Abstract Oats (Avena sativa L.) provide unique nutritional benefits and contribute to sustainable agricultural systems. Breeding high‐value oat varieties that meet milling industry standards is crucial for satisfying the demand for oat‐based food products. Test weight, thins, and groat percentage are primary traits that define oat milling quality and the final price of food‐grade oats. Conventional selection for milling quality is costly and burdensome. Multi‐trait genomic selection (MTGS) combines information from genome‐wide markers and secondary traits genetically correlated with primary traits to predict breeding values of primary traits on candidate breeding lines. MTGS can improve prediction accuracy and significantly accelerate the rate of genetic gain. In this study, we evaluated different MTGS models that used morphometric grain traits to improve prediction accuracy for primary grain quality traits within the constraints of a breeding program. We evaluated 558 breeding lines from the University of Illinois Oat Breeding Program across 2 years for primary milling traits, test weight, thins, and groat percentage, and secondary grain morphometric traits derived from kernel and groat images. Kernel morphometric traits were genetically correlated with test weight and thins percentage but were uncorrelated with groat percentage. For test weight and thins percentage, the MTGS model that included the kernel morphometric traits in both training and candidate sets outperformed single‐trait models by 52% and 59%, respectively. In contrast, MTGS models for groat percentage were not significantly better than the single‐trait model. We found that incorporating kernel morphometric traits can improve the genomic selection for test weight and thins percentage.

Abstract Einkorn wheat (Triticum monococcum) is an ancient grain crop and a close relative of the diploid progenitor (T. urartu) of polyploid wheat. It is the only diploid wheat species having both domesticated and wild forms and therefore provides an excellent system to identify domestication genes and genes for traits of interest to utilize in wheat improvement. Here, we leverage genomic advancements for einkorn wheat using an einkorn reference genome assembly combined with skim-sequencing of a large genetic population of 812 recombinant inbred lines (RILs) developed from a cross between a wild and a domesticated T. monococcum accession. We identify 15,919 crossover breakpoints delimited to a median and average interval of 114 Kbp and 219 Kbp, respectively. This high-resolution mapping resource enables us to perform fine-scale mapping of one qualitative (red coleoptile) and one quantitative (spikelet number per spike) trait, resulting in the identification of small physical intervals (400 Kb to 700 Kb) with a limited number of candidate genes. Furthermore, an important domestication locus for brittle rachis is also identified on chromosome 7A. This resource presents an exciting route to perform trait discovery in diploid wheat for agronomically important traits and their further deployment in einkorn as well as tetraploid pasta wheat and hexaploid bread wheat cultivars.

This Letter reports the first observation of the onset of fully developed turbulence in the solar corona. Long time series of white-light coronal images, acquired by Metis aboard Solar Orbiter at 2 minutes cadence and spanning about 10 hr, were studied to gain insight into the statistical properties of fluctuations in the density of the coronal plasma in the time domain. From pixel-by-pixel spectral frequency analysis in the whole Metis field of view, the scaling exponents of plasma fluctuations were derived. The results show that, over timescales ranging from 1 to 10 hr and corresponding to the photospheric mesogranulation-driven dynamics, the density spectra become shallower moving away from the Sun, resembling a Kolmogorov-like spectrum at 3 R _⊙ . According to the latest observation and interpretive work, the observed 5/3 scaling law for density fluctuations is indicative of the onset of fully developed turbulence in the corona. Metis observation-based evidence for a Kolmogorov turbulent form of the fluctuating density spectrum casts light on the evolution of 2D turbulence in the early stages of its upward transport from the low corona.

The distribution of turbulence in the heliosphere remains a mystery, due to the complexity in not only modeling the turbulence transport equations but also identifying the drivers of turbulence that vary with time and spatial location. Beyond the ionization cavity (a few astronomical units (AU) from the Sun), the turbulence is driven predominantly by freshly created pickup ions (PUIs), in contrast to the driving by stream shear and compression. Understanding the source characteristics is necessary to refine turbulence transport models and interpret measurements of turbulence and solar wind temperature in the outer heliosphere. Using a recent latitude-dependent solar wind speed model and the ionization rate of neutral interstellar hydrogen (H), we investigate the temporal and spatial variation in the strength of low-frequency turbulence driven by PUIs from 1998 to 2020. We find that the driving rate is stronger during periods of high solar activity and at lower latitudes in the outer heliosphere. The driving rates for parallel and anti-parallel propagating (relative to the background magnetic field) slab turbulence have different spatial and latitude dependences. The calculated generation rate of turbulence by PUIs is an essential ingredient to investigate the latitude dependence of turbulence in the outer heliosphere, which is important to understand the heating of the distant solar wind and the modulation of cosmic rays.

Genetic diversity found in crop wild relatives is critical to preserve and utilize for crop improvement to achieve sustainable food production amid climate change and increased demand. We genetically characterized a large collection of 1,041 Aegilops accessions distributed among 23 different species using more than 45K single nucleotide polymorphisms identified by genotyping-by-sequencing. The Wheat Genetics Resource Center (WGRC) Aegilops germplasm collection was curated through the identification of misclassified and redundant accessions. There were 49 misclassified and 28 sets of redundant accessions within the four diploid species. The curated germplasm sets now have improved utility for genetic studies and wheat improvement. We constructed a phylogenetic tree and principal component analysis cluster for all Aegilops species together, giving one of the most comprehensive views of Aegilops. The Sitopsis section and the U genome Aegilops clade were further scrutinized with in-depth population analysis. The genetic relatedness among the pair of Aegilops species provided strong evidence for the species evolution, speciation, and diversification. We inferred genome symbols for two species Ae. neglecta and Ae. columnaris based on the sequence read mapping and the presence of segregating loci on the pertinent genomes as well as genetic clustering. The high genetic diversity observed among Aegilops species indicated that the genus could play an even greater role in providing the critical need for untapped genetic diversity for future wheat breeding and improvement. To fully characterize these Aegilops species, there is an urgent need to generate reference assemblies for these wild wheats, especially for the polyploid Aegilops.

Primary systemic amyloidosis is a systemic disease characterised by the deposition of misfolded proteins extracellularly in different organs without any known cause in the background, eventually leading to multiorgan dysfunction and death. The incidence of primary amyloidosis is estimated at 5.1-12.8 cases per million, with a poor prognosis. We report a case of a 69-year male with lower back pain, shortness of breath, and anasarca diagnosed as primary systemic amyloidosis by serum-free light chain assay and kidney needle biopsy. He was started on intravenous bortezomib and dexamethasone. Though he adhered to his medications, with time he developed renal insufficiency marked by azotemia following which hemodialysis was performed. Primary systemic amyloidosis is a rare clinical condition with a very poor prognosis. Further studies are needed to understand the proper pathophysiology and treatment of the disease.

Abstract The development of next-generation sequencing (NGS) enabled a shift from array-based genotyping to directly sequencing genomic libraries for high-throughput genotyping. Even though whole-genome sequencing was initially too costly for routine analysis in large populations such as breeding or genetic studies, continued advancements in genome sequencing and bioinformatics have provided the opportunity to capitalize on whole-genome information. As new sequencing platforms can routinely provide high-quality sequencing data for sufficient genome coverage to genotype various breeding populations, a limitation comes in the time and cost of library construction when multiplexing a large number of samples. Here we describe a high-throughput whole-genome skim-sequencing (skim-seq) approach that can be utilized for a broad range of genotyping and genomic characterization. Using optimized low-volume Illumina Nextera chemistry, we developed a skim-seq method and combined up to 960 samples in one multiplex library using dual index barcoding. With the dual-index barcoding, the number of samples for multiplexing can be adjusted depending on the amount of data required, and could be extended to 3,072 samples or more. Panels of doubled haploid wheat lines (Triticum aestivum, CDC Stanley x CDC Landmark), wheat-barley (T. aestivum x Hordeum vulgare) and wheat-wheatgrass (Triticum durum x Thinopyrum intermedium) introgression lines as well as known monosomic wheat stocks were genotyped using the skim-seq approach. Bioinformatics pipelines were developed for various applications where sequencing coverage ranged from 1 × down to 0.01 × per sample. Using reference genomes, we detected chromosome dosage, identified aneuploidy, and karyotyped introgression lines from the skim-seq data. Leveraging the recent advancements in genome sequencing, skim-seq provides an effective and low-cost tool for routine genotyping and genetic analysis, which can track and identify introgressions and genomic regions of interest in genetics research and applied breeding programs.

Most stars generate winds and move through the interstellar medium that surrounds them. This movement creates a cocoon formed by the deflection of these winds that envelops and protects the stars. We call these “cocoons” astrospheres. The Sun has its own cocoon, the heliosphere. The heliosphere is an immense shield that protects the Solar System from harsh, galactic radiation. The radiation that enters the heliosphere affects life on Earth as well as human space exploration. Galactic cosmic rays are the dominant source of radiation and principal hazard affecting space missions within our Solar System. Current global heliosphere models do not successfully predict the radiation environment at all locations or under different solar conditions. To understand the heliosphere’s shielding properties, we need to understand its structure and large-scale dynamics. A fortunate confluence of missions has provided the scientific community with a treasury of heliospheric data. However, fundamental features remain unknown. The vision of the Solar wind with Hydrogen Ion charge Exchange and Large-Scale Dynamics (SHIELD) DRIVE Science Center is to understand the nature and structure of the heliosphere. Through four integrated research thrusts leading to the global model, SHIELD will: 1) determine the global nature of the heliosphere; 2) determine how pickup ions evolve from “cradle to grave” and affect heliospheric processes; 3) establish how the heliosphere interacts with and influences the Local Interstellar Medium (LISM); and 4) establish how cosmic rays are filtered by and transported through the heliosphere. The key deliverable is a comprehensive, self-consistent, global model of the heliosphere that explains data from all relevant in situ and remote observations and predicts the radiation environment. SHIELD will develop a “digital twin” of the heliosphere capable of: (a) predicting how changing solar and LISM conditions affect life on Earth, (b) understanding the radiation environment to support long-duration space travel, and (c) contributing toward finding life elsewhere in the Galaxy. SHIELD also will train the next-generation of heliophysicists, a diverse community fluent in team science and skilled working in highly transdisciplinary collaborative environments.

We study the solar cycle dependence of various turbulence cascade rates based on the methodology developed by Adhikari et al. that utilizes Kolmogorov phenomenology. This approach is extended to derive the heating rates for an Iroshnikov–Kriachnan (IK) phenomenology. The observed turbulence cascade rates corresponding to the total turbulence energy, fluctuating magnetic energy density, fluctuating kinetic energy, and the normalized cross helicity are derived from WIND spacecraft plasma and magnetometer data from 1995 through 2020. We find that (i) the turbulence cascade rate derived from a Kolmogorov phenomenology and an IK phenomenology changes with solar cycle, such that the cascade rate is largest during solar maximum and smallest during solar minimum; (ii) the turbulence energy Kolmogorov cascade rate increases from θ _UB (angle between mean magnetic field and velocity) = 0° to 90° and peaks near θ _UB = 90°, and then decreases as θ _UB tends to 180°; (iii) the 2D turbulence heating rate is larger than the slab heating rate; (iv) the 2D and slab fluctuating magnetic energy density cascade rates are larger than the corresponding cascade rates of the fluctuating kinetic energy; and (v) the total turbulence energy cascade rate is positively correlated with the solar wind speed and temperature and the normalized cross-helicity cascade rate. Finally, we find that the total turbulent energy Kolmogorov cascade rate is larger than the IK cascade rate.

Analytical solutions for 2D and slab turbulence energies in the solar corona are presented, including a derivation of the corresponding correlation lengths, with implications for the proton and electron temperatures in the solar corona. These solutions are derived by solving the transport equations for 2D and slab turbulence energies and their correlation lengths, as well as proton and electron pressures. The solutions assume background profiles for the solar wind speed, solar wind mass density, and Alfvén velocity. Our analytical solutions can be related to those obtained from joint Parker Solar Probe and Solar Orbiter Metis coronagraph observations, as reported in Telloni et al. We find that the solution for 2D turbulence energy in the absence of nonlinear dissipation decreases more slowly compared to the dissipative solution. The solution for slab turbulence energy with no dissipation exhibits a more rapid increase compared to the dissipative solution. The proton heating rate is found to be about 82% of the total plasma heating rate at 6.3 R _⊙ , which gradually decreases with increasing distance, eventually becoming ∼80% of the total plasma heating rate at ∼13 R _⊙ , consistent with that found by Bandyopadhyay et al. (2023). These analytical solutions provide valuable insight for our understanding of turbulence, and its effect on proton and electron heating rates, in the solar corona. We compare the numerically solved turbulent transport equations for the 2D and slab turbulence energies, correlation lengths, and proton and electron pressures with the analytical solutions, finding good agreement between them.

We study solar wind turbulence anisotropy in the inertial and energy-containing ranges in the inbound and outbound directions during encounters 1–9 by the Parker Solar Probe (PSP) for distances between ∼21 and 65 R _⊙ . Using the Adhikari et al. approach, we derive theoretical equations to calculate the ratio between the 2D and slab fluctuating magnetic energy, fluctuating kinetic energy, and the outward/inward Elsässer energy in the inertial range. For this, in the energy-containing range, we assume a wavenumber k ^−1 power law. In the inertial range, for the magnetic field fluctuations and the outward/inward Elsässer energy, we consider that (i) both 2D and slab fluctuations follow a power law of k ^−5/3 , and (ii) the 2D and slab fluctuations follow the power laws with k ^−5/3 and k ^−3/2 , respectively. For the velocity fluctuations, we assume that both the 2D and slab components follow a k ^−3/2 power law. We compare the theoretical results of the variance anisotropy in the inertial range with the derived observational values measured by PSP, and find that the energy density of 2D fluctuations is larger than that of the slab fluctuations. The theoretical variance anisotropy in the inertial range relating to the k ^−5/3 and k ^−3/2 power laws between 2D and slab turbulence exhibits a smaller value in comparison to assuming the same power law k ^−5/3 between 2D and slab turbulence. Finally, the observed turbulence energy measured by PSP in the energy-containing range is found to be similar to the theoretical result of a nearly incompressible/slab turbulence description.

The nature and radial evolution of solar wind electrons in the suprathermal energy range are studied. A wave–particle interaction tensor and a Fokker–Planck Coulomb collision operator are introduced into the kinetic transport equation describing electron collisions and resonant interactions with whistler waves. The diffusion tensor includes diagonal and off-diagonal terms, and the Coulomb collision operator applies to arbitrary electron velocities describing collisions with both background protons and electrons. The background proton and electron densities and temperatures are based on previous turbulence models that mediate the supersonic solar wind. The electron velocity distribution functions and electron heat flux are calculated. Comparison and analysis of the numerical results with analytical solutions and observations in the near-Sun region are made. The numerical results reproduce well the creation of the sunward electron deficit observed in the near-Sun region. The deficit of the electron velocity distribution function below the core Maxwellian fit at low velocities results from Coulomb collisions, and the excess part above the core Maxwellian fit at high velocities is determined by strong wave–particle interactions.

Interplanetary (IP) shocks are believed to play a significant role in both amplifying the background level of turbulent fluctuations and in heating the bulk solar wind (SW). This study investigates the thermodynamic properties downstream of IP shocks. We examine the temperature, density, and specific entropy changes in the shocked plasma, taking into consideration the geometric aspects of IP shock propagation within the expanding SW. Specifically, in our analysis, we account for the fact that any particular temporal range of one-point measurement may correspond to vastly different physically relevant temporal and/or spatial dimensions, such as the age of the shocked plasma and/or radial distance to the place where the plasma encountered the shock. Thus, our approach resolves the contradictions in previously reported temperature and specific entropy profiles in downstream regions and suggests that downstream regions exhibit greater turbulent heating compared to the pristine SW. This may contribute to the overall heating of the SW plasma. The paper presents a phenomenological parameter to predict specific entropy profiles and demonstrates the consistency of the proposed model with observations. We discuss the implications of these results for the thermodynamics of the SW beyond 1 au.

This paper summarizes the Seven Sisters solar wind mission concept and the outstanding science questions motivating the mission science objectives. The Seven Sisters mission includes seven individual spacecraft designed to uncover fundamental physical processes in the solar wind and provides up to ≈ 2 days of advanced space weather warnings for 550 Earth days during the mission. The mission will collect critical measurements of the thermal and suprathermal plasma and magnetic fields, utilizing, for the first time, Venus–Sun Lagrange points. The multi-spacecraft configuration makes it possible to distinguish between spatial and temporal changes, define gradients, and quantify cross-scale transport in solar wind structures. Seven Sisters will determine the 3-D structure of the solar wind and its transient phenomena and their evolution in the inner heliosphere. Data from the Seven Sisters mission will allow the identification of physical processes and the quantification of the relative contribution of different mechanisms responsible for suprathermal particle energization in the solar wind.

This Letter reports the first observational estimate of the heating rate in the slowly expanding solar corona. The analysis exploits the simultaneous remote and local observations of the same coronal plasma volume, with the Solar Orbiter/Metis and the Parker Solar Probe instruments, respectively, and relies on the basic solar wind magnetohydrodynamic equations. As expected, energy losses are a minor fraction of the solar wind energy flux, since most of the energy dissipation that feeds the heating and acceleration of the coronal flow occurs much closer to the Sun than the heights probed in the present study, which range from 6.3 to 13.3 R _⊙ . The energy deposited to the supersonic wind is then used to explain the observed slight residual wind acceleration and to maintain the plasma in a nonadiabatic state. As derived in the Wentzel–Kramers–Brillouin limit, the present energy transfer rate estimates provide a lower limit, which can be very useful in refining the turbulence-based modeling of coronal heating and subsequent solar wind acceleration.

Abstract Background Winter freezing temperature impacts alfalfa (Medicago sativa L.) persistence and seasonal yield and can lead to the death of the plant. Understanding the genetic mechanisms of alfalfa freezing tolerance (FT) using high-throughput phenotyping and genotyping is crucial to select suitable germplasm and develop winter-hardy cultivars. Several clones of an alfalfa F1 mapping population (3010 x CW 1010) were tested for FT using a cold chamber. The population was genotyped with SNP markers identified using genotyping-by-sequencing (GBS) and the quantitative trait loci (QTL) associated with FT were mapped on the parent-specific linkage maps. The ultimate goal is to develop non-dormant and winter-hardy alfalfa cultivars that can produce extended growth in the areas where winters are often mild. Results Alfalfa FT screening method optimized in this experiment comprises three major steps: clone preparation, acclimation, and freezing test. Twenty clones of each genotype were tested, where 10 samples were treated with freezing temperature, and 10 were used as controls. A moderate positive correlation (r ~ 0.36, P

Cold stress (CS) affects the survivability, geographical distribution, and yield stability of crops. Suitable management and agronomic practices can minimize the crop losses associated with cooler environments. However, agronomic practices alone can't support plants adequately to withstand the harsh cold. Therefore, exploring plants cold stress-responsive factors such as genetic, epigenetic, physiological, and cellular is crucial. This report discusses on cold stress effect, signal perception, signal transduction, gene expression, and associated molecular phenomena in plants. Three cold acclimation response pathways: Ca2+ mediated ICE1- CBF/ DREB1, hormonal, and reactive oxygen species (ROS), are elucidated. Also, this report summarizes the latest research work on genetics and genomics of forage species from the perspectives of cold tolerance improvement. In several instances, our hypotheses have been supported by a recent research output from our genetic analysis experiment on alfalfa (Medicago sativa L.) cold tolerance. We further review the importance of high-throughput genomics and phenomics for cold tolerance improvement in forage species and recommended implementing widely recognized techniques such as genomic selection (GS) and genome-wide association studies (GWAS) to develop climate-resilient cultivars. The transgenics and genome-edited cold-tolerant forage cultivars with low or no yield penalty must be the goals of future research.

This paper addresses the first direct investigation of the energy budget in the solar corona. Exploiting joint observations of the same coronal plasma by Parker Solar Probe and the Metis coronagraph aboard Solar Orbiter and the conserved equations for mass, magnetic flux, and wave action, we estimate the values of all terms comprising the total energy flux of the proton component of the slow solar wind from 6.3 to 13.3 R _⊙ . For distances from the Sun to less than 7 R _⊙ , we find that the primary source of solar wind energy is magnetic fluctuations including Alfvén waves. As the plasma flows away from the low corona, magnetic energy is gradually converted into kinetic energy, which dominates the total energy flux at heights above 7 R _⊙ . It is found too that the electric potential energy flux plays an important role in accelerating the solar wind only at altitudes below 6 R _⊙ , while enthalpy and heat fluxes only become important at even lower heights. The results finally show that energy equipartition does not exist in the solar corona.

Evidence for the presence of ion cyclotron waves (ICWs), driven by turbulence, at the boundaries of the current sheet is reported in this paper. By exploiting the full potential of the joint observations performed by Parker Solar Probe and the Metis coronagraph on board Solar Orbiter, local measurements of the solar wind can be linked with the large-scale structures of the solar corona. The results suggest that the dynamics of the current sheet layers generates turbulence, which in turn creates a sufficiently strong temperature anisotropy to make the solar-wind plasma unstable to anisotropy-driven instabilities such as the Alfvén ion cyclotron, mirror-mode, and firehose instabilities. The study of the polarization state of high-frequency magnetic fluctuations reveals that ICWs are indeed present along the current sheet, thus linking the magnetic topology of the remotely imaged coronal source regions with the wave bursts observed in situ. The present results may allow improvement of state-of-the-art models based on the ion cyclotron mechanism, providing new insights into the processes involved in coronal heating.

Abstract Background The genetic and genomic basis of flowering time and biomass yield in alfalfa (Medicago sativa L.) remains poorly understood mainly due to the autopolyploid nature of the species and the lack of adequate genomic resources. We constructed linkage maps using genotyping-by-sequencing (GBS) based single dose allele (SDA) SNP and mapped alfalfa timing of flowering (TOF), spring yield (SY), and cumulative summer biomass (CSB) in a pseudo-testcross F1 population derived from a fall dormant (3010) and a non-dormant (CW 1010) cultivars. We analyzed the quantitative trait loci (QTL) to identify conserved genomic regions and detected molecular markers and potential candidate genes associated with the traits to improve alfalfa and provide genomic resources for the future studies. Results This study showed that both fall dormant and non-dormant alfalfa cultivars harbored QTL for early and late flowering, suggesting that flowering time in alfalfa is not an indicator of its fall dormancy (FD) levels. A weak phenotypic correlation between the flowering time and fall dormancy (FD) in F1 and checks also corroborated that alfalfa FD and TOF are not the predictors of one another. The relationship between flowering time and alfalfa biomass yield was not strong, but the non-dormant had relatively more SY than dormant. Therefore, selecting superior alfalfa cultivars that are non-dormant, winter-hardy, and early flowering would allow for an early spring harvest with enhanced biomass. In this study, we found 25 QTL for TOF, 17 for SY and six QTL for CSB. Three TOF related QTL were stable and four TOF QTL were detected in the corresponding genomic locations of the flowering QTL of M. truncatula, an indication of possible evolutionarily conserved regions. The potential candidate genes for the SNP sequences of QTL regions were identified for all three traits and these genes would be potential targets for further molecular studies. Conclusions This research showed that variation in alfalfa flowering time after spring green up has no association with dormancy levels. Here we reported QTL, markers, and potential candidate genes associated with spring flowering time and biomass yield of alfalfa, which constitute valuable genomic resources for improving these traits via marker-assisted selection (MAS).
