Male gladiator frogs of Hypsiboas Wagler, 1830 build nests on available substrate surrounding ponds and streams where female spawn eggs during the breeding period. Although gladiator frogs seem to show plasticity in the way they construct their nests, there is no study reporting if these species present preferences about microhabitat conditions for nest-building (mainly under subtropical climate). Predation pressure and environmental conditions have been considered major processes shaping the great diversity of reproductive strategies performed by amphibians, but microhabitat conditions should explain where to build a nest as well as how nest looks. This study aimed to test nest site selection for nest-building by Hypsiboas faber(Wied-Neuwied, 1821), determining which factors are related to nest site selection and nest features. The survey was conducted at margins of two permanent ponds in Southern Brazil. Habitat factors were evaluated in 18 plots with nest and 18 plots in the surrounding without nest (control), describing vegetation structure and heterogeneity, and substrate characteristics. Water temperature was measured inside the nest and in its adjacency. Nest features assessed were area, depth and temperature. Habitat characteristics differed between plots with and without nest. Microhabitat selected for nest-building was characterized by great vegetation cover and height, as well as shallower water and lower cover of organic matter in suspension than in plots without nest. Differences between temperature inside nest and in its adjacency were not observed. No relationship between nest features and habitat descriptors was evidenced. Results revealed that Hypsiboas faber does not build nests anywhere. Males seem to prefer more protected habitats, probably avoiding predation, invasion of conspecific males and inclement weather. Lack of differences between temperature inside- and outside-nest suggest that nest do not improve this condition for eggs and tadpole development. Nest architecture was not related to habitat characteristics, which may be determined by other factors, as nest checking by females before amplexus. Nest site selection should increase offspring survival as well the breeding success of Hypsiboas faber. RESUMO Machos de espécies de Hypsiboas Wagler, 1830 constroem ninhos no substrato disponível no entorno de corpos d'água; as fêmeas depositam os ovos no interior destes ninhos durante a reprodução da espécie. Embora estes machos apresentem plasticidade na escolha de sítios para construção dos ninhos, ainda não existem avaliações de preferências por condições específicas de microhabitat para a nidificação (principalmente em clima subtropical). A pressão de predação e as condições ambientais são consideradas os principais processos moldando a diversidade de estratégias reprodutivas em anfíbios. Entretanto, condições de microhábitat deveriam determinar tanto onde construir um ninho quanto as variações na arquitetura do ninho. Neste estudo testamos hipóteses de seleção de microhábitats para a nidificação por sapos-ferreiros Hypsiboas faber (Wied-Neuwied, 1821), determinando características de hábitat relacionados à seleção de sítios e à arquitetura dos ninhos. O estudo foi conduzido nas margens de duas lagoas permanentes no sul do Brasil. Variáveis caracterizando o substrato e a estrutura e heterogeneidade da vegetação foram medidas em 18 parcelas com ninho e 18 parcelas adjacentes sem ninho (controle). A temperatura da água foi medida dentro do ninho e na adjacência. Atributos do ninho foram a área, a profundidade e a temperatura. Resultados demonstraram diferenças nas características de hábitat entre parcelas com e sem ninho. Parcelas com ninhos apresentaram maior cobertura e altura da vegetação, lâmina de água pouca profunda e com pouca matéria orgânica em suspensão em relação às parcelas sem ninho. Não foi observada diferença entre a temperatura dentro e fora do ninho, bem como relação entre os atributos do ninho e os descritores do habitat. Os resultados evidenciaram a seleção de hábitat para a construção de ninhos por Hypsiboas faber. Os machos parecem utilizar habitats mais protegidos, provavelmente para evitar predação, a invasão de machos coespecíficos e a ação de intempéries climáticas. A ausência de diferença entre a temperatura dentro e fora do ninho sugere que o ninho não melhora condições para o desenvolvimento dos ovos e dos girinos. A arquitetura do ninho não teve relação com fatores ambientais; comportamentos como a checagem do ninho pela fêmea antes do amplexo podem estar envolvidos. A seleção de determinados sítios para a construção de ninho deve aumentar a chance de sobrevivência da prole e o sucesso reprodutivo de Hypsiboas faber.

Animais ectotérmicos, especialmente anuros, habitam diversos lugares do nosso planeta e desenvolveram algumas estratégias para garantir essa ampla distribuição. Alguns anuros sobrevivem em ambientes frios com temperaturas muito baixas devido a alterações metabólicas, como mobilização de crioprotetores (glicose, glicerol e ureia), armazenamento de glicogênio hepático e alterações na expressão de transportadores de membrana. O Rio Grande do Sul, sul do Brasil, apresenta temperaturas muito baixas no inverno, mas não há estudos sobre a adaptação de anuros durante o inverno nessa região. Neste estudo, o objetivo foi avaliar as estratégias fisiológicas adaptativas entre as diferentes estações do ano da espécie Sul-Americana Boana pulchella a fim de identificar as possíveis vias metabólicas que estariam modificadas sazonalmente e fornecer dados para o entendimento da biologia desses animais e de possíveis estratégias para criopreservação de tecidos. Para realização desse trabalho, foram coletados 33 indivíduos machos de B. pulchella, durante as quatro estações, em três diferentes cidades do estado do Rio Grande do Sul, Brasil. Imediatamente após a coleta, os animais foram conduzidos até o Laboratório de Endocrinologia e Metabolismo Comparado da Universidade Federal do Rio Grande do Sul, eutanasiados e seus tecidos coletados. Foi realizada a dosagem de concentração de glicose, glicerol, lactato e ureia no plasma. No tecido hepático e muscular foi medida a concentração de glicogênio, concentração de lactato, oxidação de glicose à dióxido de carbono (CO2) e síntese de glicogênio a partir de glicose. No fígado, foi determinada a concentração de glicerol, ureia e triglicerídeos e no tecido adiposo a concentração de triglicerídeos. As diferenças entre os grupos foram testadas pelo teste ANOVA de uma via seguida pelo pós-teste de Tukey nas variáveis paramétricas e pelo teste Kruskal Wallis seguido de pós-teste de Dunn para as não paramétricas. Os resultados mostraram que houve diferenças na concentração plasmática de glicose entre o outono e inverno/primavera e de glicerol entre o inverno e verão bem como na concentração muscular de lactato entre o inverno e verão. No verão, a oxidação de glicose a CO2 foi maior no verão no músculo e fígado e a síntese de glicogênio a partir de glicose foi maior no verão no tecido muscular. Através dos resultados obtidos podemos concluir que provavelmente no inverno há uma maior glicólise anaeróbica com produção de lactato comparada ao verão e uma maior utilização de glicose tanto para síntese de glicogênio quanto para oxidação durante o verão no tecido muscular. No fígado, os estoques de glicogênio são mantidos constantes ao longo do ano. No músculo, há uma maior concentração de glicogênio no outono juntamente com a menor concentração de glicose plasmática, indicando que há uma preparação para aumentar as reservas de glicogênio durante o outono, deixando-as disponíveis para o inverno. A ureia hepática parece estar ligada ao catabolismo proteico e a produção parece variar ao longo das estações, onde a menor concentração de ureia ocorre quando há maior gliconeogênese hepática a partir de lactato ou maior concentração de ureia quando é necessária a síntese de glicerol a partir de aminoácidos. Ectothermic animals, especially anurans, can survive in almost all environments and developed strategies to guarantee this wide distribution. Some anurans survive in cold environments with very low temperatures due to metabolic changes, such as cryoprotectant mobilization (glucose, glycerol and urea), hepatic glycogen storage and changes in the expression of membrane transporters. Rio Grande do Sul, southern state of Brazil, show very low temperatures in winter and, however, haven't have studies on the adaptation of anurans during winter in this region. In this study, the aim was to verify if the South American tree frog B. pulchella have adaptive physiological strategies between the different seasons of the year. In order to identify the possible metabolic pathways that would be seasonally modified and provide data for understanding the biology of these animals and possible strategies for tissue cryopreservation. To carry out this study, 33 males of B. pulchella were collected, during the four seasons of the year, in three different cities in the state of Rio Grande do Sul. Immediately after the capture, animals were brought to the Laboratory of Endocrinology and Compared Metabolism at the Federal University of Rio Grande do Sul for biochemical analysis. Plasma glucose, glycerol, lactate and urea were measured in plasma. In the liver and muscle tissue were measured glycogen concentration, lactate concentration, glucose oxidation to CO2 and glycogen synthesis from glucose. In the liver was determined the concentration of glycerol, urea and triglycerides and in the adipose tissue the concentration of triglycerides. Differences between groups were tested by one-way ANOVA test followed by Tukey post-test on the parametric variables and Kruskal Wallis test followed by Dunn post-test for the non-parametric ones. The results showed differences in the plasma concentration of glucose between autumn and winter/spring, of glycerol between winter and summer and in the lactate muscle concentration between winter and summer. In summer, glucose oxidation to carbon dioxide (CO2) was higher in muscle and liver and glycogen synthesis from glucose was higher in summer in muscle tissue. We can conclude by these results that probably in winter there is a higher anaerobic glycolysis with lactate production compared to summer and a higher use of glucose for glycogen synthesis and oxidation during summer in muscle tissue. In the liver, glycogen stores are kept constant throughout the year. In the muscle, the highest concentration of glycogen in the autumn was observed together with the lowest concentration of plasma glucose, which may indicate that there is a preparation to increase the glycogen reserves during the autumn, making them available for the winter. Liver urea appears to be linked to protein catabolism and production appears to vary over the year. This variation suggests that the lowest concentration of urea occurs when there is a higher hepatic gluconeogenesis from lactate and a higher concentration of urea when glycerol synthesis from amino acids is necessary.
