En temel sinyal iletim basamaklarından biri olan Wnt/β-catenin, omurgalılarda embriyonik gelişimi ve yetişkin homeostazisini kontrol eden evrimsel olarak korunmuş bir sinyal yolağıdır. Doku homeostazında önemli bir role sahip olan Wnt/β-catenin sinyal yolağının anormal aktivasyonu birçok çeşit kanser ile bağlantılı olup, bu yolun hedef moleküllerindeki mutasyonların tümörijenik etkiye sahip olduğu belirlenmiştir.Laboratuvarımızda yürütülen daha önceki çalışmalar BRI3 (Beyin Protein 3) ve MGAT1 (Mannosil (Alfa-1,3 -) - Glikoprotein Beta-1,2-N-Asetilglukosaminiltransferaz) genlerinin Wnt/β-katenin yolağının yeni transkripsiyonel hedefleri olduğunu doğruladı. BRI3 veya MGAT1'i stabil şekilde eksprese eden hepatosellüler kanser hücreleri Huh7'lerin, doğal fenotip Huh7 hücrelerine kıyasla daha proliferatif ve istilacı yeteneklere sahip olduğu ve deri altından NUDE/SCID farelere enjekte edildiğinde daha büyük tümör oluşumu ile sonuçlandığı görülmüştür. MGAT1 ve BRI3 genlerinin hepatoselüler kanser oluşumuna katkısını aydınlatabilmek için araştırmaları daha derine indirerek RNA-dizileme yöntemine başvurulmuştur. Çeşitli hücresel yolaklarda görev alan belirli genlerde, kontrol ve BRI3 veya MGAT1 genlerinden birinin aşırı anlatımının sağlandığı hücreler arasında diferansiyel ekspresyon seviyeleri gözlemlenmiştir. Western Blot ve qRT-PCR teknikleri kullanılarak bu diferansiyel ekspresyon seviyeleri doğrulanmıştır. Kanser oluşumunun altında yatan mekanizmayı ortaya çıkarabilmek adına protein-protein fiziksel etkileşimleri biyoinformatik olarak incelenmiş ve farklı kombinasyonlarda proteinlerarası etkileşim saptanmıştır. Literatür araştırması da dahil edilerek BRI3 ve MGAT1 genlerinin hepatosellüler kanser oluşumuna katkısı seçilen genler üzerinden incelenmiş, ve aydınlatılmaya çalışılmıştır. One of the most fundamental signal transduction cascades, the Wnt/β-catenin signaling pathway, is an evolutionary conserved pathway in vertebrates which controls embryonic development and adult homeostasis. Signaling through secreted glycoproteins of Wnt family results in translocation of the transcriptional activator β-catenin into the nucleus where it controls key developmental gene expression programs. Having an important role in tissue homeostasis, aberrant activation of Wnt/β-catenin signaling pathway is often linked to a wide variety of cancers. Mutations in target molecules of this pathway are identified to have tumorigenic effect.Previous studies confirmed BRI3 (Brain Protein 3) and MGAT1 (Mannosyl (Alpha-1,3-)-Glycoprotein Beta-1,2-N-Acetylglucosaminyltransferase) genes to be novel transcriptional targets of Wnt/β-catenin pathway. Huh7 cells (hepatocellular cancer cell line) stably expressing either BRI3 or MGAT1 have greater proliferative and invasive capabilities compared to wildtype Huh7 cells, and when subcutaneously injected into NUDE/SCID mice resulted in larger tumor formation. In the light of these findings, tumors were subjected to RNA-Sequencing for further investigation. We observed differential expression of certain genes involved in several cellular pathways. Among these genes, we continued with YAP1, CSNK2B, and AFP in BRI3 expressing cells; and EDNRB, ADAM17, and HSPA8 in MGAT1 expressing cells. We validated differential expression levels by using Western Blot and qRT-PCR techniques. We further aim to shed light into the crosstalk between different signaling pathways involved in cancer progression. 113

Adipokinetik hormone (AKH) bir böcek nöropeptiti olup, insanlardaki gonadotropin-salgılayan hormonun ortoloğu olarak da bilinir. Corpora cardiaca'dan salgılanır ve enerji mobilizasyonu için genellikle depo lipitleri üzerinde etkisini gösterir. Kan hemolenfindeki trehaloz seviyesinin, kalp atışı sıklığının ve protein sentezinin artması gibi birçok çeşitli fizyolojik fonksiyonlarda da yer alırlar. Ayrıca bazı böcek türlerinde yağ asitlerinin ve RNA sentezinin inhibe edilmesinde de görev yaparlar. Kendilerine karşılık gelen adipokinetik hormone reseptörü (AKHR) olarak isimlendirilen G-protein kenetli reseptörleri (GPCR) aktive ederler. Şimdiye kadar birçok AKHR çalışılmış, ancak değnek böceği C.morosus'takiler çalışılmamıştır. Değnek böceğinin transkriptom verisini kullanılarak, bu böceğe ait adipokinetik hormone reseptör (CamAKHR) ve adipokinetik hormon (CamAKH) tanımlanmıştır. Değnek böceğine ait AKHR'nin homoloji modellemesi, kenetlenme analizi ve moleküler dinamik yöntemleri ile karakterize edilmesi ve bu sayede yeni bir nöropeptit-esaslı yeni-jenerasyon pestisit üretimine katkı sağlaması amaçlanmıştır. Çalışmanın sonunda ligandın bağlanmasında önem taşıyan on üç rezidünün, CamAKHR modelinin çoğunlukla transmembran 6, transmembran 7 ve hücre dışı ilmek 2 bölgelerinde yer aldığı gözlemlenmiştir. Bu on üç rezidüden altısı farklı böceklerde de korunmuş halde bulunduğundan, önerilen bağlanma cebi CamAKHR'nin TM2, TM3, TM6 ve TM7 bölgelerine denk gelen Glu246, Arg269, Tyr423, Tyr430, Lys446 and Phe449 rezidülerini içermektedir. Adipokinetic hormone (AKH) is an insect neuropeptide and known as gonadotropin- releasing hormone, which is orthologue to humans. It is released from corpora cardiaca and acts generally on fat body for energy mobilization. It is also involved in several different physiological functions such as increasing blood hemolymph trehalose levels, heart beat frequency, and protein synthesis. Furthermore it functions in inhibiting fatty acid and RNA synthesis in different insect species. It activates its corresponding G-Protein Coupled Receptors (GPCR), which are Adipokinetic Hormone receptors (AKHR). Many AKHRs were studied so far, but not in stick insect, Carausius morosus. Utilizing transcriptome data of stick insect, its AKHR (CamAKHR) and AKH (CamAKH) were identified. It was aimed to characterize AKHR of stick insect by homology modeling, docking analysis and molecular dynamics within this project contributing to the production of a novel neuropeptide-based next-generation pesticide. At the end, thirteen important residues mostly located in transmembrane 6, transmembrane 7 and extracellular loop 2 regions of CamAKHR model were observed contributing to ligand binding in CamAKHR- CamAKH complex. Six of them were found as the most conserved residues in different insect species, therefore proposed binding pocket includes these Glu246, Arg269, Tyr423, Tyr430, Lys446 and Phe449 residues corresponding to TM2, TM3, TM6 and TM7 of CamAKHR. 76

Evrimsel süreçte korunmuş bir yolak olan Wnt/β-katenin sinyal yolağının omurgalı erken gelişiminde, eksen oluşumunda, hücresel farklılaşma ve morfojenezde önemli görevleri vardır. Bu yolağın aktifleşmesi, β-catenin molekülünün hücre çekirdeğine taşınmasını ve orada T-hücresi faktörü/Lenfoid arttırıcı faktör (Tcf/Lef) ailesinden olan transkripsiyon faktörlerini aktifleştirerek gelişimsel ve hücre döngüsüyle alakalı genlerin anlatımlarını düzenler. Birçok farklı hücresel süreçteki görevinin dışında, Wnt/β-katenin sinyal yolağı aynı zamanda kanser gelişiminde de kritik öneme sahip hücre içi yolaklarından bir tanesidir. Bu yolağın açığa çıkarılan hedef moleküllerinden çok önemli bir kısmının mutasyona uğradıkları taktirde tümör oluşturucu karakterde oldukları bilinmektedir. Laboratuarımızda yapılan çalışmalarda, BRI3 geninin bu yolağın transkripsiyonel hedef genleri arasında bulunduğu doğrulanmıştır. Yeni hedef genlerin karakterizasyonu amacıyla birçok farklı yaklaşımdan faydalanılmıştır. Çalışmamızın ilk aşaması olarak, Maya ikili hibrit ve Co-IP teknikleri sayesinde IFITM3 ve MGAT1 proteinleri BRI3'ün bağlanma partnerleri olarak belirlenmiştir. BRI3 protein seviyesinin TNF-α muamelesine bağlı olarak yükseldiği görülmüş ve BRI3'ün yüksek anlatımının NFkB promotor aktivitesinde artışa yol açtığı belirlenmiştir. Öte yandan, Wnt/β-katenin sinyal yolağının varsayılan hedef genlerinden biri olan MGAT1'ın β-katenin aktifleşmesi sonucu anlatımının arttığı saptanmıştır. Yapılan hücre çoğalması ve yerdeğiştirimi deneylerinde BRI3 ve MGAT1 genlerinin yüksek anlatımları sağlanan Huh7 hücrelerinin kontrol Huh7 hücrelerine göre proliferatif ve invazif kapasitelerinin daha yüksek olduğu belirlenmiştir. In vivo ksenograft deneylerinde ise bu genlerin her birinin yüksek anlatımları sağlanan Huh7 hücre hatlarının NUDE/SCID farelerin deri altlarına enjekte edilmesi sonucunda tümör büyüme hızında artış görülmüştür. Oluşan tümörler RNA sekanslama yöntemi kullanılarak transkriptomik analize tabi tutulmuş ve bu sayede kanserin oluşma sürecinde hangi genlerin ve sinyal yolaklarının rolü olduğunun belirlenmesi amaçlanmıştır. The Wnt/β-catenin signaling pathway is an evolutionary conserved pathway which has important functions in vertebrate early development such as axis formation, cellular proliferation and morphogenesis. The activation of this pathway leads to translocation of the transcriptional activator β-catenin into the nucleus where it activates T-cell factor/Lymphoid enhancer factor (Tcf/Lef) family of transcription factors, which regulate expression of developmental and cell cycle-related genes. Apart from its roles in various cellular processes, Wnt/β-catenin signaling pathway is also one of the most important intracellular pathways implicated cancer progression. A significant number of identified target molecules of this pathway, are known to have tumorigenic characters when mutated.Previous studies confirmed BRI3 (Brain protein I3) gene to be one of the transcriptional target genes of Wnt/β-catenin pathway. We used various approaches for the functional characterization of its novel targets. As a first step in our study, IFITM3 and MGAT1 proteins were confirmed as interaction partners for BRI3 by Yeast-two Hybrid and Co-IP techniques. BRI3 was found to be upregulated in response to TNF-α treatment and overexpression of BRI3 resulted in an increase in NFkB promoter activity. On the other hand, MGAT1 is a putative novel target of Wnt/β-catenin signaling pathway and determined to be upregulated in response to β-catenin activation. Cell proliferation and migration assays showed that, Huh7 cells stably expressing each of the BRI3 and MGAT1 genes have greater proliferative and invasive capabilities compared to control Huh7 cells. Furthermore, in vivo xenograft experiments were performed and it was determined that the stable overexpression of both of these genes in Huh7 cell lines lead to increased rate of tumor growth in NUDE/SCID mice. The resulting tumors were subjected to transcriptomic analyses using RNA-Sequencing technique in order to determine which pathways and biological processes take part in the cancer initiation process. 151

Allatostatinler (AST), böceklerde jüvenil (gençlik) hormonu (JH) üretimini ve beslenme davranışlarını baskılayan çok işlevli nöropeptidlerdir. Allatostatinler FGLa/AST (A-türü), MIP/ASTs (B-türü) ve PISCF/ASTs (C-türü) olmak üzere üç sınıfa ayrılır. Her bir Allatostatin türü, işlevini Allatostatin reseptörü (AstR) olarak adlandırılan, kendine özgü G protein kenetli reseptör sınıfından bir proteine bağlanarak gösterir. Böceklerde JH üretiminin nasıl düzenlendiği ve AST/AstR sisteminin bu süreçteki rolü hakkında çok az bilgi mevcuttur. Ayrıca, böcek gelişimi ve beslenmesi ile ilişkili süreçlerde AstR'lerin baskılayıcı rolü nedeniyle, bu reseptörler pestisit geliştirilmesinde potansiyel bir hedeftir. Daha önce, bizim grubumuz tarafından Afrika kökenli istilacı bir böcek olan Carausius morosus'tan diğer böcek PISCF/AST reseptörleri ile yüksek bir dizi benzerliği gösteren bir transcript belirlenmiştir. Bu çalışmada, FRET yöntemi ile heterolog bir gen ifade sisteminde, bahsedilen PISCF/AST reseptörünün farmakolojik özellikleri ve erken sinyal yolakları belirlenmiştir. Reseptörü ifade eden hücrelerin PISCF/AST muamelesi ile birlikte, Gαi1 ve Gαo proteinlerinin doza bağımlı olarak etkin hale geldiği gözlenmiştir. Reseptörün Gβγ alt birimleri ile etkileşimi Gαi1 ve Gαo proteinlerinin dışında hiç bir Gα alt biriminin varlığında gözlenmemiştir. Adenilil siklaz 5 aracılı cAMP üretiminin ise PISCF/AST muamelesi ile kısmi olarak engellendiği, ve reseptörün β-arrestin2 ile doza bağımlı olarak etkileştiği belirlenmiştir. Kısaca, Carausius morosus'a ait bir PISCF/AST reseptörü işlevsel olarak nitelendirilmiş, ve Drosophila PISCF/AST peptidi ile fonksiyonel olarak etkin hale gelirken, Drosophila FGLa/AST peptidi ile etkin hale gelmediği gösterilmiştir. Allatostatins (ASTs) are multifunctional insect neuropeptides that suppress juvenile hormone (JH) synthesis and feeding behavior. Three different classes of ASTs in insects are known as FGLa/ASTs (A-type), MIP/ASTs (B-type) and PISCF/ASTs (C-type). Each type of ASTs exert its activity through binding to its cognate G protein coupled receptor families, called as Allatostatin receptors (AstRs). Very little is known on how JH synthesis is regulated and how AST/AstR system regulates this process. AstRs are also potential targets for pesticide development, since their activation negatively modulates insect growth and feeding-related processes. A transcript from an invasive African pest, Carausius morosus, which shows high sequence similarity to other insect PISCF/AST receptors was previously identified by our group. In this study, we characterized the pharmacological properties and early signaling events of this PISCF/AST receptor (CamAstRC) by means of FRET using a heterologous expression system. A dose-dependent activation of Gαi1 and Gαo were detected upon stimulation of CamAstR-C. Receptor coupling with Gβγ subunits was observed only in the presence of Gαi1 or Gαo, but not other Gα subunits. A partial inhibition of adenylyl cyclase 5-mediated cAMP production upon receptor activation with PISCF/AST and a dose-dependent interaction between β-arrestin2 and the receptor was detected. In summary, a PISCF/AST receptor from Carausius morosus has been functionally characterized, activated by Drosophila PISCF/AST, but not Drosophila FGLa/ASTs. 126
