[Objective] The construction of limonin-albumin nanoparticles provides a theoretical basis for the further application of limonin. [Methods] Limonin-albumin nanoparticles were prepared by desolvation-chemical crosslinking method using bovine serum albumin as nanocarriers. The optimal preparation process was determined by single factor experiment and response surface test with embedding rate as index, and the nanoparticles were characterized. [Results] When the amount of bovine serum albumin was 64 mg, the amount of lemon bitter was 4 mg, the specific volume ratio of alcohol to water was 5∶1, and the amount of crosslinker was 125 μL, the average particle size and polydispersion index of limonin nanoparticles were (152.2±0.5) nm and 0.196±0.026 respectively. The embedding efficiency was (79.21±0.36)%. The limonin-albumin nanoparticles observed under low-pressure transmission electron microscopy showed a spherical shape. Infrared spectroscopy and X-ray diffraction results showed that limonin was successfully encapsulated in albumin in an amorphous or disordered state. The limonin-albumin nanoparticle suspension has good storage stability. [Conclusion] The limonin-albumin nanoparticles prepared by desolvation-chemical crosslinking method had good storage stability.

Objective: This study focused on the effectively control of the spoilage and deterioration of grouper during refrigeration. Methods: Illumina high-throughput sequencing technology was used to amplify the 16S rDNA V3-V4 segment of bacteria, then analyzed the changes in the bacterial community structure of E. obliquus after being treated with 400 MPa UHP for 5 minutes and refrigerated at 4 ℃ for 3, 6, 9, and 12 d. Results: According to Alpha diversity analysis fish samples treated with and without UHP showed increasing Chao1 and Ace indices as refrigeration times were extended, but Shannon indices progressively dropped. Nonetheless, the fish samples treated with UHP had lower Chao1, Ace, and Shannon indices than the ones not treated with UHP at every refrigeration stage (3~12 days). Analysis of microbial community structure showed there was a difference in the changes in microbial community structure during the refrigeration process of the two types of fish samples. During the refrigeration period, the main dominant microbial communities in fish samples without UHP were Streptococcus (17.70%, 3 d), Plesiomonas (36.66%, 6 d)， and Peptostreptococcus (67.65%, 9 d; 43.4%, 12 d), while the main dominant microbial communities in fish samples treated with UHP were unclassified Enterobacteriaceae (27.36%, 3 d), Plesiomonas (40.31%, 6 d), Lactococcus (24.45%, 9 d), and Peptostreptococcus (77.91%, 12 d). Meanwhile, Peptostreptococcus was identified as the specific dominant bacteria for the later stage of cold storage (12th day) of two processed fish samples. PICRUSt functional gene prediction found that the relative abundance of genes related to stress response, toxin transport, DNA recombination, and signal transduction in fish samples treated with and without UHP increased significantly on the 9th and 12th days of refrigeration, respectively.Conclusion: Compared with the samples without UHP, UHP treatment can effectively reduce the abundance and diversity of bacterial communities, change the succession pattern of the main dominant bacterial communities, and affect the abundance structure changes of the bacterial communities in the fish samples during refrigeration.

Clear cell renal cell carcinoma (ccRCC) is the most common type of renal cancers. However, circ_DENND1B has not been studied yet. GSE100186 dataset was used for the level analysis of circ_DENND1B. The quantitative real-time PCR was used to verify the expression of circ_DENND1B, microRNA-122-5p (miR-122-5p) and tissue inhibitor of metalloproteinases-2 (TIMP2) in ccRCC tissues and cells. Cell proliferation, migration, invasion and apoptosis were detected by colony formation assay, thymidine analog 5-ethynyl-2′-deoxyuridine assay, 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyl tetrazolium bromide, transwell and flow cytometry. The binding of miR-122-5p to circ_DENND1B/TIMP2 was investigated by dual-luciferase reporter assay. Finally, the role of circ_DENND1B in ccRCC was detected by tumorigenesis experiment in mice. circ_DENND1B was downregulated in ccRCC and circ_DENND1B overexpression suppressed the malignant behaviors of ccRCC cells. circ_DENND1B acted as a sponge of miR-122-5p. miR-122-5p upregulation reversed the effects of circ_DENND1B on cell proliferation, migration, invasion and apoptosis. TIMP2 was a target of miR-122-5p. Overexpression of circ_DENND1B regulated TIMP2 level by inhibiting miR-122-5p expression in ccRCC cells. circ_DENND1B overexpression inhibited the tumor growth of ccRCC in vivo. circ_DENND1B inhibited ccRCC cell progression by promoting TIMP2 expression by sponging miR-122-5p, suggesting that circ_DENND1B might be an effective therapeutic target for ccRCC.

A permeable pipe pile is proposed to accelerate the dissipation of excess pore-water pressure through drainage holes around the pile circumference into the pile's cavity. This investigation generalizes the permeable pile-soil interface as an impeded boundary, based on which a mathematical model for soil consolidation around the permeable pile is derived. Subsequently, the influence of opening pattern, drainage hole-to-pipe pile radius ratio, and opening ratio on the consolidation efficiency is numerically investigated. A total of 240 numerical cases with different drainage hole-to-pipe pile radius ratios, opening ratios, and hydraulic conductivity ratios are calculated to determine the permeable pile-soil interface parameter using back-analysis, and an approximate expression for interface parameter is obtained. Comparing against experimentally measured excess pore-water pressure dissipation, the proposed technique with impeded drainage boundary can provide good evaluations for drainage characteristics at the permeable pile-soil interface and consolidation behavior of the surrounding soil. It is found that the spatial distribution pattern of drainage holes has negligible influence on the consolidation efficiency. Increasing the opening ratio (with constant opening size) or reducing the drainage hole-to-pipe pile radius ratio (with constant opening ratio) can both accelerate the consolidation efficiency. Un pieu tubulaire permeable est propose pour accelerer la dissipation de l'exces de pression d'eau interstitielle par des trous de drainage autour de la circonference du pieu dans la cavite de celui-ci. Cette etude generalise l'interface pieu permeablesol en tant que limite entravee, sur la base de laquelle un modele mathematique pour la consolidation du sol autour des pieux permeables est derive. Par la suite, l'influence de la configuration des ouvertures, du rapport entre le rayon des trous de drainage et celui des pieux tubulaires, et du rapport des ouvertures sur l'efficacite de la consolidation est etudiee numeriquement. Un total de 240 cas numeriques avec differents rapports entre le rayon du trou de drainage et le rayon du pieu, rapports d'ouverture et rapports de conductivite hydraulique sont calcules pour determiner le parametre d'interface pieu permeable-sol en utilisant la retro-analyse, et une expression approximative pour le parametre d'interface est obtenue. Par rapport a la dissipation de la pression d'eau interstitielle excedentaire mesuree experimentalement, la technique proposee avec une limite de drainage entravee peut fournir de bonnes evaluations des caracteristiques de drainage a l'interface pieu permeable-sol et du comportement de consolidation du sol environnant. On constate que le schema de distribution spatiale des trous de drainage a une influence negligeable sur l'efficacite de la consolidation. L'augmentation du rapport d'ouverture (avec une taille d'ouverture constante) ou la reduction du rapport du rayon de trou de drainage et le pieu tubulaire (avec un rapport d'ouverture constant) peuvent toutes deux accelerer l'efficacite de la consolidation.
