Eucalyptus covers approximately 7.5 million hectares in Brazil and serves as the primary woody species cultivated for commercial purposes. However, native insects and invasive pests pose a significant threat to eucalyptus trees, resulting in substantial economic losses and reduced forest productivity. One of the primary lepidopteran pests affecting eucalyptus is Thyrinteina arnobia (Stoll, 1782) (Lepidoptera: Geometridae), commonly referred to as the brown looper caterpillar. To address this issue, FuturaGene, the biotech division of Suzano S.A., has developed an insect-resistant (IR) eucalyptus variety, which expresses Cry pesticidal proteins (Cry1Ab, Cry1Bb, and Cry2Aa), derived from Bacillus thuringiensis (Bt). Following extensive safety assessments, including field trials across various biomes in Brazil, the Brazilian National Technical Commission of Biosafety (CTNBio) recently approved the commercialization of IR eucalyptus. The biosafety assessments involved the analysis of molecular genomics, digestibility, thermostability, non-target organism exposure, degradability in the field, and effects on soil microbial communities and arthropod communities. In addition, in silico studies were conducted to evaluate allergenicity and toxicity. Results from both laboratory and field studies indicated that Bt eucalyptus is as safe as the conventional eucalyptus clone for humans, animals, and the environment, ensuring the secure use of this insect-resistant trait in wood production.

En los valles irrigados de Río Negro y Neuquén, el cultivo de álamos acompañó históricamente la producción agrícola, utilizándose clones de Populus nigra L. en cortinas rompeviento y clones de Populus xcanadensis Moench. para las forestaciones en macizo. Buscando ampliar la disponibilidad de genotipos para los diferentes sitios de cultivo, se instaló un ensayo comparativo con trece clones de álamo en la localidad de Pomona, Provincia de Río Negro. Se evaluaron seis clones de P. xcanadensis ('Triplo', 'Ragonese 22 INTA', "Pangui INTA", "Pudú INTA", y los testigos comerciales 'Conti 12' e 'I-214'), seis clones de P. deltoides ('Harvard', 'Onda', 'Stoneville 67', 'Carabelas INTA', "20-82" y "C-657") y un clon de P. xcanescens. Se utilizó un diseño experimental de bloques completos al azar (DBCA) con quince repeticiones y parcelas de una planta. Luego de cuatro períodos de crecimiento, 'I-214' y 'Triplo' tuvieron los mayores incrementos de DAP, no encontrándose diferencias significativas con los valores de 'Conti 12', 'Ragonese 22 INTA', "Pangui INTA", "Pudú INTA", "20-82" y P. xcanescens. Los menores crecimientos fueron observados en los deltoides 'Stoneville 67' y "C- 657". Los clones 'Ragonese 22 INTA', 'Triplo' y "20-82" constituyen alternativas para el cultivo de álamos en macizos y cortinas rompeviento.

The slow calcium transient triggered by low-frequency electrical stimulation (ES) in adult muscle fibers and regulated by the extracellular ATP/IP3/IP3R pathway has been related to muscle plasticity. A regulation of muscular tropism associated with the MCU has also been described. However, the role of transient cytosolic calcium signals and signaling pathways related to muscle plasticity over the regulation of gene expression of the MCU complex (MCU, MICU1, MICU2, and EMRE) in adult skeletal muscle is completely unknown. In the present work, we show that 270 0.3-ms-long pulses at 20-Hz ES (and not at 90 Hz) transiently decreased the mRNA levels of the MCU complex in mice flexor digitorum brevis isolated muscle fibers. Importantly, when ATP released after 20-Hz ES is hydrolyzed by the enzyme apyrase, the repressor effect of 20 Hz on mRNA levels of the MCU complex is lost. Accordingly, the exposure of muscle fibers to 30 μM exogenous ATP produces the same effect as 20-Hz ES. Moreover, the use of apyrase in resting conditions (without ES) increased mRNA levels of MCU, pointing out the importance of extracellular ATP concentration over MCU mRNA levels. The use of xestospongin B (inhibitor of IP3 receptors) also prevented the decrease of mRNA levels of MCU, MICU1, MICU2, and EMRE mediated by a low-frequency ES. Our results show that the MCU complex can be regulated by electrical stimuli in a frequency-dependent manner. The changes observed in mRNA levels may be related to changes in the mitochondria, associated with the phenotypic transition from a fast- to a slow-type muscle, according to the described effect of this stimulation frequency on muscle phenotype. The decrease in mRNA levels of the MCU complex by exogenous ATP and the increase in MCU levels when basal ATP is reduced with the enzyme apyrase indicate that extracellular ATP may be a regulator of the MCU complex. Moreover, our results suggest that this regulation is part of the axes linking low-frequency stimulation with ATP/IP3/IP3R.

Multiple perturbations of the immune response affecting a range of cells have been reported in Trypanosoma cruzi-infected individuals and associated to clinical manifestations of chronic Chagas disease. There is a paucity of knowledge about the role of T follicular helper (Tfh) cells in this infection. Here, we sought to characterize circulating Tfh (cTfh) cells in chronic Chagas disease patients and to identify potential associations with disease severity in humans. cTfh cells were characterized by flow cytometry in freshly isolated PBMCs from 7 T. cruzi-infected asymptomatic patients (ASYMP), 5 patients with chronic chagasic dilated cardiomyopathy (CCC) and 8 healthy controls, using antibodies against chemokine receptors CXCR5, CXCR3, CCR6, and CCR7. Our results showed significant expansion of CD4+CD45RO+CXCR5+CCR6+ cells in ASYMP and CCC patients, along with a contraction of CD4+CD45RO+CXCR5+CXCR3-CCR6- (cTfh2) cells. ASYMP patients further exhibited decreased CD4+CD45RO+CXCR5+CXCR3+CCR6- (cTfh1) cells and expanded CD4+CD45RO+CXCR5+CXCR3-CCR6+ (cTfh17) cells while CCC patients exhibited significantly increased frequencies of CD4+CD45RO+CXCR5+CCR7+ cells. Linear regression analysis revealed a positive trend of CD4+CD45RO+CXCR5+CXCR3+CCR6+ (cTfh1/17) cells and negative trends of cTfh1 and cTfh2 cells as disease was more severe. There was no correlation between the frequencies of cTfh cells and circulating CD19+IgD-IgG+ cells or serum levels of T. cruzi-specific IgG. These results demonstrate that the cTfh compartment of humans chronically infected with T. cruzi comprises expanded CCR6-expressing cells and reduced cTfh2 cells. The association of discrete phenotypic changes in cTfh subsets with different clinical forms suggests the potential contribution of T follicular helper cells to Chagas heart disease progression.

Abstract In this article, we show that visual distance perception (VDP) is influenced by the auditory environmental context through reverberation-related cues. We performed two VDP experiments in two dark rooms with extremely different reverberation times: an anechoic chamber and a reverberant room. Subjects assigned to the reverberant room perceived the targets farther than subjects assigned to the anechoic chamber. Also, we found a positive correlation between the maximum perceived distance and the auditorily perceived room size. We next performed a second experiment in which the same subjects of Experiment 1 were interchanged between rooms. We found that subjects preserved the responses from the previous experiment provided they were compatible with the present perception of the environment; if not, perceived distance was biased towards the auditorily perceived boundaries of the room. Results of both experiments show that the auditory environment can influence VDP, presumably through reverberation cues related to the perception of room size.

Previous studies on the effect of spectral content on auditory distance perception (ADP) focused on the physically measurable cues occurring either in the near field (low-pass filtering due to head diffraction) or when the sound travels distances >15 m (high-frequency energy losses due to air absorption). Here, we study how the spectrum of a sound arriving from a source located in a reverberant room at intermediate distances (1–6 m) influences the perception of the distance to the source. First, we conducted an ADP experiment using pure tones (the simplest possible spectrum) of frequencies 0.5, 1, 2, and 4 kHz. Then, we performed a second ADP experiment with stimuli consisting of continuous broadband and bandpass-filtered (with center frequencies of 0.5, 1.5, and 4 kHz and bandwidths of 1/12, 1/3, and 1.5 octave) pink-noise clips. Our results showed an effect of the stimulus frequency on the perceived distance both for pure tones and filtered noise bands: ADP was less accurate for stimuli containing energy only in the low-frequency range. Analysis of the frequency response of the room showed that the low accuracy observed for low-frequency stimuli can be explained by the presence of sparse modal resonances in the low-frequency region of the spectrum, which induced a non-monotonic relationship between binaural intensity and source distance. The results obtained in the second experiment suggest that ADP can also be affected by stimulus bandwidth but in a less straightforward way (i.e., depending on the center frequency, increasing stimulus bandwidth could have different effects). Finally, the analysis of the acoustical cues suggests that listeners judged source distance using mainly changes in the overall intensity of the auditory stimulus with distance rather than the direct-to-reverberant energy ratio, even for low-frequency noise bands (which typically induce high amount of reverberation). The results obtained in this study show that, depending on the spectrum of the auditory stimulus, reverberation can degrade ADP rather than improve it.

Understanding the driving forces behind the shifts in the ecological balance of the oral microbiota will become essential for the future management and treatment of periodontitis. As the use of competitive approaches for modulating bacterial outgrowth is unexplored in the oral ecosystem, our study aimed to investigate both the associations among groups of functional compounds and the impact of individual substrates on selected members of the oral microbiome. We employed the Phenotype Microarray high-throughput technology to analyse the microbial cellular phenotypes of 15 oral bacteria. Multivariate statistical analysis was used to detect respiratory activity triggers and to assess similar metabolic activities. Carbon and nitrogen were relevant for the respiration of health-associated bacteria, explaining competitive interactions when grown in biofilms. Carbon, nitrogen, and peptides tended to decrease the respiratory activity of all pathobionts, but not significantly. None of the evaluated compounds significantly increased activity of pathobionts at both 24 and 48 h. Additionally, metabolite requirements of pathobionts were dissimilar, suggesting that collective modulation of their respiratory activity may be challenging. Flow cytometry indicated that the metabolic activity detected in the Biolog plates may not be a direct result of the number of bacterial cells. In addition, damage to the cell membrane may not influence overall respiratory activity. Our methodology confirmed previously reported competitive and collaborative interactions among bacterial groups, which could be used either as marker of health status or as targets for modulation of the oral environment.

In this work we report an illusion of proximity of a sound source created by a sonic crystal placed between the source and a listener. This effect seems, at first, paradoxical to naïve listeners since the sonic crystal is an obstacle formed by almost densely packed cylindrical scatterers. Even when the singular acoustical properties of these periodic composite materials have been studied extensively (including band gaps, deaf bands, negative refraction, and birrefringence), the possible perceptual effects remain unexplored. The illusion reported here is studied through acoustical measurements and a psychophysical experiment. The results of the acoustical measurements showed that, for a certain frequency range and region in space where the focusing phenomenon takes place, the sonic crystal induces substantial increases in binaural intensity, direct-to-reverberant energy ratio and interaural cross-correlation values, all cues involved in the auditory perception of distance. Consistently, the results of the psychophysical experiment revealed that the presence of the sonic crystal between the sound source and the listener produces a significant reduction of the perceived relative distance to the sound source.

Diverse microbial ecosystems underpin life in the sea. Among these microbes are many unicellular eukaryotes that span the diversity of the eukaryotic tree of life. However, genetic tractability has been limited to a few species, which do not represent eukaryotic diversity or environmentally relevant taxa. Here, we report on the development of genetic tools in a range of protists primarily from marine environments. We present evidence for foreign DNA delivery and expression in 13 species never before transformed and for advancement of tools for eight other species, as well as potential reasons for why transformation of yet another 17 species tested was not achieved. Our resource in genetic manipulation will provide insights into the ancestral eukaryotic lifeforms, general eukaryote cell biology, protein diversification and the evolution of cellular pathways.
