) with maturity. However, the TSS and minerals of KCK decreased significantly (p < 0.05) with maturity and a higher sucrose content was reported in KCK compared to KCW at later maturity. The Ca+2 and Mg+2 were prominent in both KCW and KCK. The study concluded that the physico-chemical characteristics varied widely with the maturity of king coconuts. The selection of optimum harvest maturity to match the preferred attributes of intended applications is suggested. [ABSTRACT FROM AUTHOR]

Introduction: The limited availability of nutritionally adequate and safe foods or inadequate accessibility to acquire foods can result in food insecurity. This study was conducted to investigate access and attitudes among private university students in a Colombo suburb, Sri Lanka to acquire healthy foods. Methods: A self-administered pre-validated questionnaire was used to interview a total of 103 undergraduate students who were conveniently recruited from private universities. Indications of food insecurity were assessed in terms of dietary habits. Relationships between demographic characteristics and dietary habits were determined. Access to healthy foods was based on availability of healthy foods such as herbal porridge, boiled grains and fruit salads at their university premises, and the students’ willingness to buy them. Results: The majority of the subjects were females (60.2%) and 66.7% of the subjects resided outside their family homes. Frequency of consuming at least one serving per week of grain, milk, fruits & vegetables showed no significant relationship with gender. Location of residence was significantly related to consumption of grains (P=<0.000). The results revealed that 73.1% of the subjects had no access to healthy foods in their respective university premises, while more than half (61.3%) were not satisfied with the foods they consumed. Conclusion: This study revealed a lack of access to healthy foods in private higher educational premises in the Colombo suburb, indicating the risk of food insecurity among university students. Microglia are resident immune cells of the central nervous system (CNS). In adulthood they are involved in surveillance and responses to pathogens and injury and prenatally they play a role in brain development. However, the role of microglia during the early postnatal period and how they impact development long-term remains poorly understood. Here, to investigate the specific role of microglia in postnatal development, we used a Cx3cr1-Dtr transgenic Wistar rat model to acutely ablate microglia from either postnatal day (P) 7 or 14. We specifically assessed how transient microglial ablation affected astrocytes and neurons acutely, during the juvenile period, and in adulthood. Hippocampal microglial numbers remained low at P21 in the P7-ablated animals and complexity remained reduced after P14-ablation. This protracted effect on these key immune cells led to a small but significant increase in CA1 mature neuron numbers and a significant increase in astrocyte density in the subgranular dentate gyrus in adults that had their microglia ablated at P14. However, these histological differences were small, and spatial and recognition memory in novel objection and place recognition tests were not affected. Overall, our data reveal for the first time that the transient depletion of microglia during the neonatal period impacts briefly on the brain but that the long-lasting effects are minimal. Neonatal microglia may be dispensable in the establishment of hippocampal brain function. These data also imply that novel therapeutic anti-inflammatories that cross the blood-brain barrier to inhibit microglia are unlikely to have long-term negative consequences if administered in the neonatal period., (Copyright © 2020 Elsevier Inc. ATPases associated with diverse cellular activities (AAA+) proteases utilize ATP hydrolysis to actively unfold native or misfolded proteins and translocate them into a protease chamber for degradation. This basic mechanism yields diverse cellular consequences, including the removal of misfolded proteins, control of regulatory circuits, and remodeling of protein conformation. Among various bacterial AAA+ proteases, FtsH is only membrane-integrated and plays a key role in membrane protein quality control. Previously, we have shown that FtsH has substantial unfoldase activity for degrading membrane proteins overcoming a dual energetic burden of substrate unfolding and membrane dislocation. Here, we asked how efficiently FtsH utilizes ATP hydrolysis to degrade membrane proteins. To answer this question, we measured degradation rates of the model membrane substrate GlpG at various ATP hydrolysis rates in the lipid bilayers. We find that the dependence of degradation rates on ATP hydrolysis rates is highly nonlinear: (i) FtsH cannot degrade GlpG until it reaches a threshold ATP hydrolysis rate; (ii) after exceeding the threshold, the degradation rates steeply increase and saturate at the ATP hydrolysis rates far below the maxima. During the steep increase, FtsH efficiently utilizes ATP hydrolysis for degradation, consuming only 40-60% of the total ATP cost measured at the maximal ATP hydrolysis rates. This behavior does not fundamentally change against water-soluble substrates as well as upon addition of the macromolecular crowding agent Ficoll 70. The Hill analysis shows that the nonlinearity stems from coupling of three to five ATP hydrolysis events to degradation, which represents unique cooperativity compared to other AAA+ proteases including ClpXP, HslUV, Lon, and proteasomes., (© 2019 The Protein Society.)

Background: In Asia, rotavirus accounts for approximately 45% of admissions due to acute gastroenteritis in children <5 years, and causes about 145,000 deaths every year. We studied the distribution of rotavirus strains from Myanmar, Sri Lanka, and Nepal during 2009-2015., Methods: Stool samples collected from children <5 years of age hospitalized with acute diarrhea in the three sites and positive for rotavirus antigen by enzyme immunoassay (EIA) were sent to the Christian Medical College, Vellore from 2009 to 2015. G and P typing of rotavirus strains were performed using reverse-transcription polymerase chain reaction (RT-PCR)., Result: Of the 2354 EIA positive samples tested, G12P[8] (36.8%), G1P[8] (30.1%), and G12P[6] (41.3%) were the most common strains isolated from Myanmar, Sri Lanka, and Nepal respectively., Conclusion: There was substantial diversity of rotavirus genotypes, and continued surveillance in developing countries of Asia will help in understanding the epidemiology of rotavirus before and after introduction of vaccines., (Copyright © 2018 Elsevier Ltd. The treatment of relapsed aggressive lymphoma remains a challenge. Platinum-containing chemotherapy is standard of care. Gemcitabine/oxaliplatin (Gem-Ox) with or without rituximab (R) is an outpatient regimen with a favorable toxicity profile. This retrospective 'real world' study reports outcomes for 44 unselected patients with relapsed/refractory aggressive lymphoma treated with Gem-Ox ± R. 41% had primary refractory disease. The overall response rate (ORR) was 43% with a complete response (CR) of 30%. Response to the prior treatment regimen significantly affected the ORR with only 8% achieving CR if prior remission was <12 months. Grade 3-4 hematological toxicity was common and 22% had febrile neutropenia. Eight patients proceeded to stem cell transplant. Overall, outcomes remain poor with a median overall survival of 8 months. In this high-risk group of patients, Gem-Ox ± R results in similar responses to other more toxic, inpatient regimens and should therefore be considered as second line therapy in relapsed lymphoma.

Restroom internal door handles have the potential to become contaminated by pathogenic bacteria, particularly because frequent breakdowns occur in hand hygiene. Cleaning these door handles periodically could reduce this cross-contamination risk. The sustained effect following cleaning with chlorhexidine could be beneficial in restroom facilities as cleaning episodes are of necessity at time intervals. The cleaning efficacies and residual effects of Sani Cloth CHG 2% wipes were investigated in a double-blinded randomized crossover controlled trial in a school setting. No significant difference occurred in initial cleaning efficacy; however, following a six-hour period of use by pupils of the restroom facilities, the internal door handles wiped with Sani-Cloth CHG 2% wipes were significantly less contaminated than those with the control wipe (14% v. 32%, p = .02). Cleaning with Sani-Cloth CHG 2% wipes demonstrated significant improvements in the continuous cleanliness of restroom door handles during use with this simple and inexpensive technique.
