Metallic nanoparticles are interesting areas of research due to their unique properties which can be advantageous for producing smart products. Silver nanoparticles (AgNPs) are remarkably used in pharmaceutical industry because oftheir strong biological activities. The aim of the present study was to investigate the cytotoxicity and wound healing capacity of the biologically prepared silver nanoparticles via green synthesis route. Cytotoxicity of the biogenic AgNPs was determined by MTT (3- [4, 5- dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium-bromide) assay against L929 fibroblast cell line. Wound healing properties of the AgNPs were evaluated using in vitro-scratch wound healing assay using 3T3 fibroblast cell line. Biosynthesized AgNPs inhibited the propagation of fibroblasts at a half maximal inhibitory concentration (IC50) of 23.507 μg/mL after 24 h incubation. In vitro wound healing assay also revealed that the biogenic AgNPs stimulated the 3T3 fibroblasts' cell proliferation. It can be suggested that biologically synthesized AgNPs can be used effectively for biomedical applications such as wound dressing materials. Metalik nanopartiküller, akıllı ürünler üretmek için avantajlı olabilecek benzersiz özelliklerinden dolayı ilgi çeken araştırma alanlarıdır. Gümüş nanopartiküller sahip oldukları güçlü biyolojik aktiviteler nedeniyle farmasötik endüstrisinde sıklıkla kullanılmaktadır. Mevcut çalışmanın amacı, yeşil sentez metodu üzerinden biyolojik olarak sentezlenmiş gümüş nanopartiküllerin sitotoksitesini ve yara iyileşme kapasitesini belirlemektir. Gümüş nanopartiküllerin sitotoksisitesi L929 fibroblast hücrelerine karşı MTT (3-[4,5-dimetiltiyazol-2-il]-2,5-difenil-tetrazolyum bromür) analiz yöntemiyle belirlenmiştir. Gümüş nanopartiküllerin yara iyileştirici özellikleri 3T3 fibroblast hücrelerine karşı in vitro çizik testi yöntemi ile belirlenmiştir. Biyosentetik gümüş nanopartiküllere ait 24 saat inkübasyon sonucunda hesaplanan IC50 değeri 23.507 μg/mL olarak belirlenmiştir. In vitro yara iyileşme testi sonucuna göre gümüş nanopartiküllerin 3T3 fibroblast hücrelerinin proliferasyonunu uyardığı belirlenmiştir. Çalışma sonucuna göre biyolojik olarak sentezlenen gümüş nanopartiküllerin yara örtüsü gibi biyomedikal uygulamalarda etkin şekilde kullanılabileceği öngörülmektedir.

Antioxidant, antimicrobial and antibiofilm activities of potassium metaborate (KBO2) was investigated within the present study. Antioxidant capacity of potassium metaborate was determined by β-carotene bleaching (BCB) assay and hydroxyl radical scavenging activity. Potassium metaborate was evaluated for its antimicrobial effects against selected Gram-positive bacteria, Gram-negative bacteria and a yeast via broth dilution method. The inhibition capability of potassium metaborate on the microbial biofilm formation of tested microorganisms was measured by microplate biofilm method using MTT (3- [4, 5- dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium-bromide). Biofilm inhibition capacity of potassium metaborate was also observed by Scanning Electron Microscope (SEM). Potassium metaborate was found to have the ability to scavenge hydroxyl radicals with an inhibition rate of 71.13% at 100 mM concentration. Antioxidant activity of potassium metaborate as determined by BCB assay gave higher result with an inhibition rate of 86.96% at the same concentration. According to the MIC (minimum inhibition concentration) values, the potassium metaborate inhibited the growth of C. albicans, S. aureus and E. coli at 62.5 mM concentrations while it was 31.25 mM for B. subtilis and 125 mM for P. aeruginosa. The highest antibiofilm activity was determined at the MIC of potassium metaborate with the reduction rate of 90.18% against C. albicans. It was concluded that, potassium metaborate have strong biological activities and can be effectively used for biomedical and environmental solutions.

Aim: The present study was aimed to characterize the surgical silk sutures coated with Styrax liquidus, Turkish sweetgum balsam obtained from Liquidambar orientalis, and to investigate their anti-adhesion capacity against oral pathogenic microorganisms.Material and Methods: Fourier Transform Infrared Spectroscopy (FTIR) was applied to determine the chemical composition of the Liquidambar orientalis styrax (LOS)-coated sutures. Thermogravimetric Analysis (TGA) was performed to compare the thermal stability of the LOS-coated sutures. Scanning Electron Microscopy (SEM) was used to evaluate the morphological structure of the sutures. Anti-adhesion activity of the LOS-coated sutures was investigated against common oral pathogenic microorganisms.Results: FTIR spectrum and SEM images revealed out that LOS was succesfully coated onto the silk sutures. TGA analysis showed that LOS coating moderately affected the thermal stability of the silk sutures. According to the anti-adhesion activity analysis, the highest activity was observed against S. aureus, a gram positive bacteria.Conclusion: Coating the surgical silk sutures with LOS might be useful to prevent the surgical site infections in oral surgery., Amaç: Bu çalışmada, Liquidambar orientalis'ten elde edilen Türk sığla balsamı olan Styrax liquidus ile kaplanmış cerrahi ipek sütürlerin karakterize edilmesi ve oral patojenik mikroorganizmalara karşı anti-adezyon kapasitelerinin araştırılması amaçlandı.Gereç ve Yöntemler: Liquidambar orientalis styrax (LOS) kaplı sütürlerin kimyasal bileşimini belirlemek için Fourier Transform Kızılötesi Spektroskopisi (FTIR) uygulandı. LOS kaplı sütürlerin termal stabilitesini karşılaştırmak için Termogravimetrik Analiz (TGA) yapıldı. Sütürlerin morfolojik yapısını değerlendirmek için Taramalı Elektron Mikroskopisi (SEM) kullanıldı. LOS kaplı sütürlerin anti-adezyon aktivitesi, yaygın oral patojenik mikroorganizmalara karşı araştırıldı.Bulgular: FTIR spektrumu ve SEM görüntüleri LOS'un ipek sütürlere başarılı bir şekilde kaplandığını ortaya koydu. TGA analizi, LOS kaplamanın ipek sütürlerin termal stabilitesini orta derecede etkilediğini gösterdi. Anti-adezyon aktivite analizine göre, en yüksek aktivitenin gram pozitif bakteri olan S. aureus'a karşı olduğu gözlendi.Sonuç: Cerrahi ipek sütürlerin LOS ile kaplanması, oral cerrahide cerrahi bölge enfeksiyonlarını önlemek için yararlı olabilmektedir.

Aerobic, non-fermentative and Gram-negative Stenotrophomonas maltophilia is a multidrug-resistant bacilli that is known to be pathogen for human and animals. S. maltophilia has been isolated from different animal species and also found in a variety of environments including soil, water, and plants. S. maltophilia, which has the ability to form biofilms on surfaces that cause environmental problems, is resistant to many antibiotic classes such as cephalosporins, carbapenems, and aminoglycosides. Here in this study, its aimed to determine the inhibition activities of natural phenolic compounds eugenol and pulegone against S. maltophilia MU69. Antibacterial activities of eugenol and pulegone were initially determined by disc diffusion method. Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were also determined by tube dilution assay. Antibiofilm activities of the compounds were investigated by crystal violet staining and also monitorized using Scanning Electron Microscopy (SEM). Zone of inhibition measurements were found to be 39 mm and 28 mm for eugenol and pulegone, respectively. MIC and MBC values were found to be 3.12 mg/mL for eugenol while these were 2.5 mg/mL and 5 mg/mL for pulegone, respectively. The highest antibiofilm activity was found to be 56.30±0.45% for MIC of eugenol. To our knowledge, this is the first report of the antibacterial and antibiofilm activity of eugenol and pulegone against S. maltophilia. According to the biological activity results, it can be concluded that these natural agents may be potentially used for veterinary sciences, food industry or pharmaceutical applications that aim to manage S. maltophilia biofilm. Aerobik, non-fermentatif ve gram-negatif Stenotrophomonas maltophilia, insan ve hayvanlar için patojen olarak bilinen çoklu ilaç direncine sahip bir basildir. Farklı hayvan türlerinden izole edilmiş S. maltophilia suşu toprak, su ve bitkiler gibi çeşitli çevresel etmenlerden de izole edilebilmektedir. Katı yüzeyler üzerinde biyofilm oluşturabilme yeteneği sayesinde S. maltophilia suşu çevre sorunlarına yol açmakla birlikte aminoglikozitler, karbapenemler ve sefalosporinler gibi çeşitli antibiyotik sınıflarına karşı direnç göstermektedir. Bu çalışmada doğal fenolik bileşikler olan eugenol ve pulegonun S. maltophila MU69 suşu üzerindeki inhibisyon aktivitelerinin belirlenmesi amaçlanmıştır. Eugenol ve pulegonun antibakteriyel aktivitesi disk difüzyon yöntemi ile araştırılmıştır. Minimum İnhibisyon Konsatrasyonu (MİK) ve Minimum Bakterisidal Konsantrasyon (MBK) ise tüp dilüsyon yöntemi ile belirlenmiştir. Antibiyofilm aktivitenin belirlenmesinde kristal viyole boyama yöntemi uygulanmış ve ayrıca Taramalı Elektron Mikroskobi (SEM) ile görüntülenmiştir. Eugenol ve pulegon için inhibisyon zonu sırasıyla 39 mm ve 28 mm olarak ölçülmüştür. MİK ve MBK değerleri eugenol için 3.12 mg/mL olarak belirlenirken, pulegon için sırasıyla 2.5 mg/mL ve 5 mg/mL olarak tespit edilmiştir. En yüksek antibiyofilm aktivite oranı % 56.30±0.45 ile eugenole ait MİK değeri için gözlenmiştir. Bu çalışma eugenol ve pulegon bileşiklerinin S. maltophilia suşuna karşı antibakteriyel ve antibiyofilm aktivitelerinin belirlendiği ilk çalışma olarak sunulmaktadır. Biyolojik aktivite çalışmalarının sonuçlarına göre bu iki doğal bileşik veteriner bilimleri, gıda endüstrisi ve farmasötik uygulamalarda S. maltophilia’ya karşı geliştirilen sistemlerde potansiyel kullanım kapasitesine sahiptir.

Narenciye esansiyel yağları cilt ve saç bakımı,uykusuzluk, sindirimin düzenlenmesi ve bağışıklık sisteminin geliştirilmesigibi pek çok alanda kullanıma sahiptir. Bu çalışma, Türkiye’de yetiştiriciliğiyapılmakta olan bir mandalina türü olan klementin’in (Citrus clementina Hort. ex Tan) kabuğundan elde edilen esansiyelyağın biyolojik aktivitelerininbelirlenmesi amacıyla planlanmıştır. Klementin kabuk esansiyel yağının (CPEO)antimikrobiyal aktivitesi E. coli, S. aureus ve bir maya türü olan C.albicans’a karşı disk difüzyon yöntemi ile belirlenmiştir. Sitotoksisitesi MTTkolorimetrik analizi ile NIH-3T3 fibroblast hücreleri kullanılarak tespitedilmiştir. Ayrıca stratch-yara iyileşme tekniği kullanılarak CPEO’nınfibroblast hücrelerinin göçü ve proliferasyonu üzerine etkinliği in vitro yaraiyileşme aktivitesi olarak değerlendirilmiştir. Sonuçlara göre CPEO’nın kaydadeğer antimikrobiyal aktiviteye sahip olduğu gözlenmiştir. En yüksek inhibisyonzonu C. albicans’a karşı 20.67±0.58mm olarak ölçülmüştür. Sitotoksik aktivite analizine göre IC50değeri 52.50 µg/mL olarakbelirlenmiştir. CPEO için IC50 değerinin altında uygulanandozlarda fibroblast hücreleri kontrol grubuna kıyasla daha hızlı süredeproliferasyon sağlayarak yapay yaranın iyileşmesine yönelik yüksek hareketgöstermiştir. Mevcut çalışma sonuçlarına göre CPEO, patojenikmikroorganizmalara karşı sahip olduğu antimikrobiyal karakterin yanı sıra ciltuygulamaları ve yara iyileşmeye yönelik ürünlerin geliştirilmesinde kullanılmapotansiyeline sahiptir., Citrus essentialoils has many area of use such as skin and hair care, insomnia, improvingdigestion, and boosting immune system. The present study is aimed to determinethe biological activities of clementine (Citrus clementina Hort. ex Tan), one of the mandarin species,peel essential oil (CPEO) that grow in Turkey. Antimicrobial activity of CPEOwas evaluated using disc diffusion method against Escherichia coli,Staphylococcus aureus, and a fungi Candida albicans. The MTT (3-[4,5-dimethylthiazol-2-yl]-2, 5 diphenyltetrazolium bromide)-colorimetric monocytemediated cytotoxicity assay was applied using NIH-3T3 cells and IC50value was calculated. CPEO was also evaluated for its in vitro wound healingeffect on the fibroblast cell migration and proliferation using scratch woundassay technique. Results of the present study indicated that CPEO hasremarkable antimicrobial activity. The highest inhibition zone was observedagainst C. albicans as 20.67±0.58 mm. IC50 value obtained for CPEO was 52.50 µg/mL. Fibroblast cells showed highermigration than the control group to heal the artificial scar after treatment ofCPEO concentrations below the IC50 dose. The results of the present study indicated that the CPEO may be useful ineffective management of skin care applications and wound healing products withits potent activity against pathogen microorganisms.
