Abstract

The yield stress and shear thinning properties of mucus are identified as critical for ciliary coordination and mucus transport in human airways. We use here numerical simulations to explore the hydrodynamic coupling of cilia and mucus with these two properties using the Herschel--Bulkley model, in a lattice Boltzmann solver for the fluid flow. Three mucus flow regimes, i.e. a poorly organized regime, a swirly regime, and a fully unidirectional regime, are observed and analysed by parametric studies. We systematically investigate the effects of ciliary density, interaction length, Bingham number and flow index on the mucus flow regime formation. The underlying mechanism of the regime formation is analysed in detail by examining the variation of two physical quantities (polarization and integral length) and the evolution of the flow velocity, viscosity and shear-rate fields. Mucus viscosity is found to be the dominant parameter influencing the regime formation when enhancing the yield stress and shear thinning properties. The present model is able to reproduce the solid body rotation observed in experiments (Loiseau et al. , Nat. Phys. , vol. 16, 2020, pp. 1158--1164). A more precise prediction can be achieved by incorporating non-Newtonian properties into the modelling of mucus as proposed by Gsell et al. ( Sci. Rep. , vol. 10, 2020, 8405).

Abstract

OPERA is a long-baseline experiment designed to search for $\nu_{\mu}\to\nu_{\tau}$ oscillations in appearance mode. It was based at the INFN Gran Sasso laboratory (LNGS) and took data from 2008 to 2012 with the CNGS neutrino beam from CERN. After the discovery of $\nu_\tau$ appearance in 2015, with $5.1\sigma$ significance, the criteria to select $\nu_\tau$ candidates have been extended and a multivariate approach has been used for events identification. In this way the statistical uncertainty in the measurement of the oscillation parameters and of $\nu_\tau$ properties has been improved. Abstract

The reactor antineutrino anomaly might be explained by the oscillation of reactor antineutrinos toward a sterile neutrino of eV mass. In order to explore this hypothesis, the STEREO experiment measures the antineutrino energy spectrum in six different detector cells covering baselines between 9 and 11 m from the compact core of the ILL research reactor. In this Letter, results from 66 days of reactor turned on and 138 days of reactor turned off are reported. A novel method to extract the antineutrino rates has been developed based on the distribution of the pulse shape discrimination parameter. The test of a new oscillation toward a sterile neutrino is performed by comparing ratios of cells, independent of absolute normalization and of the prediction of the reactor spectrum. The results are found to be compatible with the null oscillation hypothesis and the best fit of the reactor antineutrino anomaly is excluded at 97.5\% C.L.

Abstract

The STEREO experiment is a very short baseline reactor antineutrino experiment aiming at testing the hypothesis of light sterile neutrinos as an explanation of the deficit of the observed neutrino interaction rate with respect to the predicted rate, known as the Reactor Antineutrino Anomaly. The detector center is located 10 m away from the compact, highly $^{235}$U enriched core of the research nuclear reactor of the Institut Laue Langevin in Grenoble, France. This paper describes the STEREO site, the detector components and associated shielding designed to suppress the external sources of background which were characterized on site. Abstract

The OPERA experiment was designed to study $\nu_\mu\to\nu_\tau$ oscillations in appearance mode in the CNGS neutrino beam. In this letter we report the final analysis of the full data sample collected between 2008 and 2012, corresponding to $17.97\cdot 10^{19}$ protons on target. Selection criteria looser than in previous analyses have produced ten $\nu_\tau$ candidate events, thus reducing the statistical uncertainty in the measurement of the oscillation parameters and of $\nu_\tau$ properties. A multivariate approach for event identification has been applied to the candidate events and the discovery of $\nu_\tau$ appearance is confirmed with an improved significance level of 6.1 $\sigma$. $\Delta m^2_{23}$ has been measured, in appearance mode, with an accuracy of 20%. The measurement of $\nu_\tau$ CC cross-section, for the first time with a negligible contamination from $\bar{\nu}_\tau$, and the first direct observation of the $\nu_\tau$ lepton number are also reported., Comment: 7 pages, 3 figures

Abstract

The OPERA experiment has discovered the tau neutrino appearance in the CNGS muon neutrino beam, in agreement with the 3 neutrino flavour oscillation hypothesis. The OPERA neutrino interaction target, made of Emulsion Cloud Chamber, was particularly efficient in the reconstruction of electromagnetic showers. Moreover, thanks to the very high granularity of the emulsion films, showers induced by electrons can be distinguished from those induced by $\pi^0$s, thus allowing the detection of charged current interactions of electron neutrinos. In this paper the results of the search for electron neutrino events using the full dataset are reported. An improved method for the electron neutrino energy estimation is exploited. Data are compatible with the 3 neutrino flavour mixing model expectations and are used to set limits on the oscillation parameters of the 3+1 neutrino mixing model, in which an additional mass eigenstate $m_{4}$ is introduced. At high $\Delta m^{2}_{41}$ $( \gtrsim 0.1~\textrm{eV}^{2})$, an upper limit on $\sin^2 2\theta_{\mu e}$ is set to 0.021 at 90% C.L. and $\Delta m^2_{41} \gtrsim 4 \times 10^{-3}~\textrm{eV}^{2}$ is excluded for maximal mixing in appearance mode., Comment: Editors: M. Abstract

In this paper, the Immersed Boundary Method (IBM) proposed by Pinelli is implemented for finite volume approximations of incompressible Navier-Stokes equations solutions in the open source toolbox OpenFOAM version 2.2. Solid obstacles are described using a discrete forcing approach for boundary conditions. Unlike traditional approaches encompassing the presence of a solid body using conformal meshes and imposing no-slip boundary conditions on the boundary faces of the mesh, the solid body is here represented on the Eulerian Cartesian mesh through an ad-hoc body force evaluated on a set of Lagrangian markers. The markers can move across the Eulerian mesh, hence allowing for a straightforward analysis of motion or deformation of the body. The IBM method is described and implemented in PisoFOAM, whose Pressure-Implicit Split-Operator (PISO) solver is modified accordingly. The presence of the solid body and the divergence-free of the fluid velocity are imposed simultaneously by sub-iterating between IBM and the pressure correction step. This scheme allows for the use of fast optimized Poisson solvers while granting excellent accuracy with respect to the previously mentioned constraints. Various 2D and 3D well-documented test cases of flows around fixed or moving circular cylinders are simulated and carefully validated against existing data from the literature. The capability of the new solver is discussed in terms of accuracy and numerical performances.

Abstract

The STEREO experiment is a very short baseline reactor antineutrino experiment aiming at testing the hypothesis of light sterile neutrinos as an explanation of the deficit of the observed neutrino interaction rate with respect to the predicted rate, known as the Reactor Antineutrino Anomaly. The detector center is located 10 m away from the compact, highly 235U enriched core of the research nuclear reactor of the Institut Laue Langevin in Grenoble, France. This paper describes the STEREO site, the detector components and associated shielding designed to suppress the external sources of background which were characterized on site. It reports the performances in terms of detector response and energy reconstruction.

Abstract

Les paragangliomes et phéochromocytomes (PPGL) sont des tumeurs neuroendocrines rares dont les formes métastatiques sont fréquemment associées à des déficits héréditaires en succinate déshydrogénase (SDH), une enzyme majeure du cycle de Krebs et de la chaîne respiratoire mitochondriale. La ferroptose, un mécanisme de mort cellulaire régulé par le fer et caractérisé par l’accumulation toxique de peroxydes lipidiques non contrôlés émerge actuellement comme une voie de mort cellulaire potentiellement exploitable dans les PPGL. Le diméthylfumarate (DMF), un activateur du facteur nucléaire érythroïde 2 lié au facteur 2 (NRF2) connu pour son rôle dans la modulation du stress oxydatif, suscite un intérêt croissant en tant qu’inducteur de la ferroptose.

Les phéochromocytomes et paragangliomes (PPGL) sont des tumeurs neuroendocrines, métastatiques dans 15 % des cas. Aucun critère anatomopathologique ne permet de prédire leur malignité, ce qui nécessite un suivi à vie. Les PPGL présentent une forte prédisposition génétique, et une mutation SDHB entraine un risque accru de forme agressive. Ainsi, déterminer le statut génétique et le risque de malignité des patients atteints de PPGL est un défi majeur.

Abstract

The OPERA experiment was designed to study νμ→ντ oscillations in the appearance mode in the CERN to Gran Sasso Neutrino beam (CNGS). In this Letter, we report the final analysis of the full data sample collected between 2008 and 2012, corresponding to 17.97×1019 protons on target. Selection criteria looser than in previous analyses have produced ten ντ candidate events, thus reducing the statistical uncertainty in the measurement of the oscillation parameters and of ντ properties. A multivariate approach for event identification has been applied to the candidate events and the discovery of ντ appearance is confirmed with an improved significance level of 6.1σ. |Δm232| has been measured, in appearance mode, with an accuracy of 20%. The measurement of the ντ charged-current cross section, for the first time with a negligible contamination from ¯ντ, and the first direct evidence for the ντ lepton number are also reported.

To date, no effective therapies or reliable prognostic markers are available for patients who develop metastatic PPGL (mPPGL). Our aim was to discover robust prognostic markers validated through in vitro models, and define specific therapeutic options according to tumor genomic features. Methods: We analyzed three PPGL miRNome datasets (n=443), validated candidate markers and assessed them in serum samples (n=36) to find a metastatic miRNA signature. An integrative study of miRNome, transcriptome and proteome was performed to find miRNA targets, which were further characterized in vitro. Results: A signature of six miRNAs (miR-21-3p, miR-183-5p, miR-182-5p, miR-96-5p, miR-551b-3p, and miR-202-5p) was associated with metastatic risk and time to progression. A higher expression of five of these miRNAs was also detected in PPGL patients' liquid biopsies compared with controls. The combined expression of miR-21-3p/miR-183-5p showed the best power to predict metastasis (AUC=0.804, P=4.67.10(-18)), and was found associated in vitro with pro-metastatic features, such as neuroendocrine-mesenchymal transition phenotype, and increased cell migration rate. A pan-cancer multi-omic integrative study correlated miR-21-3p levels with TSC2 expression, mTOR pathway activation, and a predictive signature for mTOR inhibitor-sensitivity in PPGLs and other cancers. Likewise, we demonstrated in vitro a TSC2 repression and an enhanced rapamycin sensitivity upon miR-21-3p expression. Conclusions: Our findings support the assessment of miR-21-3p/miR-183-5p, in tumors and liquid biopsies, as biomarkers for risk stratification to improve the PPGL patients' management. We propose miR-21-3p to select mPPGL patients who may benefit from mTOR inhibitors.

An integrative study of miRNome, transcriptome and proteome was performed to find miRNA targets, which were further characterized in vitro. Results: A signature of six miRNAs (miR-21-3p, miR-183-5p, miR-182-5p, miR-96-5p, miR-551b-3p, and miR-202-5p) was associated with metastatic risk and time to progression. A higher expression of five of these miRNAs was also detected in PPGL patients’ liquid biopsies compared with controls. The combined expression of miR-21-3p/miR-183-5p showed the best power to predict metastasis (AUC=0.804, P=4.67·10-18), and was found associated in vitro with pro-metastatic features, such as neuroendocrine-mesenchymal transition phenotype, and increased cell migration rate. A pan-cancer multi-omic integrative study correlated miR-21-3p levels with TSC2 expression, mTOR pathway activation, and a predictive signature for mTOR inhibitor-sensitivity in PPGLs and other cancers. Likewise, we demonstrated in vitro a TSC2 repression and an enhanced rapamycin sensitivity upon miR-21-3p expression. Conclusions: Our findings support the assessment of miR-21-3p/miR-183-5p, in tumors and liquid biopsies, as biomarkers for risk stratification to improve the PPGL patients’ management. We propose miR-21-3p to select mPPGL patients who may benefit from mTOR inhibitors.

Abstract

The OPERA experiment has discovered the tau neutrino appearance in the CNGS muon neutrino beam, in agreement with the 3 neutrino flavour oscillation hypothesis. The OPERA neutrino interaction target, made of Emulsion Cloud Chamber, was particularly efficient in the reconstruction of electromagnetic showers. Moreover, thanks to the very high granularity of the emulsion films, showers induced by electrons can be distinguished from those induced by $��^0$s, thus allowing the detection of charged current interactions of electron neutrinos. In this paper the results of the search for electron neutrino events using the full dataset are reported. An improved method for the electron neutrino energy estimation is exploited. Data are compatible with the 3 neutrino flavour mixing model expectations and are used to set limits on the oscillation parameters of the 3+1 neutrino mixing model, in which an additional mass eigenstate $m_{4}$ is introduced. At high $��m^{2}_{41}$ $( \gtrsim 0.1~\textrm{eV}^{2})$, an upper limit on $\sin^2 2��_{��e}$ is set to 0.021 at 90% C.L. and $��m^2_{41} \gtrsim 4 \times 10^{-3}~\textrm{eV}^{2}$ is excluded for maximal mixing in appearance mode., Editors: M. Abstract

Inclusive ρ{variant}0 meson production has been measured in 120 GeV and 280 GeV muon-proton interactions. Distributions of z and pT2 are presented. Primary ρ{variant}0 production is found to be equal to that of π0 production within errors. © 1983.

Abstract

Exclusive ρ{variant}0 production has been measured in 120, 200 and 280 GeV muon-proton interactions at high Q2 (1 GeV2 < Q2 < 25 GeV2) and W (6 GeV < W < 19 GeV). The photoproduction cross section decreases as 1/Q4. A shallow t distribution, typical of a hard scattering process is observed and the ρ{variant}0 is found to be dominantly in the helicity zero spin state. The ρ{variant}0s are mainly produced by transverse photons and s-channel helicity conservation seems to be invalid. The data cannot be described by the vector meson dominance model. These data show that at high Q2 even exclusive ρ{variant}0 muoproduction is a hard scattering process and that the soft hadron-like properties of the photon have disappeared. © 1985.

Abstract

The problem of flapping filaments in an uniform incoming flow is tackled using a Lattice Boltzmann—Immersed Boundary method. The fluid momentum equations are solved on a Cartesian uniform lattice while the beating filaments are tracked through a series of markers, whose dynamics are functions of the forces exerted by the fluid, the filament flexural rigidity and the tension. The instantaneous wall conditions on the filament are imposed via a system of singular body forces, consistently discretised on the lattice of the Boltzmann equation. We first consider the case of a single beating filament, and then the case of multiple beating filaments in a side-by-side configuration, focussing on the modal behaviour of the whole dynamical systems.
